[Distribution of antibodies to DNA in the tissues of rats with sarcoma-45].

Distribution of a label was studied in nuclear fraction and in tissues of rats with transplanted sarcoma 45 within a day after an administration of immune and nonimmune 131I-gamma-globulins. When gamma-globulins, containing antibodies to one-stranded DNA, were administered the more intensive accumulation of the label was observed in nuclei of malignant tissue as compared with other tissues (liver, heart, thymus and muscles). The diffuse distribution of the label was found if nonimmune gamma-globulins were used. The phenomenon was apparently related to the active penetration of labelled antibod

Human proteome distribution atlas for tissue-specific plasma proteome dynamics.

The plasma proteome is maintained by the influx and efflux of proteins from surrounding organs and cells. To quantify the extent to which different organs and cells impact the plasma proteome in healthy and diseased conditions, we developed a mass-spectrometry-based proteomics strategy to infer the tissue origin of proteins detected in human plasma. We first constructed an extensive human proteome atlas from 18 vascularized organs and the 8 most abundant cell types in blood. The atlas was interfaced with previous RNA and protein atlases to objectively define proteome-wide protein-organ associa

Tissue distribution pharmacokinetics of intrathecal U1 adaptor oligonucleotide in mice.

U1 Adaptor is a novel gene-silencing technology, offering an innovative approach to target genes in the CNS for the treatment of diseases. Intrathecal delivery is a medically viable route of administration of CNS-bound nucleic acid drugs; therefore, it is important to investigate U1 Adaptor distribution after intrathecal drug delivery. We investigated the distribution patterns of U1 Adaptor upon intrathecal bolus administration in mice. It readily distributes to CNS tissues, including the lumbar and the cervical spinal cord, and the cerebellum. Over time, the U1 Adaptor also accumulates in the

Tissue distribution and pharmacokinetics of isoxanthohumol from hops in rodents.

Vegetables and fruits contain prenylflavonoids with biological functions that might improve human health. The prenylflavonoid isoxanthohumol (IXA) and its derivative, 8-prenylnaringenin (8-PN), have beneficial activities, including anti-cancer effects and suppression of insulin resistance. However, their pharmacokinetic profile is unclear. Previous studies suggested flavonoids have low systemic availability and are excreted via the feces. Therefore, this study investigated the tissue distribution dynamics of high-purity IXA (>90%) from hops administered orally, either singly (50 mg/kg body wei

Molecular cloning, tissue distribution and function analysis of duck TLR7.

Toll-like receptors (TLRs) play an important role in detecting pathogen-associated molecular patterns (PAMPs). Among the TLRs, TLR7 is involved in the recognition of antiviral compounds and single-stranded RNA. This study was designed to explore the structure and function of TLR7 in duck (Anas platyrhynchos), a natural host for avian influenza virus. Firstly, the full-length cDNA of Shaoxing egg-laying duck TLR7 (duTLR7) was obtained using reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE). It consisted of 38 base pairs (bp) 5'-untranslated reg

Molecular cloning, tissue distribution, and immune function of goose TLR7.

TLR7 is a transmembrane endosomal protein that plays an essential role in innate antiviral responses via the recognition of conserved viral molecular patterns. Here, we cloned the full-length cDNA of goose TLR7 and carried out a molecular characterization of goose TLR7. The goose TLR7 gene is 3900 bp and encodes a 1045 amino acid protein with high homology to poultry (93% to duck and 83% to chicken). Similar conclusions were made by phylogenetic analysis. The predicted protein secondary structure of goose TLR7 contained a conserved Toll/interleukin-1 receptor domain and characteristic leucine-

Tissue distribution and transmission routes for the tsetse fly endosymbionts.

The tsetse fly Glossina is the vector of the protozoan Trypanosoma brucei spp., which causes Human and Animal African Trypanosomiasis in sub-Saharan African countries. To supplement their unbalanced vertebrate bloodmeal diet, flies permanently harbor the obligate bacterium Wigglesworthia glossinidia, which resides in bacteriocytes in the midgut bacteriome organ as well as in milk gland organ. Tsetse flies also harbor the secondary facultative endosymbionts (S-symbiont) Sodalis glossinidius that infects various tissues and Wolbachia that infects germ cells. Tsetse flies display viviparous repro

Distribution of opiate alkaloids in brain tissue of experimental animals.

The present study examined regional distribution of opiate alkaloids from seized heroin in brain regions of experimental animals in order to select parts with the highest content of opiates. Their analysis should contribute to resolve causes of death due to heroin intake. The tests were performed at different time periods (5, 15, 45 and 120 min) after male and female Wistar rats were treated with seized heroin. Opiate alkaloids (codeine, morphine, acetylcodeine, 6-acetylmorphine and 3,6-diacetylmorphine) were quantitatively determined in brain regions known for their high concentration of µ-op

The spatial distribution of radiodense breast tissue: a longitudinal study.

