Periodontal disease in sheep and cattle: Understanding dental health in past animal populations.

OBJECTIVE: To provide a comparative baseline for future studies of oral pathology in domestic livestock and to better understand connections between lesion prevalence and biological context in past animal populations. MATERIALS: Over 1600 sheep and cattle mandibles recovered from archaeological sites in England between 500 and 1300 CE. METHODS: A comprehensive investigation of periodontal disease was conducted based on four characteristics: dental calculus; periosteal new bone formation; alveolar recession; and ante-mortem tooth loss. The anatomical position and severity of these lesions wer

Virulence beneath the fleece; a tale of foot-and-mouth disease virus pathogenesis in sheep.

Foot-and-mouth disease virus (FMDV) is capable of infecting all cloven-hoofed domestic livestock species, including cattle, pigs, goats, and sheep. However, in contrast to cattle and pigs, the pathogenesis of FMDV in small ruminants has been incompletely elucidated. The objective of the current investigation was to characterize tissue- and cellular tropism of early and late stages of FMDV infection in sheep following three different routes of simulated natural virus exposure. Extensive post-mortem harvest of tissue samples at pre-determined time points during early infection (24 and 48 hours p

Lipid composition of microdomains is altered in neuronopathic Gaucher disease sheep brain and spleen.

Gaucher disease is a lysosomal storage disorder caused by a deficiency in glucocerebrosidase activity that leads to accumulation of glucosylceramide and glucosylsphingosine. Membrane raft microdomains are discrete, highly organized microdomains with a unique lipid composition that provide the necessary environment for specific protein-lipid and protein-protein interactions to take place. In this study we purified detergent resistant membranes (DRM; membrane rafts) from the occipital cortex and spleen from sheep affected with acute neuronopathic Gaucher disease and wild-type controls. We observ

Early and progressive circadian abnormalities in Huntington's disease sheep are unmasked by social environment.

Insidious changes in behaviour herald the onset of progressive neurodegenerative disorders such as Huntington's disease (HD), sometimes years before overt symptoms are seen. Sleep and circadian disturbances are particularly disruptive symptoms in patients with neurological disorders, but they are difficult to measure in humans. Here we studied circadian behaviour in transgenic HD sheep expressing the full-length human huntingtin protein with an expanded CAG repeat mutation in the juvenile range. Young HD sheep with no other symptoms exhibited circadian behavioural abnormalities that worsened w

Acute and chronic disease associated with naturally occurring T-2 mycotoxicosis in sheep.

A flock of approximately 1,000 sheep were exposed intermittently to food contaminated with T-2 toxin (T-2), a potent type-A trichothecene mycotoxin produced primarily by Fusarium sporotrichioides and Fusarium poae. In the acute stage of the intoxication, affected sheep developed anorexia, decreased water consumption, ruminal atony, soft faeces and apathy. One hundred and ninety of the exposed sheep died. The main gross lesions observed in animals dying during the acute disease were rumenitis and ulcerative abomasitis, depletion of lymphocytes in lymphoid organs, necrosis of the exocrine pancre

Model study of placental water transfer and causes of fetal water disease in sheep.

The purpose of the computer simulation was to use experimentally measured parameters of placental water transfer to compute conceptual water acquisition during the last third of ovine pregnancy and to evaluate the possible role of each of these parameters in the pathophysiology of polyhydramnios. Total conceptual water at birth was almost insensitive to the value of the placental filtration coefficient. It was more sensitive to fetal and maternal placental blood flows, the concentrations of actively transported or metabolically produced solutes in fetal plasma (bicarbonate, fructose, alpha-ami

Fatal hemolytic disease in sheep attributed to Leptospira interrogans serotype hardjo infection.

An outbreak of acute hemolytic disease in a large flock of sheep resulted in a mortality of approximately 5% in lambs. A diagnosis of Leptospira interrogans serotype hardjo infection was established on the basis of the presence of leptospires in the urine of one lamb, serologic findings in a guinea pig inoculated with this urine, L. hardjo titers in lambs, and pathologic findings. Prominent necropsy findings included severe icterus, thin watery blood, dark red to black kidneys and severe hemoglobinuria.

Clinical characterization and genotypic identification of border disease virus in sheep breeds in Iran.

BACKGROUND: Border disease (BD), caused by Border Disease Virus (BDV), leads to reproductive losses, congenital defects, and persistently infected (PI) lambs that maintain viral circulation. In Iran, existing reports are limited, confined to one area and breed, and rarely integrate clinical, pathological, virological, and genotypic data. This study investigates BD across Iranian sheep breeds, combining clinical observation, pathology, virus detection, and genotype analysis to clarify its epidemiology and support early recognition, targeted control measures, and improved small ruminant health a

Lean Metabolic Dysfunction-Associated Steatotic Liver Disease: A Wolf in Sheep's Clothing.

