Arabidopsis AtERF014 acts as a dual regulator that differentially modulates immunity against Pseudomonas syringae pv. tomato and Botrytis cinerea.

ERF transcription factors play critical roles in plant immune responses. Here, we report the function of AtERF014, a nucleus-localized transcriptional activator, in Arabidopsis immunity. Expression of AtERF014 was induced by Pseudomonas syringae pv. tomato (Pst) and Botrytis cinerea (Bc). AtERF014-overexpressing (OE) plants displayed increased Pst resistance but decreased Bc resistance, whereas AtERF014-RNAi plants exhibited decreased Pst resistance but increased Bc resistance. After Pst infection, expression of salicylic acid (SA)-responsive genes AtPR1 and AtPR5 in AtERF014-OE plants and of

Yeast increases resistance in Arabidopsis against Pseudomonas syringae and Botrytis cinerea by salicylic acid-dependent as well as -independent mechanisms.

Cell-wall and glucopeptide components of yeast have been reported to exhibit elicitor activity. The mode of action of defense activation by yeast is not known so far. In this study, we used the model plant Arabidopsis to investigate the activation of defense responses by yeast, the effect on resistance against different pathogens, and the mode of action. Treatment of Arabidopsis plants with an autoclaved yeast suspension induced the expression of systemic acquired resistance-related genes and accumulation of the phytoalexin camalexin. Symptom development and bacterial growth after infection wi

The Arabidopsis tex1-4 mutant exhibits altered mRNA export and hormonal crosstalk in plant immunity against Pseudomonas syringae.

TEX1 contributes to mRNA export and SA-mediated defense while modulating ABA-driven susceptibility, linking RNA processing to immune regulation in Arabidopsis TEX1, a subunit of the multi-protein THO/TREX complex conserved in eukaryotes, facilitates the synthesis, splicing, and nuclear-cytoplasmic export of mRNAs. However, its mechanism in plants remains to be thoroughly established, particularly its contribution to plant immunity and the mRNA export mechanism in response to pathogen challenges. Here, we showed that TEX1 contributes to mRNA export and hormonal signaling during pathogen infecti

Pseudomonas syringae pv. tabaci 8018 is Virulent Against Tobacco Cultivars N509 and Burley21 Due to Spontaneous Mutation in hopAZ1.

The complete genome sequence of a relatively weak virulent tobacco pathogen, Pseudomonas syringae pv. tabaci 8018 (Pta8018) was determined to elucidate key genetic elements that cause disease. Genome comparison of Pta8018 to the previously available P. syringae pv. tabaci strains revealed that Pta8018 and Pta11528 had high genome synteny, with an approximately 5 Mb chromosome inversion in Pta6605. Furthermore, Pta8018 had lost the pathogenicity island GI-1 including a tabtoxin biosynthesis gene cluster and showed weak virulence on Nicotiana tabacum 'Xanthi NC' (Xanthi). When inoculated into se

The AcLFYL1-AcCSE module enhances kiwifruit resistance to Pseudomonas syringae pv. actinidiae by regulating cell density and lignin biosynthesis.

Canker disease caused by Pseudomonas syringae pv. actinidiae (Psa) is a severe bacterial infection threatening global kiwifruit production. Psa causes lignin degradation, cell wall rupture, leaf wilting, and canker formation on branches and trunks, often leading to plant death. The plant cell wall serves as a structural barrier against pathogens, with its thickness, composition, and cell density influencing disease resistance. Comparative studies between resistant germplasms Actinidia eriantha "Maohuaxiong" (A. eriantha 'MHX') and Actinidia latifolia "Kuoye" (A. latifolia 'KY') and susceptible

Sulfur-Induced Resistance against Pseudomonas syringae pv. actinidiae via Triggering Salicylic Acid Signaling Pathway in Kiwifruit.

Sulfur has been previously reported to modulate plant growth and exhibit significant anti-microbial activities. However, the mechanism underlying its diverse effects on plant pathogens has not been elucidated completely. The present study conducted the two-year field experiment of sulfur application to control kiwifruit canker from 2017 to 2018. For the first time, our study uncovered activation of plant disease resistance by salicylic acid after sulfur application in kiwifruit. The results indicated that when the sulfur concentration was 1.5-2.0 kg m-3, the induced effect of kiwifruit canker

Bacterial Leaf Infiltration Assay for Fine Characterization of Plant Defense Responses using the Arabidopsis thaliana-Pseudomonas syringae Pathosystem.

