Inflammatory mechanism of Rumenitis in dairy cows with subacute ruminal acidosis.

BACKGROUND: Subacute ruminal acidosis (SARA) is a metabolic disease in high-producing dairy cattle, and is accompanied by rumenitis. However, the mechanism of rumenitis remains unclear. Therefore, the aim of this study was to investigate the molecular mechanism of rumenitis in dairy cows with SARA. RESULTS: The results showed that SARA cows displayed high concentrations of ruminal volatile fatty acids, lactic acid and lipopolysaccharide (LPS). Furthermore, the blood concentrations of LPS and acute phase proteins haptoglobin, serum amyloid-A, and LPS binding protein were significantly higher i

Gastrointestinal spindle cell tumor of the rumen with metastasis to the liver in a goat.

Many neoplasms have been reported in goats; however, neoplasia of the rumen is rarely reported. A 9-y-old castrated male pygmy goat was presented with a history of respiratory stertor, fever, and anorexia. A respiratory diagnostic work-up including skull and thorax radiographs and endoscopy revealed minor enlargement of the arytenoids but no other abnormal findings. After a month of little improvement on symptomatic treatment and worsening general health, the goat was euthanized. On autopsy, the forestomachs, liver, spleen, diaphragm, and the ventral and lateral aspects of the cranial third of

Presence and species identity of rumen flukes in cattle and sheep in the Netherlands.

The purpose of the study was to gain knowledge about the prevalence and identity of rumen flukes (RF) in cattle and sheep in the Netherlands. Routine faecal examinations of diagnostic submissions between May 2009 and September 2014 showed a mean annual herd or flock RF prevalence of 15.8% for cattle and 8.0% for sheep. Prevalence in cattle was higher after 2012 than before, which may reflect a change in detection method as well as an increase in true prevalence. During November and December 2014, an abattoir survey was conducted to allow for scoring of rumen fluke burden and to obtain specimen

Quantitative analysis of cellulose degradation and growth of cellulolytic bacteria in the rumen.

Ruminant animals digest cellulose via a symbiotic relationship with ruminal microorganisms. Because feedstuffs only remain in the rumen for a short time, the rate of cellulose digestion must be very rapid. This speed is facilitated by rumination, a process that returns food to the mouth to be rechewed. By decreasing particle size, the cellulose surface area can be increased by up to 10(6)-fold. The amount of cellulose digested is then a function of two competing rates, namely the digestion rate (K(d)) and the rate of passage of solids from the rumen (K(p)). Estimation of bacterial growth on ce

Effect of feeding duration and rumen fill on behaviour in dairy cows.

The aim of the present experiment was to test the hypothesis that oral manipulation of feed is a behavioural need in cattle, irrespective of actual rumen load. Twelve rumen fistulated cows were used and subjected to four different treatments: low rumen content+long duration of eating (A), high rumen content+short duration of eating (B), high rumen content+long duration of eating (=positive control) (C) and low rumen content+short duration of eating (=negative control) (D). To obtain treatment A and B, rumen content was transferred by hand from cow A to B through the rumen fistulaes. Each treat

The biological assessment of vitamin D3 metabolites produced by rumen bacteria.

Biological assays were performed to evaluate 10-oxo-19-nor-vitamin D3 (10-oxo-D3) and 5(E) 25-hydroxy-10-oxo-19-nor-vitamin D3 (25-OH-10-oxo-D3) two bacterial products of vitamin D3 (D3) and 25-hydroxyvitamin D3 (25-OHD3) metabolism, respectively. The 5(Z) and 5(E) isomers of 10-oxo-D3 were, respectively, 40- and 80-fold less active than D3 in stimulating Ca+2 absorption from the gut. 25-Hydroxy-10-oxo-D3 did not stimulate Ca+2 absorption. Only 5(Z) 10-oxo-D3 induced mobilization of bone Ca+2. In addition, both 10-oxo-D3 and 25-OH-10-oxo-D3 showed poor affinities for either the plasma D3-bindi

[The rumen acidosis complex--recent knowledge and experiences (1). A review].

By "Rumen acidosis" until recently only the acute indigestion with intoxication caused by excessive production of lactic acid was understood. Meanwhile, however, it has become evident that further pathologic changes can take place in the acid-base status of the rumen contents which may also be referred to as "Rumen acidosis". Basing on new literature and own experiences the paper gives a review of that "Rumen acidosis complex": Fundamentals of the regulation of pH in the rumen; chronic latent rumen acidosis (hyper-and parakeratosis, chronic hyperplastic rumenitis, rumenitis-liver abscess compl

Impact of dietary inclusion of field peas (Pisum sativum) on milk production, blood metabolites, rumen fermentation, and composition of rumen bacterial community of lactating dairy cows.

This study determined the effects of inclusion of field peas in the diet of lactating dairy cows on milk production, blood metabolites, rumen fermentation, and the rumen bacterial community. Twelve midlactation Holstein cows were used in a replicated 3 × 3 Latin square design with three 21-d experimental periods. Cows were offered either a control TMR or a TMR containing coarsely ground peas with a geometric mean diameter of 4.2 mm at inclusion rates of 3.9% (LP) or 7.8% (HP) of DM. Increasing the dietary inclusion rate of peas from 0% to 7.8% of DM increased DMI from 23.7 to 24.3 kg/d and mil

Effects of yeast β-glucan on fermentation parameters, microbial community structure, and rumen epithelial cell function in high-concentrate-induced yak rumen acidosis in vitro.