INTRODUCTION: Mammographic breast density is one of the strongest known markers of susceptibility to breast cancer. To date research into density has relied on a single measure (for example, percent density (PD)) summarising the average level of density for the whole breast, with no consideration of how the radiodense tissue may be distributed. This study aims to investigate the spatial distribution of density within the breast using 493 mammographic images from a sample of 165 premenopausal women (~3 medio-lateral oblique views per woman). METHODS: Each breast image was divided into 48 regio

Tissue distribution and induction of human multidrug resistant protein 3.

The multidrug resistance protein (MRP) family consists of several members and, for some of these transporter proteins, distinct roles in multidrug resistance and normal tissue functions have been well established (MRP1 and MRP2) or are still under investigation (MRP3). MRP3 expression studies in human tissues have been largely restricted to the mRNA level. In this report we extended these studies and further explored MRP3 expression at the protein level. Western blot and immunohistochemistry with two MRP3-specific monoclonal antibodies, M(3)II-9 and M(3)II-21, showed MRP3 protein to be present

Tissue distribution of an inducible cystatin in isoproterenol-treated rats.

A well-characterized, monospecific rabbit antiserum directed to an isoproterenol-inducible type 2 salivary cystatin was used for immunocytochemical localization of this cystatin in rat salivary glands, as well as in other organs of normal and isoproterenol-treated rats. Immunocytochemical analysis revealed a moderate staining of secretory granules within the acinar cells of submandibular glands, which was more pronounced in tissues obtained from female rats. In addition, the inducible cystatin was readily detected within granular convoluted tubule cells and striated duct cells of submandibular

Tissue distribution and characterization of prorenin-converting enzyme in mouse.

Renin is produced from an inactive precursor, prorenin, through endoproteolytic cleavage at paired basic amino-acid residues. Using (35S)methionine-labeled prorenin, that was synthesized with Xenopus oocyte expression system, as a substrate, we have determined the tissue distribution and the nature of prorenin-converting activity in mouse. The highest activity was found in the submandibular gland of male ICR mouse. The activity of the enzyme seemed to be parallel to that of renin. This enzyme activity, with an optimal pH 8.0-8.5, was inhibited by leupeptin, antipain and benzamidine.

Tissue distribution and developmental expression of protein kinase C isozymes.

Protein kinase C is a ubiquitous enzyme found in a variety of mammalian tissues and is especially highly enriched in brain and lymphoid organs. Based on biochemical and immunological analyses, we have identified three types of protein kinase C isozyme (designated types I-III) from rat brain. Monospecific antibodies against each of the protein kinase C isozymes were prepared for the determination of tissue distribution, subcellular localization, and developmental changes of these enzymes. The various protein kinase C isozymes were found to be distinctively distributed in different tissues: the

Infrared Microimaging of Sucrose Distribution in Plant Vascular Tissues.

Sucrose is the primary transport sugar in plants, serving as an essential energy source and signaling molecule. Detection, visualization, and quantification of sucrose in various plant tissues are essential for understanding the metabolic and physiological processes that sustain plant life. Traditional metabolite-mapping techniques have struggled to visualize the quantitative distribution of sucrose at sufficient resolution to distinguish vascular bundles from surrounding tissues. Here, we present a Fourier-transform infrared (FTIR) imaging approach that can visualize sucrose in plant tissues

Tissue Distribution of [14C]-Lefamulin into the Urogenital Tract in Rats.

Lefamulin, a semisynthetic pleuromutilin antibiotic approved in the United States, Canada, and Europe for intravenous and oral treatment of community-acquired bacterial pneumonia, is highly active in vitro against bacterial pathogens that cause sexually transmitted infections (STIs), including multidrug-resistant strains of Neisseria gonorrhoeae, Chlamydia trachomatis, and Mycoplasma genitalium. This nonclinical study used quantitative whole-body autoradiography (QWBA) and qualitative tape-transfer microautoradiography (MARG) to investigate lefamulin distribution into urogenital tract tissues

Partitioning and Spatial Distribution of Drugs in Ocular Surface Tissues.

Ocular drug absorption after eye drop instillation has been widely studied, but partitioning phenomena and spatial drug distribution are poorly understood. We investigated partitioning of seven beta-blocking drugs in corneal epithelium, corneal stroma, including endothelium and conjunctiva, using isolated porcine tissues and cultured human corneal epithelial cells. The chosen beta-blocking drugs had a wide range (-1.76-0.79) of n-octanol/buffer solution distribution coefficients at pH 7.4 (Log D7.4). In addition, the ocular surface distribution of three beta-blocking drugs was determined by ma

Tissue distribution of tetrabromobisphenol A and cadmium in mixture inhalation exposure.

Tetrabromobisphenol A (TBBPA) and cadmium chloride (CdCl2) are the typical representative pollutants of brominated flame retardants and heavy metals found in the air of e-waste recycling workshops. However, their metabolic kinetics through mixture inhalation is unknown. In the present study, 8-week old Institute of Cancer Research (ICR) male mice were whole-body exposed to TBBPA and CdCl2 mixtures by inhalation. Tissue samples were collected for TBBPA and cadmium (Cd) analysis at 2, 4, 6, and 8 weeks during exposure and at 4 and 8 weeks after the completion of the 8-week exposure period. TBBPA
