Room-Temperature Noncollinear Ferroelectricity in van der Waals WO_{2}Cl_{2} with a Wide Bandgap.

Low-dimensional ferroelectrics are attractive for their promising prospects in nanoelectronics. Compared with widely used ferroelectric perovskites, most low-dimensional ferroelectrics exhibit several inborn weaknesses such as small bandgaps (mostly <2  eV, i.e., semiconductorslike) or faint polarizations (e.g., <1  μC/cm^{2} for sliding ferroelectrics even if their bandgaps can be large). Here we experimentally demonstrate the room-temperature ferroelectricity of van der Waals WO_{2}Cl_{2}. The well-tested d^{0} rule inherited from ferroelectric perovskites leads to a large dipole (∼3  eÅ) fr

Method for quality-assessing excised human corneal buttons for nanoparticle visualization.

This study introduces a methodology for analyzing light transmission and nanoparticle penetration in excised human corneas using multiphoton microscopy. By developing an accessible approach utilizing the laser from a confocal or multiphoton microscope for assessing corneal quality via light transmission, we measured transmission to be over 90% through corneal tissue across a wavelength range of 700-1060 nm. The method enables precise optical measurements crucial for validating tissue quality before further experimentation without the need for separate equipment. Additionally, the study demonst

The Effect of Diabetes Mellitus Type 1 on the Energy Metabolism of Hepatocytes: Multiphoton Microscopy and Fluorescence Lifetime Imaging.

A decrease in the regenerative potential of the liver during the development of non-alcoholic fatty liver disease (NAFLD), which is observed in the vast majority of patients with diabetes mellitus type 1, significantly increases the risk of postoperative liver failure. In this regard, it is necessary to develop new approaches for the rapid intraoperative assessment of the condition of liver tissue in the presence of concomitant liver pathology. A modern label-free approach based on multiphoton microscopy, second harmonic generation (SHG), and fluorescence lifetime imaging microscopy (FLIM) all

Revisiting the Cornea and Trabecular Meshwork Junction With 2-Photon Excitation Fluorescence Microscopy.

PURPOSE: To investigate the collagen and elastin architecture at the junction of the human cornea and trabecular meshwork (TM). METHODS: The cornea, TM, and ciliary body (CB) tendons of unfixed human corneal buttons were imaged with an inverted 2-photon excited fluorescence microscope (FluoView FV-1000; Olympus, Central Valley, PA). The laser (Ti:sapphire) was tuned to 850 nm for 2-photon excitation. Backscatter signals of second harmonic generation and autofluorescence were collected through a 425/30-nm emission filter and a 525/45-nm emission filter, respectively. The second harmonic genera

Visualization of lymphatic vascular invasion in breast cancer by multiphoton microscopy.

Lymphatic vascular invasion (LVI) is regarded as one of the independent factors which affect the prognosis of breast cancer. Once LVI is formed, it indicates the tumor has metastasized or has the possibility of metastasis. In this work, multiphoton microscopy (MPM), which relies on the two-photon excited fluorescence (TPEF) and second harmonic generation (SHG), was applied to identify the typical morphology of LVI and also visualize the histological features of LVI. Furthermore, the pixel density of collagen fibers was extracted as a quantitative parameter to differentiate LVI from the ductal

A Custom Multiphoton Microscopy Platform for Live Imaging of Mouse Cornea and Conjunctiva.

Conventional histological analysis and cell culture systems are insufficient to simulate in vivo physiological and pathological dynamics completely. Multiphoton microscopy (MPM) has become one of the most popular imaging modalities for biomedical study at cellular levels in vivo, advantages include high resolution, deep tissue penetration and minimal phototoxicity. We have designed an MPM imaging platform with a customized mouse eye holder and a stereotaxic stage for imaging ocular surface in vivo. Dual fluorescent protein reporter mouse enables visualization of cell nuclei, cell membranes, ne

Determination of extracellular matrix collagen fibril architectures and pathological remodeling by polarization dependent second harmonic microscopy.

Polarization dependence second harmonic generation (P-SHG) microscopy is gaining increase popularity for in situ quantification of fibrillar protein architectures. In this report, we combine P-SHG microscopy, new linear least square (LLS) fitting and modeling to determine and convert the complex second-order non-linear optical anisotropy parameter ρ of several collagen rich tissues into a simple geometric organization of collagen fibrils. Modeling integrates a priori knowledge of polyhelical organization of collagen molecule polymers forming fibrils and bundles of fibrils as well as Poisson ph

Polychromatic Polarization Microscopy Differentiates Collagen Fiber Signatures in Benign Pancreatic Tissue and Pancreatic Ductal Adenocarcinoma.

The orientation of collagen fibers in relation to malignant epithelium is known to carry prognostic information in a variety of tissues. The data are the strongest for breast and pancreatic ductal adenocarcinoma. However, information inherent in collagen fiber topology in malignant tissues remains untapped in daily surgical pathology practice, largely because collagen fibers within areas of desmoplasia cannot be resolved with standard diagnostic microscopy. The methodologies used to visualize collagen fiber orientation are either of insufficient resolution to consistently capture collagen fibe

Concise report: a minimal-invasive method to retrieve and identify entheseal tissue from psoriatic arthritis patients.