This study employed a completely randomized design (CRD) to evaluate the effects of graded levels of yeast β-glucan supplementation on subacute ruminal acidosis (SARA) induced by high-concentrate diets in yaks. Seven treatment groups were established: a low-concentrate control (LC; 50 % concentrate + 50 % forage), a high-concentrate control (HC; 80 % concentrate + 20 % forage), and HC diets supplemented with 0.5 %, 1 %, 2 %, 4 %, or 8 % yeast β-glucan. Rumen pH, gas production, and volatile fatty acid (VFA) profiles were measured at 3, 6, 9, 12, and 24 h of in vitro fermentation. At 9, 12, and

Bioavailability of rumen-protected L-methionine supplement in lactating dairy cows.

Rumen-protected AA are largely used to supply the MP in dairy diets. Rumen-protected methionine (RP-Met) supplements differ in their proportion of the biologically active L-form isomer and in their bioavailability. Based on the plasma free AA dose-response approach, we determined the bioavailability of L-Met60 (LipoAktiv L-methionine 60), a rumen-protected L-Met supplement, and evaluated its effects on plasma AA profile. Ten multiparous Holstein cows in mid lactation were used in a replicated 5 × 5 Latin square design with 7-d experimental periods. All treatments used L-Met60, with doses and r

Integrating Diagnostic Tools for Early Recognition of Rumenitis in a Neonatal Calf.

Rumenitis is an inflammatory condition of the rumen, typically seen in adult cattle managed on high-energy diets. In calves, it is uncommon and often linked to ruminal drinking due to esophageal groove dysfunction. Early diagnosis is challenging due to nonspecific clinical signs. A one-month-old male Limousin calf was presented with persistent non-fetid fluid regurgitation, rhythmic mastication, inappetence, and progressive neurological signs. Clinical examination revealed signs of dehydration and neurological dysfunction. Laboratory evaluation demonstrated metabolic acidosis (pH 7.16), hyperk

Transcriptomic Changes in the Rumen Epithelium of Cattle after the Induction of Acidosis.

The transition from normal forage to a highly fermentable diet to achieve rapid weight gain in the cattle industry can induce ruminal acidosis. The molecular host mechanisms that occur in acidosis are largely unknown. Therefore, the histology and transcriptome profiling of rumen epithelium was investigated in normal and acidosis animals to understand the molecular mechanisms involved in the disease. The rumen epithelial transcriptome from acidosis (n=3) and control (n=3) Holstein steers was obtained using RNA-sequencing. The mean values of clean reads were 70,975,460±984,046 and 71,

Telocytes as a Novel Structural Component in the Muscle Layers of the Goat Rumen.

Telocytes (TCs) have been identified as a distinct type of interstitial cells, but have not yet been reported in the gastrointestinal tract (GIT) of ruminants. In this study, we used transmission electron microscopy (TEM) and double-labelling immunofluorescence (IF) (antibodies: CD34, vimentin and PGP9.5) to seek TCs and investigate their potential functions in the muscle layers of the goat rumen. TCs were distributed widely in the myenteric plexus (TC-MYs) between the circular and longitudinal muscle layers, within circular muscle layers (TC-CMs) as well as in longitudinal muscle layers (TC-L

Influence of balance of protein and energy metabolism in rumen on hypomagnesaemia in cows.

This study was carried out in order to determine the influence of nitrogen excess in rumen and deficit of digestible organic matter in a feed ration for dairy cows on hypomagnesaemia at the beginning of green forage feeding. The balance of nitrogen and energy metabolism in rumen was determined using the rules of evaluation of the nutritional value of fodder (PDI system) according to the Institut National de la Recherche Agronomique (INRA), taking into consideration a potential protein synthesis from nitrogen by microorganisms in rumen (MPRN) and microbial protein synthesized from digestible or

[Electrophysiologic changes in rumen epithelium in their effect on magnesium transport--a review].

The forestomach is the main site of Mg2+ absorption in the gastrointestinal tract of ruminants and maintains Mg2+ homeostasis. It has long been known that an increase in K+ intake and, consequently, in ruminal K+ concentration ([K+]) decreases the apparent digestibility of Mg2+, which increases the risk of hypomagnesemia and tetany. The present review summarizes new findings on the mechanisms of Mg2+ absorption across the rumen epithelium. It has been shown that transcellular and active Mg2+ transport is the predominant pathway for Mg2+ transport from lumen to blood. It is well established tha

Intravenous administration of ionophores in ruminants: effects on metabolism independent of the rumen.

The effect of i.v. administration of ionophores on metabolism in ruminants was investigated in two experiments. In Exp. 1, four Angus heifers were assigned randomly to receive i.v. monensin (18 mg, n = 2) or vehicle (control, n = 2). Samples were collected from indwelling vena cava cannulas from -60 to 240 min. Concentrations of K, Mg (P less than .05) and P (P less than .10) were lower and glucose (GLU) and free fatty acids (FFA) were higher (P less than .05) in monensin-treated than in control heifers. Serum insulin (INS) initially declined and subsequently increased (P less than .05) follow
