Raised Intracellular Calcium Contributes to Ischemia-Induced Depression of Evoked Synaptic Transmission.

Oxygen-glucose deprivation (OGD) leads to depression of evoked synaptic transmission, for which the mechanisms remain unclear. We hypothesized that increased presynaptic [Ca2+]i during transient OGD contributes to the depression of evoked field excitatory postsynaptic potentials (fEPSPs). Additionally, we hypothesized that increased buffering of intracellular calcium would shorten electrophysiological recovery after transient ischemia. Mouse hippocampal slices were exposed to 2 to 8 min of OGD. fEPSPs evoked by Schaffer collateral stimulation were recorded in the stratum radiatum, and whole ce

Melatonin inhibits voltage-sensitive Ca(2+) channel-mediated neurotransmitter release.

Melatonin is involved in various neuronal functions such as circadian rhythmicity and thermoregulation. Melatonin has a wide range of pharmacologically effective concentration levels from the nanomolar to millimolar levels. Recently, the antiepileptic effect of high dose melatonin has been the focus of clinical studies; however, its detailed mechanism especially in relation to neurotransmitter release and synaptic transmission remains unclear. We studied the effect of melatonin at high concentrations on the neurotransmitter release by monitoring norepinephrine release in PC12 cells, and excita

Loss of long-term potentiation in the hippocampus after experimental subarachnoid hemorrhage in rats.

Survivors of aneurysmal subarachnoid hemorrhage (SAH) often suffer from cognitive impairment such as memory loss. However, the underlying mechanisms of these impairments are not known. Long-term potentiation (LTP) of synapses in the hippocampus is generally regarded as a molecular substrate of memory. The purpose of this study was to examine the effect of SAH on LTP in the hippocampal Schaffer collateral (CA3-CA1) pathway in a rat model of SAH. We found SAH caused significant vasospasm of the middle cerebral artery (MCA) compared to saline injected or sham controls (P<0.001). Basic neurotransm

Effects of saccharin intake on hippocampal and cortical plasticity in juvenile and adolescent rats.

The sensory system is developed and optimized by experiences given in the early phase of life in association with other regions of the nervous system. To date, many studies have revealed that deprivation of specific sensory experiences can modify the structure and function of the central nervous system; however, the effects of sensory overload remains unclear. Here we studied the effect of overloading the taste sense in the early period of life on the synaptic plasticity of rat hippocampus and somatosensory cortex. We prepared male and female Sprague Dawley rats with ad libitum access to a 0.1

Synaptic transmission and hippocampal long-term potentiation in transgenic mice expressing FAD-linked presenilin 1.

Mutations in two related genes, presenilin 1 and presenilin 2 (PS1 and PS2), cause a subset of early-onset familial Alzheimer's disease (FAD). PS1 is expressed in a variety of neuronal and peripheral tissues, including neuronal populations known to be at risk in Alzheimer's disease such as CA1 hippocampal neurons. To examine whether FAD-linked mutations in PS1 directly influence the physiology of learning and memory, we measured the field excitatory postsynaptic potential (fEPSP) at the Schaffer collateral-CA1 synapse in hippocampal slices. Basal synaptic transmission and long-term potentiatio

Long-term potentiation deficits and excitability changes following traumatic brain injury.

The effects of traumatic brain injury (TBI) on hippocampal long-term potentiation (LTP) and cellular excitability were assessed at postinjury days 2, 7, and 15. TBI was induced using a well-characterized central fluid-percussion model. LTP of the Schaffer collateral/commissural system was assessed in vivo in urethane-anesthetized rats. Significant LTP of the population excitatory postsynaptic potential (EPSP) slope was found only in controls, and no recovery to control levels was observed for any postinjury time point. Four measurement parameters reflecting pyramidal cell discharges (populatio

Chronic theophylline treatment in vivo increases high affinity adenosine A1 receptor binding and sensitivity to exogenous adenosine in the in vitro hippocampal slice.

