Restoration-related salivary Streptococcus mutans level: a dental caries risk factor?

OBJECTIVES: The salivary level of Streptococcus mutans related to filled teeth was compared with the levels related to decayed and sound teeth, in order to establish whether the presence of restorations may increase the risk of infection of other teeth by Streptococcus mutans. METHODS: The sound, decayed and filled teeth were recorded in 809, 6-7-year-old school-children. Salivary Streptococcus mutans detection (i.e. more than 1 x 10(4) CFU/ml) and counts were evaluated. Streptococcus mutans log count means and prevalence values of subjects with only sound teeth (group 1), with filled, withou

Streptococcus mutans CcpA Promotes Biofilm Exopolysaccharide Production and Virulence Gene Expression.

Streptococcus mutans is a key cariogenic pathogen of dental caries due to its strong ability to synthesize extracellular glucans and form biofilms. Glucosyltransferases, encoded by gtfB/C/D genes in S. mutans, are responsible for producing biofilm exopolysaccharides (EPS) and are considered to be critical virulence factors. Previous studies have highlighted the roles of various regulatory factors of gtf genes in S. mutans. Here, we investigated the role of the global transcriptional regulator CcpA encoded by ccpA in regulating the EPS synthesis and biofilm formation of S. mutans. A ccpA in-fra

Enzymatic Galactosylation of Erythritol Enhances Antibacterial Activity against Cariogenic Streptococcus mutans.

Erythritol is a widely used sweetener with beneficial properties and bioactivities, including the inhibition of Streptococcus mutans, a bacterium that induces dental caries. Galactosylation of compounds is an attractive method for improving antimicrobial activity. In this study, a novel metagenomic β-galactosidase gene, bga7, was successfully expressed in Escherichia coli. The recombinant enzyme was used to galactosylate erythritol, generating a high yield (93.6%) of galactoside product at a concentration of 2 U/mL upon incubation with 20 mM o-nitrophenyl-β-d-galactopyranoside and 0.5 M erythr

Patchouli Alcohol as a Potential Anti-Cariogenic Compound Targeting Streptococcus mutans.

Patchouli alcohol, a natural tricyclic sesquiterpene isolated from Pogostemon cablin, exhibits promising antimicrobial properties, yet its activity against Streptococcus mutans (S. mutans), a major cariogenic bacterium associated with dental caries, has not been characterized. This study determined the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of patchouli alcohol against S. mutans to be 40 and 80 μg/mL, respectively. A series of phenotypic assays showed that patchouli alcohol delayed bacterial growth and culture acidification, reduced extracellular po

Antibiofilm potential of plant extracts: inhibiting oral microorganisms and Streptococcus mutans.

INTRODUCTION: A range of disinfectant mouthwashes are available for oral hygiene. The gold standard is Chlorhexidine digluconate (CHX), which, like other available products, cannot be used without side effects in the long term. However, in recent years, therapy with herbal products, often considered antiquated, has regained considerable interest. Therefore, the search for plant compounds as an alternative to existing oral disinfectants is meaningful. METHODS: In this study, eleven Mediterranean plant extracts were tested for their antimicrobial effect in vitro. Methanol extracts of the follow

Inhibition of Streptococcus mutans growth and biofilm formation through protein acetylation.

Numerous cellular processes are regulated in response to the metabolic state of the cell, and one such regulatory mechanism involves lysine acetylation. Lysine acetylation has been proven to play an important role in the virulence of Streptococcus mutans, a major cariogenic bacterial species. S. mutans' glucosyltransferases (Gtfs) are responsible for synthesizing extracellular polysaccharides (EPS) and contributing to biofilm formation. One of the most common nonsteroidal anti-inflammatory drugs is acetylsalicylic acid (ASA), which can acetylate proteins through a nonenzymatic transacetylation

SepM mutation in Streptococcus mutans clinical isolates and related function analysis.

BACKGROUND: Streptococcus mutans (S. mutans) is an important pathogenic bacterium that causes dental caries, while Streptococcus gordonii (S. gordonii) is a non-cariogenic bacterium that inhibits the growth of S. mutans. The SepM protein can promote the inhibitory ability of S. mutans against S. gordonii by cleaving CSP-21 and activating the ComDE two-component system. This study was designed to explore sepM mutation in S. mutans clinical isolates and related function in the regulation of interactions with S. gordonii. METHODS: The S. mutans clinical strains that can inhibit the growth of S.

F-type proton-pumping ATPase mediates acid tolerance in Streptococcus mutans.

AIMS: Streptococcus mutans is highly sensitive to inhibitors of proton-pumping F-type ATPase (F-ATPase) under acidic conditions. Herein, we investigated the role of S. mutans F-ATPase in acid tolerance using a bacterium expressing the F-ATPase β subunit at lower levels than the wild-type strain. METHODS AND RESULTS: We generated a mutant S. mutans expressing the catalytic β subunit of F-ATPase at lower levels than the wild-type bacterium. The mutant cells exhibited a significantly slower growth rate at pH 5.30, whereas the rate was essentially the same as that of wild-type cells at pH 7.40. I

Dlt operon regulates physiological function and cariogenic virulence in Streptococcus mutans.

Streptococcus mutans is one of the major cariogenic pathogens in the oral cavity. The dlt operon is responsible for the process of D-alanylation of lipoteichoic acid and is related to the virulence of S. mutans. The dlt operon contributes to the adhesion, biofilm formation, stress response, interspecies competitiveness and autolysis of S. mutans. In addition, we have summarized the possible regulatory networks of the dlt operon. This review highlights the significant role of the dlt operon in S. mutans and provides new ideas for ecological caries prevention.

