Tapeworm infection affects sleep-like behavior in three-spined sticklebacks.

Sleep is a complex and conserved biological process that affects several body functions and behaviors. Evidence suggests that there is a reciprocal interaction between sleep and immunity. For instance, fragmented sleep can increase the probability of parasitic infections and reduce the ability to fight infections. Moreover, viral and bacterial infections alter the sleep patterns of infected individuals. However, the effects of macro-parasitic infections on sleep remain largely unknown, and measuring sleep in non-model organisms remains challenging. In this study, we investigated whether macro-

Locomotor effects of a fibrosis-based immune response in stickleback fish.

The vertebrate immune system provides an impressively effective defense against parasites and pathogens. However, these benefits must be balanced against a range of costly side-effects including energy loss and risks of auto-immunity. These costs might include biomechanical impairment of movement, but little is known about the intersection between immunity and biomechanics. Here, we show that a fibrosis immune response to Schistocephalus solidus infection in freshwater threespine stickleback (Gasterosteus aculeatus) has collateral effects on their locomotion. Although fibrosis is effective at

The secretome of a parasite alters its host's behaviour but does not recapitulate the behavioural response to infection.

Parasites with complex life cycles have been proposed to manipulate the behaviour of their intermediate hosts to increase the probability of reaching their final host. The cause of these drastic behavioural changes could be manipulation factors released by the parasite in its environment (the secretome), but this has rarely been assessed. We studied a non-cerebral parasite, the cestode Schistocephalus solidus, and its intermediate host, the threespine stickleback (Gasterosteus aculeatus), whose response to danger becomes significantly diminished when infected. These altered behaviours appear o

Morphological and Molecular Identification of Stellantchasmus dermogenysi n. sp. (Digenea: Heterophyidae) in Thailand.

We tried a series of morphological and molecular approaches to identify a new species of Stellantchasmus (Digenea: Heterophyidae) originating from the wrestling half-beaked fish, Dermogenys pusillus of Thailand. Adult worm samples of the new species were recovered from hamsters experimentally infected with the metacercariae from D. pusillus in Thailand. Two isolates (Thai and Korean) of Stellantchasmus falcatus were used as comparative control groups. Worm samples of 3 Stellantchasmus groups were morphologically observed and molecularly analyzed with the mitochondrial cytochrome c oxidase 1 ge

Differential miRNA expression in the three-spined stickleback, response to environmental changes.

miRNAs play important role in the various physiological and evolutionary processes, however, there is no data allowing comparison of evolutionary differences between various ecotypes adapted to different environmental conditions and specimen demonstrating immediate physiological response to the environmental changes. We compared miRNA expression profiles between marine and freshwater stickleback populations of the three-spined stickleback to identify the evolutionary differences. To study the immediate physiological response to foreign environment, we explored the changes induced by transfer o

Pygidiopsis summa (Digenea: Heterophyidae): Status of Metacercarial Infection in Mullets from Coastal Areas in the Republic of Korea.

To know the infection status of zoonotic trematode metacercariae in brackish water fish, we surveyed mullets collected from 18 coastal areas in the Republic of Korea. The metacercariae of Pygidiopsis summa were detected in 236 (68.2%) out of 346 mullets examined. They were found in mullets from 15 areas except for those from Boseong-gun (Jeollanam-do), Pohang-si, and Uljin-gun (Gyeongsangbuk-do). Especially in mullets from Taean-gun (Chungcheongnam-do) and Geoje-si (Gyeongsangnam-do), their prevalences were 100% and 95.5%, and the average metacercarial density was more than 1,000 per fish. Stellantchasmus falcatus (Digenea: Heterophyidae) in Cambodia: Discovery of Metacercariae in Mullets and Recovery of Adult Flukes in an Experimental Hamster.

Stellantchasmus falcatus (Digenea: Heterophyidae) is first reported from Cambodia through recovery of the metacercariae from mullet fish and adult flukes from an experimentally infected hamster. We purchased 7 mullets, Chelon macrolepis, in a local market of Phnom Penh, Cambodia, and each of them was examined by the artificial digestion method on May 2010. The metacercariae of S. falcatus were detected in all mullets (100%) examined, and their average density was 177 per fish. They were elliptical, 220×168 μm in average size. They were orally infected to an hamster to obtain adult flukes. Adul

Centromere inactivation on a neo-Y fusion chromosome in threespine stickleback fish.

