Metabolic health and cardiovascular outcomes in treated hypertension: a secondary analysis of SPRINT.

BACKGROUND: Metabolic abnormalities frequently coexist with hypertension and independently contribute to cardiovascular disease (CVD) risk. Whether metabolic dysfunction predicts incident CVD events and/or modifies the efficacy of intensive blood pressure control in primary prevention remains uncertain. METHODS: Data from Systolic Blood Pressure Intervention Trial participants without baseline CVD or diabetes were analyzed. Metabolic health was classified based on triglycerides ≥ 150 mg/dL, high-density lipoprotein cholesterol < 40 mg/dL in men or < 50 mg/dL in women, fasting glucose ≥ 100 mg

Therapeutic targeting of local metabolic dysfunction in secondary lymphedema: mechanisms and opportunities.

Secondary lymphedema is a chronic and progressive disorder resulting from impaired lymphatic drainage after surgery, radiotherapy, infection, trauma, or other lymphatic insults. Its pathological features include chronic inflammation, adipose deposition, fibrosis, and structural and functional damage to lymphatic vessels. Previous studies have mainly explained disease progression from the perspectives of lymphatic injury and tissue remodeling, while local metabolic dysfunction and the associated immune alterations remain insufficiently defined. Recent evidence indicates that lymphedematous tiss

Environmental chemicals and metabolic disruption in primary and secondary human parathyroid tumors.

BACKGROUND: The incidence of primary hyperparathyroidism has increased 300% in the United States in the past 30 years, and secondary hyperparathyroidism is almost universal in patients with end-stage renal disease. We assessed the presence of environmental chemicals in human hyperplastic parathyroid tumors as possible contributing factors to this increase. METHODS: Cryopreserved hyperplastic parathyroid tumors and normal human parathyroids were analyzed by gas chromatography and liquid chromatography coupled to ultra-high-resolution mass spectrometry, bioinformatics, and biostatistics. RNA-protein complexes as key regulators of plant secondary metabolism: Biophysical interactions and functional significance.

RNA-protein complexes (RNPs) are central regulators of post-transcriptional gene expression and are increasingly implicated in controlling when, where and to what extent specialised (secondary) metabolic pathways operate in plants. Relative to transcription factor-centred regulation, RNPs can modify pathway output rapidly by modulating mRNA processing, stability, localisation and translation, thereby enabling plants to adjust metabolite production on developmentally and environmentally relevant timescales. Many regulatory RNPs assemble into membraneless biomolecular condensates (e.g., stress g

Exogenous melatonin modulates growth, antioxidant defense, and secondary metabolism of Matthiola incana under drought stress.

Drought stress is a major environmental constraint that adversely affects plant growth, physiological performance, and the accumulation of bioactive compounds. The use of biostimulants such as melatonin (MT) has emerged as a promising strategy to enhance plant tolerance to abiotic stress. In this study, the effects of exogenous MT on growth, physiological traits, antioxidant defense, and secondary metabolism of Matthiola incana under different irrigation regimes (100, 70, 40, and 25% field capacity, FC) were investigated. The experiment was conducted as a factorial design with four MT concentr…

Nano-selenium alleviates cadmium stress in cherry tomato via modulating subcellular distribution and secondary metabolism.

Cadmium (Cd), a toxic heavy metal, harms plant growth and human health. This study shows that under hydroponic conditions foliar nano-seleni … hich was absent in the sodium selenate treatment. These changes were primarily routed through plant secondary metabolism, ABC transporters, and amino acid (tryptophan, glycine, serine, phenylalanine) pathways. This study offers new mechanistic insights into mitigating heavy metal stress in cherry tomatoes via nanomaterial‑based metabolic modulation.

Comparative genomics of Brucella species reveals key determinants of secondary metabolism, antimicrobial resistance, and virulence.

The genomes of 40 Brucella strains were retrieved from the NCBI database to investigate Brucellosis at the genomic level, focusing on secondary metabolites, resistance genes, and virulence factors. Genome analysis software, secondary metabolite mining tools, and relevant gene databases were employed for detailed analysis. The genome sizes of these strains range from 4.88 to 6.00 MB, with G+C content between 53.5 and 60.5%. Phylogenetic analysis classified the strains into three distinct clades: Brucella anthropi CCUG 34461, Brucella sp. NBRC 13694, and Brucella anthropi MAG47. Pan-genome analy

Microbial diversity and secondary metabolism potential in relation to dark alterations in Paleolithic Lascaux Cave.

Tourism in Paleolithic caves can cause an imbalance in cave microbiota and lead to cave wall alterations, such as dark zones. However, the mechanisms driving dark zone formation remain unclear. Using shotgun metagenomics in Lascaux Cave's Apse and Passage across two years, we tested metabarcoding-derived functional hypotheses regarding microbial diversity and metabolic potential in dark zones vs unmarked surfaces nearby. Taxonomic and functional metagenomic profiles were consistent across years but divergent between cave locations. Aromatic compound degradation genes were prevalent inside and

Anti-Klebsiella pneumoniae activity of secondary metabolism of Achromobacter from the intestine of Periplaneta americana.

