Symbiotic effectiveness and ecologically adaptive traits of native rhizobial symbionts of Bambara groundnut (Vigna subterranea L. Verdc.) in Africa and their relationship with phylogeny.

Bambara groundnut (Vigna subterranea L. Verdc.) is an indigenous, drought-tolerant, underutilized African food legume, with the ability to fix atmospheric N2 in symbiosis with soil bacteria called rhizobia. The aim of this study was to assess the morpho-physiological, symbiotic and phylogenetic characteristics of rhizobia nodulating Bambara groundnut in Ghana, Mali and South Africa. The morpho-physiologically diverse isolates tested were also found to exhibit differences in functional efficiency and phylogenetic positions. Based on Enterobacterial Repetitive Intergenic Consensus (ERIC)-PCR ban

Inoculation with Bradyrhizobium ottawaense Effectively Reduces N2O Emissions from Degraded Soybean Nodules in Andosols Dominated by nosZ-lacking Bradyrhizobia.

Nitrous oxide (N2O) is a potent greenhouse gas, and its largest anthropogenic source is agricultural soils. In these soils, N2O emissions are produced from nitrogen fertilization and nitrogen-rich crop residues, such as soybean nodules. Previous studies demonstrated that inoculation with Bradyrhizobium diazoefficiens possessing the N2O reductase gene (nosZ) effectively mitigated N2O emissions from soybean fields at harvest. More recently, Bradyrhizobium ottawaense, which exhibits higher nosZ expression, reduced N2O emissions more effectively than B. diazoefficiens during the degradation of soy

Metabolic Snorkeling - The Metabolic Interaction Between Plant Organs and Tissues with different Oxygen Availabilities.

Plants constantly experience partial hypoxia due to varying external oxygen availability and the existence of hypoxic niches, such as the shoot apical meristem, phloem or root nodules. Waterlogging experiments indicate that hypoxic stress at the root does not only lead to a local metabolic response, such as the accumulation of hypoxia-related metabolites, but also causes metabolic alterations in the normoxic shoot. Moreover, hypoxia-related metabolites are exported from the hypoxic root towards the normoxic shoot, where they can be recycled. Maintaining import of glycolytic substrates from the

Alfalfa varieties can weakly choose beneficial nitrogen-fixing bacteria from a population isolated from a single field.

In natural and agricultural systems, legumes recruit rhizobia from diverse soil populations to fix nitrogen in root nodules. A few legumes, including the model legume Medicago truncatula, can select and enrich beneficial rhizobia. Here, we investigated whether its perennial relative, Medicago sativa (alfalfa), a globally important forage crop, also possesses this ability. We developed a genetically variable collection of 117 Sinorhizobium meliloti strains sampled from three field-grown alfalfa varieties, performed multi-strain and single-strain inoculations in a nitrogen-free greenhouse experi

Medicago truncatula Gaertn. as a model for understanding the mechanism of growth promotion by bacteria from rhizosphere and nodules of alfalfa.

The present study showed all the 16 strains isolated and identified from the alfalfa rhizosphere and nodules, and registered in GenBank, to be good candidates for targeted use in studies addressing the rather weak known mechanism of plant growth promotion, including that of Medicago truncatula, a molecular crop model. Based on physiological, biochemical and molecular analysis, the 16 isolates obtained were ascribed to the following five families: Bacillaceae, Rhizobiaceae, Xantomonadaceae, Enterobacteriaceae and Pseudomonadaceae, within which 9 genera and 16 species were identified. All these

Lotus japonicus CLV1-Like Receptor HAR1 Promotes Nitrogen Utilisation and Growth Under Non-Symbiotic Conditions.

Legumes establish mutualistic symbiosis with nitrogen (N)-fixing bacteria, which allows them to utilise atmospheric N2. Because the maintenance of symbiosis requires abundant carbon (C) sources, legumes regulate the balance between carbon consumption and nitrogen acquisition by systemically controlling the nodule number through CLAVATA1 (CLV1)-like receptors. In Lotus japonicus, the CLV1-like receptor HYPERNODULATION ABERRANT ROOT FORMATION1 (HAR1) acts in shoots to regulate root nodulation and contributes to symbiotic C/N coordination. This raises the possibility that HAR1 may also influence

Rhizobia block Cd absorption in peanut via rhizosphere microbiome assembly and N-regulated host defense.

