Purpura fulminans due to Staphylococcus aureus.

BACKGROUND: Purpura fulminans is an acute illness commonly associated with meningococcemia or invasive streptococcal disease, and it is typically characterized by disseminated intravascular coagulation (DIC) and purpuric skin lesions. In this article, we report the first 5 cases (to our knowledge) of purpura fulminans directly associated with Staphylococcus aureus strains that produce high levels of the superantigens toxic shock syndrome toxin-1 (TSST-1), staphylococcal enterotoxin serotype B (SEB), or staphylococcal enterotoxin serotype C (SEC). METHODS: Cases were identified in the Minneapo

[Devastating community-acquired Staphylococcus aureus bacteremia].

Three patients, two men aged 52 and 62 years and a woman aged 83 years, experienced non-specific flu-like symptoms for a few days but subsequently developed a complicated disease with multiple metastatic infections and endocarditis due to bacteraemia. Only the man aged 62 years had a port of entry, a finger wound. Two patients died. Community-acquired S. aureus bacteraemia is a much more serious disease than nosocomial bacteraemia. The community-acquired bacteraemia is characterised by the absence of a primary focus and non-specific flu-like symptoms in the beginning. This results in a long-du

Inhibitory Effect of Staphylococcus Epidermidis Fermentation Broth on Staphylococcus Aureus.

In this study, Staphylococcus epidermidis (S. epidermidis) fermentation broth with an inhibitory effect on Staphylococcus aureus (S. aureus) was produced by optimizing cultivation conditions. S. epidermidis fermentation broth cultivated in NA medium (SFB) does not inhibit the growth of S. aureus, whereas Gly-SFB or Glu-SFB, prepared by incorporating glycerol or glucose into NA medium, significantly inhibited S. aureus growth. GC‒MS and HPLC revealed several fatty acids in Gly-SFB and Glu-SFB, which inhibited the growth of S. aureus. LC‒MS/MS combined with untargeted metabolomics was used to ex

Effects of isosorbide mononitrate loaded nanoparticles conjugated with anti-Staphylococcus aureus α-toxin on Staphylococcus aureus biofilms.

Staphylococcus aureus (S. aureus) is associated with recalcitrant chronic infection, especially in chronic rhinosinusitis (CRS). S. aureus infection and biofilms cause poorer postsurgical outcomes. We developed isosorbide mononitrate (ISMN) loaded nanoparticles conjugated with an anti-Staphylococcus aureus alpha-toxin (anti-S. aureus α-toxin) antibody that could target biofilms and investigated their anti-biofilm effect. Anti-S. aureus α-toxin antibody coupled immunoliposomes were generated. The effect of ISMN immunoliposomes on S. aureus biofilm formation and their anti-biofilm efficacy were

Prevalence and Characterization of Staphylococcus aureus and Methicillin-Resistant Staphylococcus aureus Isolated from Guangxi Dairy Farms.

Staphylococcus aureus (S. aureus) is a major pathogen responsible for mastitis in dairy cows and can contaminate raw milk, thereby posing significant health risks to consumers. The emergence of methicillin-resistant S. aureus (MRSA) has further heightened public health concerns due to its antibiotic resistance and infectious potential. In this study, we examined the prevalence, virulence genes, antimicrobial resistance, spa types, and biofilm formation of S. aureus isolates from dairy farms in Guangxi Province, China. Among 242 randomly selected samples, 37 S. aureus strains were identified (1

Rapid Detection of Staphylococcus aureus and Methicillin-Resistant Staphylococcus aureus by Multiplex Real-Time Fluorescent PCR.

Staphylococcus aureus (S.aureus) is a common Gram-positive human pathogen that causes community-acquired and nosocomial infections, with diverse clinical manifestations ranging from local superficial lesions and food poisoning to fatal systemic infections. The discovery of antibiotics significantly reduced the mortality rate, but the problem of drug resistance has since become increasingly prominent. Since the first identification of MRSA in 1960, this strain has emerged as a global public health threat. MRSA is a major pathogen of nosocomial infections, capable of causing various severe disea

Staphylococcus aureus host interactions and adaptation.

Invasive Staphylococcus aureus infections are common, causing high mortality, compounded by the propensity of the bacterium to develop drug resistance. S. aureus is an excellent case study of the potential for a bacterium to be commensal, colonizing, latent or disease-causing; these states defined by the interplay between S. aureus and host. This interplay is multidimensional and evolving, exemplified by the spread of S. aureus between humans and other animal reservoirs and the lack of success in vaccine development. In this Review, we examine recent advances in understanding the S. aureus-hos

Staphylococcus aureus bacteriuria: implications and management.

