Detection of Lyme Disease and Q Fever Agents in Wild Rodents in Central Italy.

The maintenance of tick-borne disease agents in the environment strictly depends on the relationship between tick vectors and their hosts, which act as reservoirs for these pathogens. A pilot study aimed to investigate wild rodents as reservoirs for zoonotic tick-borne pathogens (Borrelia burgdorferi sensu lato (s.l.), Coxiella burnetii, Francisella tularensis, and Anaplasma phagocytophilum) was carried out in an area of Gran Sasso e Monti della Laga National Park (Abruzzi Region, central Italy), a wide protected area where, despite sporadic reports of infection in humans and animals, eco-epid

pH as a biomarker of neurodegeneration in Huntington's disease: a translational rodent-human MRS study.

Early diagnosis and follow-up of neurodegenerative diseases are often hampered by the lack of reliable biomarkers. Neuroimaging techniques like magnetic resonance spectroscopy (MRS) offer promising tools to detect biochemical alterations at early stages of degeneration. Intracellular pH, which can be measured noninvasively by (31)P-MRS, has shown variations in several brain diseases. Our purpose has been to evaluate the potential of MRS-measured pH as a relevant biomarker of early degeneration in Huntington's disease (HD). We used a translational approach starting with a preclinical validation

Tularemia, plague, yersiniosis, and Tyzzer's disease in wild rodents and lagomorphs in Canada: a review.

Information related to infection of wild rodents or lagomorphs in Canada by Francisella tularensis, Yersinia pestis, other Yersinia spp., and Clostridium piliforme was searched for this study. Reports on tularemia in humans linked to these species came from diagnostic databases, literature, wildlife health specialists, and public health agencies. Tularemia has been diagnosed in 8 species of wild rodent and 2 species in the genus Lepus in Canada. Tularemia occurred in wild animals, or in humans associated with these species, in all jurisdictions except the Yukon and Nunavut. Tularemia was diagn

Alteration of dopaminergic markers in gastrointestinal tract of different rodent models of Parkinson's disease.

Parkinson's disease (PD) is a progressive neurological disorder that is often associated with various gastrointestinal (GI) symptoms. The link between the alteration of dopaminergic system and the symptoms of the GI tract in PD is complicated. To determine the changes in the dopaminergic system in the GI tract in PD, two kinds of rodent PD models were used in the present study. One was 6-hydroxydopamine (6-OHDA) -treated rats in which 6-OHDA was microinjected in the bilateral substantia nigra (SN). The other was 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) -treated mice in which MPTP wa

Antiparkinsonian action of a delta opioid agonist in rodent and primate models of Parkinson's disease.

The opioid peptides localized in striatal projection neurons are of great relevance to Parkinson's disease, not only as a consequence of their distribution, but also due to the pronounced changes in expression seen in Parkinson's disease. It has long been suspected that increased expression of enkephalin may represent one of the many mechanisms that compensate for dopamine (DA) depletion in Parkinson's disease. Here we demonstrate that a systemically delivered, selective delta opioid agonist (SNC80) has potent antiparkinsonian actions in both rat and primate models of Parkinson's disease. In r

Validation of a rodent model of Parkinson's Disease: evidence of a therapeutic window for oral Sinemet.

Behavioral measures of parkinsonism that are more clinically relevant than rotometry have been developed for rats with severe unilateral dopamine depletions, and the validity of these measures is supported by reports that these parkinsonian symptoms are attenuated by drugs that are effective in the clinical setting. Although the therapeutic gold standard, L-DOPA:carbidopa (Sinemet), effectively attenuates parkinsonian symptoms, the beneficial effects of this drug are limited by the dyskinesias that it produces at higher doses. The range of effective doses, from the minimum dose that produces b

Non-transgenic rodent models associated with Alzheimer's disease: applications, evaluations, and perspectives.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder. Familial AD accounts for less than 1% of cases, while sporadic AD (SAD) accounts for over 95%. Mild cognitive impairment (MCI) is the critical transition phase from normal aging to AD dementia. Understanding the pathological progression from MCI to AD and the mechanisms underlying SAD is essential. Rodent models, including transgenic and non-transgenic models, are vital tools for developing effective AD therapies. However, transgenic models primarily mimic familial AD and poorly replicate MCI and the complex pathological fea

Altered Expression and Function of Hepatic Transporters in a Rodent Model of Polycystic Kidney Disease.