Cadmium (Cd) contamination severely threatens peanut (Arachis hypogaea L.) production and symbiotic nitrogen fixation (SNF). Although rhizobia inoculation can alleviate heavy metal toxicity in plants, how nitrate (NO3-) regulates Cd translocation and the defense of the "rhizobia-root-nodule" system remains unclear. Here, we investigated the mechanisms by which the peanut rhizobium, Rhizobium sp. HM13, mitigates Cd toxicity and sustains SNF under varying NO3- levels (N- and N+) via field and pot trials. Field trials showed that HM13 reduced rhizosphere bioavailable Cd and decreased seed Cd accu

The chromosome-level genome assembly of Astragalus sinicus and comparative genomic analyses provide new resources and insights for understanding legume-rhizobial interactions.

The legume species Astragalus sinicus (Chinese milk vetch [CMV]) has been widely cultivated for centuries in southern China as one of the most important green manures/cover crops for improving rice productivity and preventing soil degeneration. In this study, we generated the first chromosome-scale reference genome of CMV by combining PacBio and Illumina sequencing with high-throughput chromatin conformation capture (Hi-C) technology. The CMV genome was 595.52 Mb in length, with a contig N50 size of 1.50 Mb. Long terminal repeats (LTRs) had been amplified and contributed to genome size expansi

Rhizobia-mediated soybean rhizosphere and nodule endophytic microorganisms reduce bioavailability of Cd and Cu in soil.

Low-level bioavailable cadmium (Cd) and copper (Cu) in agricultural soils poses a severe threat to soil health and food safety; however, the mechanisms by which indigenous Cu-Cd tolerant rhizobia modulate plant-microbe-soil interactions remain poorly understood. In this field trial, the effects of two Cu-Cd tolerant strains, Sinorhizobium xinjiangense YN5 (RB) and Rhizobium pusense GF4 (RD), when inoculated individually and in combination (RC), were assessed with respect to soybean growth, heavy‑metal partitioning, rhizosphere and nodule endophytic microbiomes, and soil functional genes under

Identification and functional characterization of soybean root hair microRNAs expressed in response to Bradyrhizobium japonicum infection.

Three soybean [Glycine max (L) Merr.] small RNA libraries were generated and sequenced using the Illumina platform to examine the role of miRNAs during soybean nodulation. The small RNA libraries were derived from root hairs inoculated with Bradyrhizobium japonicum (In_RH) or mock-inoculated with water (Un_RH), as well as from the comparable inoculated stripped root samples (i.e. inoculated roots with the root hairs removed). Sequencing of these libraries identified a total of 114 miRNAs, including 22 novel miRNAs. A comparison of miRNA abundance among the 114 miRNAs identified 66 miRNAs that

Sucrose and pipecolic acid-mediated enrichment of rhizosphere Rhizobiale Burkholderiale bacteria improve soybean growth under low light conditions.

Low light (LL) is a major constraint on the productivity of intercropped legumes and dense planting crops, and also affects the root-associated microbial communities. However, how LL reshapes the root-microbe interactions and whether the root microbiota can mitigate LL-induced damage in legumes remain unclear. Here, a meta-analysis based on field observations revealed that negative effects predominated (>70%) in intercropped and dense planting legume systems, with the light intensity emerging as the primary determinant of yield variation. Using soybean as a model, we found that LL suppressed p

Effect of selenium hyperaccumulation on the root endophytic and rhizosphere microbiome of two Astragalus species.

Metal hyperaccumulation is prevalent throughout plant evolution, particularly in the legume family (Fabaceae), and acts as a presumed chemical defence against herbivory. However, metal hyperaccumulation can have non-target impacts on other biological interactors, including plant-microbe interactions. This information is important given the interest in utilizing hyperaccumulating plants for phytoremediation of anthropogenically contaminated soils. Here we employ a greenhouse experiment manipulating selenium level along with 16S rRNA gene amplicon sequencing methods to explore the effect of sele

Identification of the DOG1 Gene Family in Cultivated Peanut and Negative Regulation of Ralstonia solanacearum Infection by AhDOG1-L4.

