Genetic diversity and DNA barcoding of the black fly (Diptera: Simuliidae) vectors of parasites causing human onchocerciasis in Guatemala.

Genetic variation based on mitochondrial cytochrome c oxidase I (COI) and II (COII) sequences was investigated for three black fly nominal species, Simulium metallicum Bellardi complex, S. callidum Dyar and Shannon, and S. ochraceum Walker complex, which are vectors of human onchocerciasis from Guatemala. High levels of genetic diversity were found in S. metallicum complex and S. ochraceum complex with maximum intraspecific genetic divergences of 11.39% and 4.25%, respectively. Levels of genetic diversity of these nominal species are consistent with species status for both of them as they are

Stability and change in the distribution of cytospecies of the Simulium damnosum complex (Diptera: Simuliidae) in southern Ghana from 1971 to 2011.

BACKGROUND: Simulium damnosum s.l., the most important vector of onchocerciasis in Africa, is a complex of sibling species that have been described on the basis of differences in their larval polytene chromosomes. These (cyto) species differ in their geographical distributions, ecologies and epidemiological roles. In Ghana, distributional changes have been recorded as a consequence of vector control and environmental change (e.g. deforestation), with potential disease consequences. We review the distribution of cytospecies in southern Ghana and report changes observed with reference to histori

Anticoagulant activity in salivary gland homogenates of Thyrsopelma guianense (Diptera: Simuliidae), the primary vector of onchocerciasis in the Brazilian Amazon.

In this study, anticoagulant activity was detected in salivary gland homogenates (SGHs) of Thyrsopelma guianense (Diptera: Simuliidae). The SGH yielded 1.07 microg +/- 0.03 (n = 15) of total soluble protein per pair of glands. In addition, following SDS-PAGE (12.5% gel) and silver nitrate staining, 12 polypeptides with molecular weights ranging from 14-69 kDa were detected in all physiological ages analyzed (12 h, 24 h, 48 h and 72 h following emergence). Coagulation bioassays showed that the SGHs had activities that interacted at all levels of coagulation (the intrinsic, extrinsic and common

Construction and characterisation of a BAC library made from field specimens of the onchocerciasis vector Simulium squamosum (Diptera: Simuliidae).

A Bacterial Artificial Chromosome (BAC) library was made from wild-caught Simulium squamosum, which is an important vector of human onchocerciasis. The library is composed of 12,288 BACs, with an average insert size of 128 kb, and is expected to contain ~1.54 GB of cloned DNA. Random BAC-end sequencing generated over 95 kb of DNA sequence data from which putative S. squamosum gene sequences and novel repetitive DNA families were identified, including DNA transposons, retrotransposons and simple sequence repeats (SSRs). The sequence survey also provided evidence of DNA of microbial origin, and

Evaluation of Partially Submerged Sticky Traps On Lake Spillways For Adult Black Fly (Diptera: Simuliidae) Surveillance and Arbovirus Detection.

Sentinel surveillance systems demonstrate an improved ability to supplement monitoring data and anticipate arbovirus outbreaks (i.e., sentinel avian species). Management complications can arise during unpredictable or unseasonal disease detections, especially in rural areas where resident distribution is patchy. Using spillways near residential lake communities as static surveillance locations, we tested a novel partially submerged sticky trapping technique and screened wild populations of adult female black flies (Diptera: Simuliidae) for West Nile virus (WNV) and eastern equine encephalitis

Simuliid blackflies (Diptera: Simuliidae) and ceratopogonid midges (Diptera: Ceratopogonidae) as vectors of Mansonella ozzardi (Nematoda: Onchocercidae) in northern Argentina.

Mansonella ozzardi, a relatively nonpathogenic filarial parasite of man in Latin America, is transmitted by either ceratopogonid midges or simuliid blackflies. In the only known focus of the disease in north-western Argentina the vectors have never been incriminated. This study investigated the potential vectors of M. ozzardi in this area. The only anthropophilic species of these Diptera families biting man at the time of the investigation were Simulium exiguum, S. dinellii, Culicoides lahillei and C. paraensis. Using experimentally infected flies S. exiguum and both species of Culicoides allo

Integrative taxonomy reveals a new species of black fly (Diptera: Simuliidae) in the Simulium varicorne species-group from Thailand.