Autosomal dominant polycystic kidney disease (ADPKD) is a common form of inherited polycystic kidney disease (PKD) and is a leading cause of kidney failure. Fluid-filled cysts develop in the kidneys of patients with ADPKD, and cysts often form in their liver and other organs. Previous data have shown that bile acids are increased in the liver of polycystic kidney (PCK) rats, a rodent model of PKD; these changes may be associated with alterations in liver transporter expression and function. However, the impact of PKD on hepatic transporters has not been characterized. Therefore, this preclinic

A novel approach to adenine-induced chronic kidney disease associated anemia in rodents.

To date, good experimental animal models of renal anemia are not available. Therefore, the purpose of this study was to establish a novel approach to induce chronic kidney disease (CKD) with severe anemia by oral administration of adenine in rodents. Adenine was administered to 6-week-old male C57BL/6 mice (25 and 50 mg/kg body weight) by oral gavage daily for 28 days. Serum creatinine and BUN as well as hematocrit, hemoglobin (Hb) and plasma erythropoietin (EPO) levels were monitored to assess renal function and anemia, respectively. Adenine at 25 mg/kg for 28 days slightly increased plasma c

Lyme disease bacterium does not affect attraction to rodent odour in the tick vector.

BACKGROUND: Vector-borne pathogens experience a conflict of interest when the arthropod vector chooses a vertebrate host that is incompetent for pathogen transmission. The qualitative manipulation hypothesis suggests that vector-borne pathogens can resolve this conflict in their favour by manipulating the host choice behaviour of the arthropod vector. METHODS: European Lyme disease is a model system for studying this conflict because Ixodes ricinus is a generalist tick species that vectors Borrelia pathogens that are specialized on different classes of vertebrate hosts. Avian specialists like

Study of oxidative damage and antioxidant systems in two Huntington's disease rodent models.

Huntington's disease (HD) is a hereditary neurodegenerative disorder, characterized by motor, psychiatric, and cognitive symptoms. The genetic defect responsible for the onset of the disease, expansion of CAG repeats in exon 1 of the gene that codes for huntingtin, has been unambiguously identified. The mechanisms by which the mutation causes the disease are not completely understood yet. However, defects in the energy metabolism of affected cells, which may cause oxidative damage, have been proposed as underlying molecular mechanisms that participate in the etiology of the disease. In this ch

Neurological approaches for investigating West Nile virus disease and its treatment in rodents.

West Nile virus (WNV) has had a major public health impact since its emergence in the Western Hemisphere; in 2012, nearly 3000 cases of WN neuroinvasive disease were identified in the United States. The underlying mechanisms of WN neurologic disease can only be studied to a limited extent in patients, but can be investigated in much greater detail in animal models. In this paper, we describe how we and others have employed a variety of electrophysiological and neurological techniques to study experimental WNV infections in hamsters and mice. The methods have included electrophysiological motor

The use of rodent skilled reaching as a translational model for investigating brain damage and disease.

Neurological diseases, including Parkinson's disease, Huntington's disease, and brain damage caused by stroke, cause severe motor impairments. Deficits in hand use are one of the most debilitating motor symptoms and include impairments in body posture, forelimb movements, and finger shaping for manipulating objects. Hand movements can be formally studied using reaching tasks, including the skilled reaching task, or reach-to-eat task. For skilled reaching, a subject reaches for a small food item, grasps it with the fingers, and places it in the mouth for eating. The human movement and its assoc

Behavioral analysis of motor and non-motor symptoms in rodent models of Parkinson's disease.

