Bacopa monniera Attenuates Scopolamine-Induced Impairment of Spatial Memory in Mice.

Scopolamine, an anticholinergic, is an attractive amnesic agent for discerning the action of candidate antiamnesic drugs. Bacopa monniera Linn (Syn. Brahmi) is one such antiamnesic agent that is frequently used in the ancient Indian medical system. We have earlier reported the reversal of diazepam-induced amnesia with B. monniera. In this study we wanted to test if scopolamine-induced impairment of spatial memory can also be ameliorated by B. monniera using water maze mouse model. The objective of study was to study the effect of B. monniera on scopolamine-induced amnesia. We employed Morris w

Vestibular loss causes hippocampal atrophy and impaired spatial memory in humans.

The human hippocampal formation plays a crucial role in various aspects of memory processing. Most literature on the human hippocampus stresses its non-spatial memory functions, but older work in rodents and some other species emphasized the role of the hippocampus in spatial learning and memory as well. A few human studies also point to a direct relation between hippocampal size, navigation and spatial memory. Conversely, the importance of the vestibular system for navigation and spatial memory was until now convincingly demonstrated only in animals. Using magnetic resonance imaging volumetry

Vitamin A deficiency produces spatial learning and memory impairment in rats.

Vitamin A and its derivatives (retinoids) play important roles in many physiological processes. The recent finding of high levels of cellular retinol-binding protein type 1 immunoreactivity, cellular retinoic acid-binding protein type 1 immunoreactivity and the presence of nuclear retinoid receptors in the central nervous system of adult rodents suggests that retinoids may carry out important roles in the adult brain. In consideration of the role of the hippocampus in spatial learning and memory we evaluated the effect of vitamin A deprivation in adult rats on these functions. Following 12 wee

Orientation-dependent spatial memories for scenes viewed on mobile devices.

We examined whether spatial representations for scenes experienced on the screens of mobile devices are orientation dependent and whether the type of movement (physical vs. simulated) during learning affects the encoding and the retrieval of spatial information. Participants studied a spatial layout depicted on a tablet and then carried out perspective-taking trials in which they localized objects from imagined perspectives. Depending on condition, participants either rotated the tablet along with their body or remained stationary and swiped with their finger on the screen to change their view

Retrograde amnesia in rats with lesions to the hippocampus on a test of spatial memory.

The present study examined remote spatial memory in a test that spans several months to determine whether remote memories are spared relative to more recent ones, as predicted by models of memory consolidation. At 3, 6 or 12 months of age, groups of rats received forced-choice training as to the location of food reward in a cross maze. At 12.5 months, rats received bilateral neurotoxic lesions to the hippocampus or a control surgical procedure and 2 weeks later their memory for the spatial location was tested. Their performance was compared to that of rats with hippocampal or control lesions w

When is the perirhinal cortex necessary for the performance of spatial memory tasks?

The perirhinal cortex and hippocampus have close anatomical links and it has, therefore, been proposed that they have important, coordinated roles in memory. This review examines the relative role of these structures in spatial memory tasks that are known to be hippocampal-dependent. The published lesion data gives a mixed picture, as only some studies detect spatial deficits after perirhinal cortex lesions. The possible reasons for these inconsistencies are reviewed, along with electrophysiological data that indicate how perirhinal cortex lesions may alter neuronal activity in the hippocampus

Visual but not spatial working memory deficit in children with spina bifida.

Twenty children with spina bifida and twenty controls were assessed on a battery of visuospatial working memory tests. Children with spina bifida performed as well as the control group in the visuospatial test and in the forward and backward versions of the Corsi test, but were impaired in the 'House Visual Span.' This test, designed in our lab, provides specific information regarding visual ability to discriminate between pictorial targets and lures, and requires specific visual processing of stimuli. These results are discussed in terms of working memory models and are viewed as supporting t

Rightward Saccades Distort the Scaffold of Spatial Working Memory in ADHD.

Attention-deficit/hyperactivity disorder (ADHD) is typically characterized by executive dysfunction, yet emerging evidence points to spatially asymmetrical features of cognition. We examined whether transsaccadic working memory (tWM), the mechanism that integrates visual and motor signals across eye movements, shows directional vulnerability in ADHD. Participants with ADHD completed a spatial tWM task in which memory arrays (one or three items) were presented before leftward, rightward, or fixation trials, and repositioned a probe to the remembered target location. For comparison, data from a

Sleep benefits cross-dimensional binding of spatial context in episodic memory.

The active systems consolidation hypothesis predicts that sleep compared to wakefulness benefits context binding in episodic memory for retention intervals of up to 12 h. Extending previous research on sleep benefits in item-context associative memory, we investigated whether sleep also improves bound source memory for cross-dimensional context associations. In a source-monitoring experiment, we used intentionally learned pictures as items and incidentally learned horizontal and vertical screen positions as context dimensions to compare cross-dimensional binding in spatial source memory for re

Ketamine Alters Tuning of Neural and Behavioral Spatial Working Memory Precision.