BACKGROUND: Staphylococcus aureus is isolated in around 0.2%-4% of positive urinary cultures, more commonly in the contexts of long-term care, urological abnormalities and procedures, male sex, older age and comorbidities. Isolation may represent contamination, colonization, urinary tract infection or bacteraemic seeding from another site, and may be linked to S. aureus bacteraemia. However, there is little guidance on investigation and management of S. aureus bacteriuria. We performed a retrospective analysis of cases in our service, including clinical characteristics, investigations and trea

Methicillin-resistant Staphylococcus aureus in Nepal.

Staphylococcus aureus is both a frequent commensal and a leading cause of endocarditis, bacteremia, osteomyelitis and skin and soft tissue infections and device-related infections. We performed this minireview to summarize the prevalence of Staphylococcus aureus among clinical samples and estimate the proportion of methicillin-resistant Staphylococcus aureus. The prevalence of Staphylococcus aureus among clinical isolates in Nepal is 34.5%. On average, the proportion of multi-drug resistance in Staphylococcus aureus is 57.1%. Methicillin-resistant Staphylococcus aureus accounts for a total of

[Endogenous endophthalmitis following staphylococcus aureus meningitis].

Endogenous endophthalmitis is a rare complication of acute meningitis. A healthy 82-year-old woman suddenly developed septic bacteremia and meningitis owing to Staphylococcus aureus (methicillin-susceptible Staphylococcus aureus); she presented with a high fever and drowsiness. Improvement of these initial symptoms unveiled impaired bilateral visual acuity. Detailed ophthalmological assessments diagnosed it as probable endogenous endophthalmitis, which was immediately treated with vitrectomy. Subsequently, the patient totally recovered without any neurologic or ophthalmic sequela.

Crystal Structure of Staphylococcus aureus Cas9.

The RNA-guided DNA endonuclease Cas9 cleaves double-stranded DNA targets with a protospacer adjacent motif (PAM) and complementarity to the guide RNA. Recently, we harnessed Staphylococcus aureus Cas9 (SaCas9), which is significantly smaller than Streptococcus pyogenes Cas9 (SpCas9), to facilitate efficient in vivo genome editing. Here, we report the crystal structures of SaCas9 in complex with a single guide RNA (sgRNA) and its double-stranded DNA targets, containing the 5'-TTGAAT-3' PAM and the 5'-TTGGGT-3' PAM, at 2.6 and 2.7 Å resolutions, respectively. The structures revealed the mechanis

Staphylococcus aureus sinus infections in children.

OBJECTIVE: Staphylococcus aureus can cause sinusitis in children. The predominant MRSA clone in the United States, USA300, has been associated with skin and soft tissue as well as invasive diseases. USA300 has increased among CA methicillin-susceptible S. aureus (CA-MSSA) isolates. We describe the clinical characteristics of pediatric patients with S. aureus cultured from sinus specimens, treated at Texas Children's Hospital (TCH), and characterized their isolates by molecular methods. METHODS: This was a retrospective study of children with endoscopic sinus surgery (ESS) cultures positive fo

Food poisoning and Staphylococcus aureus enterotoxins.

Staphylococcus aureus produces a wide variety of toxins including staphylococcal enterotoxins (SEs; SEA to SEE, SEG to SEI, SER to SET) with demonstrated emetic activity, and staphylococcal-like (SEl) proteins, which are not emetic in a primate model (SElL and SElQ) or have yet to be tested (SElJ, SElK, SElM to SElP, SElU, SElU2 and SElV). SEs and SEls have been traditionally subdivided into classical (SEA to SEE) and new (SEG to SElU2) types. All possess superantigenic activity and are encoded by accessory genetic elements, including plasmids, prophages, pathogenicity islands, vSa genomic isl

Community-associated methicillin-resistant Staphylococcus aureus.

Community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) has emerged over the last decade across the United States and the world, becoming a major pathogen in many types of community-acquired infections. Although most commonly associated with minor skin and soft tissue infections, such as furuncles, CA-MRSA also can cause necrotizing fasciitis, pyomyositis, osteoarticular infections, and community-acquired pneumonia. This article discusses the epidemiology, diagnosis, and management of these infections from the perspective of the emergency physician.

Nisin-bacteriophage crossresistance in Staphylococcus aureus.

The combined effect of nisin and two lytic phages against Staphylococcus aureus was assessed. In short-time challenge experiments performed in pasteurized milk, a synergistic effect was observed. However, the development of nisin-adapted cells seriously compromised bacteriophage activity. A nisin-adapted strain became partially resistant to both phages. Efficiency of plaquing as well as adsorption values differed. Changes on the bacterial surface, often linked to nisin resistance, could account for the phenotypes observed, most likely by interfering with binding/recognition of phage receptors.