Black flies are a group of hematophagous insects with high species diversity, driven by a combination of ecological, evolutionary, and life-history factors. By integrating DNA barcoding with traditional morphological method, a new black fly species was discovered. Simulium (Gomphostilbia) wiangpingense sp. nov. is described based on females, males, pupae, and mature larvae collected from Chiang Mai province, Thailand. This new species is assigned to the S. burtoni subgroup of the S. varicorne species-group within the subgenus Gomphostilbia Enderlein, 1921. It is characterized in the female and

Cytotaxonomic description of Simulium kaffaense, a new member of the S. damnosum complex (Diptera: Simuliidae) from south-western Ethiopia.

Although Ethiopia is one of the countries worst affected by human onchocerciasis, the exact taxonomic identity of the blackflies acting as the main vectors in the endemic areas has never been determined. A cytotaxonomic analysis of Simulium damnosum s.l. collected from three endemic sites in south-western Ethiopia has now revealed the existence of the 'Kisiwani' form (a non-anthropophilic cytoform that is common in East Africa) and a newly recognized species, Simulium kaffaense. Simulium kaffaense sp. nov. is differentiated from other members of the S. damnosum complex by six fixed inversions

Morphological description and genetic analysis of a new black fly species (Diptera: Simuliidae) in the subgenus Asiosimulium from central Thailand.

BACKGROUND: Black flies are among the most medically and veterinary important insects, as adult females of certain species are the sole vector of Onchocerca volvulus. Here, a new black fly species belonging to the subgenus Asiosimulium Takaoka & Choochote, 2005, is described and formally named as Simulium (Asiosimulium) kittipati sp. nov. METHODS: Pupae and larvae of black flies were collected from available substrates in the stream from central Thailand. Pupae were individually separated in plastic tubes and maintained until adult flies emerged. The emerged adult flies associated with their

Morphological redescription of Simulium takahasii (Rubtsov), the first species of the subgenus Wilhelmia Enderlein (Diptera: Simuliidae) recognized in East Asia.

Simulium takahasii (Rubtsov), which was originally described from Japan, and recorded from Korea and China, is the first among the 19 species of the subgenus Wilhelmia Enderlein recorded from East Asia. It is striking in mating, blood-feeding and ovipositing in captivity and in experimentally transmitting Dirofilaria immitis (Leidy) and Brugia pahangi (Buckley & Edeson), and it is a severe biter of cattle and horses, rarely of humans. Nevertheless, updated information about its morphological characteristics was lacking, making comparisons with related species described from China difficult, si

Identification of two West African black flies (Diptera: Simuliidae) of the Simulium damnosum species complex by analysis of cuticular paraffins.

Simulium damnosum is known to be a sibling species complex with at least 24 cytotaxonomically distinct forms in Africa. Of these, six occur in the area of West Africa covered by the World Health Organization Onchocerciasis Control Programme. They are all vectors of Onchocerca volvulus, and several are capable of migrating great distances. We report here a comparatively simple chemical method of species identification using individual adult females of pooled samples that have been collected and stored dry in glass vials. Females of two species, S. sirbanum and S. squamosum, were identified by e

Uptake of Onchocerca volvulus (Nematoda: Onchocercidae) by Simulium (Diptera: Simuliidae) is not strongly dependent on the density of skin microfilariae in the human host.

The relation between the number of microfilariae (mf) ingested by host-seeking vectors of human onchocerciasis and skin mf load is an important component of the population biology of Onchocerca volvulus, with implications for disease control and evaluation of the risk of transmission recrudescence. The microsimulation model ONCHOSIM has been used to assess such risk in the area of the Onchocerciasis Control Program (OCP) in West Africa, based on a strongly nonlinear relation between vector mf uptake and human mf skin density previously published. However, observed levels of recrudescence have

The cytotaxonomy of Simulium damnosum s.l. (Diptera: Simuliidae) from the Thyolo onchocerciasis focus in Malawi and description of a new member of the complex.

Larvae of the Simulium damnosum complex were collected in southern Malawi, in particular from the Thyolo onchocerciasis focus. Cytological identifications revealed the occurrence of S. (Edwardsellum) thyolense sp.nov., S. kilibanum Gouteux, 'Ketaketa', a new cytoform tentatively designated 'Mwamphanzi', and two unassigned specimens. Simulium thyolense sp.nov. is presumably the major vector of Onchocerca volvulus in the area, but it remains possible that the other cytospecies are secondary vectors. Simulium thyolense sp.nov. is most appropriately classified near the 'Sanje' subcomplex, whereas
