A novel DNA biosensor based on enzyme-enhanced fluorescence detection on etched optical fibers was developed. The hybridization complex of DNA probe and biotinylated target was formed on the etched optical fiber, and was then bound with streptavidin labeled horseradish peroxidase (streptavidin-HRP). The target DNA was quantified through the fluorescent detection of bi-p,p'-4-hydroxyphenylacetic acid (DBDA) generated from the substrate 4-hydroxyphenylacetic acid (p-HPA) under the catalysis of HRP, with a detection limit of 1 pM and a linear range from 1.69 pM to 169 pM. It is facile to regenera

A novel organoarsenic probe, desthiobiotin-phenylarsenic oxide (DesBiotin-PAO(III)), was synthesized by coupling desthiobiotin with p-aminophenylarsenic acid (PAO(III)) via a mixed anhydride method. This probe enables the in situ capture of intracellular arsenic binding proteins, which are subsequently adsorbed onto streptavidin-coated magnetic beads (SA-MBs) through the desthiobiotin-streptavidin interaction, thereby achieving selective separation of arsenic-binding proteins from other intracellular components. Because the binding affinity of desthiobiotin for streptavidin is considerably wea

BACKGROUND: High-efficiency and highly reliable analysis of microRNAs (miRNAs) in bodily fluids highlights its significance to be extensively utilized as candidates for non-invasive "liquid biopsy" approaches. DNA biosensors based on strand displacement amplification (SDA) methods have been successfully designed to detect miRNAs given the efficiently amplified and recycled of the target sequences. However, the unpredictable DNA framework and heavy reliance on free diffusion or random reactant collisions in existing approaches lead to delayed reaction kinetics and inadequate amplification. Thus

We describe the development of a novel, robust assay system for determining the changes in activity of proprotein converting enzymes. An assay for prolyl oligopeptidase (POP) activity was constructed using a peptide-streptavidin substrate coupled to magnetic microspheres and cleavage was detected by loss of streptavidin on the MAGPIX reader. Test analysis of postmortem pituitary extracts from schizophrenia patients showed an increase in POP activity compared to controls. The results were validated using both fluorometric and Western blot analyses for POP activity and immunoreactivity, respecti

In this study, we report a new three-dimensional (3D), bead-based microfluidic chip developed for rapid, sensitive and specific detection of influenza hemagglutinin. The principle of microfluidic chip is based on implementation of two-step procedure that includes isolation based on paramagnetic beads and electrochemical detection. As a platform for isolation process, streptavidin-modified MPs, which were conjugated via biotinylated glycan (through streptavidin-biotin affinity) followed by linkage of hemagglutinin to glycan, were used. Vaccine hemagglutinin (HA vaxi) was labeled with CdS quantu

The streptavidin-biotin system underpins numerous immunoassays for immobilization, labeling, and signal amplification, yet it is highly susceptible to interference from free biotin in biosamples, leading to inaccurate results and potential clinical misdiagnosis. Current countermeasures remain unsatisfactory, as antibody- or streptavidin-based scavenging strategies suffer from limited depletion capacity, poor assay compatibility, or separation-dependent workflows, making them difficult to broadly integrate into automated immunoassays. Herein, we present a rationally engineered nano-trap that in

Solid-state nanopores enable single-molecule detection of long double-stranded nucleic acids and can resolve the position of site-specific molecular labels attached to a DNA carrier. These labels are employed in many applications, such as molecular barcoding, protein mapping, and structure-based DNA data storage. Often, it is implicitly assumed that these labels do not significantly perturb molecular transport through the pore. However, the magnitude of such perturbations and their potential impact on positional readout remain largely unquantified. Here, we systematically quantify how dense mo

Optical resonator-based biosensors are important for advancing medical diagnostics and environmental monitoring due to their high sensitivity and label-free detection capabilities. In this study, we present a systematic comparison of three 3-aminopropyltriethoxysilane (APTES) functionalization methods - ethanol-based, methanol-based, and vapor-phase - on an Optical Cavity-based Biosensor (OCB) designed to detect streptavidin. The APTES process is an important first step for surface functionalization to form a linker to immobilize receptor molecules on the sensor surface. Our aim was to identif

RNA-binding proteins (RBPs) can bind and mediate RNA-RNA contacts. However, identifying specific RBP-organized RNA-RNA contacts remains challenging. Here, we present a capture RIC-seq (CRIC-seq) technique to map specific RBP-associated RNA-RNA contacts globally. We describe steps for formaldehyde cross-linking to fix RNA in situ conformation, pCp-biotin labeling to mark RNA juncture, and in situ proximity ligation to join proximal RNAs. We then detail immunoprecipitation to isolate specific RBP-associated RNA-RNA contacts, biotin-streptavidin selection to enrich chimeric RNAs, and library cons

The 3-phenoxybenzoic acid (3-PBA) metabolized from pyrethroids is more toxic and has a longer half-life to degradation in a natural environment compared to its parent compounds. Few reports have focused on the environmental detection of 3-PBA. In this study, anti-3-PBA nanobodies in trivalent form (Nb3) were biotinylated. A sensitive enzyme-linked immunosorbent assay (ELISA) based on the combination of Nb3-biotin and streptavidin-horseradish peroxidase (SA-HRP) was developed for the environmental detection of 3-PBA. After optimization, the ELISA showed a half-maximum signal inhibition concentr