BACKGROUND: Working memory (WM) is a key element of cognition that depends on accurate internal representation of stimuli and often impaired in psychiatric disorders, including schizophrenia. Prior human neuropharmacological studies with ketamine suggest that glutamate N-methyl-D-aspartate receptors (NMDARs) are critical for spatial WM (sWM), with NMDAR antagonism attenuating task-evoked neural activation and reducing WM accuracy. Animal studies, at the cellular level, and cortical microcircuit models hypothesize that NMDAR antagonism impairs sWM by decreasing the specificity of neuronal respo

Age-dependent vulnerability to spatial memory interference in APP/PS1 mice.

BACKGROUND: While Alzheimer's disease (AD) is well known for progressive memory impairment, less is understood about how amyloid pathology affects flexible updating of competing spatial representations. Here, we used the Objects in Updated Locations (OUL) paradigm to investigate how amyloidosis influences spatial memory updating and the handling of competing mnemonic information in APP/PS1 mice. METHODS: APP/PS1 and wild-type control mice were tested in the OUL paradigm, which probes memory for object displacement and subsequent updating of overlapping spatial representations. Aerobic exercise improves spatial memory in a rat model of meningitis.

Infections of the nervous system, such as acute bacterial meningitis, pose serious health problems that require immediate intervention. In experimental animals, exposure to lipopolysaccharide (LPS) is used to induce meningitis. Aside from drug intervention to reduce inflammation in meningitis, aerobic exercise helps to maintain the regulatory mechanisms of brain homeostasis through anti-inflammatory mechanisms. The aim of this study was to evaluate the effect of aerobic exercise on malondialdehyde (MDA), nuclear factor-kappa B (NF-κB), insulin-like growth factor 1 (IGF-1), endothelial nitric o

A comparison of behavioral paradigms assessing spatial memory in tree shrews.

Impairments in spatial navigation in humans can be preclinical signs of Alzheimer's disease. Therefore, cognitive tests that monitor deficits in spatial memory play a crucial role in evaluating animal models with early stage Alzheimer's disease. While Chinese tree shrews (Tupaia belangeri) possess many features suitable for Alzheimer's disease modeling, behavioral tests for assessing spatial cognition in this species are lacking. Here, we established reward-based paradigms using the radial-arm maze and cheeseboard maze for tree shrews, and tested spatial memory in a group of 12 adult males in

A proposed attention-based model for spatial memory formation and retrieval.

Animals use sensory information and memory to build internal representations of space. It has been shown that such representations extend beyond the geometry of an environment and also encode rich sensory experiences usually referred to as context. In mammals, contextual inputs from sensory cortices appear to be converging on the hippocampus as a key area for spatial representations and memory. How metric and external sensory inputs (e.g., visual context) are combined into a coherent and stable place representation is not fully understood. Here, I review the evidence of attentional effects alo

The olfactory bulb modulates entorhinal cortex oscillations during spatial working memory.

Cognitive functions such as working memory require integrated activity among different brain regions. Notably, entorhinal cortex (EC) activity is associated with the successful working memory task. Olfactory bulb (OB) oscillations are known as rhythms that modulate rhythmic activity in widespread brain regions during cognitive tasks. Since the OB is structurally connected to the EC, we hypothesized that OB could modulate EC activity during working memory performance. Herein, we explored OB-EC functional connectivity during spatial working memory performance by simultaneous recording local fiel

Noisy galvanic vestibular stimulation modulates spatial memory in young healthy adults.

Hippocampal and striatal circuits play important roles in spatial navigation. These regions integrate environmental information and receive intrinsic afferent inputs from the vestibular system. Past research indicates that galvanic vestibular stimulation (GVS) is a non-invasive technique that modulates hippocampal and striatal activities. There are also evidences for enhanced motor and cognitive functions through GVS. This study extends previous research to investigate whether noisy GVS may improve hippocampal- and striatal-associated aspects of spatial navigation performance. Using a virtual

Dissociable spatial memory systems revealed by typical and atypical human development.

Rodent lesion studies have revealed the existence of two causally dissociable spatial memory systems, localized to the hippocampus and striatum that are preferentially sensitive to environmental boundaries and landmark objects, respectively. Here we test whether these two memory systems are causally dissociable in humans by examining boundary- and landmark-based memory in typical and atypical development. Adults with Williams syndrome (WS)-a developmental disorder with known hippocampal abnormalities-and typical children and adults, performed a navigation task that involved learning locations

An intrinsic association between olfactory identification and spatial memory in humans.

It was recently proposed that olfaction evolved to aid navigation. Consistent with this hypothesis, olfactory identification and spatial memory are linked to overlapping brain areas which include the orbitofrontal cortex and hippocampus. However, the relationship between these two processes has never been specifically investigated. Here, we show that olfactory identification covaries with spatial memory in humans. We also found that the cortical thickness of the left medial orbitofrontal cortex, and the volume of the right hippocampus, predict both olfactory identification and spatial memory.

Human parietal cortex lesions impact the precision of spatial working memory.

The neural mechanisms that support working memory (WM) depend on persistent neural activity. Within topographically organized maps of space in dorsal parietal cortex, spatially selective neural activity persists during WM for location. However, to date, the necessity of these topographic subregions of human parietal cortex for WM remains unknown. To test the causal relationship of these areas to WM, we compared the performance of patients with lesions to topographically organized parietal cortex with those of controls on a memory-guided saccade (MGS) task as well as a visually guided saccade (
