The Delta subunit of RNA polymerase is required for virulence of Streptococcus agalactiae.

Group B streptococci (GBS) remain the most significant bacterial pathogen causing neonatal sepsis, pneumonia, and meningitis in the United States despite the chemoprophylaxis strategies for preventing infection recommended by the Centers for Disease Control and Prevention. Using signature-tagged transposon mutagenesis to screen for novel virulence factors, we identified the rpoE gene as essential for development of sepsis in a neonatal rat model of GBS infection. An rpoE allelic replacement mutant displayed attenuated virulence in the sepsis model of infection identical to that of the transpos

Effect of uncoupling agents and respiratory inhibitors on the growth of Streptococcus agalactiae.

2,4-Dinitrophenol, dicoumarol, carbonylcyanide, m-chlorophenyl-hydrazone and pentachlorophenol all depressed aerobic molar growth yields of Streptococcus agalactiae to values equal to, or less than, those supported by substrate level phosphorylation. When the only source of energy was from substrate phosphorylation (anaerobic growth conditions), there was also a severe depression of the molar growth yield by the same four uncoupling agents. These results indicate that the effect of these agents is to uncouple both substrate and oxidative phosphorylation in S. agalactiae. Amytal inhibited gluco

Towards a holistic epidemiology of Streptococcus agalactiae using the BakRep repository.

BACKGROUND: Streptococcus agalactiae is a versatile multi-host pathogen that can cause severe neonatal disease in humans and mastitis in dairy animals. Its ability to infect a wide range of hosts is largely driven by its high genomic plasticity and the acquisition of distinct accessory genes. The global population of S. agalactiae is characterized by multiple capsular serotypes and clonal complexes that differ in their propensity to cause invasive disease, including hypervirulent CC17 (often serotype III) associated with neonatal meningitis, whereas CC1/CC19/CC23 are more often colonizing line

Pyomyositis Caused by Streptococcus agalactiae: A Case Report and Review of the Literature.

Pyomyositis is a subacute bacterial infection of the skeletal muscle that is more common in the tropics. Staphylococcus aureus is the most common pathogen involved in pyomyositis, but Streptococcus agalactiae (group B Streptococcus (GBS)) can cause pyomyositis. We herein present a case of pyomyositis of the right gluteus maximus caused by GBS bacteremia (primary bacteremia). An 81-year-old man with a history of diabetes mellitus presented with warmth and painful swelling of the right lateral buttock. Computed tomographic (CT) images revealed swelling of the right gluteus maximus without an abs

Macrolide and lincosamide resistance of Streptococcus agalactiae in pregnant women in Poland.

Knowing about the antibiotic resistance, serotypes, and virulence-associated genes of Group B Streptococcus for epidemiological and vaccine development is very important. We have determined antimicrobial susceptibility patterns, serotype, and virulence profiles. The antibiotic susceptibility was assessed for a total of 421 Streptococcus agalactiae strains, isolated from pregnant women and neonates. Then, 89 erythromycin and/or clindamycin-resistant strains (82 isolates obtained from pregnant women and seven isolates derived from neonates) were assessed in detail. PCR techniques were used to id

Interaction of Streptococcus agalactiae and Cellular Innate Immunity in Colonization and Disease.

Streptococcus agalactiae (Group B streptococcus, GBS) is highly adapted to humans, where it is a normal constituent of the intestinal and vaginal flora. Yet, GBS has highly invasive potential and causes excessive inflammation, sepsis, and death at the beginning of life, in the elderly and in diabetic patients. Thus, GBS is a model pathobiont that thrives in the healthy host, but has not lost its potential virulence during coevolution with mankind. It remains incompletely understood how the innate immune system contains GBS in the natural niches, the intestinal and genital tracts, and which mol

Transport of multidrug resistance substrates by the Streptococcus agalactiae hemolysin transporter.

