TOR1AIP2 as a candidate gene for dystonia-hemichorea/hemiballism.

Dystonia is a movement disorder characterized by genetic and clinical heterogeneity. A recurring p.(Glu303del)-deletion in TOR1A is a well-established cause for DYT-TOR1A (DYT1), an autosomal dominant early-onset isolated dystonia. TOR1A encodes TorsinA, an AAA + ATPase located in the nuclear envelope. By whole exome analyses of a family with a novel dystonia-hemichorea-/hemiballism phenotype, we identified a TOR1AIP2 NM_001199260.2 c.1234A > G p.(Arg412Gly) variant. The variant is very rare in databases and was absent from whole exome data from >1000 dystonia patients. TOR1AIP2 encodes LULL1,

Abnormal movements in Rett syndrome are present before the regression period: a case study.

The suspicion of a diagnosis of Rett syndrome (RTT) is based on clinical criteria that are often not present in the first two stages of the disease, as many of its symptoms will appear at a later age. This sometimes postpones the genetic diagnosis and an early clinical intervention. We present the case of 19-months-old girl who came to the consultation because of an arrest of psychomotor development noticed 5 months earlier without change in sleep pattern, behavior, or social communication. In the observation of 1 hour videotape, she presented subtle stereotypic movements of the face and hands

Atypical head banging developed in teens persisting into adulthood as sleep-related rhythmic movement disorder.

UNLABELLED: Head banging is one subtype of sleep-related rhythmic movement disorder, characterized by stereotyped, repetitive rhythmic movements, such as lifting the head or entire upper body, banging the frontal area hard on the pillow, or slamming the occiput against a headboard. An atypical form of head banging with punching or slapping the head with a hand is extremely rare, with only 4 such cases reported so far. Herein, we present a young adult male who, since his early teens, has had atypical head banging, also called head-slapping, and discuss the neuropsychological and polysomnographi

Paraneoplastic autoimmune movement disorders.

PURPOSE OF REVIEW: To provide an overview of paraneoplastic autoimmune disorders presenting with various movement disorders. RECENT FINDINGS: The spectrum of paraneoplastic autoimmune disorders has been expanding with the discovery of new antibodies against cell surface and intracellular antigens. Many of these paraneoplastic autoimmune disorders manifest as a form of movement disorder. With the discovery of new neuronal antibodies, an increasing number of idiopathic or neurodegenerative movement disorders are now being reclassified as immune-mediated movement disorders. These include anti-N-

Rethinking Stereotypies in Autism.

Stereotyped movements ("stereotypies") are semi-voluntary repetitive movements that are a prominent clinical feature of autism spectrum disorder. They are described in first-person accounts by people with autism as relaxing and that they help focus the mind and cope in overwhelming sensory environments. Therefore, we generally recommend against techniques that aim to suppress stereotypies in individuals with autism. Further, we hypothesize that understanding the neurobiology of stereotypies could guide development of treatments to produce the benefits of stereotypies without the need to genera

An analogue assessment of hand stereotypies in two cases of Rett syndrome.

Stereotype hand movements are one behavioural manifestation of Rett syndrome, a neurodegenerative disorder resulting in severe mental retardation. They are neuropathological in origin, and constitute a barrier to the acquisition and exhibition of more developmentally appropriate behaviours, as well as posing a health risk for some individuals. The present study used an analogue assessment procedure, in which experimental conditions were constructed varying in the consequences delivered contingent upon the exhibition behaviour, to examine environmental correlates to the moment-to-moment express

Sex-Based Differences in Autism Symptoms in a Large, Clinically-Referred Sample of Preschool-Aged Children with ASD.

Previous research has identified possible sex-based differences in restricted and repetitive behaviors in autism spectrum disorder (ASD). However, this finding is mixed, particularly among preschool-aged children. We investigated the presence of sex-based differences in parent-rated ASD symptomatology, using the Autism Spectrum Rating Scale (ASRS). Participants consisted of a large (n = 481,100 female), clinically-referred sample of preschoolers (ages 2-5) diagnosed with ASD (NVIQ: M = 67.11, SD = 21.79). Females had less severe symptoms on the Total, Unusual Behaviors, DSM-5, and Stereotypy s

Amphetamine stereotypy, the basal ganglia, and the "selection problem".

Amphetamine and other stimulant drugs induce stereotyped head movements in rats, which interfere with normal goal-directed behavior. However, rats given access to food while intoxicated learn to suppress these movements in order to feed. This suggests that the suppression of stereotypy is an instrumentally learned response reinforced by the ingestion of food. Consistent with this interpretation, rats learn to suppress stereotyped head movements when intraoral infusions of milk are made contingent on maintaining a stationary head position, but not when such infusions are given noncontingently.

Psychiatric aspects of abnormal movement disorders.

