The role of mitochondrial Na⁺/Ca²⁺ exchanger in brain cell aging.

Considering the world's population aging and the importance of mitochondrial calcium regulation for all brain pathologies, researchers cannot neglect the role of mitochondrial sodium/calcium exchanger (NCLX) not only in pathologies like Alzheimer's and Parkinson's diseases, but in physiological "Работает" aging as well. Despite its critical role in mitochondrial metabolism upon neurodegeneration, the role of NCLX in physiological aging of CNS is almost unknown. NCLX interacts with regulatory partners like TMEM65 and signal pathways of PKA and HIF, connecting to broader metabolic networks of hyp

Ionic mechanisms for the transduction of acidic stimuli in rabbit carotid body glomus cells.

1. The release of [3H]dopamine (DA) in response to inhibition of the Na+ pump or to intracellular acid load was studied in rabbit carotid bodies (CB) previously incubated with the precursor [3H]tyrosine. The ionic requirements of the release response and the involvement of specific ion transport systems were investigated. 2. Inhibition of the Na+ pump, by incubating the CB with ouabain or in K(+)-free medium, evokes a DA release response which requires the presence of Na+ and Ca2+ in the medium and is insensitive to nisoldipine. This suggests that the response is triggered by entry of external

Empagliflozin Attenuates Myocardial Sodium and Calcium Dysregulation and Reverses Cardiac Remodeling in Streptozotocin-Induced Diabetic Rats.

Diabetes mellitus (DM) has significant effects on cardiac calcium (Ca2+) and sodium (Na⁺) regulation. Clinical studies have shown that empagliflozin (Jardiance™) has cardiovascular benefits, however the mechanisms have not been fully elucidated. This study aimed to investigate whether empagliflozin modulates cardiac electrical activity as well as Ca2+/Na⁺ homeostasis in DM cardiomyopathy. Electrocardiography, echocardiography, whole-cell patch-clamp, confocal microscopic examinations, and Western blot, were performed in the ventricular myocytes of control and streptozotocin-induced DM rats, wi

Mitochondrial cardiomyopathies feature increased uptake and diminished efflux of mitochondrial calcium.

Calcium (Ca2+) influx into the mitochondrial matrix stimulates ATP synthesis. Here, we investigate whether mitochondrial Ca2+ transport pathways are altered in the setting of deficient mitochondrial energy synthesis, as increased matrix Ca2+ may provide a stimulatory boost. We focused on mitochondrial cardiomyopathies, which feature such dysfunction of oxidative phosphorylation. We study a mouse model where the main transcription factor for mitochondrial DNA (transcription factor A, mitochondrial, Tfam) has been disrupted selectively in cardiomyocytes. By the second postnatal week (10-15day ol

Klotho attenuates isoproterenol-induced hypertrophic response in H9C2 cells by activating Na+/K+-ATPase and inhibiting the reverse mode of Na+/Ca2+-exchanger.

Cardiac hypertrophy plays a major role in heart failure and is related to patient morbidity and mortality. Calcium overloading is a main risk for cardiac hypertrophy, and Na+/K+-ATPase (NKA) has been found that it could not only regulate intracellular Na+ levels but also control the intracellular Ca2+ ([Ca2+]i) level through Na+/Ca2+-exchanger (NCX). Recent studies have reported that klotho could affect [Ca2+]i level. In this study, we aimed at exploring the role of klotho in improving isoproterenol-induced hypertrophic response of H9C2 cells. The H9C2 cells were randomly divided into control

Potential New Agents for the Management of Hyperkalemia.

Hyperkalemia is a common electrolyte disturbance with multiple potential etiologies. It is usually observed in the setting of reduced renal function. Mild to moderate hyperkalemia is usually asymptomatic, but is associated with poor prognosis. When severe, hyperkalemia may cause serious acute cardiac arrhythmias and conduction abnormalities, and may result in sudden death. The rising prevalence of conditions associated with hyperkalemia (heart failure, chronic kidney disease, and diabetes) and broad use of renin-angiotensin-aldosterone system (RAAS) inhibitors and mineralocorticoid receptor an

Regulation of Na/Ca exchange by cytoplasmic protons modifies intracellular calcium dynamics and the cardiac response to ischemia.

