BACKGROUND: Armadillo repeat-containing X-linked 1 (ARMCX1) has been reported to exhibit a suppressive effect in a variety of solid tumors. However, neither its biological role nor its potential mechanism of action has yet been reported in nasopharyngeal carcinoma (NPC). METHODS: Immunohistochemical staining of an NPC tissue microarray was performed to evaluate the clinicopathologic association between ARMCX1 and NPC patients. The effect of ARMCX1 on the growth, migratory, and invasive capacities of NPC cells was assessed in vitro using colony formation, Cell Counting Kit-8 (CCK-8), 5-ethynyl-

The Vpx protein encoded by HIV-2/simian immunodeficiency virus (SIV) can antagonize the restriction of the host intrinsic restriction factor, SAMHD1, in nondividing cells by promoting its polyubiquitination and subsequent degradation, thereby facilitating viral replication and immune evasion. However, the role of deubiquitinating enzymes (DUBs) in the dynamics of virus and host remains poorly understood. Here, we demonstrate that DUB USP37 significantly reverses the Vpx-mediated degradation of SAMHD1 in various HIV-2/SIV subtypes by interacting with SAMHD1 and removing its ubiquitin chains. No

We previously identified the protein Lbh as necessary for cranial neural crest (CNC) cell migration in Xenopus through the use of morpholinos. However, Lbh is a maternally deposited protein and morpholinos achieve knockdowns through prevention of translation. In order to investigate the role of Lbh in earlier embryonic events, we employed the new technique "Trim-Away" to degrade this maternally deposited protein. Trim-Away utilizes the E3 ubiquitin ligase trim21 to degrade proteins targeted with an antibody and was developed in mammalian systems. Our results show that Xenopus is amenable to th

BACKGROUND AND PURPOSE: EZH2 (enhancer of zeste homologue 2) inhibitors are an emerging class of drugs that target epigenetic regulation. However, their efficacy in solid tumours has been limited, partly due to drug-induced upregulation of fatty acid synthesis. Combining lipid metabolic modulation with EZH2 inhibition may offer a promising strategy to enhance antitumor activity.
EXPERIMENTAL APPROACH: We conducted a screen of clinically approved lipid-lowering drugs to identify candidates that could enhance the efficacy of EZH2 inhibitors and found that fenofibrate significantly potentiated t

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by progressive motor neuron loss, with limited effective therapies. Docosahexaenoic acid (DHA) exhibits neuroprotective effects, but its limited transport across the blood-brain barrier (BBB) restricts clinical utility. Major facilitator superfamily domain-containing protein 2A (MFSD2A) is the primary transporter of DHA into the central nervous system, yet its role in ALS remains unclear. This study investigated the therapeutic potential and mechanisms of MFSD2A overexpression combined with DHA supplementat

Deprivation of nutrients in the tumor microenvironment drives malignant progression, yet the molecular mechanisms linking metabolic stress to metastasis in bladder cancer remain incompletely understood. Here, we report that nutrient-deprivation stress promotes metastasis by orchestrating a post-translational modification cascade centered on Beclin-1. Clinical analysis revealed that acetylation of Beclin-1 at lysine residues K430 and K437 was significantly reduced in muscle-invasive bladder cancer (MIBC) compared with non-muscle-invasive bladder cancer (NMIBC), a molecular signature inversely c

The genetic basis of most human disease cannot be explained by common variants. One solution to this 'missing heritability problem' may be rare missense variants, which are individually scarce but collectively abundant. However, the phenotypic impact of rare variants is under-appreciated as gene function is normally studied in the context of a single 'wild-type' sequence. Here, we explore the impact of naturally occurring missense variants in the human population on the cytosolic antibody receptor TRIM21, using volunteer cells with variant haplotypes, CRISPR gene editing and functional reconst

BACKGROUND: Intratumoral heterogeneity is the primary challenge in the treatment of glioblastoma (GBM). The presence of glioma stem cells (GSCs) and their conversion between different molecular phenotypes contribute to the complexity of heterogeneity, culminating in preferential resistance to radiotherapy. ARP2/3 (actin-related protein-2/3) complexes (ARPs) are associated with cancer migration, invasion and differentiation, while the implications of ARPs in the phenotype and resistance to radiotherapy of GSCs remain unclear.
METHODS: We screened the expression of ARPs in TCGA-GBM and CGGA-GBM

We investigate the accumulated microbial and autoantigen antibody repertoire in adult-onset dermatomyositis patients sero-positive for TIF1γ (TRIM33) autoantibodies. We use an untargeted high-throughput approach which combines immunoglobulin disease-specific epitope-enrichment and identification of microbial and human antigens. We observe antibodies recognizing a wider repertoire of microbial antigens in dermatomyositis. Antibodies recognizing viruses and Poxviridae family species are significantly enriched. The identified autoantibodies recognise a large portion of the human proteome, includi

Ro52 antigen has recently been identified as TRIM21 protein, but the clinical significance of anti-Ro52/TRIM21 antibodies remains controversial. The aim of this multicentric study was to investigate the significance of anti-Ro52 antibodies without anti-SSA/Ro60 antibodies in various connective diseases. Sera were selected by each laboratory using its own method (ELISA, immunodot or Luminex technology), and then performed with ANA Screen BioPlex™ reagent (BIO-RAD). Among the 247 screened sera, 155/247 (63%) were confirmed as anti-Ro52 positive and anti-SSA/Ro60 negative. These sera were analyze