Hydrogel-Encapsulated Biofilm Inhibitors Abrogate the Cariogenic Activity of Streptococcus mutans.

We designed and synthesized analogues of a previously identified biofilm inhibitor IIIC5 to improve solubility, retain inhibitory activities, and to facilitate encapsulation into pH-responsive hydrogel microparticles. The optimized lead compound HA5 showed improved solubility of 120.09 μg/mL, inhibited Streptococcus mutans biofilm with an IC50 value of 6.42 μM, and did not affect the growth of oral commensal species up to a 15-fold higher concentration. The cocrystal structure of HA5 with GtfB catalytic domain determined at 2.35 Å resolution revealed its active site interactions. The ability o

Inhibition of Streptococcus mutans biofilms with bacterial-derived outer membrane vesicles.

BACKGROUND: Biofilms are microbial communities surrounded by a self-produced extracellular matrix which protects them from environmental stress. Bacteria within biofilms are 10- to 1000-fold more resistant to antibiotics, making it challenging but imperative to develop new therapeutics that can disperse biofilms and eradicate infection. Gram-negative bacteria produce outer membrane vesicles (OMV) that play critical roles in communication, genetic exchange, cargo delivery, and pathogenesis. We have previously shown that OMVs derived from Burkholderia thailandensis inhibit the growth of drug-sen

Saliva from subjects harboring Actinobacillus actinomycetemcomitans kills Streptococcus mutans in vitro.

BACKGROUND: Previous research indicated that patients with localized aggressive periodontitis (LAgP) had minimal proximal decay. We speculated that differences in these two proximal dental diseases (LAgP and proximal decay) in LAgP could be due to the effect of saliva on the growth of key microorganisms related to these two infections. Carbon dioxide (CO(2)) is required for growth of Actinobacillus actinomycetemcomitans (Aa), the reputed cause of LAgP. Bicarbonate, a source of CO(2), buffers acid production by Streptococcus mutans (Sm), a key organism associated with caries. The purpose of thi

Antibacterial and Antibiofilm Effects of L-Carnitine-Fumarate on Oral Streptococcal Strains Streptococcus mutans and Streptococcus sobrinus.

Streptococcus mutans is a major pathogenic habitant of oral caries. Owing to its physiological and biochemical features, it prevails in the form of plaque biofilm together with another important mutans streptococci species, Streptococcus sobrinus. Both species are considered as initiators of cavity lesions, and biofilm is essential to the dental caries process. Compared with the planktonic populations, the biofilm form has higher resistance to environmental conditions and antibiotics. Dental plaques also secure the long-term survival of microorganisms and protection from any stress conditions.

[Prevention of cariogenic dental plaque. Study of the structures implicated in the adhesion and coaggregation in Streptococcus mutans and Streptococcus sobrinus].

Cariogenic dental plaque may be assimilated to a biofilm resulting from the adhesion of S. mutans, then from the coaggregation of other streptococci, or other genus. We used a static monospecific biofilm model. Supports or bacteria were treated with inhibitors before adhesion in order to clarify the nature of adhesins responsible for the primary adhesion of S. mutans and S. sobrinus on Tygon. To determine the bindings of coaggregation, inhibitors were applied on one-day-old biofilms. Analysis of effects were performed by automatic inoculator Spiral (Interscience) for microbiological methods, a

A Rapid and Specific RAA-CRISPR/Cas13a Assay for Detecting Streptococcus mutans.

Streptococcus mutans is recognized as a major etiological agent of dental caries, a highly prevalent chronic global health issue. The early identification of such cariogenic pathogens is therefore critical for implementing effective preventive strategies. To address this need, we established an integrated detection platform coupling recombinase-aided amplification (RAA) with CRISPR-Cas13a for S. mutans detection and evaluated its performance against conventional polymerase chain reaction (PCR). The proposed RAA-CRISPR-Cas13a assay exhibited superior sensitivity and faster turnaround time compa

Antibacterial and antibiofilm activities of Zn2+ against Staphylococcus aureus and Streptococcus mutans.

STATEMENT OF PROBLEM: Staphylococcus aureus (S aureus) and Streptococcus mutans (S mutans) are pathogenic bacteria associated with denture-related stomatitis. Zinc (Zn), a widely used metallic antibacterial agent, can induce the production of reactive oxygen species (ROS) in cells, thereby inhibiting microbial growth and proliferation; studies, however, are lacking. PURPOSE: The purpose of this in vitro study was to determine the antibacterial and antibiofilm function of Zn2+ against S aureus and S mutans and assess gene expression changes to clarify its inhibitory mechanisms. Effects of epigallocatechin gallate on the development of matrix-rich Streptococcus mutans biofilm.

In this study, we evaluated the impact of Epigalocatechin-3-gallate (EGCG) on S. mutans biofilm development for 24 and 46 h using high-resolution confocal laser scanning microscopy. EGCG treatment led to the formation of interspaced exopolysaccharide (EPS)-microcolony complexes unevenly distributed on the surface of hydroxyapatite disc, forming a thinner and less complex biofilm structure with significantly reduced biomass, matrix volume, and thickness compared to the NaCl treated group (negative control). At 46 h, the biofilm of the EGCG-treatment group failed to form the bacterial-EPS supers
