Non-specific biodegradation of the organophosphorus pesticides, cadusafos and ethoprophos, by two bacterial isolates.

An enrichment culture technique was used for the isolation of microorganisms responsible for the enhanced biodegradation of the nematicide cadusafos in soils from a potato monoculture area in Northern Greece. Mineral salts medium supplemented with nitrogen (MSMN), where cadusafos (10 mg l(-1)) was the sole carbon source, and soil extract medium (SEM) were used for the isolation of cadusafos-degrading bacteria. Two pure bacterial cultures, named CadI and CadII, were isolated and subsequently characterized by sequencing of 16S rRNA genes. Isolate CadI showed 97.4% similarity to the 16S rRNA gene

Uncommon non-fermentative gram-negative superbugs - A surge in bacteraemia patients at an apex super speciality hospital.

BACKGROUND AND OBJECTIVES: Gram negative bacteria are most common cause of bacteraemia, with one fourth being attributed to non-fermentive Gram negative bacteria (NFGNB). Contribution of NFGNB to antimicrobial resistance burden is also high. Commonly isolated NFGNB Pseudomonas aeruginosa and Acinetobacter baumannii have been well studied however there is paucity of data of uncommon species including, Stenotrophomonas maltophilia, Burkholderia spp., Sphingomonas paucimobilis in our region. This study was thus designed to explore implication of antimicrobial resistance and associated determinant

Biochar-microbe-hydrogel with cyclic hydraulic leaching establishing niches for integrated soil-groundwater bioremediation.

Bioremediation of soil and groundwater contaminated by persistent organic pollutants (e.g., chlorinated organics) using functional bacteria faces inherent challenges, such as competition from indigenous microorganisms (exogenous bacteria is difficult to establish microbial niches in soil) and poor pollutant accessibility. This study developed an integrated strategy that combines Biochar-Microbe-Hydrogel (BMH) matrix with Cyclic Hydraulic Leaching (CHL) for the concurrent bioremediation of soil-groundwater. The BMH is active gel beads encapsulated functional bacteria in porous biochar with hydr

Monitoring the coral disease, plague type II, on coral reefs in St. John, U.S. Virgin Islands.

In July 1997, conspicuous white patches of necrotic tissue and bare skeleton began to appear on scleractinian corals in several bays around St. John, US Virgin Islands. Analysis of diseased coral tissue from five different species confirmed the presence of a Sphingomonas-like bacterium, the pathogen for plague type II. To date, 14 species of hard corals have been affected by plague type II around St. John. This disease was monitored at Haulover and Tektite Reefs at depths of 7-12 meters. The study site at Tektite Reef has > 50% cover by scleractinian corals with 90% of hard corals being compos

Antarctic moss fairy rings serve as reservoirs for plant growth-promoting bacteria.

BACKGROUND: Antarctic terrestrial vegetation and microbial communities are undergoing climate-driven changes, which in some cases have been linked to the emergence of plant diseases. Ring-shaped plant disease structures, known as fairy rings (FRs), have been observed in moss fields dominated by Sanionia uncinata. Previous studies suggest that FRs promote vegetation development through interactions with bacterial communities. This raises the possibility that the bacterial communities in Antarctic FRs may also have ecologically significant functions. To address this, we analyzed the bacterial co

Identification of bacteria from clinical samples using Bartonella alpha-Proteobacteria growth medium.

In an effort to overcome historical problems associated with the isolation of Bartonella species from animal and human blood samples, our laboratory developed a novel, chemically modified, insect-based, liquid culture medium (Bartonella alpha-Proteobacteria growth medium, BAPGM). In this study, we describe the isolation of non-Bartonella bacteria from aseptically obtained human blood and tissue samples that were inoculated into BAPGM pre-enrichment culture medium, and were obtained during attempts to define each individuals Bartonella infection status. After incubation for at least 7 days in l

Impact of inoculation protocols, salinity, and pH on the degradation of polycyclic aromatic hydrocarbons (PAHs) and survival of PAH-degrading bacteria introduced into soil.

Degradation of polycyclic aromatic hydrocarbons (PAHs) and survival of bacteria in soil was investigated by applying different inoculation protocols. The soil was inoculated with Sphingomonas paucimobilis BA 2 and strain BP 9, which are able to degrade anthracene and pyrene, respectively. CFU of soil bacteria and of the introduced bacteria were monitored in native and sterilized soil at different pHs. Introduction with mineral medium inhibited PAH degradation by the autochthonous microflora and by the strains tested. After introduction with water (without increase of the pore water salinity),

Comparison of MALDI TOF with conventional identification of clinically relevant bacteria.

