Oxygen is indispensable for sustaining tissue and cell physiological functions, yet a hypoxic microenvironment often arises in tissue injury and regeneration, impairing graft scaffold performance and hindering repair processes. Thus, oxygen-releasing scaffolds have become a pivotal research frontier in tissue engineering. Via elaborate spatial constructions (e.g. electrospun fibers, hydrogels, microspheres) and diverse modification strategies, these scaffolds enable sustained, spatiotemporally controllable oxygen release at injury sites, alleviating local hypoxia and boosting tissue regenerati

Aging is a gradual process leading to the decline of physiological functions across cells, organs, tissues, systems, and the surrounding microenvironment, particularly affecting the musculoskeletal system. Bone aging often presents with osteoporosis and impaired osteogenic niche, thereby increasing fracture risk and decreasing regenerative capacity. Therefore, bone aging and osteoporotic bone defects have become a significant challenge in clinical practice. Tissue-engineered scaffolds are of significant importance in managing osteoporotic bone defects by providing mechanical support, facilitat

Tissue engineering provides a path forward for emerging personalized medicine therapies as well as the ability to bring about cures for diseases or chronic injuries. Traumatic spinal cord injuries (SCIs) are an example of a chronic injury in which no cure or complete functional recovery treatment has been developed. In part, this has been due to the complex and interconnected nature of the central nervous system (CNS), the cellular makeup, its extracellular matrix (ECM), and the injury site pathophysiology. One way to combat the complex nature of an SCI has been to create functional tissue-eng

We present a novel method for preparing bioactive and biomineralized calcium phosphate (mCP)-loaded biopolymer composite scaffolds with a porous structure. Two types of polymers were investigated as matrices: one natural, cellulose acetate (CA), and one synthetic, polycaprolactone (PCL). Biomineralized calcium phosphate particles were synthesized via wet chemical precipitation, followed by the addition of organic biominerals, such as magnesium gluconate and zinc gluconate, to enhance the bioactivity of the pure CP phase. We compared the morphological and chemical characteristics of the two typ

Owing to dysfunction of the uterus, millions of couples around the world suffer from infertility. Different from conventional treatments, tissue engineering provides a new and promising approach to deal with difficult problems such as human tissue or organ failure. Adopting scaffold-based tissue engineering, three-dimensional (3D) porous scaffolds in combination with stem cells and appropriate biomolecules may be constructed for uterine tissue regeneration. In this study, a hierarchical tissue engineering scaffold, which mimicked the uterine tissue structure and functions, was designed, and th

Human placenta is a source of extracellular matrix for tissue engineering. In this study, placenta powder (PP), made from decellularized human placenta, was physically incorporated into synthetic poly(ethylene glycol) (PEG)-based hydrogels via UV-initiated thiol-ene coupling (TEC). The PP-incorporated PEG hydrogels (MoDPEG+) showed tunable storage moduli ranging from 1080 ± 290 to 51,400 ± 200 Pa. The addition of PP (1, 4, or 8 wt %) within the PEG hydrogels increased the storage moduli, with the 8 wt % PP hydrogels showing the highest storage moduli. PP reduced the swelling ratios compared wi

Maxillofacial bone defects caused by congenital malformations, trauma, tumors, and inflammation can severely affect functions and aesthetics of maxillofacial region. Despite certain successful clinical applications of biomaterial scaffolds, ideal bone regeneration remains a challenge in maxillofacial region due to its irregular shape, complex structure, and unique biological functions. Scaffolds that address multiple needs of maxillofacial bone regeneration are under development to optimize bone regeneration capacity, costs, operational convenience. etc. In this review, we first highlight the

Tissue engineering solutions have been widely explored for enhanced healing of skin wounds. Diabetic foot ulcers (DFU) are particularly challenging wounds to heal for a variety of reasons, including aberrant ECM, dysregulation of vascularization, and persistent inflammation. Tissue engineering approaches, such as porous collagen-based scaffolds, have shown promise in replacing the current treatments of surgical debridement and topical treatments. Collagen-glycosaminoglycan scaffolds, which are FDA approved for diabetic foot ulcers, can benefit from further functionalization by incorporation of

Osteochondral tissue (OCT) related diseases, particularly osteoarthritis, number among the most prevalent in the adult population worldwide. However, no satisfactory clinical treatments have been developed to date to resolve this unmet medical issue. Osteochondral tissue engineering (OCTE) strategies involving the fabrication of OCT-mimicking scaffold structures capable of replacing damaged tissue and promoting its regeneration are currently under development. While the piezoelectric properties of the OCT have been extensively reported in different studies, they keep being neglected in the des

The osteochondral defect repair has been most extensively studied due to the rising demand for new therapies to diseases such as osteoarthritis. Tissue engineering has been proposed as a promising strategy to meet the demand of simultaneous regeneration of both cartilage and subchondral bone by constructing integrated gradient tissue-engineered osteochondral scaffold (IGTEOS). This review brought forward the main challenges of establishing a satisfactory IGTEOS from the perspectives of the complexity of physiology and microenvironment of osteochondral tissue, and the limitations of obtaining t

Efficient vascularization within a scaffold is an essential criterion for evaluating the success of volumetric bone formation. Various strategies using angiogenic growth factors and cell-based approaches to induce effective osteogenic and angiogenic activities have been investigated. In this study, we propose a new highly porous multiple-cell-laden collagen/hydroxyapatite scaffold fabricated using a whipped bioink. After in vitro culturing of cells in the porous scaffolds for an extended culture period, osteogenic and angiogenic activities were significantly enhanced owing to the well-develope

Chitosan and gelatin have been extensively used in tissue engineering for a wide range of different applications, such as wound healing or bone regeneration, due to their advantages: excellent biocompatibility (promoting cell adhesion and proliferation), low price and biodegradability. Nonetheless, their main drawback is that they have poor mechanical properties, consequently restricting their use in bone tissue engineering. In previous studies, both materials were cross-linked, with added calcium minerals, which led to an improvement in both mechanical and biological properties. Therefore, th

The scaffolds, which morphologically and physiologically mimic natural features of the bone, are of high demand for regenerative medicine. To address this challenge, we have developed innovative bioactive porous silicon- wollastonite substrates for bone tissue engineering. Additive manufacturing through selective laser melting approach has been exploited to fabricate scaffolds of different architecture. Unique material combining osteoinductivity, osteoconductivity and bioactive elements allows flexibility in design. As the porous structure is required for the ingrowth of the bone tissue, the C

Naturally bone is a hierarchical and highly integrative dynamic tissue that is continuously remodeled by osteoblasts and osteoclasts. Deformities in bone due to trauma and/or disease are highly prevalent and mostly need surgical intervention. However, the methods of surgical treatments are associated with donor site morbidity, infection and/or complete rejection. Bone tissue-engineering provides a platform for growth of new bone tissue by fabricating scaffolds along with cells, growth factors and other dynamic forces. The polymeric materials especially natural polymers in their nanofibrous for

Cartilage is an avascular, aneural, and alymphatic connective tissue with a limited capacity caused by low mitotic activity of its resident cells, chondrocytes. Natural repair of full thickness cartilage defects usually leads to the formation of fibrocartilage with lower function and mechanical force compared with the original hyaline cartilage and further deterioration can occur. Tissue engineering and regenerative medicine is a promising strategy to repair bone and articular cartilage defects and rehabilitate joint functions by focusing on the optimal combination of cells, material scaffolds
