Detection of Zoonotic Protozoa Toxoplasma gondii and Sarcocystis suihominis in Wild Boars from Spain.

Food safety regulations require the control of the presence of protozoa in meats destined for human consumption. Wild boar (Sus scrofa) meat may constitute a source of zoonoses. A 23.8% (688/2881) seroprevalence of anti-Toxoplasma gondii antibodies and 72.2% (662/910) Sarcocystis sarcocysts prevalence were detected among wild boars hunted in Southwestern areas of Spain. Identity of Sarcocystis spp. was performed by RFLP-PCR and sequencing, detecting S. miescheriana (7/8) and the zoonotic S. suihominis (1/8). Risk assessment studies of these coccidian in meats destined to human consumption are

Light and electron microscope study of Sarcocystis sp. from the fallow deer (Cervus dama).

By means of light and electron microscopy a study was made of Sarcocystis sp. from 11 fallow deer (Cervus dama). Cysts of Sarcocystis sp. were found in the tongue and abdominal muscle of 3 of 11 deer from forests near Bonn (FRG). These measured 212-560 micron in length and 54-120 micron in width and contained metrocytes and merozoites. The cyst wall, which had narrow band-like protrusions, is compared with other Sarcocystis sp. from Cervidae.

MOLECULAR CHARACTERIZATION OF SARCOCYSTIS SPOROCYSTS IN INTESTINES OF BOBCAT (LYNX RUFUS) FROM MINNESOTA.

Sarcocystis species are heteroxenous protozoan parasites of major veterinary and public health relevance, yet the role of wild felids in their natural transmission cycles remains poorly understood. In this study, we investigated Sarcocystis infections in 189 bobcats (Lynx rufus) harvested from Minnesota during 2024-2025. Intestinal homogenates of 179 were examined microscopically, and Sarcocystis sporocysts were found in 38 bobcats (21.2%). DNA was extracted from all 38 positive samples, of which 19 were PCR-positive, and 14 yielded high-quality sequences for molecular characterization using p

Identifying Sarcocystis spp. infection in goats: a combined morphological, serological, and molecular approach.

Domestic goats (Capra aegagrus hircus) are intermediate hosts for three species of Sarcocystis: S. capracanis, S. hircicanis, and S. moulei. The present study aimed to describe and identify Sarcocystis spp. in goat/kid muscles utilizing optical and transmission electron microscopy, a serological method, and PCR-sequencing of individual cysts. Both S. capracanis and S. hircicanis were identified by light and transmission electron microscopy and molecular methods. No macroscopic cysts were detected in any of the examined animals (n = 90, from two Argentinean provinces); only microscopic sarcocys

Molecular identification of Sarcocystis aucheniae in the wild South American camelid Vicugna vicugna.

Vicuñas (Vicugna vicugna) are wild South American camelids (SACs) protected by law in Argentina, and information on pathogens that infect them is scarce. In this study, an adult vicuña found dead in the province of Salta was examined, and evidence of infection by Sarcocystis sp. protozoans was sought. Infection of skeletal muscles by S. aucheniae, with the production of macroscopic sarcocysts, a disease known as SAC sarcocystosis, has been described in the other three SACs - llamas, alpacas, and guanacos - but its occurrence in vicuñas has so far remained unknown. In the analyzed individual, m

Gray fox (Urocyon cinereoargenteus) identified as a new intermediate host for Sarcocystis neurona.

The protozoan Sarcocystis neurona can cause severe disease in horses, marine mammals, and several other animal species in North America. The Virginia opossum (Didelphis virginiana) is its definitive host, and the raccoon (Procyon lotor) has been considered its primary intermediate host in the USA. Although canids have not previously been identified as important intermediate hosts for this parasite, we here report several natural cases in the gray fox (Urocyon cinereoargenteus). We identified muscular infections in 11 (23.9 %) of 46 gray foxes from Pennsylvania, USA. In hematoxylin and eosin (H

Molecular characterisation of five Sarcocystis species in domestic sheep (Ovis aries) from Spain.

The major aim of the present study was to determine by molecular methods whether the wide and narrow types of macroscopic sarcocysts in Spanish sheep belonged to different species, that is, Sarcocystis gigantea and Sarcocystis medusiformis, respectively. Additionally, we wanted to identify and characterize molecularly the species forming microscopic sarcocysts and determine the phylogenetic placement of all species found. Portions of the oesophagus, diaphragm and hind legs containing macroscopic sarcocysts were collected from slaughtered culled ewes at an abattoir in the Province of Madrid, Ce

High-throughput screen of drug repurposing library identifies inhibitors of Sarcocystis neurona growth.

The apicomplexan parasite Sarcocystis neurona is the primary etiologic agent of equine protozoal myeloencephalitis (EPM), a serious neurologic disease of horses. Many horses in the U.S. are at risk of developing EPM; approximately 50% of all horses in the U.S. have been exposed to S. neurona and treatments for EPM are 60-70% effective. Advancement of treatment requires new technology to identify new drugs for EPM. To address this critical need, we developed, validated, and implemented a high-throughput screen to test 725 FDA-approved compounds from the NIH clinical collections library for anti

Sheep experimentally infected with Sarcocystis from dogs. II. Abortion and disease in ewes.

