Thin layer chromatography convulsant screen extended by gas chromatography-mass spectrometry.

Acute onset convulsive disorders in the canine may result from exposure to a variety of toxicants including strychnine, insecticides, metaldehyde, zinc phosphide, methylxanthines, drugs of abuse, bromethalin, and the tremorgenic mycotoxins (roquefortine and penitrem A). Although several of the above can be identified in a single gas chromatography-mass spectrometry (GC-MS) screen most have to be determined by separate tests. This report describes a modification of the strychnine extraction procedure, which allows thin layer chromatographic (TLC) identification of strychnine, bromethalin, roque

Influence of morphine and cyclazocine on the cortical epileptic foci in rabbits.

The effects of morphine, cyclazocine and naloxone on penicillin- and strychnine-induced epileptic foci were studied in rabbits. The intracortical injection of penicillin (75, 150 and 300 units) elicited isolated spikes followed by repeated ictal events. The application of strychnine (0.062 and 0.125%) over the cortical surface of one side induced appearance of ipsilateral spiking spreading to the contralateral cortex. Administration of morphine (0.25-0.75 mg/kg i.v.) or cyclazocine (0.05-3.0 mg/kg i.v.) inhibited the occurrence or the duration of the EEG and motor manifestations induced by pen

Action and localization of glycine and taurine in the cat retina.

The effects on retinal ganglion cells of iontophoretically applied glycine, taurine and strychnine were studied in the optically intact eye of the cat. Glycine and taurine suppressed the light-evoked discharge of all on-centre and off-centre brisk ganglion cells, regardless of the visual stimulus used. Strychnine blocked the action of externally applied glycine and taurine. The light-evoked response of all ganglion cells was raised by strychnine. The tonic discharge of the light response was suppressed or raised by the drugs more than the phasic response. A population of amacrine cells, which

The action of local anaesthetics on the spinal cord of the cat.

The effects of intravenously injected lignocaine, procaine and amethocaine on the monosynaptic and polysynaptic reflexes and the cord dorsum potentials elicited by selective electrical stimulation of appropriate afferent fibres have been investigated in decerebrate cats before and after strychnine. The three drugs depress the amplitude of the reflex responses in the basal preparation for 40 to 50 min. Smaller doses eliminate the reflex augmentation produced by strychnine for comparable periods. Very small doses selectively abolish the augmentation of the P component of the cord dorsum response

Blunted response to low oxygen of rat respiratory network after perinatal ethanol exposure: involvement of inhibitory control.

Acute ethanol depresses respiration, but little is known about chronic ethanol exposure during gestation and breathing, while the deleterious effects of ethanol on CNS development have been clearly described. In a recent study we demonstrated that pre- and postnatal ethanol exposure induced low minute ventilation in juvenile rats. The present study analysed in juvenile rats the respiratory response to hypoxia in vivo by plethysmography and the phrenic (Phr) nerve response to ischaemia in situ. Glycinergic neurotransmission was assessed in situ with strychnine application and [(3)H]strychnine b

Involvement of GABA and glycine in recurrent inhibition of spinal motoneurons.

1. Recurrent inhibitory postsynaptic potentials (IPSPs) were recorded intracellularly from chloride-loaded motoneurons in the isolated lumbar spinal cord of neonatal rats (day 5-day 12). This in vitro preparation exhibited an intact and functional recurrent inhibitory pathway that displayed characteristics previously described for this pathway in other species. 2. Although strychnine (1-5 microM) depressed the chloride-dependent recurrent synaptic potentials evoked by ventral root stimulation by 48.2 +/- 2.7% (mean +/- SE, n = 13), confirming that part of the recurrent IPSP is mediated by a gl

Interactions between allosteric modulators and 4-DAMP and other antagonists at muscarinic receptors: potential significance of the distance between the N and carboxyl C atoms in the molecules of antagonists.