The cardiac sodium-calcium exchanger (NCX1) is a critical regulator of intracellular calcium (Ca) in cardiomyocytes. Although strongly regulated by pH, the role of NCX1 pH sensitivity in cardiac function and intracellular Ca regulation is not understood. We used CRISPR/Cas9 to produce a pH-resistant NCX1 mouse by replacing histidine 165 with alanine (H165A). H165A mice live into adulthood and are fertile. Comparative studies in isolated ventricular myocytes demonstrated that NCX1 current (INCX) was significantly inhibited by lowering pHi to ~6.5 in the wild type (WT) but was unaffected in myoc

The modulation of pacing-induced changes in intracellular sodium levels by extracellular Ca2+ in isolated perfused rat hearts.

This study demonstrates the inverse relationship between extracellular free calcium ([Ca(o)]f) and intracellular sodium ([Na(i)]) in isolated perfused rat hearts and thus supports the role of [Na(i)] in the "calcium paradox". It also shows that the extent of the increase in [Na(i)] (delta[Na(i)]), and the extent of the decrease in left ventricular developed pressure (deltaLVDP) in isolated perfused rat hearts, induced by pacing, is modulated by [Ca(o)]f. At low (0.24 mM) as well as normal (1.15 mM) [Ca(o)]f, [Na(i)] increased with pacing, progressively and significantly (P<0.01 and P<0.05, res

Overexpression of human mutated G93A SOD1 changes dynamics of the ER mitochondria calcium cycle specifically in mouse embryonic motor neurons.

Motor neurons vulnerable to the rapidly progressive deadly neurodegenerative disease amyotrophic lateral sclerosis (ALS) inherently express low amounts of calcium binding proteins (CaBP), likely to allow physiological motor neuron firing frequency modulation. At the same time motor neurons are susceptible to AMPA receptor mediated excitotoxicity and internal calcium deregulation which is not fully understood. We analysed ER mitochondria calcium cycle (ERMCC) dynamics with subsecond resolution in G93A hSOD1 overexpressing motor neurons as a model of ALS using fluorescent calcium imaging. When c

The effect of sodium transport and calcium channel inhibitors on phorbol ester-induced contraction of bovine airway smooth muscle.

We studied the role of sodium transport and calcium channels in protein kinase C mediated signal-transduction pathways in bovine airway smooth muscle. 4-beta phorbol 12,13 dibutyrate (PDB), an activator of protein kinase C, caused dose-related slowly developing contraction in bovine bronchial rings with a peak effect at 60-90 min. 4-alpha PDB, an inactive analogue, was without effect. Mean peak PDB-induced contraction (measured as a percentage of the maximum response to methacholine) was reduced from 122% to 20% when experiments were carried out in calcium-free fluids +EDTA (10(-3) M). Selective extraction of lithium over alkali and alkaline earth ions by synergistic solvent extraction.

Direct lithium extraction (DLE) from natural surface and geothermal brines is very challenging due to the low ratio of lithium to other metals, and the lack of suitable materials that bind lithium with sufficiently high selectivity. In this paper, a synergistic solvent extraction system is described that comprises a liquid ion exchanger (saponified bis(2-ethylhexyl)dithiophosphoric acid) and a lithium-selective ligand (2,9-dibutyl-1,10-phenanthroline) in an aliphatic diluent. The extraction mechanism was investigated and was confirmed to involve the binding of lithium to the selective ligand,

Analysis of cardiac mitochondrial Na+-Ca2+ exchanger kinetics with a biophysical model of mitochondrial Ca2+ handling suggests a 3:1 stoichiometry.

Calcium is a key ion and is known to mediate signalling pathways between cytosol and mitochondria and modulate mitochondrial energy metabolism. To gain a quantitative, biophysical understanding of mitochondrial Ca(2+) regulation, we developed a thermodynamically balanced model of mitochondrial Ca(2+) handling and bioenergetics by integrating kinetic models of mitochondrial Ca(2+) uniporter (CU), Na(+)-Ca(2+) exchanger (NCE), and Na(+)-H(+) exchanger (NHE) into an existing computational model of mitochondrial oxidative phosphorylation. Kinetic flux expressions for the CU, NCE and NHE were devel

Scorpion polypeptide BmK IT2 alleviates epileptic seizures and neuronal pyroptosis by regulating sodium channel Nav1.6.