Metabolic dysfunction-associated steatotic liver disease (MASLD) affects approximately 32.4% of the global population. It is expected to be the primary cause of end-stage liver disease and liver transplantation. Obesity is a major risk factor for MASLD. However, a subset of patients with MASLD does not exhibit obesity-related traits, and this group is frequently neglected in clinical workups. Therefore, these patients do not receive timely diagnosis and treatment. Research has shown that MASLD is a multisystemic disease that is linked not only to type 2 diabetes mellitus but also to end-stage

Generation and Characterisation of Monoclonal Antibodies against Nairobi Sheep Disease Virus Nucleoprotein.

Nairobi sheep disease (NSD), caused by the viral agent NSD virus (NSDV), is a haemorrhagic fever disease affecting and inducing high mortality in sheep and goat populations. NSDV belongs to the genus Orthonairovirus of the Nairoviridae family from the order Bunyavirales. Other viruses circulating in livestock such as Crimean-Congo haemorrhagic fever virus (CCHFV) and Dugbe virus (DUGV) are members of the same genus and are reported to share antigenic features. There are very few available materials to study NSDV infection both in vitro and in vivo. In the present work, we characterised two mon

Cross-species efficacy of enzyme replacement therapy for CLN1 disease in mice and sheep.

CLN1 disease, also called infantile neuronal ceroid lipofuscinosis (NCL) or infantile Batten disease, is a fatal neurodegenerative lysosomal storage disorder resulting from mutations in the CLN1 gene encoding the soluble lysosomal enzyme palmitoyl-protein thioesterase 1 (PPT1). Therapies for CLN1 disease have proven challenging because of the aggressive disease course and the need to treat widespread areas of the brain and spinal cord. Indeed, gene therapy has proven less effective for CLN1 disease than for other similar lysosomal enzyme deficiencies. We therefore tested the efficacy of enzyme

Adeno-Associated Virus Gene Therapy in a Sheep Model of Tay-Sachs Disease.

Tay-Sachs disease (TSD) is a fatal neurodegenerative disorder caused by a deficiency of the enzyme hexosaminidase A (HexA). TSD also occurs in sheep, the only experimental model of TSD that has clinical signs of disease. The natural history of sheep TSD was characterized using serial neurological evaluations, 7 Tesla magnetic resonance imaging, echocardiograms, electrodiagnostics, and cerebrospinal fluid biomarkers. Intracranial gene therapy was also tested using AAVrh8 monocistronic vectors encoding the α-subunit of Hex (TSD α) or a mixture of two vectors encoding both the α and β subunits se

An EEG Investigation of Sleep Homeostasis in Healthy and CLN5 Batten Disease Affected Sheep.

UNLABELLED: Sheep have large brains with human-like anatomy, making them a useful species for studying brain function. Sleep homeostasis has not been studied in sheep. Here, we establish correlates of sleep homeostasis in sheep through a sleep deprivation experiment. We then use these correlates to elucidate the nature of sleep deficits in a naturally occurring ovine model of neuronal ceroid lipofuscinosis (NCL, Batten disease) caused by a mutation in CLN5 In humans, mutations in this gene lead to cortical atrophy and blindness, as well as sleep abnormalities. We recorded electroencephalograms

Antibody-induced anemia in fetal sheep: model for hemolytic disease of the fetus and newborn.

OBJECTIVE: Our purpose was to produce a condition analogous to alloimmune hemolytic disease of the fetus and newborn by infusing antierythrocyte antibodies in fetal sheep. METHODS: Antierythrocyte antibodies were infused intravascularly into late-gestation ovine fetuses over a 10-day period. Fetal blood was sampled daily for complete blood cell counts, blood gases, iron, erythropoietin (EPO), and electrolyte concentrations. Red cell mass (RCM) and blood volume were determined every other day using indicator dilution techniques. Results were compared with eight similarly aged control animals.

Autoantibodies, sheep cell agglutinins and anti-albumin antibodies in alcoholic liver disease.

The frequency of serum autoantibodies including anti-albumin antibodies was investigated for patients with various categories of alcoholic liver disease (ALD). The 95 patients were grouped into 3 categories: fatty liver (25 cases), alcoholic hepatitis (29 cases), and alcoholic cirrhosis (41 cases), and each group was matched with healthy controls by age, sex, ethnic origin and socio-economic status. For all patients with ALD, there was a 5-fold greater frequency over the controls of positive tests for antinuclear antibody (ANA), but titres were low; the pattern of ANA reactions was speckled in

Creutzfeldt-Jakob disease and sheep brain. A report from central and Southern Italy.

The places of origin of 7 out of 8 patients with CJD coincide with the distribution of sheep-rearing in central and southern Italy, confirming the suggested link between this disease and eating and-or handling sheep CNS tissue. Since 6 of the 8 cases were women, it seems more likely that the virus responsible for CJD enters the body through a break in the skin of the hands and forearms in the process of foodhandling rather than via the Digestive tract. This aspect should be borne in mind when investigating the problems of CJD transmission.
