Sleep Disorders in Huntington's Disease.

Huntington's chorea (Huntington's disease, HD) is a genetic disorder caused by autosomal dominant mutation, leading to progressive neurodegenerative changes in the central nervous system. Involuntary movements such as chorea occur typically in HD patients, accompanied by progressive cognitive and psychiatric disturbances. Other common symptoms of HD are circadian and sleep abnormalities, which are observed from the earliest stages of the disease or even before the occurrence of clinical symptoms. The most common sleep problems reported by HD patients include insomnia, difficulties in falling a

Circadian rhythm disorder in a rare disease: Smith-Magenis syndrome.

Smith-Magenis syndrome (SMS) is a clinically recognizable contiguous gene syndrome, caused by interstitial deletion of chromosome 17p11.2. The SMS phenotype include distinctive facial features, developmental delay and neurobehavioral abnormalities. The patients present major sleep disturbances ascribed to a phase shift of their circadian rhythm of melatonin with a paradoxical diurnal secretion of the hormone. Treatment with morning beta-blockers and evening melatonin reinstated a normally timed melatonin circadian rhythm, improved daytime behavior and restored normal sleep habits, resulting in

Exploring the nexus: Sleep disorders, circadian dysregulation, and Alzheimer's disease.

We reviewed the connections among Alzheimer's disease (AD), sleep deprivation, and circadian rhythm disorders. Evidence is mounting that disrupted sleep and abnormal circadian rhythms are not merely symptoms of AD, but are also involved in accelerating the disease. Amyloid-beta (Aβ) accumulates, a feature of AD, and worsens with sleep deprivation because glymphatic withdrawal is required to clear toxic proteins from the brain. In addition, disturbances in circadian rhythm can contribute to the induction of neuroinflammation and oxidative stress, thereby accelerating neurodegenerative processes

Advances in the pharmacological management of non-24-h sleep-wake disorder.

Introduction: Melatonin, a hormone that regulates circadian rhythms and the sleep-wake cycle, is produced mainly during the dark period in the pineal gland and is suppressed by light exposure. Patients with non-24-h sleep-wake disorder (non-24) fail to entrain the master clock with the 24-h light-dark cycle due to the lack of light perception to the suprachiasmatic nucleus typically in totally blind individuals or other organic disorders in sighted individuals, causing a progressive delay in the sleep-wake cycle and periodic insomnia and daytime sleepiness.Areas covered: Herein, the authors re

Towards improving the prognosis of stroke through targeting the circadian clock system.

Rhythmicity of the circadian system is a 24-hour period, driven by transcription-translation feedback loops of circadian clock genes. The central circadian pacemaker in mammals is located in the hypothalamic suprachiasmatic nucleus (SCN), which controls peripheral circadian clocks. In general, most physiological processes are regulated by the circadian system, which is modulated by environmental cues such as exposure to light and/or dark, temperature, and the timing of sleep/wake and food intake. The chronic circadian disruption caused by shift work, jetlag, and/or irregular sleep-wake cycles

The Importance of Melatonin and Mitochondria Interaction in Mood Disorders and Schizophrenia: A Current Assessment.

Mitochondria play a critical role in regulating cellular functions, such as redox signaling, calcium homeostasis, and apoptosis. Also, mitochondria are crucial for neurogenesis and neuronal functions. Melatonin is an indole analog hormone, which is generally produced by the pineal gland. It plays a vital role in circadian rhythm and act as a powerful antioxidant by scavenging free radicals, immunomodulators, and anticancer agents. Schizophrenia and mood disorders are the two major psychiatric disorders. Disturbances of sleep and circadian rhythms are well-known symptoms of schizophrenia and mo

Light, melatonin and the sleep-wake cycle.

Blood levels of the pineal hormone melatonin are high at night and low during the day. Its secretion is regulated by a rhythm-generating system located in the suprachiasmatic nucleus of the hypothalamus, which is in turn regulated by light. Melatonin is regulated not only by that circadian oscillator but acts as a darkness signal, providing feedback to the oscillator. Melatonin has both a soporific effect and an ability to entrain the sleep-wake rhythm. It also has a major role in regulating the body temperature rhythm. Melatonin rhythms are altered in a variety of circadian rhythm disorders.

Sleep, circadian, and mental health in neurodivergent neurotypes: Lived experience perspective on the research landscape and roadmaps.

Neurodivergent neurotypes refer to brains that function differently from what is considered the societal standard or "stereotypical" norm, and include autism and attention deficit hyperactivity disorder (ADHD). Currently, global estimates suggest over 15% to 20% of the global population is neurodivergent. Autistic people and those with ADHD often experience mental health problems and have a heightened likelihood of anxiety and depression. Epidemiological evidence shows that sleep and circadian rhythm disruption, such as low self-reported sleep quality, insomnia symptoms, and delayed sleep-wake

"Circadian misalignment and the gut microbiome. A bidirectional relationship triggering inflammation and metabolic disorders"- a literature review.