Alongside the classical motor symptoms, non-motor symptoms are increasingly recognised to play a major role in the disability associated with Parkinson's disease in humans. Animal models based on experimental depletion of forebrain dopamine have traditionally focussed on the simple and easy to measure motor impairments, and they reproduce well the bradykinesia, rigidity and impairments in the initiation and sequencing of voluntary goal-directed movement. However, a more comprehensive analysis is now urgently required. In this chapter we summarise the predominant unilateral and bilateral dopami

[Climate change in spain and risk of infectious and parasitic diseases transmitted by arthropods and rodents].

Due to Spain's being located near Africa, being a stopping-off point for migrating birds and individuals and due to its climate conditions, nearing those of areas where there are vector-borne diseases, this is a country where this type of diseases could taken on greater importance due to the climate change. The possible risk would result from the geographical spread of already established vectors or due to subtropical vectors adapted to surviving in cooler, dried climates being imported and taking up residence. Hypothetically, the vector-borne diseases subject to be influenced by the climate c

Neuroprotective actions of the ginseng extract G115 in two rodent models of Parkinson's disease.

The herbal remedy, ginseng, has recently been demonstrated to possess neurotrophic and neuroprotective properties, which may be useful in preventing various forms of neuronal cell loss including the nigrostriatal degeneration seen in Parkinson's disease (PD). In these studies, we examine the potential neuroprotective actions of the ginseng extract, G115, in two rodent models of PD. Animals received oral administration of G115 prior to and/or following exposure to the parkinsonism-inducing neurotoxin, 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP), in mice, or its toxic metabolite, 1-methy

Spontaneous cholangiofibrosis in Long-Evans Cinnamon rats: a rodent model for Wilson's disease.

The Long-Evans Cinnamon (LEC) rat is a rodent model of Wilson's disease characterized by ceruloplasmin deficiency, hepatic copper accumulation, and hepatocellular injury. So far, the LEC rat appears to be the only strain in which cholangiofibrosis develops spontaneously. The aim of the study reported here was to characterize the time course of development and investigate the structural and ultrastructural features of cholangiofibrosis and their possible relationship to hepatic copper and iron content. The livers of 54 rats (22 males), ages 5 to 113 weeks, were examined by light microscopy and

The role of rodents as reservoirs for some diseases in new reclaimed areas in north Sinai.

In a trial to determine the role of Sinai rodents as reservoirs to rickettsial diseases, the IFA technique was used to detect the presence of R. typhi and spotted fever group in blood of some commensal and wild rodent species collected in Sinai during the last five years (1985-1989). Out of 277 Rattus spp. collected in El Arish (47.3%) were positive to R. typhi and only (5.9%) of 35 Mus musculus were positive to R. typhi. As for Gerbillus spp. only (1.5%) of 206 animals were positive for R. typhi. The positive cases of spotted fever group were (34.6%) in Rattus spp. and (18.8%) in Gerbillus sp

Spatiotemporal dynamics of the capture rate of Rattus tanezumi and its implications for rodent-borne diseases in the Three Gorges Reservoir Area, China.

Rattus tanezumi (R. tanezumi) is a major amplifying host for plague and an important reservoir host for multiple zoonotic pathogens, including leptospirosis and hantavirus. Its population dynamics in the Three Gorges Reservoir (TGR) Area of China are influenced by climate and reservoir-induced environmental changes, thereby impacting disease transmission risks. Based on rodent capture rate data collected using cage traps during night surveys in January, March, May, July, September, and November each year from 2015 to 2021, this study investigated the spatiotemporal dynamics of R. tanezumi and

Arthropod- and rodent-borne viral and rickettsial diseases in the WHO South-East Asia and Western Pacific regions: memorandum from a WHO meeting.

Arthropod- and rodent-borne viral and rickettsial diseases continue to be one of the most important causes of morbidity and mortality in the WHO South-East Asia and Western Pacific Regions. Both epidemics and sporadic cases occur. In some years, outbreaks of dengue haemorrhagic fever and Japanese encephalitis reached alarming proportions. The significance of other arthropod- and rodent-borne viral and rickettsial diseases has still to be determined. Therefore, continuous epidemiological surveillance, diagnosis, and control of these groups of diseases remains an urgent task. The objectives, targ
