The role of collagen cross-linker in dentin mineralization: a scoping review.

OBJECTIVES: Dentin remineralization is a complex process that involves the crystallization of exposed collagen to restore the mechanical properties of demineralized dentin. It plays an essential role in managing dental caries and erosive tooth wear. Collagen cross-linkers have emerged as a promising strategy to enhance the stability of exposed collagen while facilitating mineral deposition. This scoping review evaluated the role of collagen cross-linkers in dentin remineralization. DATA: This scoping review was conducted following the PRISMA-ScR guidelines. SOURCES: The databases Medline, Sc

Hypomineralisation or hypoplasia?

Enamel hypomineralisation is a qualitative defect, with reduced mineralisation resulting in discoloured enamel in a tooth of normal shape and size. Because the enamel is weaker, teeth can undergo post eruptive breakdown, resulting in missing enamel. Enamel hypoplasia is a quantitative defect of the enamel presenting as pits, grooves, missing enamel or smaller teeth. It can sometimes be difficult to differentiate between the two. In this review paper, we aim to explain the importance of differentiating between the two conditions, and how to manage patients presenting with enamel defects.

Dendritic polyglutamic acid-chelerythrine nanocomplex for the reversal of bacterial tooth decay.

Caries begin with an imbalance between demineralization and remineralization due to the continuous acid production by cariogenic bacteria. However, the development of dental materials that could prevent and treat caries via a simple and efficient mechanism has always been a challenge. To address this issue, therapeutic nanoparticles composed of a dendritic polyglutamic acid (DPGlu) and chelerythrine (CHE) complex (DPGlu@CHE) were developed via hydrophobic interactions and hydrogen bonds. DPGlu@CHE could be adsorbed onto the tooth surface, releasing CHE rapidly under acidic conditions to remove

Immunolocalization of gamma-carboxyglutamic acid-containing proteins in developing molar tooth germs of the rat.

The localization of dentin gamma-carboxyglutamic acid containing proteins (DGP's) was examined in rat molar tooth germs at various stages of development by immunostaining with the peroxidase-antiperoxidase method. Odontoblasts and their processes stained positive, whereas all other cells were negative. No staining was found in predentin. Dentin stained only after a mild demineralization. The results support the concept that DGP's are deposited by odontoblasts directly into the mineralizing dentin.

Attachment to and phagocytosis of mineral by alveolar bone osteoclasts.

The mechanisms by which the osteoclast attaches to and resorbs bone are not fully understood. Morphologic techniques have primarily been used to examine these mechanisms, but many studies have been based on decalcified material. In this study, the attachment of the osteoclast to alveolar bone and its relationship to the mineral component during the active resorption associated with tooth eruption in the rat was examined using transmission electron microscopy and techniques designed to minimize demineralization effects during processing. Large conglomerates of bone mineral were found within bot

Dental Caries in Children: An Update.

Dental caries, also known as cavities, are the most prevalent dental problem in children. The etiology is mostly multifactorial and a result of an imbalance between the constant mineralization and demineralization on the tooth surface. It is important to assess oral health risks, counsel caregivers, and encourage oral hygiene. Recent guidelines by the American Academy of Pediatrics (AAP) underscore the evolving role of the pediatrician in initiating early dental health interventions.

The use of hydroxyapatite toothpaste to prevent dental caries.

Dissolution of hydroxyapatite from the tooth structure at low pH can lead to the irreversible destruction of enamel and dentin, which if left untreated can result in pain and tooth loss. Hydroxyapatite toothpastes contain hydroxyapatite particles in micro- or nanocrystalline form that have been shown to deposit and restore demineralized enamel surfaces. As such, they are currently being explored as a fluoride-free anti-caries agent. This narrative review article aims to summarize the recent findings of the research investigating the remineralization potential of HAP toothpaste in vitro, in sit

Evaluation of Caries-Free Restorations Bonded with Various Adhesive Systems: In Vitro Study.

PURPOSE: Secondary caries originate from a leakage pathway where oral acids can penetrate faster and demineralize the tooth substrate deeper which can be visualized by dye penetration. The ability to prevent secondary caries by contemporary adhesive systems was evaluated in this study. Dye penetration distance through leakage and into the tooth substrate adjacent to Class V restorations after artificial caries exposure was compared. MATERIALS AND METHODS: Previously frozen extracted human molars were used to prepare the Class V cavities at the CEJ on axial surfaces. All cavities were restored

Differential induction of surface chemical compositional change on tooth structure by glass ionomer restorative materials.

