Static Magnetic Field Stimulation over Parietal Cortex Enhances Somatosensory Detection in Humans.

The role of neuronal oscillations in human somatosensory perception is currently unclear. To address this, here we use noninvasive brain stimulation to artificially modulate cortical network dynamics in the context of neurophysiological and behavioral recordings. We demonstrate that transcranial static magnetic field stimulation (tSMS) over the somatosensory parietal cortex increases oscillatory power specifically in the alpha range, without significantly affecting bottom-up thalamocortical inputs indexed by the early cortical component of somatosensory evoked potentials. Critically, we next s

Monosynaptic inputs to ErbB4-expressing inhibitory neurons in mouse primary somatosensory cortex.

Previous reports have described inputs to the somatosensory cortex (S1) in mouse or rat using retrograde or anterograde tracers. Such studies do not, however, reveal which particular cell types within the S1 cortex receive direct monosynaptic connections from these input sources. Here we describe the monosynaptic inputs to a subpopulation of mouse S1 inhibitory neurons that express ErbB4. We used a previously described "bridge protein," composed of the ErbB4 ligand, neuregulin (NRG1), fused to the avian viral receptor TVB (TVB-NRG1), along with EnvB pseudotyped lentivirus (LV) and rabies virus

Functional reorganization of adult raccoon somatosensory cerebral cortex following neonatal digit amputation.

Amputation of a forepaw digit in raccoons 2-8 weeks of age produced dramatic changes in the functional organization of somatosensory cortex examined electrophysiologically 9-12 months later. The cortical region normally representing the digit that was amputated received widely overlapping input from the entire forepaw, with local disruption of somatotopic organization. Compared with normal animals, the receptive fields of cortical neurons in amputated animals were larger, often included both glabrous and hairy skin, sometimes involved discontinuous skin regions, and were much more variable in

Experience-driven plasticity in the secondary somatosensory cortex in people born without hands.

In congenital handlessness, where individuals rely on their feet for compensatory manual functions, reorganization in the primary somatosensory cortex (SI) appears to be limited to local shifts by adjacent body parts. It remains unclear how congenital handlessness and lifelong compensatory foot use shape the sensorimotor system beyond SI, and whether compensatory abilities can drive broader functional reorganization. Here, we used task-based and resting-state functional MRI (fMRI) to investigate how compensatory foot use changes body representations in the secondary somatosensory cortex (SII)

Cerebellar climbing fibers impact experience-dependent plasticity in the mouse primary somatosensory cortex.

In the cerebellum, climbing fibers (CFs) provide instructive signals for supervised learning at parallel fiber to Purkinje cell synapses. It has not been tested so far whether CF signaling may also influence plasticity in other brain areas. Here, we show that optogenetic CF activation suppresses potentiation of whisker responses in layer 2/3 pyramidal cells in the primary somatosensory (S1) cortex of awake mice that is observed after repeated whisker stimulation. Using two-photon imaging and chemogenetics, we find that CFs control plasticity by modulating SST- and VIP-positive interneurons in

Conveying tactile object characteristics through customized intracortical microstimulation of the human somatosensory cortex.

Microstimulation of the somatosensory cortex can evoke tactile percepts in people with spinal cord injury, providing a means to restore touch. While location and intensity can be reliably conveyed, two issues that prevent creating more complex naturalistic sensations are a lack of methods to effectively scan the large stimulus parameter space and difficulties with assessing percept quality. Here, we address both challenges with an experimental paradigm that enables three male individuals with tetraplegia to control their stimulation parameters in a blinded fashion to create sensations for diff

Neural Correlation between Evoked Tactile Sensation and Central Activities in the Somatosensory Cortex.

Evoked tactile sensation (ETS) is induced with stimulation of areas in the projected finger map (PFM) in the stump skin of forearm amputees. The aim of the present study is to further explore the neural correlation of the ETS with central activities in the somatosensory cortex (SI). Two forearm amputees were recruited to participate in this study. The neuroimaging technique of Magnetoencephalography (MEG) was employed to reveal the activities in the somatosensory cortex (SI), while the thumb and little finger areas in the PFM and the thumb and little finger of the contralateral hand were stimu

Transcranial magnetic stimulation over contralateral primary somatosensory cortex disrupts perception of itch intensity.

Itch, a complex unpleasant sensation causing the desire to scratch, results from the activity of a network of brain regions. However, the specific functional contributions of individual regions within this network remain poorly understood. We investigated whether contralateral primary and secondary somatosensory cortices (S1, S2) and ipsilateral inferior frontal gyrus (IFG) are critically involved in the cortical processing of acute itch. Continuous theta burst transcranial magnetic stimulation (cTBS) was applied to either S1, S2 or IFG, followed by itch induction using a histamine prick. Resu

Expectation violation and attention to pain jointly modulate neural gain in somatosensory cortex.

The neural processing and experience of pain are influenced by both expectations and attention. For example, the amplitude of event-related pain responses is enhanced by both novel and unexpected pain, and by moving the focus of attention towards a painful stimulus. Under predictive coding, this congruence can be explained by appeal to a precision-weighting mechanism, which mediates bottom-up and top-down attentional processes by modulating the influence of feedforward and feedback signals throughout the cortical hierarchy. The influence of expectation and attention on pain processing can be m

Personalized, bilateral whole-body somatosensory cortex stimulation to relieve fatigue in multiple sclerosis.