An in situ nanozyme assembly amplification strategy is reported based on the programmable bioaffinity-driven assembly of streptavidin-Pd@Pt and biotin-Pd@Pt nanozymes, enabling ultrasensitive colorimetric immunoassay of protein biomarkers at sub-femtomolar levels. In this design, streptavidin-Pd@Pt nanozymes are first anchored onto the immunocomplexes through standard biotinylated antibody recognition, followed by the recruitment of biotin-Pd@Pt nanozymes via strong streptavidin-biotin interactions. This stepwise and localized assembly process induces the formation of high-density Pd@Pt nanozy

The small amount of cell-free fetal DNA (cffDNA) can be a useful biomarker for early non-invasive prenatal diagnosis (NIPD) of achondroplasia. In this study, a novel non-invasive electrochemical DNA sensor for ultrasensitive detecting FGFR3 mutation gene, a pathogenic gene of achondroplasia, based on biocatalytic signal materials and the biotin-streptavidin system are presented. Notably encapsulation of hemin in metal-organic frameworks-based materials (hemin-MOFs) and platinum nanoparticles (PtNPs) were used to prepare hemin-MOFs/PtNPs composites via a one-beaker-one-step reduction. In this work, using electrochemical impedance spectroscopy (EIS), we have, for the first time, label-free monitored protein immobilization on a gold surface through a strategy of electroaddressing, compatible with the production of microarrays for multi-detection. This functionalization is achieved via the alkyne/azide cycloaddition, better known as the "click" reaction. The electroaddressing was applied to a polythiol hexynyl derivative previously grafted onto the gold surface. This compound consists of two dithiol phosphate groups and a hexynyl function and was synthesized through a supporte

Passive immunotherapy against scorpion envenomations is facilitated by the preliminary titration of circulating toxins in envenomed patients. Currently, routinely used ELISA tests allow only the titration of the whole venom, without reference to the toxins which compose the venom and spread variably within the tissue. Taking as a model one of the three toxins responsible for the lethal effects of Androctonus australis hector (Aahl) venom, we developed an ELISA sandwich test based on a fragment of recombining antibody (scFv) consisting of the variable chains of the monoclonal IgG 9C2 coupled to

In microscale surface ion conduction (μSIC) sensing, binding of charged analytes to functionalized bead surfaces alters interfacial ion transport, making sensor performance highly dependent on surface electrostatics. Previously, probe immobilization relied on commercially available streptavidin-coated beads, which exhibit large intrinsic zeta potentials and fixed biotin-based coupling chemistry. Here, we report an in-house surface functionalization strategy for silica microbeads that enables controlled probe immobilization and substantially reduced baseline surface charge. We investigated the expression of cytokeratins 7 (CK7) and 20 (CK20) in nasal polyps on a group of 106 patients with chronic rhinosinusitis with nasal polyposis (45 women - 42% and 61 men - 58%) who benefited from surgical procedures. Harvested biological material was analyzed in the pathology laboratory through two methods: histopathological and immunohistochemical analysis. Classical histopathological method of processing the tissues initially fixed in 10% formalin was used. The tissues were then processed by paraffin impregnation, sectioned and stained with Hematoxylin-Eosin. The immunohist

Plasma membrane proteins are crucial for signal transduction, trafficking, and cell-cell interactions, all of which are vital for cell survival. These proteins, including G-protein coupled receptors, ion channels, transporters, and receptors, are key drug targets due to their central role in receiving and amplifying cellular signals. However, the isolation and purification of plasma membrane proteins pose significant challenges because of their integration with phospholipid bilayers and the small fraction of these proteins present in the plasma membrane. Biotinylation, in combination with stre

Oblique-incidence reflectivity difference (OIRD) is a novel optical technique for protein microarray detection with the characteristics of being real-time, label-free, high-throughput and compatible with arbitrary chip substrates. It is necessary yet challenging to improve the sensitivity of the OIRD microarray and gain a clear understanding of the enhancement mechanism for practical applications. In this study, we report a microarray chip specifically designed for OIRD to improve its sensitivity by using an electrochemically etched nanostructured fluorine-doped tin oxide (FTO) slide as the su

Toxoplasmosis is a zoonotic disease of economic importance found worldwide, and it is caused by Toxoplasma gondii, which affects a wide range of hosts. High prevalence of toxoplasmosis has been reported in rodents, and they are considered very important in the circulation and maintenance of the disease. However, epidemiologic studies of the disease in rodents are generally scarce in the Tropics. This study utilized the immunohistochemical (IHC) technique to detect Toxoplasma gondii in wild rats sampled from across the North Central Nigeria. The brain, intestine, diaphragm, lungs and kidney tis

Native mass spectrometry (nMS), particularly in conjunction with gas-phase ion mobility spectrometry measurements, has proven useful as a structural biology tool for evaluating the stoichiometry, conformation, and topology of protein complexes. Here, we demonstrate the combination of trapped ion mobility spectrometry (TIMS) and surface-induced dissociation (SID) on a Bruker SolariX XR 15 T FT-ICR mass spectrometer for the structural analysis of protein complexes. We successfully performed SID on mobility-selected protein complexes, including the streptavidin tetramer and cholera toxin B with b