The present investigation examined the effects of chronic treatment with the adenosine receptor antagonist theophylline in vivo, on in vitro hippocampal electrophysiology and adenosine A1 receptor binding in the same animals. Adult rats were injected once daily (i.p.) with theophylline for 1 week at 75 mg/kg, followed by an additional week at 100 mg/kg, or with saline for the same 2-week period. Two days following the last injection, hippocampal slices were prepared and population spikes recorded from the pyramidal cell layer of area CA1 were elicited by Schaffer collateral-commissural fiber s

Interstitial PCO2 and pH in rat hippocampal slices measured by means of a novel fast CO2/H(+)-sensitive microelectrode based on a PVC-gelled membrane.

We describe here the construction and properties of a double-barrelled microelectrode (tip diameter 4-10 microns) which permits simultaneous measurements of PCO2 and pH, and which has a 90% response time of only one or a few seconds for a step change in PCO2. The fast response of the CO2-sensitive barrel is due to (i) the use of a PVC-gelled (tridodecylamine-containing) membrane solution which enables the construction of extremely short (> or = 4 microns), yet mechanically stable, membrane columns, and (ii) the presence of carbonic anhydrase in the filling solution. [Development of the neuronal structure of the hippocampus during pre- and post-natal ontogenesis in the albino rat. I. Neurohistological demonstration of the development of lung-axonal neurons in the CA3 and CA4 regions].

In this paper qualitative investigations by means of light microscopy (Golgi impregnation) considering the ontogenesis of efferent neurons of the CA3, CA4a, CA4b regions in the rat hippocampus are presented following the already described development of CA1 pyramids (MINKWITZ and HOLZ 1975). After the migration of neuroblasts the types of projective neurons can be distinguished in the mentioned regions at the end of the prenatal period. During the first postnatal time particularly the basal dendritic tree develops expressing more distinctly the typical forms of pyramids (CA3, CA4a) and the of

Autism spectrum disorder related phenotypes in a mouse model lacking the neuronal actin binding protein profilin 2.

INTRODUCTION: Profilin 2 (PFN2) is an actin binding protein highly expressed in the brain that participates in actin dynamics. It has been shown in vitro and in vivo that in neurons it functions both post-synaptically to shape and maintain dendritic arborizations and spine density and plasticity, as well as pre-synaptically to regulate vesicle exocytosis. PFN2 was also found in protein complexes with proteins that have been implicated in or are causative of autism spectrum disorder. METHODS: We employ a genetically engineered knock-out mouse line for Pfn2 that we previously generated to study

Conditional deletion of KCC2 impairs synaptic plasticity and both spatial and nonspatial memory.

The postsynaptic inhibition through GABAA receptors (GABAAR) relies on two mechanisms, a shunting effect due to an increase in the postsynaptic membrane conductance and, in mature neurons, a hyperpolarization effect due to an entry of chloride into postsynaptic neurons. The second effect requires the action of the K+-Cl- cotransporter KCC2 which extrudes Cl- from the cell and maintains its cytosolic concentration very low. Neuronal chloride equilibrium seems to be dysregulated in several neurological and psychiatric conditions such as epilepsy, anxiety, schizophrenia, Down syndrome, or Alzheim

Cytoarchitecture of the Hippocampal Formation in the African Giant Rat (Cricetomys gambianus, Waterhouse).

The African Giant Rat, AGR is an indigenous nocturnal rodent noted for its unique olfactory and cognitive abilities. They have been deployed more recently in the detection of landmines and diagnosis of tuberculosis - two scourges that have had a tremendous negative impact on the African landscape. This olfactory-aided cognition has been linked to the hippocampus. While the anatomical infrastructure of the olfactory bulb of the AGR has been elucidated, little is known about the adaptive cytoarchitecture of the AGR hippocampal formation. This study describes the histological features, including

Activity-Dependent Changes in Axonal Action Potential Latency Coordinated with Synaptic Potentiation in Individual Hippocampal Neurons.

Neural plasticity enables the nervous system to adapt its structure and function in response to experience. Although synaptic plasticity is a central cellular mechanism underlying learning and memory, action potential propagation along axons is also subject to plastic regulation and critically shapes neural computation. However, how these distinct forms of plasticity are coordinated within individual neurons remains poorly understood. Here, we simultaneously monitored synaptic responses and antidromically evoked action potentials in hippocampal CA1 pyramidal neurons from male rats using whole-

Biohybrid restoration of the hippocampal loop re-establishes the non-seizing state in anin vitromodel of limbic seizures.