OBJECTIVES: To establish a minimally invasive biopsy technique for the analysis of entheseal tissue in patients with psoriatic arthritis (PsA). METHODS: Human cadavers were used for establishing the technique to retrieve tissue from the lateral humeral epicondyle enthesis (cadaveric biopsies). After biopsy, the entire enthesis was surgically resected (cadaveric resections). Biopsies and resections were assessed by label-free second harmonic generation (SHG) microscopy. The same technique was then applied in patients with PsA with definition of entheseal tissue by SHG, staining of CD45+immune

Longitudinal Single-Axon-Resolution Imaging of Peripheral Nerve Injury Response in Mice Using an Optical Window Implant.

Chronic, high-resolution imaging of peripheral nerve injury and regeneration remains a major technical challenge due to the optical inaccessibility of peripheral nerves embedded in soft tissue. Here, we present a broadly applicable approach for longitudinal, high-resolution optical imaging of peripheral nerves in vivo that combines a flexible, optically transparent polydimethylsiloxane (PDMS) window along with multimodal microscopy. The PDMS window is implanted in the skin above the mouse sciatic nerve, providing stable, motion-tolerant optical access for over one month without impairing nerve

Collagen organization and structure in FLBN5-/- mice using label-free microscopy: implications for pelvic organ prolapse.

Pelvic organ prolapse (POP) is a gynecological disorder described by the descent of superior pelvic organs into or out of the vagina as a consequence of disrupted muscles and tissue. A thorough understanding of the etiology of POP is limited by the availability of clinically relevant samples, restricting longitudinal POP studies on soft-tissue biomechanics and structure to POP-induced models such as fibulin-5 knockout (FBLN5-/-) mice. Despite being a principal constituent in the extracellular matrix, little is known about structural perturbations to collagen networks in the FBLN5-/- mouse cerv

Three-dimensional mapping of the orientation of collagen corneal lamellae in healthy and keratoconic human corneas using SHG microscopy.

Keratoconus is an eye disorder that causes the cornea to take an abnormal conical shape, thus impairing its refractive functions and causing blindness. The late diagnosis of keratoconus is among the principal reasons for corneal surgical transplantation. This pathology is characterized by a reduced corneal stiffness in the region immediately below Bowman's membrane, probably due to a different lamellar organization, as suggested by previous studies. Here, the lamellar organization in this corneal region is characterized in three dimensions by means of second-harmonic generation (SHG) microscop

A diffraction-limited scanning system providing broad spectral range for laser scanning microscopy.

Diversified research interests in scanning laser microscopy nowadays require broadband capability of the optical system. Although an all-mirror-based optical design with a suitable metallic coating is appropriate for broad-spectrum applications from ultraviolet to terahertz, most researchers prefer lens-based scanning systems despite the drawbacks of a limited spectral range, ghost reflection, and chromatic aberration. One of the main concerns is that the geometrical aberration induced by off-axis incidence on spherical mirrors significantly deteriorates image resolution. Here, we demonstrate

Enhanced piezoresponse in van der Waals 2D CuCrInP2S6 through nanoscale phase segregation.

van der Waals metal chalcogen thiophosphates have drawn elevated interest for diverse applications, including energy harvesting, electronics and optoelectronics. Despite this progress, the role of nanoscale ion migration in complex intermediary thiophosphate compounds has not been well understood, resulting in their structure-property characteristics remaining elusive. Herein, we focus on copper-deficient CuCrInP2S6 as a prototypic layered thiophosphate compound to address this shortcoming. Piezo force microscopy reveals that this material exhibits unusual cage-like domain networks with an enh

Dentin-cement interfacial interaction: calcium silicates and polyalkenoates.

The interfacial properties of a new calcium-silicate-based coronal restorative material (Biodentine™) and a glass-ionomer cement (GIC) with dentin have been studied by confocal laser scanning microscopy (CLSM), scanning electron microscopy (SEM), micro-Raman spectroscopy, and two-photon auto-fluorescence and second-harmonic-generation (SHG) imaging. Results indicate the formation of tag-like structures alongside an interfacial layer called the "mineral infiltration zone", where the alkaline caustic effect of the calcium silicate cement's hydration products degrades the collagenous component of

Label-free structural and mechanical characterization of rat uterosacral ligaments.

This study presents quantitative applications of label-free imaging methods to characterize the structure of the uterosacral ligaments (USLs) before, during, and after loading. Rat USLs (n=14) were excised with their spinal and cervical attachments, clamped at these attachment sites, and pulled uniaxially in a custom-built tensile testing machine along their main in vivo loading direction. During uniaxial testing, optical coherence tomography (OCT) images were recorded, revealing the re-arrangement and failure of the structural components of the USLs. Before and after uniaxial testing, second

Epitaxially Integrated Electro-Optic KNbO3 Thin Films on Silicon.

The absence of a native electro-optic effect in silicon remains a fundamental limitation for integrated photonics, motivating the integration of ferroelectric oxides as active materials. Here, we report the first epitaxial integration of KNbO3 thin films on silicon, establishing a new materials platform for silicon-integrated electro-optics. Silicon integration of KNbO3 has remained unexplored due to potassium volatility challenges during synthesis. Employing suboxide molecular-beam sources assisted by in situ RHEED monitoring and guided by thermodynamic simulations, we establish an adsorption

Multiomic analysis of uterine leiomyomas in self-described Black and White women: molecular insights into health disparities.

BACKGROUND: Black women are at an increased risk of developing uterine leiomyomas and experiencing worse disease prognosis than White women. Epidemiologic and molecular factors have been identified as underlying these disparities, but there remains a paucity of deep, multiomic analysis investigating molecular differences in uterine leiomyomas from Black and White patients. OBJECTIVE: To identify molecular alterations within uterine leiomyoma tissues correlating with patient race by multiomic analyses of uterine leiomyomas collected from cohorts of Black and White women.
