Toxocara canis: proteolytic enzymes secreted by the infective larvae in vitro.

Second-stage larvae of the dog nematode Toxocara canis are infective to man and cause the syndromes of visceral larva migrans and ocular toxocariasis. Larvae cultured in vitro secrete proteases which degrade components of a model of extracellular matrix and basement membranes. These enzymes have been characterized using a variety of techniques. Multiple enzyme activities were demonstrated by substrate gel electrophoresis, associated with proteins of molecular weights of 120 and 32 kDa. The enzyme activity was inhibited both in substrate gels and in a radiogelatin microplate assay by phenylmeth

The impact of single versus repeated exposure of mice to Toxocara canis.

Infection with T. canis can alter dramatically the brain and behavior of the host. Previous results suggest that if the mammalian host is exposed either simultaneously to lead, or has a history of prior exposure to that toxic substance, the magnitude of the behavioral reaction to T. canis may be modified or even reduced. The present data suggest that the magnitude of both the behavioral and tissue/immune reactions may be less if the organism has multiple, instead of a single exposure, to T. canis. Lead, and perhaps other environmental toxicants may alter neurotropic products of the parasite, t

Radiolabeling and autoradiographic tracing of Toxocara canis larvae in male mice.

Artificially hatched infective larvae of Toxocara canis were labeled with 75Se in Medium 199 (Gibco) containing 75Se-methionine. Male CD-1 mice were infected with radiolabeled larvae by intragastric intubation or by intraperitoneal injection. At intervals of 3-56 days mice were killed and the organs prepared for compressed organ autoradiography. Radioactivity of parasitic larvae showed an exponential decrease with time, reflecting catabolism of label with a biological half life of 26 days (effective half life of 21 days) making possible experiments lasting several months. Total body larva coun

Effects of irradiation on the biology of the infective larvae of Toxocara canis in the mouse.

Mice were infected with either 2,000 normal or irradiated embryonated eggs of Toxocara canis and the number of larvae in their livers, lungs, brains, and carcasses investigated at 5, 20, and 33 days of infection. Mortality of mice infected with normal eggs was 33% between day 4 and 8 postinfection but there was no mortality among mice infected with irradiated eggs. Irradiation with 60, 90, or 150 kr of X-rays inhibited the migration of larvae from the livers and lungs and their accumulation in brain and carcass in proportion to the irradiation dose. By day 33 of infection, the ratio of larvae

[The larva precipitation test with Ascaris suum and Toxocara canis larvae].

Investigations were made by means of a larvae precipitation test (LPT) on different laboratory animals (pigs and white mice, and pigs, rabbits, guinea pigs and white mice respectively) experimentally infected with Ascaris suum and Toxocara canis respectively for the serological evidence of specific A. suum and T. canis antibodies. Living A. suum larvae isolated from the lungs of experimentally infected white mice forming oral, excretion porus and cuticula precipitates proved to be a suitable antigen for the intravital evidence of prepatent A. suum infections in pigs and mice. By using living T

Effect of fenbendazole on Toxocara canis larvae in tissues of infected dogs.

The effect of fenbendazole therapy was studied in six dogs fed 10,000 embryonated Toxocara canis eggs. At 47 days after they were fed T canis, four dogs were given fenbendazole in two divided doses totaling 50 mg/kg of body weight each day for 14 days. Two infected dogs were not given fenbendazole. All dogs were necropsied at the end of treatment and the foci were counted in the lungs; their skeletal muscles were digested in 1% trypsin for the recovery of larvae. The T canis larvae were not recovered from the skeletal muscle of the four infected dogs treated with fenbendazole; 15 and 42 larvae

Species-specific and common epitopes on the secreted and surface antigens of Toxocara cati and Toxocara canis infective larvae.

It is widely accepted that the major cause of visceral larva migrans (VLM) in man is Toxocara canis infection. This has been largely based on the detection of antibodies to this species. We have compared the antigens of T. canis and Toxocara cati in order to establish whether assay for the former might be compromised by infection with the latter. Comparisons were made by radioiodination of the surface and excretory/secretory (ES) glycoproteins of the infective larvae of both species, immunoprecipitation with poly- and monoclonal reagents, and SDS-PAGE. The SDS-PAGE profiles of surface antigens

In vitro Evaluation of the Nematicidal Efficacy of Quercetin on Adult Toxocara canis.

