Reagents for detection of Rift Valley fever virus infection in sheep.

Rift Valley fever virus (RVFV) causes Rift Valley fever (RVF), resulting in morbidity and mortality in humans and ruminants. Evidence of transboundary outbreaks means that RVFV remains a threat to human health and livestock industries in countries that are free from the disease. To enhance surveillance capability, methods for detection of RVFV are required. The generation of reagents suitable for the detection of RVFV antigen in formalin-fixed, paraffin-embedded tissues from infected animals have been developed and are described herein. Recombinant nucleoprotein (rNP) was expressed in Escheric

Rift Valley fever virus infections in Egyptian cattle and their prevention.

Rift Valley fever virus (RVFV) causes consistently severe outbreaks with high public health impacts and economic losses in livestock in many African countries and has also been introduced to Saudi Arabia and Yemen. Egypt with its four large outbreaks in the last 40 years represents the northernmost endemic area of RVFV. The purpose of this study was to provide an insight into the current anti-RVFV antibody status in immunized as well as non-immunized dairy cattle from the Nile Delta of Egypt. During 2013-2015, a total of 4,167 dairy cattle from four governorates including Dakahlia, Damietta, G

Interplay between the Virus and Host in Rift Valley Fever Pathogenesis.

Rift Valley fever virus (RVFV) belongs to the genus Phlebovirus, family Bunyaviridae, and carries single-stranded tripartite RNA segments. The virus is transmitted by mosquitoes and has caused large outbreaks among ruminants and humans in sub-Saharan African and Middle East countries. The disease is characterized by a sudden onset of fever, headache, muscle pain, joint pain, photophobia, and weakness. In most cases, patients recover from the disease after a period of weeks, but some also develop retinal or macular changes, which result in vision impairment that lasts for an undefined period of

Ultrastructural study of Rift Valley fever virus in the mouse model.

Detailed ultrastructural studies of Rift Valley fever virus (RVFV) in the mouse model are needed to develop and characterize a small animal model of RVF for the evaluation of potential vaccines and therapeutics. In this study, the ultrastructural features of RVFV infection in the mouse model were analyzed. The main changes in the liver included the presence of viral particles in hepatocytes and hepatic stem cells accompanied by hepatocyte apoptosis. However, viral particles were observed rarely in the liver; in contrast, particles were extremely abundant in the CNS. Despite extensive lymphocyt

Potential vectors of Rift Valley fever virus in the Mediterranean region.

We evaluated the ability of three mosquito species (Aedes caspius, Aedes detritus, Culex pipiens), collected in southern France and Tunisia, and of different laboratory-established colonies (Aedes aegypti, Aedes albopictus, Aedes vexans, Anopheles gambiae, Culex pipiens, Culex quinquefasciatus) to disseminate two strains of Rift Valley fever virus (RVFV), the virulent ZH548 and the avirulent Clone 13. After feeding on an infectious blood meal at 10(8.5) plaque-forming units/mL, females were maintained at 30 degrees C for 14 days. Surviving females were tested for the presence of virus on head

Detection of Rift Valley fever virus in mosquitoes by RT-PCR.

A reverse transcriptase-polymerase chain reaction (RT-PCR) assay to detect Rift Valley fever (RVF) virus RNA in experimentally infected mosquitoes was developed. The specificity of the assay was evaluated with three other phleboviruses; sandfly fever Sicilian (Sabin), sandfly fever Naples (Sabin) and Punta Toro (MSP 3) viruses. The relative sensitivity of the assay, determined by using RVF virus RNA extracted from serial dilutions of virus culture, was approximately 50 plaque forming units. This sensitivity level was 100-fold higher when a nested PCR procedure was used. When the RT-PCR assay w

Replication and dissemination of Rift Valley fever virus in Culex pipiens.

Following ingestion of 10(4.2) to 10(7.2) plaque-forming units (PFU) of Rift Valley fever (RVF) virus, 662 of 850 female Culex pipiens (78%) became infected. Those mosquitoes that became infected separated into two distinct groups: 1) those with a nondisseminated infection limited to the gut, and 2) those with a disseminated infection. The former group contained a median of 10(3.2) PFU, while those females with a disseminated infection contained a median of 10(5.5) PFU. Only those females with a disseminated infection were capable of transmitting virus by bite to a susceptible hamster. This is

Neutralizing Antibodies Against Rift Valley Fever Virus: Current Status and Advances.