Having one and only one centromere per chromosome is essential for proper chromosome segregation during both mitosis and meiosis. Chromosomes containing two centromeres are known as dicentric and often mis-segregate during cell division, resulting in aneuploidy or chromosome breakage. Dicentric chromosome can be stabilized by centromere inactivation, a process which reestablishes monocentric chromosomes. However, little is known about this process in naturally occurring dicentric chromosomes. Using a combination of fluorescence in situ hybridization (FISH) and immunofluorescence combined with

Genotoxic effects of metal pollution in two fish species, Oreochromis niloticus and Mugil cephalus, from highly degraded aquatic habitats.

In Egypt, Lake Qaroun and its neighbouring fish farms are in a serious environmental situation as a result of pollution by agricultural sewage and domestic non-treated discharges. The present study aims to evaluate genotoxic effects of toxic metals in cultured and wild Nile tilapia, Oreochromis niloticus and mullet, Mugil cephalus collected from these contaminated aquatic habitats, in comparison with fish from a non-polluted reference site. Heavy-metal concentrations (Cu(2+), Zn(2+), Pb(2+), Fe(2+) and Mn(2+)) in water and sediment samples were recorded. The condition factor (CF) was taken as

Low-frequency alternating electric field superchilling mitigates ice crystal growth and preserves taste quality in Asian swamp eel (Monopterus albus).

This study investigated the effects of low-frequency alternating electric field-assisted superchilling (ESF) on the quality of Asian swamp eels. Compared to conventional superchilling (SF), ESF treatment significantly reduced ice crystal size, decreasing the equivalent diameters and cross-sectional areas by 19.6% and 35.5%, respectively, at day 8, while maintaining more regular crystal morphology. ESF also effectively suppressed the conversion of immobilized water to free water, delayed umami loss, and mitigated bitterness development, as confirmed by nuclear magnetic resonance and electronic

Eco-Evolutionary Dynamics of Infection and Immunity in Experimentally Founded Lake Populations of Threespine Stickleback.

AbstractHost-parasite interactions are likely to involve eco-evolutionary feedbacks. Parasites can impose strong selection on host immune traits. Evolution of these traits can suppress parasite abundance, which alters the strength of selection. Such eco-evo feedbacks can be hard to document in natural settings, where long-term interactions may have converged on relatively stable equilibria. However, when a species disperses into an unoccupied habitat, it can experience altered parasite abundance (e.g., enemy release) or encounter new parasite genotypes. These perturbations may be followed by o

Population-level immunologic variation in wild threespine stickleback (Gasterosteusaculeatus).

Wild organisms are regularly exposed to a wide range of parasites, requiring the management of an effective immune response while avoiding immunopathology. Currently, our knowledge of immunoparasitology primarily derives from controlled laboratory studies, neglecting the genetic and environmental diversity that contribute to immune phenotypes observed in wild populations. To gain insight into the immunologic variability in natural settings, we examined differences in immune gene expression of two Alaskan stickleback (Gasterosteus aculeatus) populations with varying susceptibility to infection

Molecular mechanisms of Eda-mediated adaptation to freshwater in threespine stickleback.

A main goal of evolutionary biology is to understand the genetic basis of adaptive evolution. Although the genes that underlie some adaptive phenotypes are now known, the molecular pathways and regulatory mechanisms mediating the phenotypic effects of those genes often remain a black box. Unveiling this black box is necessary to fully understand the genetic basis of adaptive phenotypes, and to understand why particular genes might be used during phenotypic evolution. Here, we investigated which genes and regulatory mechanisms are mediating the phenotypic effects of the Eda haplotype, a locus r

Selection on an extreme-yet-conserved larval life-history strategy in a tapeworm.

