Degradation of cyanobacterial biosignatures by ionizing radiation.

Primitive photosynthetic microorganisms, either dormant or dead, may remain today on the martian surface, akin to terrestrial cyanobacteria surviving endolithically in martian analog sites on Earth such as the Antarctic Dry Valleys and the Atacama Desert. Potential markers of martian photoautotrophs include the red edge of chlorophyll reflectance spectra or fluorescence emission from systems of light-harvesting pigments. Such biosignatures, however, would be modified and degraded by long-term exposure to ionizing radiation from the unshielded cosmic ray flux onto the martian surface. In this i

Putative functional redundancy in cyanobacterial microbiomes allows stable bioplastic production despite shifts in dominant populations.

This study investigates whether a single initial cyanobacterial microbiome, originating from a mixed community, retains its capacity for polyhydroxybutyrate (PHB) production under non-sterile long-term operation when subjected to cultivation conditions that promote different dominant cyanobacterial populations. Distinct selective pressures were applied in two photobioreactors: one subjected to high pH (>10) and nutrient limitation, which favored the dominance of Synechococcus sp., and one operating under controlled pH (8.0-8.25) and sufficient nutrient supplementation, which promoted the enric

Reduced mediators released by cyanobacteria during exoelectrogenesis detected using differential pulse voltammetry.

Cyanobacteria generate electrical current through exoelectrogenesis (extracellular electron transfer) downstream of photosynthesis, yet the identity of the endogenous redox mediator(s) responsible remains unresolved. Here, we apply differential pulse voltammetry (DPV) to detect and characterise redox-active species released by Synechocystis sp. PCC 6803 under illumination. To enable sensitive detection, we developed an electrolyte intermediate between BG11 (Blue-Green-11) growth medium and MOPS (3-(N-morpholino)propanesulfonic acid) buffer that minimises background electrochemical interference

Design, synthesis and activity evaluation of pseudilin analogs against cyanobacteria as IspD inhibitors.

The discovery of safe, effective, and selective chemical algicides is the stringent need for the algicides development, and it is also one of the effective routes to control cyanobacteria harmful algal blooms and to meet the higher requirements of environmental and ecological. In this work, a series of novel bromo-N-phenyl-5-o-hydroxyphenylpyrazole-3-carboxyamides were rationally designed as pseudilin analogs by bioisosteric replacement and molecular hybridization strategies, in which the pyrrole unit of pseudilin was replaced with pyrazole and further combined with the dominant structural fra

Differential requirement of two homologous proteins encoded by sll1214 and sll1874 for the reaction of Mg protoporphyrin monomethylester oxidative cyclase under aerobic and micro-oxic growth conditions.

The two open reading frames in the Synechocystis sp. PCC 6803 genome, sll1214 and sll1874, here designated cycI and cycII, respectively, encode similar proteins, which are involved in the Mg protoporphyrin monomethylester (MgProtoME) cyclase reaction. The impairment of tetrapyrrole biosynthesis was examined by separate inactivation of both cyclase encoding genes followed by analysis of chlorophyll contents, MgProtoME levels and several enzyme activities of tetrapyrrole biosynthesis. We additionally addressed the question, whether the two isoforms can complement cyclase deficiency under normal

Retention of a SulP-family bicarbonate transporter in a periplasmic N2-fixing cyanobacterial endosymbiont of an open ocean diatom.

Symbioses between diatoms and the N2-fixing, heterocyst-forming cyanobacteria Richelia spp. are widespread and contribute to primary production. Unique to these symbioses is a variation in the symbiont location: one lives in the host cytoplasm (endobiont) vs. residing between the host frustule and plasmalemma (periplasmic endobiont). Both partners are photosynthetic, yet how the partners acquire, share, or compete for bicarbonate necessary for their photosynthesis is unknown. The genomes of both endobionts (ReuHH01 and RintRC01, respectively) contain genes encoding SulP-family proteins, which

Control of photosynthetic and high-light-responsive genes by the histidine kinase DspA: negative and positive regulation and interactions between signal transduction pathways.