BACKGROUND: Colorectal cancer (CRC) ranks among the most prevalent malignancies globally, and radiotherapy remains a critical treatment modality. However, its efficacy is frequently compromised by acquired radioresistance. The endoplasmic reticulum chaperone protein BIP plays a pivotal role in regulating radioresistance by coordinating the balance between protective autophagy and apoptosis, though the regulatory roles of circular RNAs (circRNAs) in this process remain poorly understood.
MATERIALS: Differentially expressed circRAB5A (hsa-circ-0123297) was identified from the GSE186940 dataset.

Acute pancreatitis (AP) involves acinar cell death and severe inflammation. Although the E3 ubiquitin ligase TRIM21 regulates inflammation, its role in the pathogenesis of AP remains undefined. This study aims to explore the role of TRIM21 in regulating inflammation during AP. In this study, TRIM21 levels show a severity-dependent increase in patients with AP, which is more than that in healthy controls. Consistently, increased TRIM21 expression is observed in the murine models of AP and exhibits spatial co-localization with macrophages. Macrophage-specific Trim21 ablation mitigates pancreatic

Tau protein truncated at Asp 421 is a characteristic feature of Alzheimer's disease and other tauopathies. Here, we show that a monoclonal antibody against Asp421, 5G2, cleared insoluble tau in the brains of JNPL3 mice, decreased tau levels in brain interstitial fluid in awake JNPL3 mice, improved in vivo neuronal function, and reduced microglial Iba-1 expression in PS19 mice, in which neuronal tau aggregation and dysfunction occurred earlier than microglial activation. For mechanistic insight using culture models, 5G2 prevented tau-mediated toxicity, cleared extra- and intracellular tau, and

INTRODUCTION: Breast cancer is the most common malignant tumor worldwide, and its high heterogeneity limits the efficacy of current clinical therapies. R-spondin 2 (RSPO2) expression is closely associated with breast cancer subtypes, but the precise mechanisms by which it drives disease progression, particularly its relationship with ferroptosis, remain unclear.
METHODS: RSPO2 expression was examined in primary breast cancer specimens and in MCF-7 and MDA-MB-231 cell lines. Functional assays, including proliferation, migration, and invasion tests, were performed following RSPO2 overexpression

Antibodies mediate the majority of their effects in the extracellular domain, or in intracellular compartments isolated from the cytosol. Under a growing list of circumstances, however, antibodies are found to gain access to the cytoplasm. Cytosolic immune complexes are bound by the atypical antibody receptor TRIM21, which mediates the rapid degradation of the immune complexes at the proteasome. These discoveries have informed the development of TRIM-Away, a technique to selectively deplete proteins using delivery of antibodies into cells. A range of related approaches that elicit selective pr

Trim-Away is an antibody-based method for the degradation of endogenous cellular proteins without prior genetic manipulation. From the effect that protein degradation has on the phenotype, one can infer the function of the protein-coding gene. Among the strengths of Trim-Away, the authors highlight the possibility to target embryonic proteins that have not yet been produced or are not yet functionally required at the time of antibody delivery. Conversely, a limitation of Trim-Away is the possibility that while the target protein is being degraded, de novo translation may replenish it, whereby

Cathepsin D (CTSD) is a lysosomal aspartic protease that plays vital roles in regulating the properties of solid tumors, including proliferation, apoptosis, migration, metastasis, and angiogenesis. However, the function of CTSD in haematological malignancies remains largely elusive. Here we show that CTSD is highly expressed in acute myeloid leukemia (AML) and that high CTSD expression is associated with unfavourable prognosis. Knockdown of CTSD in AML cells inhibits cell proliferation and anti-apoptotic activity. Mechanistically, CTSD decreased the expression of the E3 ubiquitin ligase TRIM21

INTRODUCTION: The fat mass and obesity-associated protein (FTO), a key RNA N6-methyladenosine (m6A) demethylase, has been highlighted for its important role in inflammatory response. Emerging evidences link the O-GlcNAcylation to numerous human diseases, particularly inflammation. However, the specific role and underlying mechanism of FTO O-GlcNAcylation in inflammation remain elusive.
METHODS: The FTO O-GlcNAcylation modification was determined by co-immunoprecipitation (Co-IP) assay, metabolic glycan labeling combined with click reaction, and liquid chromatography-tandem mass spectrometry (

Kinesin superfamily plays pivotal role in tumor progression. Our previous study identified KIF26B as a novel biomarker for bladder cancer diagnosis and prognosis. Herein, we illustrated another role of KIF26B in bladder cancer progression through stimulating the NF-κB signaling pathway. Mechanically, KIF26B activated the NF-κB signaling pathway by enabling nuclear localization of NF-κB subunit p65 without influencing its protein stability. Instead, KIF26B could recruit TRIM21 and add K63-linked ubiquitin to p65, which allowed its nuclear localization. Further, PKCε responded to TNFα and mediat