Bacterial wilt, caused by Ralstonia solanacearum, is a devastating soilborne disease often referred to as plant cancer. It threatens hundreds of species of plants and causes losses in yield and a deterioration in the quality of infected plants. Delay of Germination 1 (DOG1) plays vital roles in plant growth, development, and abiotic and biotic stress responses. However, the functions of DOG1 in biotic stress remain unexplored in cultivated peanut (Arachis hypogaea L.). In this study, we systematically identified and characterized the AhDOG1 gene family in peanut using bioinformatics approaches

Integrative Multi-Omics Analysis Reveals Transcriptomic and Metabolic Remodeling Associated with Enhanced Peanut Nodulation Under Arbuscular Mycorrhizal Fungal Inoculation and Calcium Application.

Peanut (Arachis hypogaea L.) yield depends on biological nitrogen fixation, but the molecular mechanisms underlying the combined effects of arbuscular mycorrhizal fungi (AMF) and calcium fertilizer on nodulation remain unclear. Here, we used integrated transcriptomic and metabolomic analyses to investigate potential mechanisms in peanut roots. Compared with the non-inoculated control, AMF inoculation alone was associated with a 22.1% higher nodule number per plant. The combined application of AMF and CaO showed a 35.9% higher nodulation than AMF alone, and a 30.9% higher AMF colonization rate

Synergistic interaction of Rhizobium leguminosarum bv. viciae and arbuscular mycorrhizal fungi as a plant growth promoting biofertilizers for faba bean (Vicia faba L.) in alkaline soil.

Egyptian soils are generally characterized by slightly alkaline to alkaline pH values (7.5-8.7) which are mainly due to its dry environment. In arid and semi-arid regions, salts are less concentrated and sodium dominates in carbonate and bicarbonate forms, which enhance the formation of alkaline soils. Alkaline soils have fertility problems due to poor physical properties which adversely affect the growth and the yield of crops. Therefore, this study was devoted to investigating the synergistic interaction of Rhizobium and arbuscular mycorrhizal fungi for improving growth of faba bean grown in

Climate Change and Nitrogen-Fixing Legumes: Investigating Stress-Modulated Dynamics of Carbon Fixation and Root Nodulation.

In the Anthropocene era, climate change is increasingly subjecting the crops to overlapping abiotic stressors such as drought, elevated temperatures, and air pollution, thereby disrupting their physiological integrity and functional performance. This review synthesises current knowledge on responses of N2-fixing plants to such stressors, focusing on core physiological processes and symbiotic nitrogen fixation via nodulation. The intricate interdependence between these traits is explored through the lens of altered source-sink relationships, which are highly sensitive to multifactorial environm

Combined application of rhizobacteria, organic and inorganic amendments reduce lead and cadmium uptake and improve growth of chickpea by modulating physiology and antioxidant status.

Due to a lack of high-quality water, farmers have been compelled to use sewage water for irrigation, contaminating agricultural soils with multiple heavy metals. For the remediation of contaminated soil, plant growth-promoting rhizobacteria (PGPR), pressmud (PM), and iron (III) oxide were used to improve the growth and phytostabilization potential of chickpea grown in contaminated soil. Contaminated soil was collected from a nearby field, receiving sewage and factory water over the last 60 years. Chickpea seeds were inoculated with metal-tolerant (lead and cadmium) rhizobacterial and rhizobial

Strigolactones in Plants and Their Interaction with the Ecological Microbiome in Response to Abiotic Stress.

Phytohormones play an essential role in enhancing plant tolerance by responding to abiotic stresses, such as nutrient deficiency, drought, high temperature, and light stress. Strigolactones (SLs) are carotenoid derivatives that occur naturally in plants and are defined as novel phytohormones that regulate plant metabolism, growth, and development. Strigolactone assists plants in the acquisition of defensive characteristics against drought stress by initiating physiological responses and mediating the interaction with soil microorganisms. Nutrient deficiency is an important abiotic stress facto

Identification and characterization of RibN, a novel family of riboflavin transporters from Rhizobium leguminosarum and other proteobacteria.

Rhizobia are symbiotic bacteria able to invade and colonize the roots of legume plants, inducing the formation of nodules, where bacteria reduce atmospheric nitrogen (N2) to ammonia (NH3). Riboflavin availability influences the capacity of rhizobia to survive in the rhizosphere and to colonize roots. In this study, we identified the RL1692 gene of Rhizobium leguminosarum downstream of a flavin mononucleotide (FMN) riboswitch. RL1692 encodes a putative transmembrane permease with two EamA domains. The presence of an FMN riboswitch regulating a transmembrane protein is usually observed in ribofl