In the absence of specialized mobile immune cells, plants utilize their localized programmed cell death and Systemic Acquired Resistance to defend themselves against pathogen attack. The contribution of a specific Arabidopsis gene to the overall plant immune response can be specifically and quantitatively assessed by assaying the pathogen growth within the infected tissue. For over three decades, the hemibiotrophic bacterium Pseudomonas syringae pv. maculicola ES4326 (Psm ES4326) has been widely applied as the model pathogen to investigate the molecular mechanisms underlying the Arabidopsis im

Multiple lessons from the multiple functions of a regulator of type III secretion system assembly in the plant pathogen Pseudomonas syringae.

The assembly of type III secretion systems (T3SSs), which inject bacterial effector proteins into the cytosol of animal and plant hosts, is a highly regulated process. Animal pathogens use a length-control protein to produce T3SS needles of fixed length and then a second regulator, such as YopN in Yersinia spp, to mediate host contact-dependent effector delivery. For Pseudomonas syringae and other plant pathogens, regulation of the assembly process differs because the T3SS pilus must grow through variably thick plant cell walls before contacting the host plasma membrane. Characterization of a non-ribosomal peptide synthetase-associated diiron arylamine N-oxygenase from Pseudomonas syringae pv. phaseolicola.

The regiospecific oxidation of aromatic amines to aryl nitro compounds is critical to the synthesis of several natural products having pharmacological importance. The arylamine N-oxygenase (AAO) from Streptomyces thioluteus (AurF) selectively oxidizes p-aminobenzoic acid to p-nitrobenzoic acid and has been the subject of investigation for its unique chemistry and substrate preferences. Little, however, is known about the biochemistry and substrate specificities of AurF homologues, which are often associated with non-ribosomal peptide synthetases or polyketide synthases and have substrate bindi

A Biostimulant Based on Ecklonia maxima and Yeast Extract Increases the Resistance of Tomato Plants Toward Pseudomonas syringae pv. tomato DC3000.

Plant biostimulants represent a promising option to improve agricultural production and stress resistance while reducing the use of fertilizers and pesticides. Despite various evidence demonstrating the beneficial role of biostimulants in preventing the negative effects of abiotic stress on plants, the ability of biostimulants to bolster defense mechanisms has been brought to light only recently. In this work, the impact of a biostimulant based on Ecklonia maxima and yeast extracts (S/Y) on the response of tomato infected with Pseudomonas syringae pv. tomato DC3000 (Pst DC3000) was assessed. A transcription factor SlWRKY71 activated the H2S generating enzyme SlDCD1 enhancing the response to Pseudomonas syringae pv DC3000 in tomato leaves.

H2S is a well-known gaseous signaling molecule that plays important roles in plant response to biotic stresses. Pseudomonas syringae pv tomato (Pst) could cause enormous loss, while whether H2S could modulate plant defense against Pst is still unclear. By CRISPR/Cas9, the Sldcd1 gene editing mutant showed reduced endogenous H2S content and attenuated resistance, whereas treatment with exogenous H2S could enhance the resistance. A transcription factor, SlWRKY71, was screened and identified to promote the transcription of SlDCD1 via yeast one-hybrid, dual-luciferase reporter system, electrophore

Control of Bacterial Canker caused by Pseudomonas syringae pv. actinidiae (Psa) in kiwifruit plants by resinous exudates and extracts of Adesmia balsamica.

BACKGROUND: Pseudomonas syringae pv. actinidiae (Psa) is the pathogen responsible for kiwifruit's bacterial canker, which produces significant economic losses. Effective treatments and measures to control this disease have not yet been found; it is currently controlled with many products containing heavy metals (especially copper) or antibiotics (especially streptomycin), which can cause serious phytotoxicity and antibacterial resistance problems. Therefore, it is essential to develop new strategies to control this disease by obtaining a product of natural origin of sustainable use, which is b