ETHNOPHARMACOLOGICAL RELEVANCE: Epilepsy is the fourth most common neurological disorder, and nearly 30 % of patients develop seizures that are refractory to conventional anticonvulsant drugs. The etiology of epilepsy is diverse and includes a variety of genetic channelopathies that result in neuronal hyperexcitability, seizure generation, maladaptive circuit plasticity, intracellular calcium dysregulation, and neurodegeneration. For instance, mutations in SCN8A encoding the voltage-gated sodium channel Nav1.6 are known to disrupt calcium homeostasis in neurons, induce neuronal death, and caus

Cronkhite-Canada Syndrome- The first case from Pakistan: a case report.

Hyperkalemia is a common electrolyte disorder in patients with chronic kidney disease (CKD) that increases in prevalence with the decline of glomerular fltration rate (GFR). Another risk of hyperkalemia is the use of renin-angiotensin-aldosterone system inhibitors (RAASi) and/or mineralocorticoid receptor antagonists (MRAs) in managing CKD and proteinuria. The treatment of chronic hyperkalemia is challenging especially for outpatients. Treatment options for hyperkalemia include the potassium exchange resins of which two new potassium binders, Patiromer Sorbitex Calcium, and Sodium Zirconium Cy

Amiloride and its analogs as tools in the study of ion transport.

Amiloride inhibits most plasma membrane Na+ transport systems. We have reviewed the pharmacology of inhibition of these transporters by amiloride and its analogs. Thorough studies of the Na+ channel, the Na+/H+ exchanger, and the Na+/Ca2+ exchanger, clearly show that appropriate modification of the structure of amiloride will generate analogs with increased affinity and specificity for a particular transport system. Introduction of hydrophobic substituents on the terminal nitrogen of the guanidino moiety enhances activity against the Na+ channel; whereas addition of hydrophobic (or hydrophilic

A mechanistic investigation of the effects of lactic acidosis on myocardial contractility in the Neotropical fast-swimming freshwater fish Brycon amazonicus.

Lactic acidosis commonly impairs myocardial contractility in vertebrates, limiting cardiac performance under metabolic stress. However, the rheophilic Neotropical species matrinxã (Brycon amazonicus) appears to exhibit physiological adaptations that support cardiac resilience. In this study, ventricular muscle strips were exposed to extracellular lactic acidosis (LA, 22 mM lactic acid, pH 6.9) and evaluated for twitch contractility and the roles of calcium handling and pH regulation. Isometric ventricular preparations were utilized to assess: (a) the effects of LA; (b) the combined effects of

Endogenous ouabain in renal Na(+) handling and related diseases.

The Na(+) pump and its Endogenous modulator Ouabain (EO) can be considered as an ancestral enzymatic system, conserved among species ranging from Drosophila to humans, related to Na handling. In this review, we examine how EO is linked with vascular function in hypertension and if it impacts the pathogenesis of heart and renal failure. Moreover, the molecular mechanism of endogenous ouabain-linked hypertension involves the sodium pump/sodium-calcium exchanger duet. Biosynthesis of EO occurs in adrenal glands and is under the control of angiotensin II, ACTH and epinephrine. Elevated concentrati

Mechanisms of disease: what can mouse models tell us about the molecular processes underlying Dent disease?

Two knockout mouse models of Dent disease are similar with regard to the characteristics of Fanconi syndrome, but differ markedly with respect to vitamin D and renal calcium handling. One model exhibits hypercalciuria, renal calcifications and renal failure; the other does not. Data from such experimental models have greatly advanced our understanding of the molecular mechanisms underlying Dent disease. This Review summarizes some of the important phenotypic characteristics shared by mouse models and people with Dent disease. Experimental data are used to predict the molecular mechanisms under
