Ovarian Tissue Collection for Fertility Preservation in Children: The Need for Standardised Surgical Practice Guidance.

BACKGROUND: Chemotherapy, pelvic radiotherapy (including total body irradiation) and novel compounds used to treat children and teenagers with benign or malignant diseases can lead to impaired fertility. For prepubertal female patients at high risk of treatment-related infertility, upfront storage of ovarian tissue is increasingly being recognised as standard of care. No surgical guidelines exist to ensure best practice technique. We reviewed current UK practice to assess surgical management. METHODS: A ten-item, anonymous multiple-choice survey was distributed to the lead surgeons in all pae

scRNA-seq assessment of the human lung, spleen, and esophagus tissue stability after cold preservation.

BACKGROUND: The Human Cell Atlas is a large international collaborative effort to map all cell types of the human body. Single-cell RNA sequencing can generate high-quality data for the delivery of such an atlas. However, delays between fresh sample collection and processing may lead to poor data and difficulties in experimental design. RESULTS: This study assesses the effect of cold storage on fresh healthy spleen, esophagus, and lung from ≥ 5 donors over 72 h. We collect 240,000 high-quality single-cell transcriptomes with detailed cell type annotations and whole genome sequences of donors,

Patient Reported Outcomes Following Lower Extremity Soft Tissue Sarcoma Resection with Microsurgical Preservation of Ambulation.

BACKGROUND: Lower extremity soft tissue sarcoma treatment has evolved from primarily amputation procedures toward limb salvage. This series assesses whether soft tissue sarcoma tissue defects, extensive enough to require microsurgical reconstruction, can reliably result in preservation of ambulation, as well as objectively evaluate functional outcomes utilizing a patient-reported validated scale. It will also look at whether immediate functional muscle reconstructions and tendon transfers can be successful at restoring ambulation, potentially expanding the indications for limb salvage procedur

In vitro preservation of morphofunctional integrity of domestic cat testicular tissue up to 22 days.

Domestic cat testicular tissue culture may support fertility preservation when mature spermatozoa are unavailable, but its evaluation requires suitable fixation and culture conditions. This study compared neutral buffered formalin, Bouin's fluid, modified Davidson's fluid, and form-acetic acid for the histological and immunohistochemical assessment of feline testicular tissue, and then evaluated juvenile testicular fragments cultured for 10 or 22 days in a rodent-derived basal medium alone or supplemented with retinol, taurine, or both. Fixation markedly affected tissue morphology and immunohi

Adipose Tissue Palmitoylation Cycling Mediates Insulin Resistance and Preservation of β-Cell Function in Mice.

UNLABELLED: Excess adiposity is associated with insulin resistance and pancreatic β-cell dysfunction. We show that palmitoylation cycling in white adipose tissue (WAT) balances insulin sensitivity and insulin secretion. WAT and brown adipose tissue (BAT) ablation of acyl-protein thioesterase 1 (APT1) in APT1-deficient (knockout) mice (APT1-AdipoKO) resulted in increased body weight and exacerbated insulin resistance when the mice were fed a high-fat diet (HFD). APT1 ablation limited to BAT decreased body weight without affecting insulin resistance or glucose metabolism in HFD-fed mice. Insulin

Comparative Evaluation of APAFIX and 10% Formalin for Human Histologic Tissue Preservation: A Pilot Study.

Formalin has long served as the standard fixative in histopathology; however, its limitations-such as antigen masking, nucleic acid degradation, and carcinogenicity-have prompted the search for safer alternatives. APAFIX, a formalin-free fixative, was developed to maintain diagnostic integrity while improving safety in routine tissue handling. This pilot study was conducted to compare the efficiency of the APAFIX solution with formalin in preserving human histologic tissue architecture and cellular morphology as assessed by routine Hematoxylin and Eosin (H and E) staining. Ten human biopsies f

Enhanced RNA Preservation in Mouse Brain Tissue: A Strategy Combining Cardiac Perfusion with Hypersaline Immersion.

One of the key research methods for low-density spatial transcriptomics involves collecting and sequencing microregions of interest from tissue sections. However, prolonged capillary microneedle sampling poses a risk of RNA degradation. To address this issue, we developed a preservation strategy consisting of: (1) mouse cardiac perfusion with phosphate-buffered saline (PBS) to reduce blood-borne RNase activity; (2) brain tissue immersion in a hypersaline solution prior to sectioning; and (3) microregion sampling under low-temperature conditions. Assessment of tissue section RNA integrity and d

Improvement and development of immersion fixatives for enhanced tissue preservation in the whole eyeball specimens.

