To clarify the major factor caused by oxygen-enhancing charge production of Shewanella decolorationis NTOU1 towards a polarized anode, a series of experimental runs (i.e., with/without ambient air flushing and with/without ammonia addition as nitrogen source) were conducted in this study. Within 6-day of operation at +0.4 V vs. Ag|AgCl and starting with 35 mM of lactate, consistently the electrical charge production under the aerobic condition was higher than that under the anaerobic condition. In all the experimental runs, the values of nicotinamide adenine dinucleotide (NADH) production were

A 58-year-old male with severe psoriasis on Risankizumab presented with painful, left leg swelling with erythema and blisters concerning for necrotizing fasciitis. Intraoperative findings showed non-necrotizing bullous cellulitis. The blood and tissue cultures grew Shewanella algae. A handful case of non-necrotizing bullous cellulitis has been reported but this is the first documented case of non-necrotizing bullous cellulitis and bacteremia in PubMed.

Antibiotic resistance is increasing at an alarming rate worldwide, in large part due to their misuse and improper disposal. Antibiotics administered to treat human and animal diseases, including feed supplements for the treatment or prevention of disease in farm animals, have contributed greatly to the emergence of a multitude of antibiotic-resistant pathogens. Shewanella is one of many bacteria that have developed antibiotic resistance, and in some species, multiple-antibiotic resistance (MAR). Shewanella is a rod-shaped, Gram-negative, oxidase-positive, and H2S-producing bacterium that is na

Shewanella algae (S. algae) is a rare bacterium that causes infectious diseases in humans. Herein, we present a case of an 84-year-old man with S. algae-induced bacteremia and performed a review of 12 cases identified via a literature search and this case. Literature review of previous reports in Japan have revealed that 69.2% of patients with S. algae-induced bacteremia had a history of contact with fresh fish. Appropriate interviews of patients, especially in the hot season, and the accurate identification of the causative bacterium, by using techniques such as MALDI-TOF-MS and genetic testi

Cerebral venous sinus thrombosis in children is a rare complication of acute mastoiditis that can potentially be fatal. Clinical expertise is essential for early diagnosis and management due to its subtle course. We present the first known case of paediatric acute mastoiditis with venous sinus thrombosis caused by Shewanella algae and Actinomyces europaeus . A 17-year-old male presented clinical signs of right acute otitis media and mastoiditis. Brain computed tomography showed mastoid opacification, cerebral sinus thrombosis and an extradural collection. Microbiology revealed the presence of

The effects of the biological dissolution and reduction of goethite on mercury methylation by Shewanella oneidensis MR-1 were studied under laboratory conditions. The results showed that S. oneidensis MR-1 could dissolve goethite, and subsequently dissolved Fe3+ which was reduced to Fe2+. S. oneidensis MR-1 was also a kind of mercury methylation bacterial, which had a potential to transform inorganic mercury to methylmercury. The biological dissolution and reduction of goethite declined with the increasing initial dosage of goethite. The dissimilatory reduction of goethite could promote mercur

The toxic effects of dissolved versus bioconcentrated tributyl tin (TBT) on oyster larvae were compared. Water column TBT levels, which had no effect in solution, inhibited natural attachment and metamorphosis of oyster larvae on bottom surfaces due to bioconcentration by biofilms. This mechanism should be considered when evaluating heavy metal toxicity in the environment.

Pyrogenic black carbon, such as biochar and wildfire char, possesses a reversible electron storage capacity (ESC), allowing it to serve as an electron donor or acceptor for microorganisms. However, not all the ESC is bioavailable, and the proportion available varies by culture. We hypothesized microbes capable of producing soluble redox mediators could access the ESC in biochar interior and thus utilize a higher percentage of the ESC than organisms without a mediator. To test this hypothesis, we grew wild-type Shewanella oneidensis (MR-1) and Δ bfe MR-1, a mutant incapable of exporting flavins

Shewanella algae (S. algae) is an emerging marine zoonotic pathogen capable of causing various infections with a high mortality rate. Existing detection methods for S. algae tend to be nonspecific, complicated, and time-consuming. Here, we developed a CRISPR-based detection method, named MCTOS (Multiple Cross Displacement Amplification CRISPR/Cas12b-based Testing in One-Step), for S. algae detection. Through pan-genome analysis of 193 S. algae genomes against 103 other Shewanella genomes, we identified a S. algae-specific gene designated SA, as the detection target. MCTOS combines MCDA for tar

Antibiotic tolerance enables bacterial survival during antibiotic exposure and can promote the evolution of resistance, thereby threatening therapeutic efficacy. However, the molecular mechanisms underlying tolerance and its relationship with resistance remain incompletely understood. In this study, we show that a class D β-lactamase-deficient strain of Shewanella oneidensis (∆blaA) is highly susceptible to β-lactams, but retains the ability to survive lethal antibiotic concentrations. Under β-lactam stress, ∆blaA cells form reversible spheroplasts that revert to rod-shaped morphology and resu

