Antibacterial and efflux pump inhibitors of thymol and carvacrol against food-borne pathogens.

In this study thymol (THY) and carvacrol (CAR), two monoterpenic phenol produced by various aromatic plants, was tested for their antibacterial and efflux pump inhibitors potencies against a panel of clinical and foodborne pathogenes. Our results demonstrated a substantial susceptibility of the tested bacteria toward THY and CAR. Especially, THY displayed a strong inhibitory activity (MIC's values ranged from 32 to 64 μg/mL) against the majority of the tested strains compared to CAR. Moreover, a significant reduction in MIC's of TET and benzalkonium chloride (QAC) were noticed when tested in c

Thyme extract, but not thymol, inhibits endothelin-induced contractions of isolated rat trachea.

Thyme extract is known to posses spasmolytic effects on isolated trachea and to increase ciliary activity. A small part of this effect is mediated via beta (2) receptors, but other receptors are supposed to be involved. Endothelin has a pathophysiological impact in asthma with respect to hyper-reagibility and contracting isolated trachea smooth muscles and was, therefore, investigated. Thyme extract inhibited endothelin-induced contraction. Emax decreased depending on the concentration of thyme extract indicating a non-competitive inhibition. Bosentan, an endothelin receptor antagonist (positi

Electrochemical quantification of thymol in Thymus vulgaris essential oil with a 3D-printed electrode.

Thymol (TM), a monoterpenoid phenol with diverse antioxidant and antimicrobial properties, is of significant interest to the pharmaceutical, food, and agricultural sectors. Due to its importance, this study developed an electrochemical method for its quantification using a 3D-printed electrode made from a conductive carbon black/PLA filament (CB/PLA electrode). Electrochemical measurements were performed using square-wave voltammetry (SWV) in 0.1 mol L⁻1 Na2SO4 (pH 13). The potential was scanned from 0 to +0.80 V, exploiting the oxidation process of TM at approximately +0.30 V (vs. Ag|AgCl|KCl

Thymol as a natural monoterpenoid antibacterial agent: a comprehensive review of applications and current advances.

Thymol, a monoterpenoid phenolic compound, exhibits broad-spectrum antibacterial activity and is gaining increasing attention as a natural alternative to synthetic agents. Thymol appears to demonstrate high antibacterial efficacy against Gram-negative and Gram-positive bacteria, particularly resistant pathogens, through disruption of bacterial cell membranes, protein and enzyme inhibition, influencing DNA synthesis, and triggering oxidative stress mechanisms. Given these reported mechanisms and its broad-spectrum activity, thymol has been the subject of increasing research interest in recent y

In vitro Testing of Trichophyton indotineae Susceptibility to Essential Oil Components Citral and Thymol.

Tinea corporis caused by Trichophyton indotineae infection is a worldwide epidemic and represents a growing challenge, given that many strains of this dermatophyte are resistant to the antifungal drugs (allylamine and azoles) typically used to treat dermatophytosis. Phytochemicals isolated from essential oils are effective alternatives for inhibiting microbial pathogens. In the present study, we performed in vitro susceptibility testing of the phytochemicals citral and thymol against T. indotineae strains, including a multi-drug-resistant isolate. The susceptibility of T. indotineae to citral,

Use of Thymol in Nosema ceranae Control and Health Improvement of Infected Honey Bees.

Nosema ceranae is the most widespread microsporidian species which infects the honey bees of Apis mellifera by causing the weakening of their colonies and a decline in their productive and reproductive capacities. The only registered product for its control is the antibiotic fumagillin; however, in the European Union, there is no formulation registered for use in beekeeping. Thymol (3-hydroxy-p-cymene) is a natural essential-oil ingredient derived from Thymus vulgaris, which has been used in Varroa control for decades. The aim of this study was to investigate the effect of thymol supplementati

Voltage-dependent block of neuronal and skeletal muscle sodium channels by thymol and menthol.

BACKGROUND AND OBJECTIVE: Thymol is a naturally occurring phenol derivative used in anaesthetic practice as a stabilizer and preservative of halothane, usually at a concentration of 0.01%. Although analgesic effects have long been described for thymol and its structural homologue menthol, a molecular basis for these effects is still lacking. We studied the blocking effects of thymol and menthol on voltage-activated sodium currents in vitro as possible molecular target sites. METHODS: Whole cell sodium inward currents via heterologously (HEK293 cells) expressed rat neuronal (rat type IIA) and

Computational insight into the antioxidant activity of thymol in diverse solvent environments: a DFT approach.

Natural phenolic compounds are a good source of antioxidant. Thymol, a naturally occurring substance containing a phenolic OH group and non-phenolic CHₓ (x = 1, 3) groups, are expectedly reported to possess antioxidant activity. In this study, its antioxidant potential was systematically studied using DFT along with the B3LYP hybrid functional and 6-311G ++ (d,p) basis set in gas and several variable solvent media. The major antioxidant mechanisms (HAT, SPLET, and SETPT) were calculated. Frontier molecular orbitals, energy gap, physico-chemical reactivity descriptors, Fukui functions, MEP surf

Potentials and Challenges in Development of Vesicular Phospholipid Gel as a Novel Dermal Vehicle for Thymol.

Background/Objectives: Thymol, one of the main compounds of thyme essential oil, has shown promising effects in treating various skin disorders owing to its anti-inflammatory, antimicrobial and antioxidative activities. Due to its limited solubility in water, thymol is commonly used in higher concentrations to achieve a suitable therapeutic effect, which can consequently lead to skin irritation. To overcome these limitations, we incorporated thymol into a vesicular phospholipid gel (VPG), a novel semisolid dermal vehicle consisting of highly concentrated dispersion of phospholipid vesicles (li

Development of amphipathic derivatives of thymol and carvacrol as potent broad-spectrum antibacterial agents.

In the current study, to discover novel antibacterial agents, we designed and synthesized 72 carvacrol and thymol derivatives by biomimicking the structure and function of cationic antimicrobial peptides (AMPs). Many of the derivatives showed good antibacterial activity, and compound thy2I exhibited the most potent antibacterial activity with minimum inhibitory concentration (MIC) values ranging from 0.5 μg/mL to 8 μg/mL. Compound thy2I could kill both gram-positive and gram-negative bacteria via a membrane-targeting mechanism of action with a low frequency of resistance. In addition, thy2I ha