Allosteric enhancement of the affinity of muscarinic receptors for their ligands offers a new way to influence cholinergic neurotransmission. The structure of the allosteric binding domain(s) and the features of agonists, antagonists and modulators which determine the occurrence of either positive or negative cooperativity require clarification. We tested interactions between allosteric modulators alcuronium, strychnine and brucine and eight antagonists at muscarinic receptors expressed in CHO cells. In experiments with unlabeled antagonists, all three modulators enhanced the affinity for 4-di

A simple, cost-effective and rapid method for simultaneous determination of Strychnos nux-vomica alkaloids in blood and Ayurvedic medicines based on ultrasound-assisted dispersive liquid-liquid microextraction-thin-layer chromatography-image analysis.

A simple, rapid, cost-effective and green analytical method is developed based on ultrasound-assisted dispersive liquid-liquid microextraction (US-DLLME) coupled to thin-layer chromatography (TLC)-image analysis for the simultaneous determination of two major alkaloids of Strychnos nux-vomica L i.e., strychnine and brucine. The method is composed of three steps, namely (i) US-DLLME by injecting a mixture of 100-μL chloroform (extraction solvent) and 1-mL methanol (disperser solvent) in 5 mL of aqueous sample, followed by ultrasonication and centrifugation, (ii) TLC of 20 μL of sedimented phase

Trigeminal reflex regulation of the glottis depends on central glycinergic inhibition in the rat.

In an unanesthetized decerebrate in situ arterially perfused brain stem preparation of mature rat, strychnine (0.05-0.2 microM) blockade of glycine receptors caused postinspiratory glottal constriction to occur earlier, shifting from early expiration to inspiration. This resulted in a paradoxical inspiratory-related narrowing of the upper airway. Stimulation of the trigeminal ethmoidal nerve (EN5; 20 Hz, 100 micros, 0.5-2 V) evoked a diving response, which included a reflex apnea, glottal constriction, and bradycardia. After strychnine administration, this pattern was converted to a maintained

Tonic GABA and Glycine Inhibition Control Pain Hypersensitivity via Limiting α2δ-1- and mGluR5-Dependent NMDA Receptor Activity at Primary Afferent→Excitatory Neuron Synapses.

Impaired synaptic inhibition by GABA and glycine contributes to excitatory-inhibitory imbalance in the spinal cord associated with chronic neuropathic pain; however, the underlying mechanisms remain unclear. Here, we investigated how GABAergic and glycinergic inputs regulate synaptic N-methyl-d-aspartate receptor (NMDAR) activity in excitatory and inhibitory neurons of the spinal dorsal horn in male and female mice. Vesicular glutamate transporter 2 (VGluT2)-expressing excitatory neurons and vesicular γ-aminobutyric acid transporter (VGAT)-expressing inhibitory neurons exhibited comparable mix

The role of medial olivocochlear activity in contralateral suppression of auditory steady-state responses.

OBJECTIVE: The auditory steady-state response (ASSR) amplitudes fall in the presence of contralateral noise. However, whether and to what extent medial olivocochlear (MOC) activity involves in contralateral suppression of ASSR remain unclear. Therefore, we assess the role of MOC activity in contralateral suppression of ASSR. METHODS: Mice were treated with strychnine to completely eliminate MOC activity and then measured ASSR amplitudes in the presence of contralateral noise. RESULTS: The contralateral noise reduces ASSR amplitudes at some stimulus intensity. After treating with the strychni

Anticonvulsant properties of some new derivatives of phensuccinimide.

Anticonvulsant properties of two new derivatives, of phenylsuccinimide, N-morpholinomethyl-3-bromophenyl-succinimide(IL-7) and N-amino-3-bromophenylsuccinimide (IL-16), were studied. It was stated that both compounds have similar acute toxicity and act longer than phensuximide. They are especially active in maximal electroshock seizure (MES) test. Their effectiveness against convulsions produced by pentetrazole is less than that of phensuximide. Their effectiveness against procaine and strychnine is similar, only one of tested compounds has very high potency against strychnine. This compound,

[Comparative evaluation of the antitoxic activity of central nervous system analeptics and their combinations in barbamyl poisoning].