Evolutionary stasis characterizes many phenotypes, even ones that seem suboptimal. Among tapeworms, Schistocephalus solidus and its relatives have some of the shortest developmental times in their first intermediate hosts, yet their development still seems excessively long considering they can grow faster, larger, and safer in the next hosts in their complex life cycles. I conducted 4 generations of selection on the developmental rate of S. solidus in its copepod first host, pushing a conserved-but-counterintuitive phenotype toward the limit of known tapeworm life-history strategies. Faster pa

Complete Depletion of Primordial Germ Cells Results in Masculinization of Monopterus albus, a Protogynous Hermaphroditic Fish.

Primordial germ cells (PGCs) play an important role in sexual fate determination and gonadal development in gonochoristic fish, such as zebrafish and medaka. However, little is known about the function of PGCs in hermaphroditic fish. Rice field eel (Monopterus albus), a protogynous hermaphroditic fish, is an economically valuable aquaculture species. We eliminated PGCs in rice field eels during embryogenesis via morpholino-mediated knockdown dead end (dnd). The PGCs-depleted gonads developed into testis-like structures with Sertoli cells and Leydig cells. The gene expression pattern of 15-mont

Climate change facilitates a parasite's host exploitation via temperature-mediated immunometabolic processes.

Global climate change can influence organismic interactions like those between hosts and parasites. Rising temperatures may exacerbate the exploitation of hosts by parasites, especially in ectothermic systems. The metabolic activity of ectotherms is strongly linked to temperature and generally increases when temperatures rise. We hypothesized that temperature change in combination with parasite infection interferes with the host's immunometabolism. We used a parasite, the avian cestode Schistocephalus solidus, which taps most of its resources from the metabolism of an ectothermic intermediate

The first record of Ascocotyle (Phagicola) longa (Digenea: Heterophyidae) metacercariae as the fish-borne zoonotic trematode in Chelon auratus and Chelon saliens (Pisces: Mugilidae) around the cage culture in the Caspian Sea, Iran.

As a matter of fact, there has not been any reportage of the parasite of Ascocotyle (Phagicola) longa Ransom, 1920 from the Caspian Sea, Iran. During two years, April 2017 until 2018, 158 specimens of mullet fish, Chelon auratus and Chelon saliens (Risso, 1810) were collected and carefully examined considering the standard parasitological methods. Interestingly, the metacercaria was found in the stomach and intestine of the mullets. Morphological and morphometric analyses indicated that this parasite species belongs to Ascoctyle (Phagicola) longa. Overall infection mean intensity (52.1±128.3),

Ultrastructural and molecular characteristics of Kudoa crenimugilis n. sp. infecting intestinal smooth muscle of fringelip mullet Crenimugil crenilabis in the Red Sea.

This study describes infection of intestinal smooth muscle in fringelip mullets Crenimugil crenilabis with Kudoa crenimugilis n. sp. Of 30 individuals sampled from the Red Sea off Saudi Arabia, 6 (20%) were infected. Ovoid plasmodia (279-412 × 157-295 µm) in the smooth muscle of the intestine were packed with only mature myxospores with 4 valves. Specifically, light and transmission electron microscopy revealed quadrate myxospores with 4 equal, rounded, spore valves uniting at thin delicate suture lines. The mature myxospores were 8 (7-9) µm long, 5.2 (5-6) µm thick and 7.8 (7-8) µm wide. The

Sexual isolation promotes divergence between parapatric lake and stream stickleback.

Speciation can be initiated by adaptive divergence between populations in ecologically different habitats, but how sexually based reproductive barriers contribute to this process is less well understood. We here test for sexual isolation between ecotypes of threespine stickleback fish residing in adjacent lake and stream habitats in the Lake Constance basin, Central Europe. Mating trials exposing females to pairings of territorial lake and stream males in outdoor mesocosms allowing for natural reproductive behaviour reveal that mating occurs preferentially between partners of the same ecotype.

The Spatial and Temporal Repeatability of Genomic Responses to Natural Selection as Demonstrated in Stickleback Populations Experiencing Highly Dynamic Environments.

The evolution of genotypic parallelism under shared environmental conditions provides strong evidence for the role of natural selection. However, analyses typically examine genomic signatures of selection long after the putative selection event and only assess the repeatability of responses across spatial population replicates. This impedes our ability to attribute a particular response to a given selection pressure and to distinguish non-parallel responses caused by stochastic processes from those caused by local selection. As such, the consistency of natural selection over space and time is