Glass ionomer cement (GIC) is a restorative dental material capable of promoting mineral deposition on surrounding tooth substrates. However, it is unclear as to whether demineralization and remineralization due to an oral pH change of the tooth affect the dissolution pattern of tooth crown and root differently. It also remains to be elucidated how GIC alters superficial chemical compositions of tooth crown structurally known as enamel, in relation to the root. In this study, we investigated an effect of pH challenge on chemical compositional change of tooth crown and root, as well as the cont

Ion channels, channelopathies, and tooth formation.

The biological functions of ion channels in tooth development vary according to the nature of their gating, the species of ions passing through those gates, the number of gates, localization of channels, tissue expressing the channel, and interactions between cells and microenvironment. Ion channels feature unique and specific ion flux in ameloblasts, odontoblasts, and other tooth-specific cell lineages. Both enamel and dentin have active chemical systems orchestrating a variety of ion exchanges and demineralization and remineralization processes in a stage-dependent manner. An important role

[Changes in the microhardness of the enamel and dentin in pulp inflammation].

Clinical and experimental studies of the dental enamel and dentin microhardness were carried out. A comparative analysis has made it possible to detect parallels in the changes of dental tissue microhardness in humans and dogs. The studies have revealed a focal pattern of demineralization, with the foci located mainly in the neck of the tooth and the pericervical portion of the root, indicating a considerable reduction of the functional resistance of pulpless teeth.

APDDM mixed with i-PRF as a graft material for bone regeneration - A case report.

Demineralized dentine matrix (DDM) has both osteoconductive and osteoinductive properties, and has porous structure which helps in cell and blood vessel penetration and the release of various growth factors from the dentinal tubules. The first human dentine autograft case was done in 2002 in Japan for maxillary sinus lifting. In this clinical report, we use a hand-operated order made stainless steel apparatus to crush the tooth and prepare the DDM chair side. Chemical treatment of DDM particulate was done for demineralization and sterilisation purpose, and used immediately as a graft material

An engineered dual-functional peptide with high affinity to demineralized dentin enhanced remineralization efficacy in vitro and in vivo.

Dental caries continues to be a major global public health problem. Remineralization of demineralized dentin is regarded as one of the hotspots in the current study in the treatment of dental caries. However, traditional remineralization agents, which usually lack the ability to bind to demineralized dentin collagen, are easily removed by the fluids in the oral cavity, thus decreasing the remineralization efficacy. Non-collagenous proteins (NCPs) have significant effects on the biomineralization of dentin due to their dual high binding capacity to the collagen fibers and minerals. But NCPs are

Changes in heated and in laser-irradiated human tooth enamel and their probable effects on solubility.

Enamel of intact human teeth laser irradiated in vitro under certain conditions is known to have less subsurface demineralization than unirradiated enamel on exposure to acid; consequently, the potential use of laser irradiance to reduce caries is apparent. The laser-induced physical and/or chemical changes that cause this reduced subsurface demineralization are not known. A laser-irradiated tooth enamel surface will have a temperature gradient that decreases towards the dentin junction. Dependent on irradiant conditions, the temperature may range from greater than 1400 degrees C at the surfac

The treatment of deep dentine caries lesions.

UNLABELLED: This paper discusses the excavation of deep caries lesions, focusing on indirect pulp-capping and the stepwise excavation approach. The concept of a modified and less invasive stepwise excavation is presented, based on considerations of caries pathology. The aim of the first excavation is primarily to make a change within the cariogenic environment, and not to remove carious dentine close to the pulp because this risks an iatrogenic pulp exposure. Microbiological and clinical studies have shown that the number of bacteria decrease during stepwise excavation procedures, and that les

Histochemical localization of alkaline pyrophosphate- phosphohydrolase in tooth-forming cells of rat.

The activity of a pyrophosphate-splitting tissue factor in jaws, teeth and intestinal mucosa has been studied by means of histochemistry. Freeze-cut, unfixed sections of whole animals were incubated in a buffered medium (pH 8.6) containing inorganic pyrophosphate (PP) Pb2+, Mg2+, and Zn2+ at various concentrations. The effects of compound R 8231, heat, aldehyde fixation, and demineralization with EDTA were also investigated. In sections showing optimal staining, deposition of incubation products was found in the stratum intermedium and the subodontoblastic cells of the developing tooth, in the

Deciduous Teeth Hypomineralisation and Perinatal Risk Factors.

Hypomineralisation defects with demarcated opacities are also observed in the deciduous dentition and have been found to be a predictive factor for hypomineralisation defects in the permanent dentition. Deciduous molar hypomineralisation (DMH) represents a qualitative enamel defect primarily afflicting deciduous second molars, albeit its presence is not limited solely to deciduous second molars, as it can manifest in deciduous canines and first molars. Thus, the presence of demarcated hypomineralisation defects in deciduous teeth could be called as deciduous teeth hypomineralisation. Clinicall