BACKGROUND: A completely new approach to diagnose microbial agents at least one day earlier based on mass spectrometric analysis becomes possible in the microbiology laboratory: MALDI TOF: matrix-assisted laser desorption/ionisation time-of-flight. Comparison between results of the new procedure with those obtained by conventional testing is mandatory. METHODS: 204 clinical isolates grown on agar plates were analysed both, by the MALDI TOF Bruker microflex apparatus and by conventional identification using the VITEK II and API systems, both from bioMérieux. RESULTS: Of the identified isolate

Identification of unknown ocular pathogens in clinically suspected eye infections using ribosomal RNA gene sequence analysis.

DNA sequence-based identification of pathogens from ocular samples of patients with clinically suspected eye infections was accomplished using 16S and internal transcribed spacer (ITS) ribosomal RNA gene sequence analysis. PCR was positive for 24 of 99 samples tested. Both culture and 16S rDNA sequence analysis identified Pseudomonas aeruginosa, streptococci and Enterobacteriaceae. Isolates misidentified as Burkholderia cepacia by biochemical tests were identified as Ralstonia mannitolilytica by 16S rDNA sequence analysis. Sequence analysis identified the following microorganisms from 19 cultu

Chemotaxis-associated biotechnology for the bacterial production of biofuel using blue carbon alginate.

Brown algae are promising sources of blue carbon for biofuel production. The nonmotile Sphingomonas sp. strain A1 imports alginate, a major component of brown algae, by alginate-binding proteins AlgQ1/Q2 and an ATP-binding cassette transporter. Previously, we showed that metabolically engineered ethanol-fermentative strains A1 (A1-EPv14 and A1-EPv104) produced bioethanol from alginate and that repeated subculturing of strain A1 on soft agar plates inducibly activated motility, resulting in expression of chemotaxis toward alginate. This study examined the possibility of bioethanol production di

Loss of salivary agglutinin induces changes in the salivary microbiome and accelerates development of oral cancer.

BACKGROUND: Salivary agglutinin, also known as deleted in malignant brain tumors 1 (DMBT1), is an anti-microbial protein. DMBT1 is low in saliva from patients with oral squamous cell carcinoma (OSCC) and dramatically increases after treatment, with accompanying microbial changes. While this suggests an association between DMBT1 suppression and changes in the oral microbiota, causation has not been established. DMBT1 is also a tumor suppressor protein; its loss promotes OSCC progression, but its role in OSCC development is unknown. In this study, OSCC development was investigated in a murine ca

Effects of the polyhalogenated carbazoles 3-bromocarbazole and 1,3,6,8-tetrabromocarbazole on soil microbial communities.

Soil ecosystems are being more contaminated with polyhalogenated carbazoles (PHCZs), which raising much attention about their impact on soil microorganisms. 3-Bromocarbazole (3-BCZ) and 1,3,6,8-tetrabromocarbazole (1,3,6,8-TBCZ) are two typical PHCZs with high detection rates in the soil environment. However, ecological risk research on these two PHCZs in soil is still lacking. In the present study, after 80 days of exposure, the ecological influence of 3-BCZ and 1,3,6,8-TBCZ was investigated based on 16S rDNA sequencing, ITS sequencing, gene (16S rDNA, ITS, amoA, nifH, narG and cbbL) abundanc

Bacterial diversity and community along the succession of biological soil crusts in the Gurbantunggut Desert, Northern China.

Biological soil crusts (BSCs) are common and play critical roles in semi-arid and arid ecosystems. Bacteria, as an important community in BSCs, play critical roles in biochemical processes. However, how bacterial diversity and community change in different successional stages of BSCs is still unknown. We used 454 pyrosequencing of 16S rRNA to investigate the bacterial composition and community, and the relationships between bacterial composition and environmental factors were also explored. In different successional stages of BSCs, the number of bacteria operational taxonomic units (OTUs) dete

Foliar Application of Bacillus thuringiensis Regulates Soil Physicochemical Properties and Microbial Ecological Functions in Tea Plantations.

Bacillus thuringiensis (Bt) is widely employed as a biological control agent against pests in tea plantations, yet its impacts on soil health and microbial ecology remain insufficiently understood. This study investigated the effects of two Bt application regimes, namely moderate-frequency conventional Bt application (Bt1, 3 sprays over 21 days) and high-frequency intensive Bt application (Bt2, 6 sprays over 42 days), on soil physicochemical properties, enzyme activities, and microbial community structure and function in tea soils. The moderate-frequency conventional Bt1 significantly improved