It has been postulated that some movement disorders are secondary to unresolved, unconscious mental conflict; however, psychotherapeutic intervention has been unsuccessful and psychoanalytic formulations have not been shown to be valid. In addition, there is the interesting observation that some medications, stereotactic surgery and biofeedback have been successful in treating movement disorders. Moreover, as in the cases of amphetamine-induced stereotyped behavior, Parkinsonism, the acute dyskinesias, and Tardive Dyskinesia, there is evidence that some involuntary disorders of movement are bi

Dystonia secondary to electrical injury: surface electromyographic evaluation and implications for the organicity of the condition.

We describe a patient who developed right arm dystonia following an electrical injury. The patient's arm remained adducted, and flexed at the elbow and wrist, with all movement resulting in pain and tremor. Surface electromyographic evaluation revealed constant tonic activity of multiple upper and lower arm muscles at rest, that was not distractible. Voluntary and passive movement of the elbow or wrist resulted in high amplitude EMG activity, with motor grouping at 11 Hz at the elbow and 8 Hz at the wrist. Although a diagnosis of psychogenic dystonia was entertained, the stereotyped nature of

Evidence-based behavioral interventions for repetitive behaviors in autism.

Restricted and repetitive behaviors (RRBs) are a core symptom of autism spectrum disorders (ASD). There has been an increased research emphasis on repetitive behaviors; however, this research primarily has focused on phenomenology and mechanisms. Thus, the knowledge base on interventions is lagging behind other areas of research. The literature suggests there are evidence-based practices to treat "lower order" RRBs in ASD (e.g., stereotypies); yet, there is a lack of a focused program of intervention research for "higher order" behaviors (e.g., insistence on sameness). This paper will (a) disc

Cluster headache associated with a sixth nerve palsy: a case report.

BACKGROUND: Cluster headache is a rare primary headache disorder characterized by recurrent, stereotyped short-lasting attacks of severe, unilateral head pain accompanied by autonomic symptoms. METHODS/RESULTS: Ophthalmic features such as conjunctival injection, lacrimation, ptosis and miosis occur in the vast majority of patients with cluster headache, whereas co-existent ocular movement disorders are rare. CONCLUSIONS: To the best of our knowledge, only two documented cases of cluster headache with external ocular movement disorders have been reported. We describe herein an additional case

The Effectiveness of the Snug Vest on Stereotypic Behaviors in Children Diagnosed With an Autism Spectrum Disorder.

Various reviews of the effects of sensory integration therapy (SIT) have concluded that such interventions fail to reduce stereotypy. However, a new, and as yet untested, SIT iteration, an inflatable wearable vest known as the Snug Vest purports to decrease such repetitive behavior. In the current study, three children who emitted different forms of stereotypy participated in an alternating treatments design in which each participant wore a fully inflated vest and either a fully deflated vest or no vest. The results of the study show that the Snug Vest failed to reduce any participants' stereo

Recent advances in Tourette syndrome research.

Tourette syndrome (TS) is a developmentally regulated neurobehavioral disorder characterized by involuntary, stereotyped, repetitive movements. Recent anatomical and neuroimaging studies have provided evidence for abnormal basal ganglia and dopaminergic function in TS. Basic research on striatal inhibitory mechanisms and dopaminergic function complements the recent neuroimaging and anatomical data. Parallel studies of basal ganglia participation in the normal performance and learning of stereotyped repetitive behaviors or habits has provided additional insight. These lines of research have pro

Different effects of adenosine A1 agonist ribavirin on amphetamine-induced total locomotor and stereotypic activities in rats.

Pretreatment with a single dose of ribavirin (10, 20, and 30 mg/kg b.w., i.p.) decreased amphetamine (1.5 mg/kg b.w., i.p.)-induced total locomotor activity (distance traveled) compared to amphetamine alone, but the most significant effect was observed with 30 mg/kg dose. In contrast, total stereotypic activities (such as sniffing, self-grooming, licking, and head weaving) of these animals were unchanged. These findings demonstrate the ability of ribavirin to modulate in a different way central regulation of locomotor and stereotypic activities that in the future could have a practical applica

Behavioural disorders induced by external globus pallidus dysfunction in primates II. Anatomical study.

The anatomical organization of the basal ganglia supports their involvement in movement and behavioural disorders. Thus dyskinesia, attention deficit with or without hyperactivity, and stereotyped behaviour can be induced by microinjections of bicuculline, a GABAergic antagonist, into different parts of the external globus pallidus (GPe) in monkeys. The aim of the present study was to determine the anatomo-functional circuits inside the basal ganglia which are specifically related to each of these behavioural changes. For that, axonal tracers were injected in the same pallidal sites where abno

Crack dancing in the United Kingdom: apropos a video case presentation.

We report an adult patient presenting with choreiform movements 4 days after a large intravenous dose of cocaine. These movements were transitory and they normalized a week after admission. We believe this to be the first video case of acute chorea secondary to cocaine--a phenomenon popularly known as "crack dancing. " Cocaine abuse is associated with a wide range of movement disorders, including dystonia and exacerbation of Tourette's syndrome, multifocal tics, opsoclonus-myoclonus, choreiform movements, and stereotyped behavior known as "punding." Transient choreiform movements with a typica