BACKGROUND: Rift Valley fever virus (RVFV) is a mosquito-borne zoonotic pathogen that has caused repeated epidemics across Africa and the Arabian Peninsula, posing a severe and growing threat to public health and livestock. Infection in ruminants causes high neonatal mortality and catastrophic abortion storms; human disease ranges from self-limiting febrile illness to hemorrhagic fever, encephalitis, and permanent blindness. No licensed human vaccines or specific antiviral therapeutics are available, creating an urgent unmet medical need. METHODS: We systematically reviewed the peer-reviewed

Rift Valley fever virus activates multiple cell death pathways in neurons.

Neuronal injury and death contribute to long-term impairments and lethality during viral encephalitis. Rift Valley fever virus (RVFV), an arbovirus with epidemic potential, can manifest as late-onset encephalitis in humans, yet this disease outcome remains understudied. A lethal rodent model of RVF encephalitis is characterized by a dysregulated immune response, neuronal necrosis, and blood-brain barrier breakdown that precedes lethality. In this study, we built upon this prior work using both in vivo and in vitro models to interrogate the mechanism of cell death in neurons during RVFV infecti

Rift Valley Fever Virus Primes Immune Responses in Aedes aegypti Cells.

The ongoing global emergence of arthropod-borne (arbo) viruses has accelerated research into the interactions of these viruses with the immune systems of their vectors. Only limited information exists on how bunyaviruses, such as Rift Valley fever virus (RVFV), are sensed by mosquito immunity or escape detection. RVFV is a zoonotic phlebovirus (Bunyavirales; Phenuiviridae) of veterinary and human public health and economic importance. We have shown that the infection of mosquitoes with RVFV triggers the activation of RNA interference pathways, which moderately restrict viral replication. Here,

Mechanistic models of Rift Valley fever virus transmission: A systematic review.

Rift Valley fever (RVF) is a zoonotic arbovirosis which has been reported across Africa including the northernmost edge, South West Indian Ocean islands, and the Arabian Peninsula. The virus is responsible for high abortion rates and mortality in young ruminants, with economic impacts in affected countries. To date, RVF epidemiological mechanisms are not fully understood, due to the multiplicity of implicated vertebrate hosts, vectors, and ecosystems. In this context, mathematical models are useful tools to develop our understanding of complex systems, and mechanistic models are particularly s

Host-based processes as therapeutic targets for Rift Valley fever virus.

Rift Valley fever virus (RVFV) is an enveloped, segmented, negative sense RNA virus that replicates within the host's cytoplasm. To facilitate its replication, RVFV must utilize host cell processes and as such, these processes may serve as potential therapeutic targets. This review summarizes key host cell processes impacted by RVFV infection. Specifically the influence of RVFV on host transcriptional regulation, post-transcriptional regulation, protein half-life and availability, host signal transduction, trafficking and secretory pathways, cytoskeletal modulation, and mitochondrial processes

Rift valley fever virus infection among livestock handlers in Ibadan, Nigeria.

Livestock handlers are occupationally exposed to Rift Valley fever virus (RVFV) because of frequent and longer contact with mosquito vectors, animal blood and other tissues. We determined the seroprevalence of RVF virus infection among livestock handlers in Ibadan, Nigeria. Blood samples were collected from 265 workers in two major livestock markets and a livestock farm. Questionnaires were administered to obtain information on risk factors that may be associated with RVF virus transmission. The blood samples were tested using enzyme-linked immunosorbent assays (ELISA) to detect multispecies a

Data-driven modeling to assess receptivity for Rift Valley Fever virus.

Rift Valley Fever virus (RVFV) is an enzootic virus that causes extensive morbidity and mortality in domestic ruminants in Africa, and it has shown the potential to invade other areas such as the Arabian Peninsula. Here, we develop methods for linking mathematical models to real-world data that could be used for continent-scale risk assessment given adequate data on local host and vector populations. We have applied the methods to a well-studied agricultural region of California with [Formula: see text]1 million dairy cattle, abundant and competent mosquito vectors, and a permissive climate th