We have deleted a gene for a sensor histidine kinase, dspA (or hik33), in the cyanobacterium Synechocystis sp. strain PCC6803. In low and moderate light, the mutant grew slowly under photoautotrophic conditions, with a doubling time of approximately 40 h, and had severely reduced photosynthetic oxygen evolution. When the mutant was maintained in low or moderate light in the presence of glucose, its growth rate was only somewhat lower than that of wild-type cells. However, the mutant was light sensitive and rapidly died in high light. Furthermore, levels of many transcripts encoding genes assoc

Transient electron paramagnetic resonance of the triplet state of P700 in photosystem I: evidence for triplet delocalization at room temperature.

Spin-polarized EPR spectra of the triplet state of P700, the primary electron donor in photosystem I (PS I), have been measured for the first time at room temperature. The measurements were performed on intact PS I from Synechococcus sp. after prereduction of all iron-sulfur centers and on vitamin K1 depleted PS I from Synechocystis 6803. The two preparations give similar spectra with a polarization pattern which indicates that the triplet state is formed via recombination of a radical pair. The axial and nonaxial zero-field splitting (zfs) parameters are found to be magnitude of D = (284 +/-

The generation of voltage (ΔV) by purified photosynthetic protein-pigment complex of photosystem I (PS I) from the cyanobacterium Synechocystis sp. PCC 6803 immobilized onto a porous Millipore nitrocellulose membrane filter (MF) and contacting with indium tin oxide (ITO) semiconductor glass slides, termed ITO|PS I - MF|ITO have been directly measured. In the presence of the ascorbate/2,6-dichlorophenolindophenol redox couple and disaccharide trehalose, photoinduced generation of ΔV with a maximum amplitude was observed upon light irradiation for more than 1 h. In Situ, High-Resolution Quantification of CO2 Uptake Rates via Automated Off-Gas Analysis Illuminates Carbon Uptake Dynamics in Cyanobacterial Cultures.

Quantification of cyanobacterial CO2 fixation rates is vital to determining their potential as industrial strains in a circular bioeconomy. Currently, however, CO2 fixation rates are most often determined through indirect and/or low-resolution methods, resulting in an incomplete picture of both dynamic behaviors and total carbon fixation potential. To address this, we developed the "Automated Carbon and CO2 Experimental Sampling System" (ACCESS); a low-cost system for in situ off-gas analysis that supports the automated acquisition of high-resolution volumetric CO2 uptake rates from multiple c

Asymmetric distribution of type IV pili triggered by directional light in unicellular cyanobacteria.

The type IV pili (T4P) system is a supermolecular machine observed in prokaryotes. Cells repeat the cycle of T4P extension, surface attachment, and retraction to drive twitching motility. Although the properties of T4P as a motor have been scrutinized with biophysics techniques, the mechanism of regulation remains unclear. Here we provided the framework of the T4P dynamics at the single-cell level in Synechocystis sp. PCC6803, which can recognize light direction. We demonstrated that the dynamics was detected by fluorescent beads under an optical microscope and controlled by blue light that in

Structure optimization and bioactivity evaluation of ThDP analogs targeting cyanobacterial pyruvate dehydrogenase E1.

Harmful cyanobacteria bloom (HCB) has occurred frequently in recent years and it is urgent to develop novel algicides to deal with this problem. In this paper, a series of novel thiamin diphosphate (ThDP) analogs 5a-5g were designed and synthesized targeting cyanobacterial pyruvate dehydrogenase complex E1 (Cy-PDHc E1). Our results showed that compounds 5a-5g have higher inhibitory activities against Cy-PDHc E1 (IC50 9.56-3.48 µM) and higher inhibitory activities against two model cyanobacteria strains Synechocystis sp PCC6803 (EC50 2.03-1.58 µM) and Microcystis aeruginosa FACHB905 (EC50 1.86-

Proteome Mapping of a Cyanobacterium Reveals Distinct Compartment Organization and Cell-Dispersed Metabolism.

Cyanobacteria are complex prokaryotes, incorporating a Gram-negative cell wall and internal thylakoid membranes (TMs). However, localization of proteins within cyanobacterial cells is poorly understood. Using subcellular fractionation and quantitative proteomics, we produced an extensive subcellular proteome map of an entire cyanobacterial cell, identifying ∼67% of proteins in Synechocystis sp. PCC 6803, ∼1000 more than previous studies. Assigned to six specific subcellular regions were 1,712 proteins. Proteins involved in energy conversion localized to TMs. The majority of transporters, with

Synthesis of cyclic di-nucleotidic acids as potential inhibitors targeting diguanylate cyclase.