The wall of eyeball limits the penetration of the common fixatives, raising inadequate fixation or degeneration of internal tissue and posing challenges for preparing thin sections of the whole eyeball. In addition, many formaldehyde-containing fixatives exhibit the strong cross-linking effect that can excessively mask antigenic epitopes, thereby hampering immunological analyses. This study tried to develop an available fixative that exerts high permeability to mouse whole eyeballs and applicability to immunostaining. Glyoxal is known to have milder cross-linking capacity compared to formaldeh

Tissue Preservation Using Socket-Shield Technique in Lower Molar Site: A Proof of Principle Report.

Background/Objectives: The socket-shield technique (SST) was developed to preserve the facial/buccal portion of a tooth root to prevent post-extraction ridge resorption. It has gained attention for use in anterior implant sites, but its application in posterior sites remains unexplored. The aim of this case report was to report a proof-of-principle case using SST in a lower molar site and evaluate its effectiveness in preserving tissues. Methods: A 34-year-old non-smoking patient with a non-restorable mandibular first molar (tooth #36) underwent immediate implant placement with the SST. The to

Differences in the mechanical properties of intestinal tissue based on preservation freezing duration and temperature.

In this study, tissue samples were stress tested to determine if freezing duration and temperature alter their mechanical properties. Tissue samples taken from the small intestine of pigs were assigned to 5 groups: fresh tissue, -28.9 °C for 7 days, -62.2 °C for 7 days, -28.9 °C for 30 days, and -62.2 °C for 30 days. Tissue was stored in PBS for the assigned temperature and duration until testing occurred with the exception of fresh tissue which was tested at sample collection. Before testing, samples were thawed in a room temperature bath, and the thickness was measured. Samples were then mou

Simultaneous Assessment of Soft and Hard Tissue Behaviors After Alveolar Ridge Preservation Using Bone Substitutes.

This study aimed to simultaneously assess hard and soft tissues alterations and their proportions after alveolar ridge preservation (ARP). Participants (n = 65) who were previously enrolled in a clinical trial investigating ARP healing were selected. The CBCT DICOM (Digital Imaging and Communications in Medicine) and the cast STL (stereolithographic) files of each subject were imported, segmented, and superimposed. A cross-section view of the superimposed image presented the outlines from each DICOM and STL file. The center of preserved ridge was selected in the superimposed image and used to

Subperiosteal connective tissue grafts for pocket reduction and preservation of gingival esthetics: a case report.

BACKGROUND: Clinical studies and recent histological evidence following mucogingival surgery for the treatment of gingival recession have documented that when closely adapted to a previously exposed root surface, connective tissue is capable of forming a new attachment. Despite these findings, no clinical tests have been conducted to examine the ability of connective tissue to reduce probing depth (PD) and increase clinical attachment levels (CAL) when it is implanted into periodontal osseous defects. The purpose of this paper is to report the clinical results on a patient following 2 subperio

Microscaled Cell Surface Proteomics for Cryo-preserved Cells and Tissue Samples.

Cell surface proteins (CSPs) regulate key cellular functions and represent valuable targets for diagnostics and therapeutics. Despite advances in proteomic workflows, CSP analysis from cryopreserved or low-input clinical samples remains limited by technical constraints, including reduced membrane integrity, inefficient labeling, and high background. To address these challenges, we optimized and benchmarked two complementary surface enrichment strategies compatible with low-input applications (fewer than 1 million cells) and real-world sample types, including fresh, viably cryopreserved, and di

Dual Th1 and tissue-repair Treg cells accumulate in skeletal muscle preserving tissue integrity during infection.

UNLABELLED: Chronic infections require mechanisms that limit tissue damage while preserving pathogen control, yet the contribution of regulatory T (Treg) cells to this balance remains unclear. In this study, we characterized Treg cell responses during a chronic parasitic infection using experimental Trypanosoma cruzi infection as a model of persistent low-level parasitism and chronic tissue inflammation. We found that, although Treg cell numbers decline in the spleen, they accumulate in parasite-affected tissues such as skeletal muscle, where they adopt a combined Th1-associated and tissue-rep