Over the last decade, emerging studies have related the gut microbiome and gut dysbiosis to sleep and sleep disorders. For example, intermittent hypoxia associated with obstructive sleep apnea was shown to reproducibly alter the gut microbiome. Circadian rhythm disorders (CRD) (eg, shift work disorders, delayed sleep phase syndrome, and advanced sleep phase syndrome) constitute another group of conditions that might be influenced by gut dysbiosis. Indeed, both central and peripheral clocks can affect and be affected by gut microbiota and their metabolites. In addition, the tight rhythmic regul

[The association between circadian rhythm disruption-related behaviors and myopia in children and adolescents].

Child and adolescent myopia has become a critical global public health concern, with its prevalence steadily rising worldwide. Shifts in circadian behaviors, including irregular sleep schedules and increased nighttime exposure to artificial light, have contributed to widespread circadian rhythm disruptions manifested as sleep disorders, social jetlag, and excessive nighttime blue light exposure. Accumulating evidence suggests that circadian rhythm-disrupting behaviors are closely linked to the onset and progression of myopia in children and adolescents, although the underlying mechanisms remai

The impact of pharmacotherapy for premenstrual dysphoric disorder on sleep.

Premenstrual dysphoric disorder (PMDD) affects a subset of women of reproductive age, characterized by severe mood disturbances and physical symptoms during the luteal phase of the menstrual cycle. Symptoms include mood swings, irritability, anxiety, fatigue, physical discomfort, and disruptions to sleep and circadian rhythms, such as altered melatonin secretion. Despite the prevalence of these symptoms, the impact of PMDD treatments on sleep and circadian markers, like melatonin, remains insufficiently understood. This review examines how dysregulated serotonin signaling, disrupted allopregna

Sleep disorders in perinatal women.

Insufficient sleep is common in the general population, and can result from environmental and psychosocial factors, medical and psychiatric disorders, and sleep disorders, such as insomnia, circadian rhythm disorders, sleep apnoea and restless legs. Women are particularly at risk for sleep disorders, and complaints of sleep disturbance are more prevalent among women than men across the life span. During the perinatal period, many common sleep disorders, such as obstructive sleep apnoea or restless legs may be exacerbated, or in the case of insomnia or narcolepsy, treatment options may change.

Melatonin: Some Aspects of Biochemistry, Physiology, and Pharmacology.

Melatonin is a hormone synthesized by the pineal gland from tryptophan as a precursor and serotonin as an intermediate. It is secreted during the night and displays a peak around 3 a.m., depending on the individual chronotype. This rhythm is generated by an endogenous biological clock located in the suprachiasmatic nuclei (SCN) of the hypothalamus and entrained to the light/dark cycle (circadian rhythm). The roles of melatonin are numerous, but this hormone is described as a messenger of nighttime to the body, a modulator of circadian rhythmicity in mammals (including humans), and a regulator

[Chronobiological aspects of bipolar disorder].

BACKGROUND: Numerous symptoms of bipolar disorder are regulated by the circadian rhythm. Because of this association it is assumed that disruption of the circadian rhythm may be part of the pathomechanism of bipolar disorder. OBJECTIVES: A comparison and subsequent critical discussion of the current data situation on chronobiological aspects of bipolar disorder are presented. METHODS: A narrative literature search was carried out and the main findings are presented in a summarized form. RESULTS: There are a large number of animal and human studies investigating the connection between disord

Chronotherapeutics and psychiatry: setting the clock to relieve the symptoms.

Circadian rhythms are near 24-h cycles in a number of physiological and behavioural parameters and the underpinning circadian timing systems is one of the key homeostatic regulatory systems in mammalian physiology. Many common psychiatric conditions are associated with disrupted sleep, including a common occurrence of delayed or advanced phase sleep syndromes, which in themselves may be indicative of dysregulated circadian timing in these disorders. In this article we discuss the evidence for abnormal circadian rhythms in seasonal affective disorder, bipolar disorder and attention deficit/hype

Jet lag and other sleep disorders relevant to the traveler.

Sleep and wakefulness are governed by homeostatic and circadian regulatory processes, and perturbations therein are primarily responsible for the sleep disturbances associated with travel. Misalignment between endogenous rhythms and the light/dark cycle can result in circadian rhythm sleep disorders, including jet lag. This condition will be the primary focus of this review, with an emphasis on predisposing factors, preventative options, and treatment strategies.