The continuous outbreak of drug-resistant bacterial and viral infections imposes the need to search for new drug candidates. Natural products from marine bacteria still inspire the design of pharmaceuticals. Indeed, marine bacteria have unique metabolic flexibility to inhabit each ecological niche, thus expanding their biosynthetic ability to assemble unprecedented molecules. The One-Strain-Many-Compounds approach and tandem mass spectrometry allowed the discovery of a Shewanella aquimarina strain as a source of novel imidazolium alkaloids via molecular networking. The alkaloid mixture was sho

Microorganisms, iron minerals, and humic acid are widely common in the soil and water environment and closely interact within environmental processes. In this study, the Cr(VI) removal by Shewanella oneidensis MR-1 (S. oneidensis) was examined in the presence of goethite and humic acid (HA) to mimic the real environment situation. Scanning electron microscopy (SEM) combined with energy disperse spectroscopy (EDS), X-ray photoelectron spectroscopy (XPS), and electron paramagnetic resonance (EPR) technologies were used to probe the Cr(VI) reduction mechanism. Our results showed that S. oneidensi

C-type cytochromes located on the outer membrane (OMCs) of genus Shewanella act as the main redox-active species to mediate extracellular electron transfer (EET) from the inside of the outer membrane to the external environment: the central challenge that must be met for successful EET. The redox states of OMCs play a crucial role in dictating the rate and extent of EET. Here, we report that the surface wettability of the electrodes strongly influences the EET activity of living organisms of Shewanella loihica PV-4 at a fixed external potential: the EET activity on a hydrophilic electrode is m

Nitric oxide (NO) signaling in animals controls processes such as smooth muscle relaxation and neurotransmission by activation of soluble guanylate cyclase (sGC). Prokaryotic homologues of the sGC heme domain, called H-NOX domains, have been identified and are generally found in a predicted operon in conjunction with a histidine kinase. Here, we show that an H-NOX protein (SO2144) from Shewanella oneidensis directly interacts with the sensor histidine kinase (SO2145), binds NO in a 5-coordinate complex similar to mammalian sGC, and in that form inhibits the activity of a histidine kinase (SO21

Although biodegradable plastics have been developed to mitigate the environmental impact of conventional plastics, their timely degradation remains challenging. In this study, we aimed to accelerate the degradation of bioplastics using an electricity-assisted approach by engineering Shewanella oneidensis MR-1 to express triacylglycerol lipase (TGL) from Bacillus sp. JY35. The engineered strain was evaluated for its ability to degrade bioplastics, such as polybutylene succinate (PBS), polycaprolactone (PCL), and polybutylene adipate terephthalate (PBAT). Clear zone assays confirmed the degradat

Bacteria naturally alter the redox state of many compounds and perform atom-by-atom nanomaterial synthesis to create many inorganic materials. Recent advancements in synthetic biology have spurred interest in using biological systems to manufacture nanomaterials, implementing biological strategies to specify the nanomaterial characteristics such as size, shape, and optical properties. Here, we combine the natural synthetic capabilities of microbes with engineered genetic control circuits toward biogenically synthesized semiconductor nanomaterials. Shewanella can transfer electrons to various extracellular electron acceptors. We electrochemically investigated the pathway of extracellular electron transfer from Shewanella strain Hac319 to electrodes. A resting cell suspension of Shewanella strain Hac319 containing lactate produced a steady-state sigmoidal wave in the presence of flavin mononucleotide (FMN) in cyclic voltammetry, but not in the absence of FMN. A harvested cell suspension without cell-washing also produced a similar catalytic wave without any external addition of free FMN. The midpoint potentials of the two sigmoidal waves

β-Lactam antibiotics were the earliest discovered and are the most widely used group of antibiotics that work by inactivating penicillin-binding proteins to inhibit peptidoglycan biosynthesis. As one of the most efficient defense strategies, many bacteria produce β-lactam-degrading enzymes, β-lactamases, whose biochemical functions and regulation have been extensively studied. A signal transduction pathway for β-lactamase induction by β-lactam antibiotics, consisting of the major peptidoglycan recycling enzymes and the LysR-type transcriptional regulator, AmpR, has been recently unveiled in so

Bioremediation of chromium through the reduction of hexavalent chromium (as the chromate ion, CrO42-) is based on the notion that the product, trivalent chromium (Cr(III)), is less toxic than chromate. In this study, we show that soluble Cr(III), present at pH 6-8 as the Cr3+ ion and/or hydroxyl complexes (henceforth referred to as uncomplexed Cr(III)), can be found transiently in significant concentrations and has a deleterious effect on Shewanella sp. MR-4. However, Cr(lll) complexed to an organic ligand or precipitated as Cr(OH)3(s) has little or no effect on cells. Similarly, during the re