PURPOSE: Toxocara canis is a globally distributed zoonotic parasite found in dogs' intestines, leading to various pathological damages, particularly to the intestinal flora. The larval stage causes human toxocariasis, especially in children, and may result in neurological disorders and blindness. Quercetin is a flavonoid with strong secondary metabolites and possesses medicinal advantages and antiparasitic qualities. METHODS: The assay involved  four groups, each of  10 young adult T. canis; Group I was incubated in concentrations of an ethanolic extract of quercetin, Group II in albendazole

Intestine proteomic and metabolomic alterations in dogs infected with Toxocara canis.

Toxocariasis is an important zoonotic parasitic disease. Toxocaris canis adults live and reproduce in the intestinal tract of dogs and other canine hosts, and the infectious eggs are continuously excreted in feces, which causes environmental contamination and has an important public health significance. In this study, TMT proteomic and untargeted metabolomic methods were used to explore the physiological and pathological effects on the intestinal tract of dogs which infected with T. canis, and a series of bioinformatics analyses were conducted to identify differentially expressed proteins (DEP

Cyclosporine A increases the intensity of Toxocara canis infection in swiss mice.

Toxocariasis is a zoonotic disease of worldwide distribution. The connection between parasitic diseases and conditions that depress the immune system, such as the use of immunosuppressive drugs, has been studied. The purpose of this study was to evaluate the effect of Cyclosporine A (CsA) on the intensity of infection, humoral response and gene transcription of interleukins IL-4, IL-10 and IL-12 in mice experimentally infected with Toxocara canis. To this end, mice were divided into two groups treated with CsA (G1: 10 mg/Kg and G2: 50 mg/kg), the G3 and G4 group received PBS. After the last ad

The visceral larva migrans caused by Toxocara canis: a case report.

Hepatic toxocarosis is caused by the dog´s roundworm, Toxocara canis. Responsible for an eosinophilic inflammatory syndrome causing liver damage that can be detected on ultrasound, computed tomography and sometimes magnetic resonance imaging. We report the case of a nine-year-old child, living in countryside, with a notion of cohabitation with canids. He presented a digestive symptomatology revealed by abdominal pain, with a hemeosinophilia in the hemogram. The etiological assessment of hyper eosinophilia objectified a positive Toxocara canisserology. The imaging assessment in search of digest

Immune response to chronic Toxocara canis infection in a mice model.

AIMS: The zoonotic nematode Toxocara canis causes larva migrans syndrome that induces an immune response characterized by the production of antibodies and eosinophilia. A Th2 polarization has been associated with the infection, but there are still details of the cellular and humoral immune response that need to be described. Thus, the aim of this study was to describe the systemic host immune response to T canis chronic infection in a mouse model. METHODS AND RESULTS: BALB/c mice were inoculated once with 500 T canis embryonated eggs, per os. After 49 days, the amounts of larval found in brai

Is routine disinfection efficient in preventing contamination with Toxocara canis eggs?

Toxocara canis (Werner, 1782) is a zoonotic nematode commonly parasitizing dogs worldwide with great public health importance as the aetiological agent of human toxocariasis. In this respect, the aim of this study was to evaluate the effect of six disinfectant products commonly used in kennels, veterinary clinics and as household cleaning products on the embryogenesis and viability of T. canis eggs. The composition of active ingredients in these commercial disinfectants was sodium hypochlorite (A); a mix of N-(3-aminopropyl)-N-dodecylpropan-1.3-diamine and didecyldimethylammonium chloride (B);

Detection of Toxocara canis DNA in tissues of experimentally infected mice.

The main etiological agent of toxocariasis is the helminth Toxocara canis. Several difficulties are found in the diagnosis of this disease, because of nonspecific clinical signs and possible cross-reactions that may occur in the available test, the indirect ELISA. Therefore, molecular diagnosis has been indicated as an alternative to conventional diagnosis. The purpose of this study was to evaluate the polymerase chain reaction (PCR) technique for the identification of T. canis in tissues of experimentally infected mice. To this end, nine mice were inoculated with 1500 embryonated eggs and wer