Streptococcus agalactiae (group B streptococcus [GBS]) causes neonatal sepsis, pneumonia, and meningitis, as well as infections of the bovine udder. The S. agalactiae hemolysin is regarded as an important virulence factor, and hemolysin expression is dependent on the cyl gene cluster. cylA and cylB encode the ATP binding and transmembrane domains of a typical ATP binding cassette (ABC) transporter. The deduced proteins contain the signature sequence of a multidrug resistance (MDR) transporter, and mutation of the genes results in a nonhemolytic and nonpigmented phenotype. To further elucidate

Regulation of purine biosynthesis by a eukaryotic-type kinase in Streptococcus agalactiae.

Group B streptococci (GBS) are the principal causal agents of human neonatal pneumonia, sepsis and meningitis. We had previously described the existence of a eukaryotic-type serine/threonine kinase (Stk1) and phosphatase (Stp1) in GBS that regulate growth and virulence of the pathogen. Our previous results also demonstrated that these enzymes reversibly phosphorylated an inorganic pyrophosphatase. To understand the role of these eukaryotic-type enzymes on growth of GBS, we assessed the stk1-mutants for auxotrophic requirements. In this report, we describe that in the absence of the kinase (Stk

Antimicrobial susceptibility patterns of Streptococcus agalactiae in a German university hospital.

Group B streptococci (GBS) are the major cause of neonatal sepsis and meningitis. GBS infection in neonates is usually treated with a combination of penicillin and gentamicin. According to consensus guidelines, pregnant women at risk receive intrapartum prophylaxis with either ampicillin or penicillin or, in case of allergy, with erythromycin or clindamycin. We investigated the susceptibility patterns of 190 GBS strains from neonates isolated from 1993-1999 and 150 GBS strains collected from adult women in 1997 and 1999. All isolates were susceptible to penicillin, ampicillin and cefotaxime. E

[Bacteremia caused by Streptococcus agalactiae in adults. Presentation of 6 cases].

The incidence of Streptococcus agalactiae (SA) bacteremia in adults has increased in recent years, particularly in patients with severe underlying diseases. However SA is still an unusual pathogen in adults. We report 6 cases of SA bacteremia in adults, seen during 18 months. The microorganism was only isolated from the blood in 3 cases, and from blood and other locations in the remaining three cases. The age of the patients ranged from 35 to 70 years. Both sex were affected in the same proportion. In all cases except in one there was a previous underlying disease.

Adaptive microevolution in CC17 Streptococcus agalactiae from a mother-neonate infection.

BACKGROUND: The hypervirulent CC17 GBS is a major cause of neonatal invasive disease. Although this lineage can acquire mutations during host adaptation, the functional significance of such microvariations for bacterial pathogenesis remains unclear. METHODS: The GBS strains were isolated from a mother-neonate pair, with clinical data obtained from electronic medical record. MALDI-TOF MS was used for species identification. Antimicrobial susceptibility testing was performed using K-B method and interpreted by CLSI criteria. Serotyping and MLST were used for molecular epidemiological analysis.

Murine Model of Candida albicans and Streptococcus agalactiae Vaginal Co-Colonization.

Diverse microbes are isolated from the human vaginal tract, and many of the organisms that inhabit this niche can switch between commensal and pathogenic lifestyles. Factors, including community composition and the host environment, influence disease outcomes; however, the mechanisms determining these outcomes are not well understood. In this manuscript, we describe a murine model to investigate interkingdom interactions and the host response to Candida albicans and Streptococcus agalactiae (Group B Streptococcus, GBS), pathobionts that colonize the female genital tract (FGT). Emerging evidenc

Lumbar osteomyelitis and knee septic arthritis caused by Streptococcus agalactiae bacteraemia.

A man in his late 70s presented to the emergency department endorsing a week of malaise. He was recently hospitalised for 2 days for new back pain and was discharged with non-opioid pain medications but continued to seek care as he felt unwell. On presentation, he was afebrile with a leukocytosis. Physical examination revealed a painful left knee with no evidence of trauma. Arthrocentesis revealed purulent fluid with elevated white blood cell consistent with septic arthritis. He was started on broad-spectrum antibiotics and underwent irrigation and synovectomy of the left knee. Aspirate and bl