This is the first study of Sarcocystis-induced abortion in sheep. Eleven pregnant ewes were experimentally inoculated with 50,000, 100,000, or 500,000 Sarcocystis ovicanis sporocytis from dogs. Eight ewes either aborted, died, or became moribund before term; they produced 15 fetuses, 11 of normal appearance and 4 necrotic. No evidence of intrauterine transmission was obtained. All infected ewes became anemic, inappetent, and lost weight. Ewes inoculated with the greatest numbers of sporocysts exhibited the most striking signs of acute illness. At necropsy of acutely ill ewes the heart was the

Molecular characterization of Sarcocystis spp. in intestines of coyotes and foxes from Pennsylvania, USA.

BACKGROUND: Wild canids serve as definitive hosts for numerous Sarcocystis species infecting livestock and wildlife. However, species-level identification in free-ranging carnivores remains difficult, impeded by the morphological resemblance among the sporocysts of various parasite species. METHODS: Intestinal mucosal scrapings from 40 wild canids: red fox (Vulpes vulpes, n = 8), gray fox (Urocyon cinereoargenteus, n = 6) and coyote (Canis latrans, n = 26) in Pennsylvania, USA, sampled in 2024, were tested for Sarcocystis sporocysts. RESULTS: Sporocysts were detected microscopically in intes

Zoonotic potential of bovine Sarcocystis species: a challenge for public health and meat inspection.

Foodborne parasitic diseases remain a neglected public health issue worldwide. Sarcocystosis, caused by the ingestion of tissue cysts of Sarcocystis species, represents a growing challenge for food safety and public health. Cattle serve as intermediate hosts for several Sarcocystis species, including those with confirmed zoonotic potential such as S. hominis, S. heydorni, and the recently described S. sigmoideus (2024/2025). In humans, intestinal sarcocystosis often presents with gastrointestinal distress, whereas extraintestinal (muscular) infections can lead to severe clinical manifestations

The first molecular characterization of Sarcocystis cameli in the Indian dromedary camels (Camelus dromedarius).

In the present study, tissue samples (tongue, esophagus and heart) were investigated from dromedary camels of India for identification and characterization of Sarcocystis spp. using histopathology, PCR and gene sequencing. Genomic DNA extracted from these tissue samples was used for PCR amplification of the cytochrome c oxidase subunit I gene (cox1) of Sarcocystis spp. and the partial sequence of small subunit ribosomal RNA (18S rRNA) gene of the S. cameli. The PCR products were purified, sequenced and analyzed using bioinformatics tools. Based on phylogenetic analysis of the cox1 gene, the se

Molecular Prevalence of Sarcocystis spp. and Toxoplasma gondii in Slaughtered Equids in Northern Tunisia.

Sarcocystis spp. and Toxoplasma gondii are two apicomplexan protozoa that infect a broad range of vertebrates, however, little is known about the infection of equids with these parasites. A total of 184 slaughtered equids from slaughterhouses of Bizerte and Tunis located in Northern Tunisia, were examined for meat infections with Sarcocystis spp. and T. gondii by PCR. The prevalence of infections with Sarcocystis spp. and T. gondii were 38% (95% CI: 31-45) and 39.7% (95% CI: 32.6-46.7), respectively. The highest prevalence of infection with Sarcocystis spp. was observed in donkeys (48.6%; 95%

First molecular characterization of Sarcocystis neurona causing meningoencephalitis in a domestic cat in Brazil.

Sarcocystis neurona is the main agent associated with equine protozoal myeloencephalitis (EPM). Apart from horses, S. neurona has been occasionally described causing neurologic disease in several other terrestrial animals as well as mortality in marine mammals. Herein, we describe the clinical, pathological, and molecular findings of a fatal case of S. neurona-associated meningoencephalitis in a domestic cat. The causing agent was analyzed by multilocus genotyping, confirming the presence of S. neurona DNA in the tissue samples of the affected animal. Significant molecular differences were fou

In silico identification of immunotherapeutic and diagnostic targets in the glycosylphosphatidylinositol metabolism of the coccidian Sarcocystis aucheniae.

Meat of the South American camelids (SACs) llama and alpaca is an important source of animal protein and income for rural families in the Andes, and a product with significant growth potential for local and international markets. However, infestation with macroscopic cysts of the coccidian protozoon Sarcocystis aucheniae, a parasitosis known as SAC sarcocystosis, significantly hampers its commercialization. There are no validated methods to diagnose the presence of S. aucheniae cysts other than carcass examination. Moreover, there are no available drugs or vaccines to cure or prevent SAC sarco

Molecular and Microscopic Identification of Sarcocystis spp. in the Intestines of the Tawny Owl (Strix aluco) in Lithuania.

The role of birds of the Strigiformes order in the transmission of globally distributed Sarcocystis parasites remains insufficiently studied. In the present work we aimed to evaluate the diversity of Sarcocystis species in the intestines of 22 naturally infected Tawny Owls (Strix aluco) sampled in Lithuania. The Sarcocystis infection was established using combined light microscopy and DNA sequence analysis. Samples were analyzed using Sarcocystis-genus and species-specific primers in nested PCR followed by Sanger sequencing. Sporocysts of Sarcocystis spp. were detected in 45.5% of samples, whi