BACKGROUND: The patients suffering from multiple sclerosis (MS) often consider fatigue the most debilitating symptom they experience, but conventional medicine currently offers poorly efficacious therapies. OBJECTIVE: We executed a replication study of an innovative approach for relieving MS fatigue. METHODS: According to the sample size estimate, we recruited 10 fatigued MS patients who received 5-day transcranial direct current stimulation (tDCS) in a randomized, double-blind, Sham-controlled, crossover study, with modified Fatigue Impact Scale (mFIS) score reduction at the end of the trea

Parallel pathways from motor and somatosensory cortex for controlling whisker movements in mice.

Mice can gather tactile sensory information by actively moving their whiskers to palpate objects in their immediate surroundings. Whisker sensory perception therefore requires integration of sensory and motor information, which occurs prominently in the neocortex. The signalling pathways from the neocortex for controlling whisker movements are currently poorly understood in mice. Here, we delineate two pathways, one originating from primary whisker somatosensory cortex (wS1) and the other from whisker motor cortex (wM1), that control qualitatively distinct movements of contralateral whiskers.

Altered somatosensory barrel cortex refinement in the developing brain of Mecp2-null mice.

Rett syndrome (RTT) is a neurodevelopmental disorder caused by mutations in the methyl-CpG binding protein 2 (MeCP2) gene. In previous studies, monoaminergic dysfunctions have been detected in patients with RTT and in a murine model of RTT, the Mecp2-null mouse. Therefore, the pathogenesis of RTT is thought to involve impairments in the monoaminergic systems. However, there have been limited data showing that the impairment of monoamines leads to early symptoms during development. We used histochemistry to study the somatosensory barrel cortex in the B6.129P2(C)-Mecp2(tm1.1Bird) mouse model of

Amplitude and timing of somatosensory cortex activity in task-specific focal hand dystonia.

OBJECTIVE: Task-specific focal hand dystonia (tspFHD) is a movement disorder diagnosed in individuals performing repetitive hand behaviors. The extent to which processing anomalies in primary sensory cortex extend to other regions or across the two hemispheres is presently unclear. METHODS: In response to low/high rate and novel tactile stimuli on the affected and unaffected hands, magnetoencephalography (MEG) was used to elaborate activity timing and amplitude in the primary somatosensory (S1) and secondary somatosensory/parietal ventral (S2/PV) cortices. MEG and clinical performance measure

Temporal dynamics of plastic changes in human primary somatosensory cortex after finger webbing.

The primary somatosensory cortex (SI) exhibits a detailed topographic organization of the hand and fingers, which has been found to undergo plastic changes following modifications of the sensory input. Although the spatial properties of these changes have been extensively investigated, little is known about their temporal dynamics. In this study, we adapted the paradigm of finger webbing, in which 4 fingers are temporarily webbed together, hence modifying their sensory feedback. We used magnetoencephalography, to measure changes in the hand representation in SI, before, during, and after finge

Ventrolateral and dorsomedial somatosensory association cortex damage produces distinct somesthetic syndromes in humans.

Five somatosensory cortices have distinctive somatotopic representations, cytoarchitecture, and connectivity: primary somatosensory cortex (SI), ventrolateral association cortices (SII, SIII, and SIV), and dorsomedial association cortex (supplementary sensory area). Patients with focal lesions of ventrolateral (n = 5) and dorsomedial (n = 6) somatosensory association cortices (SACs) and hemiparetic (n = 8) and neurologically normal control patients (n = 14) underwent detailed somesthetic testing that encompassed basic, intermediate, and complex (tactile object recognition) somesthetic function

Neurologic Outcome After Resection of Parietal Lobe Including Primary Somatosensory Cortex: Implications of Additional Resection of Posterior Parietal Cortex.

OBJECTIVE: Postoperative neurologic outcomes after primary somatosensory cortex (S1) resection have not been well documented. This study was designed to evaluate the neurologic deterioration that follows resection of the S1 areas and to assess the risk factors associated with these morbidities. METHODS: We reviewed 48 consecutive patients with medically intractable epilepsy who underwent resection of the S1 and/or the adjacent cortex. The 48 patients were categorized into 4 groups according to the resected area as seen on postoperative magnetic resonance images: group 1 (resection of S1 only;

Light-Modulated Circadian Synaptic Plasticity in the Somatosensory Cortex: Link to Locomotor Activity.

The circadian clock controls various physiological processes, including synaptic function and neuronal activity, affecting the functioning of the entire organism. Light is an important external factor regulating the day-night cycle. This study examined the effects of the circadian clock and light on synaptic plasticity, and explored how locomotor activity contributes to these processes. We analyzed synaptic protein expression and excitatory synapse density in the somatosensory cortex of mice from four groups exposed to different lighting conditions (LD 12:12, DD, LD 16:8, and LL). Hierarchical unimodal processing within the primary somatosensory cortex during a bimodal detection task.

Do sensory cortices process more than one sensory modality? To answer these questions, scientists have generated a wide variety of studies at distinct space-time scales in different animal models, and often shown contradictory conclusions. Some conclude that this process occurs in early sensory cortices, but others that this occurs in areas central to sensory cortices. Here, we sought to determine whether sensory neurons process and encode physical stimulus properties of different modalities (tactile and acoustic). For this, we designed a bimodal detection task where the senses of touch and he

Attenuation of sensory processing in the primary somatosensory cortex during rubber hand illusion.

The neural representation of the body is easily altered by the integration of multiple sensory signals in the brain. The "rubber hand illusion" (RHI) is one of the most popular experimental paradigms to investigate this phenomenon. During this illusion, a feeling of ownership of the rubber hand is created. Some studies have shown that somatosensory processing in the brain is attenuated when RHI occurs. However, it is unknown where attenuation of somatosensory processing occurs. Here, we show that somatosensory processing is attenuated in the primary somatosensory cortex. We found that the earl