Tests conducted on rats poisoned with increasingly lethal doses of sodium amytal demonstrated the antitoxic activity of an analeptic mixture to be superior to that of picrotoxin, strichnine, corasol and caffeine entering into its composition and also to the activity of bemegride. As to the degree of antitoxic activity in poisoning of mice with sodium amytal (one LD50) the CNS analeptics are arranged in the following descending oder of their action: analeptic mixture, picrotoxin, bemegride, corasol, strychnine. In poisoning with higher doses of sodium amytal (LD84) corasol, strychnine and caffe

Regulation of glycine receptor binding in the mouse hypoglossal nucleus in response to axotomy.

Recent studies have shown that muscarinic receptors in brain hypoglossal nuclei exhibit a loss of specific ligand binding in response to axotomy of the hypoglossal nerve. The mouse hypoglossal nucleus contains a high level of receptors for the inhibitory neurotransmitter, glycine; the ligand [3H]strychnine binds to the glycine receptor with high affinity. In the present study [3H]strychnine binding in mouse hypoglossal nuclei was examined at 1 to 150 days after unilateral lesions of the hypoglossal nerve. Brains were sectioned on a cryostat, thaw-mounted onto microscope slides, incubated with

Chemical modification of the glycine receptor with fluorescein isothiocyanate specifically affects the interaction of glycine with its binding site.

Fluorescein 5'-isothiocyanate (FITC) was used to modify lysine residues of the strychnine-sensitive glycine receptor. Pretreatment of rat spinal cord synaptic plasma membranes with FITC specifically affected the ability of glycine to displace [3H]strychnine binding. Glycine completely prevented the effect of FITC modification, suggesting the existence of lysine group(s) either at or in the vicinity of the agonist binding site. Labeling of purified glycine receptor with FITC indicates that such lysine residue(s) are located in the 48,000 daltons polypeptide. Chemical cleavage of the FITC-labele

The anticonvulsant and CNS activity of N-butyryl-N-butylurea in mice.

As a further extension of our studies related to CNS activity of substituted butylureas, a number of derivatives of butylurea were synthesized. Among these a derivative of n-butylurea, N-butyryl-N-butylurea (NBNB) was prepared by acylation of n-butylurea with butyryl chloride in pyridine. The compound was found to possess considerable sedative-hypnotic action. Sleeping time of pentobarbital and barbital was significantly potentiated by NBNB. The compound also exhibited moderate anti-tremorine action and produced significant reduction in the activity ratio for Treadmill experiments. Significant

Role of synaptic inhibition in turtle respiratory rhythm generation.

In vitro brainstem and brainstem-spinal cord preparations were used to determine the role of synaptic inhibition in respiratory rhythm generation in adult turtles. Bath application of bicuculline (a GABA(A) receptor antagonist) to brainstems increased hypoglossal burst frequency and amplitude, with peak discharge shifted towards the burst onset. Strychnine (a glycine receptor antagonist) increased amplitude and frequency, and decreased burst duration, but only at relatively high concentrations (10-100 microM). Rhythmic activity persisted during combined bicuculline and strychnine application (

Disorder of the inhibitory glycine receptor: inherited myoclonus in Poll Hereford calves.

Inherited congenital myoclonus in Poll Hereford calves is characterized by hyperesthesia and myoclonic jerks of the skeletal musculature that occur spontaneously and in response to sensory stimuli. The symptoms of the disorder suggest a failure of spinal inhibition and are similar to those in subconvulsive strychnine poisoning. Strychnine is a high-affinity antagonist of the synaptic actions of glycine. Our recent biochemical studies revealed a specific and marked deficit in [3H]strychnine binding sites in brain stem and spinal cord membranes from myoclonic calves compared with unaffected cont