Five analogs of cyclic di-nucleotidic acid including c-di-GMP were synthesized and evaluated for their biological activities on Slr1143, a diguanylate cyclase of Synechocystis sp. Slr1143 was overexpressed from the recombinant plasmid which contained the gene of interest and subsequently purified by affinity chromatography. A new HPLC method capable of separating the compound and product peaks with good resolution was optimized and applied to the analysis of the compounds. Results obtained show that cyclic di-inosinylic acid 1b demonstrates a stronger inhibition on Slr1143 than c-di-GMP and is

Function of iron-stress-induced protein A in cyanobacterial cells with monomeric and trimeric photosystem I.

The acclimation of cyanobacteria to iron deficiency is crucial for their survival in natural environments. In response to iron deficiency, many cyanobacterial species induce the production of a pigment-protein complex called iron-stress-induced protein A (IsiA). IsiA proteins associate with photosystem I (PSI) and can function as light-harvesting antennas or dissipate excess energy. They may also serve as chlorophyll storage during iron limitation. In this study, we examined the functional role of IsiA in cells of Synechocystis sp. PCC 6803 grown under iron limitation conditions by measuring t

Quantification of NAD(P)H in cyanobacterial cells by a phenol extraction method.

In photosynthetic organisms, it is recognized that the intracellular redox ratio of NADPH is regulated within an appropriate range for the cooperative function of a wide variety of physiological processes. However, despite its importance, there is large variability in the values of the NADPH fraction [NADPH/(NADPH + NADP+)] quantitatively estimated to date. In the present study, the light response of the NADPH fraction was investigated by applying a novel NADP(H) extraction method using phenol / chloroform / isoamyl alcohol (PCI) in the cyanobacterium Synechocystis sp. Production of Bioplastic Compounds by Genetically Manipulated and Metabolic Engineered Cyanobacteria.

Direct conversion of carbon dioxide to valuable compounds is a desirable way to reduce the environmental burden and switch from fossil to renewable fuels. Cyanobacteria are photosynthetic bacteria that perform oxygenic photosynthesis and are able to produce valuable compounds from carbon dioxide in the air. Synechocystis and Synechococcus species, model unicellular cyanobacteria, can produce succinate and lactate, which are commodity chemicals used to generate bioplastics. Several cyanobacteria are also able to produce polyhydroxybutyrate, a biodegradable polyester that accumulates under nitro

Dynamic photosynthetic labeling and carbon-positional mass spectrometry monitor in vivo RUBISCO carbon assimilation rates.

RIBULOSE-1,5-BISPHOSPHATE CARBOXYLASE/OXYGENASE (RUBISCO) is the most abundant enzyme and CO2 bio-sequestration system on Earth. Its in vivo activity is usually determined by 14CO2 incorporation into 3-phosphoglycerate (3PGA). However, the radiometric analysis of 3PGA does not distinguish carbon positions. Hence, RUBISCO activity that fixes carbon into the 1-C position of 3PGA and Calvin-Benson-Bassham (CBB) cycle activities that redistribute carbon into its 2-C and 3-C positions are not resolved. This study aims to develop technology that differentiates between these activities. In source fra

Removal of Photosystem I Subunit PsaE Leads to Photosynthetic Hydrogen Production in a Cyanobacterial PsaF-HoxYH Fusion Strain.

Photosynthetic hydrogen (photoH2) production is a promising approach for the utilisation of solar energy. Fusion of a hydrogenase (H2ase) to photosystem I (PSI) provides a direct light-driven route for photoH2 generation. We established the lateral transmembrane subunit PsaF of PSI as a new fusion partner for the NiFe-hydrogenase (HoxYH) in the cyanobacterium Synechocystis sp. PCC 6803. Gradual reduction of PsaC expression in a PsaF-HoxYH strain was established to access the most redox negative FeS-cluster FX of PSI, but this did not result in photoH2 production. In another approach PSI subuni