BACKGROUND: Klebsiella pneumoniae is one of the main pathogens of clinical isolation and nosocomial infections, as K. pneumoniae show broad-spectrum resistance to β-lactam and carbapenem antibiotics. It is emerging clinical need for a safe and effective drug to anti-K. pneumoniae. At present, Achromobacter mainly focused on its degradation of petroleum hydrocarbons, polycyclic aromatic hydrocarbons, assisting insects to decompose, degrade heavy metals and utilize organic matter, but there were few reports on the antibacterial activity of the secondary metabolites of Achromobacter. RESULTS: In

Cinacalcet lowers FGF-23 level together with bone metabolism in hemodialyzed patients with secondary hyperparathyroidism.

PURPOSE: Secondary hyperparathyroidism (sHPT) is associated with elevated levels of FGF-23, which in turn are connected to adverse outcomes in ESRD patients. The relationship between FGF-23 and bone metabolism in patients with sHPT treated with cinacalcet has not been studied. METHODS: Thirty-four stable chronically hemodialyzed patients with sHPT were prospectively followed during the treatment with cinacalcet without any changes in concurrent vitamin D or phosphate binder dose. Blood samples were collected at the start and after 6 months of study. Levels of osteocalcin (OC), cross-linked C-

Regional cerebral glucose metabolism in patients with secondary depressive episodes after fatal pancreatic cancer diagnosis.

BACKGROUND: Secondary depression is common in the clinical oncology setting after pancreatic cancer diagnosis, following which the patients have to face the fact that they have a cancer with an extremely poor prognosis. However, the specific pathophysiology remains unclear. The present study examined the regional cerebral glucose metabolism using F18-fluorodeoxyglucose (F18-FDG) positron emission tomography (PET) in antidepressant-naïve pancreatic cancer patients with a depressive episode after their cancer diagnosis and before their cancer treatment. METHODS: Regional cerebral glucose metabo

The impact of calcimimetics on mineral metabolism and secondary hyperparathyroidism in end-stage renal disease.

The impact of calcimimetics on mineral metabolism and secondary hyperparathyroidism in end-stage renal disease. Secondary hyperparathyroidism is often complicated by elevations in calcium and phosphorus either as a result of the disease per se or due to toxicity from current therapeutic options. These disturbances in mineral metabolism limit the successfulness of therapy and have been implicated as contributing to the development and progression of vascular calcification, an important and often overlooked component of cardiovascular disease in patients on dialysis. Phosphorus, active vitamin D

Secondary metabolism of theophylline biotransformation products in man--route of formation of 1-methyluric acid.

The route of formation of 1-methyluric acid (1MU) from theophylline has been investigated by i.v. administration of 1-methylxanthine (1MX) before and after allopurinol treatment and of 1,3-dimethyluric acid (1,3DMU) to healthy male volunteers. 1,3DMU was recovered unchanged in urine and was not demethylated to 1MU. By contrast after administration of 1MX the major part of the dose was recovered as 1MU. After allopurinol this was reversed with most of the 1MX dose being excreted unchanged and only a minor amount as 1MU. The results show that 1MU derived from theophylline is formed by an initial

StuPPO9 Enhances Drought Tolerance in Potato (Solanum tuberosum) by Coordinating Photosynthetic Stability and Secondary Metabolism Reprogramming.

Drought stress is a primary abiotic constraint limiting potato productivity. While polyphenol oxidase (PPO) is known to participate in stress responses, the specific role of StuPPO9 in drought tolerance remains poorly understood. In this study, we generated StuPPO9-overexpressing (OE) and CRISPR/Cas9-mediated knockout (C4) lines in the potato cultivar 'Atlantic'. Under sustained drought stress, OE lines exhibited significantly superior growth phenotypes compared to wild-type (WT) and C4 plants, characterized by increased leaf and root relative water content, root number and enhanced photosynth

Pathotype-specific roles of putative conidiation regulators in development, secondary metabolism, and pathogenicity in Fusarium fujikuroi.

Fusarium fujikuroi, the causal agent of rice bakanae disease, comprises two pathotypes with distinct symptoms: the fumonisin-producing blight type (B14) and the gibberellin-producing elongation type (B20). Asexual spore production and secondary metabolism are essential for disease transmission and development in F. fujikuroi, and conidiation regulators frequently influence secondary metabolism and other phenotypes in filamentous fungi. To investigate the genetic basis underlying differences between the two pathotypes, we generated deletion mutants for 14 genes corresponding to well-characteriz…