Objective. The compromise of the hippocampal loop is a hallmark of mesial temporal lobe epilepsy (MTLE), the most frequent epileptic syndrome in the adult population and the most often refractory to medical therapy. Hippocampal sclerosis is found in >50% of drug-refractory MTLE patients and primarily involves the CA1, consequently disrupting the hippocampal output to the entorhinal cortex (EC). Closed-loop deep brain stimulation is the latest frontier to improve drug-refractory MTLE; however, current approaches do not restore the functional connectivity of the hippocampal loop, they are design

Phenobarbital does not worsen outcomes of neonatal hypoxia on hippocampal LTP on rats.

INTRODUCTION: Neonatal hypoxia is a common cause of early-life seizures. Both hypoxia-induced seizures (HS), and the drugs used to treat them (e.g., phenobarbital, PB), have been reported to have long-lasting impacts on brain development. For example, in neonatal rodents, HS reduces hippocampal long-term potentiation (LTP), while PB exposure disrupts GABAergic synaptic maturation in the hippocampus. Prior studies have examined the impact of HS and drug treatment separately, but in the clinic, PB is unlikely to be given to neonates without seizures, and neonates with seizures are very likely to

Pre-synaptic release deficits in a DYT1 dystonia mouse model.

DYT1 early-onset generalized torsion dystonia (DYT1 dystonia) is an inherited movement disorder caused by mutations in one allele of DYT1 (TOR1A), coding for torsinA. The most common mutation is a trinucleotide deletion (ΔGAG), which causes a deletion of a glutamic acid residue (ΔE) in the C-terminal region of torsinA. Although recent studies using cultured cells suggest that torsinA contributes to protein processing in the secretory pathway, endocytosis, and the stability of synaptic proteins, the nature of how this mutation affects synaptic transmission remains unclear. We previously reporte

Group 1 and 2 metabotropic glutamate receptors play differential roles in hippocampal long-term depression and long-term potentiation in freely moving rats.

This study examined the role of metabotropic glutamate receptors (mGluRs) in hippocampal long-term depression (LTD) in vivo. The group 1 mGluR antagonist (S)4-carboxyphenylglycine (4CPG), group 1/2 antagonist (RS)-alpha-methyl-4-carboxyphenylglycine (MCPG), and group 2 antagonists (RS)-alpha-methylserine-O-phos-phate monophenyl ester (MSOPPE) and (2S)-alpha-ethylglutamic acid (EGLU) were used. The NMDA receptor antagonist D(-)-2-amino-5-phosphonopentanoic acid (AP5) was used to examine the NMDA receptor contribution to the observed LTD. ATF4 Modulates Group 1 mGluR-Dependent molecular signaling, hippocampal plasticity and behavior.

Group 1 metabotropic glutamate receptors (Gp1 mGluRs) are critical effectors in behavior. Abnormal Gp1 mGluR signaling is frequently observed in neurodevelopmental and cognitive disorders. It is established that activation of Gp1 mGluR elicits translation-dependent neuronal plasticity mechanisms, such as long-term synaptic depression (LTD). However, whether and how transcription is involved is not clear. In this study, we study a cellular stress associated transcription factor, activation transcription factor 4 (ATF4). We first employed RNA sequencing and revealed that knocking down ATF4 using

Modulation of temporoammonic-CA1 synapses by neuropeptide Y is through Y1 receptors in mice.

Reduced levels of neuropeptide Y (NPY), an abundant neuromodulator in the brain, are linked to multiple neuropsychiatric disorders, including post-traumatic stress disorder (PTSD). The CA1 region of hippocampus is important for anxiety regulation and highly expresses NPY. Injecting NPY into CA1 is anxiolytic and alleviates behavioral symptoms in a model of traumatic stress; these anxiolytic effects are blocked by a Y1 receptor antagonist. However the location of Y1Rs that mediate NPY's anxiolytic effects in CA1 remains unclear. CA1 receives inputs from entorhinal cortex through the temporammon
