Diversity for endoribonuclease nsp15-mediated regulation of alpha-coronavirus propagation and virulence.

Understanding the role of the endoribonuclease non-structural protein 15 (nsp15) (EndoU) in coronavirus (CoV) infection and pathogenesis is essential for vaccine target discovery. Whether the EndoU activity of CoV nsp15, as a virulence-related protein, has a diverse effect on viral virulence needs to be further explored. Here, we found that the transmissible gastroenteritis virus (TGEV) and feline infectious peritonitis virus (FIPV) nsp15 proteins antagonize SeV-induced interferon-β (IFN-β) production in human embryonic kidney 293 cells. Interestingly, compared with wild-type infection, infect

Preparation and characterization of polyclonal antibody against severe acute respiratory syndrome-associated coronavirus spike protein.

A truncated gene (designated S1) encoding the receptor-binding domain (RBD) in the spike (S) protein of severe acute respiratory syndrome-associated coronavirus (SARS-CoV) was amplified by PCR. The gene was cloned into prokaryotic expression vector pGEX-6P-1, resulting in a recombinant plasmid pGEX-SARS-S1. Subsequently, pGEX-SARS-S1 was transformed into host cells BL21(DE3)pLysS, and the expression of the S1 protein was induced by isopropyl β-D-thiogalactoside (IPTG). Polyclonal antibody against SARS-CoV S1 protein was generated in a rabbit immunized with the purified S1 protein. The reactivi

Comparison of vesicular stomatitis virus pseudotyped with the S proteins from a porcine and a human coronavirus.

The surface proteins S of severe acute respiratory syndrome coronavirus (SARS-CoV) and transmissible gastroenteritis virus (TGEV) were compared for their ability to mediate infection of viral pseudotypes based on vesicular stomatitis virus (VSV). The cell tropism of the respective pseudotypes corresponded to the tropism of the viruses from which the S protein was derived. Higher infectivity values were obtained with the SARS-CoV S protein than with the TGEV S protein. Differences were observed with respect to the importance of the cytoplasmic tail and the membrane anchor of the S proteins. In

Development of colloidal gold and carbon immunochromatographic strips for rapid detection of PDCoV.

Porcine deltacoronavirus (PDCoV) is an enteric coronavirus that causes diarrhea in suckling piglets, leading to serious economic losses in the swine industry. In this study, we report the development of competitive immunochromatographic strips for PDCoV detection using both colloidal gold and carbon nanoparticles, which also represents the novel reported application of carbon nanoparticles in a viral immunochromatographic assay. The established strips had visual detection limits of 1:128 (gold) and 1:64 (carbon) at 1 × 108 TCID50/mL. Both assays showed excellent specificity, exhibiting no cros

Generation of APN-chimeric gene-edited pigs by CRISPR/Cas9-mediated knock-in strategy.

The prevalence of porcine enteric coronaviruses (PECs), including transmissible gastroenteritis virus (TGEV), swine acute diarrhea syndrome coronavirus (SADS-CoV), porcine delta coronavirus (PDCoV), and porcine epidemic diarrhea virus (PEDV), poses a serious threat to animal and public health. Here, we aimed to further optimize the porcine aminopeptidase N (pAPN) gene editing strategy to explore the balance between individual antiviral properties and the biological functions of pAPN in pigs. Finally, APN-chimeric gene-edited pigs were produced through a CRISPR/Cas9-mediated knock-in strategy.

Establishment of a stable CHO cell line with high level expression of recombinant porcine IFN-β.

A CHO cell clone (CHO-PoIFN-β) with stable porcine IFN-β expression under control of CMV promoter was selected under G418 pressure. In a 25cm(2) cell culture flask (5 ml culture medium), the cumulative protein yield of recombinant PoIFN-β reached 2.3×10(6) IU/ml. This cells clone maintained stable expression for at least 20 generations even in the absence of G418 selection pressure. The expressed recombinant PoIFN-β could induce the expression of porcine Mx protein in PK15 cells, and activate the chicken Mx promoter-controlled luciferase reporter gene expression, confirming that the recombinan

Establishment of a blocking ELISA for assessing neutralizing antibody levels against porcine epidemic diarrhea virus.

A highly specific blocking ELISA was developed for serological diagnosis of porcine epidemic diarrhea (PED) using monoclonal antibodies (mAbs) against the S1 protein of PEDV. The S1 protein was expressed in HEK293F cells and used to immunize BALB/c mice. Through hybridoma screening, a high-affinity monoclonal antibody designated 3E3 was obtained. Indirect ELISA, Western blot, and immunofluorescence assays confirmed its significant neutralizing activity, with a neutralization titer of 1:2¹⁰. A blocking ELISA was subsequently established using HRP-labeled 3E3. The assay showed no cross-reactivit

Design and synthesis of APN and 3CLpro dual-target inhibitors based on STSBPT with anticoronavirus activity.

Coronaviruses (CoVs) have continuously posed a threat to human and animal health. However, existing antiviral drugs are still insufficient in overcoming the challenges caused by multiple strains of CoVs. And methods for developing multi-target drugs are limited in terms of exploring drug targets with similar functions or structures. In this study, four rounds of structural design and modification on salinomycin were performed for novel antiviral compounds. It was based on the strategy of similar topological structure binding properties of protein targets (STSBPT), resulting in the high-efficie

Receptor tyrosine kinase inhibitors block proliferation of TGEV mainly through p38 mitogen-activated protein kinase pathways.

Emerging coronaviruses (CoVs) primarily cause severe gastroenteric or respiratory diseases in humans and animals, and no approved therapeutics are currently available. Here, A9, a receptor tyrosine kinase inhibitor (RTKI) of the tyrphostin class, is identified as a robust inhibitor of transmissible gastroenteritis virus (TGEV) infection in cell-based assays. Moreover, A9 exhibited potent antiviral activity against the replication of various CoVs, including murine hepatitis virus (MHV), porcine epidemic diarrhea virus (PEDV) and feline infectious peritonitis virus (FIPV). We further performed a

Luteolin exhibits broad-spectrum antiviral activity against swine enteric coronaviruses by targeting 3CLpro.

Swine enteric coronaviruses (SECoVs), including porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (TGEV), and porcine deltacoronavirus (PDCoV), pose a major threat to global swine production, with current control measures remaining suboptimal. These infections are often characterized by mixed infections, highlighting the urgent need for anti-SECoV drugs. This study elucidates the potential of luteolin as a broad-spectrum antiviral agent targeting the 3 C like protease (3CLpro). Molecular docking and dynamics simulations indicated a stable and high-affinity interaction

TGEV infection activates pro‑inflammatory signaling via the YY1/HSP40/NF‑κB pathway in intestinal epithelial cells and organoids.

Transmissible gastroenteritis virus (TGEV) is a porcine enteropathogenic coronavirus that causes severe intestinal inflammation and diarrhea in pigs, leading to substantial economic losses to the swine industry worldwide. Inflammation is a major cause of death in pigs infected with TGEV. However, the role of host-mediated inflammatory responses in TGEV pathogenesis remains poorly understood. Here, we report for the first time that the heat shock protein HSP40 (DNAJB6) is highly expressed in the jejunum and ileum of Wuzhishan pigs during TGEV infection, and that its transcription is activated b

Etiological role of viruses in outbreaks of acute gastroenteritis in The Netherlands from 1994 through 2005.

Acute gastroenteritis is one of the most common diseases worldwide. In developed countries, viruses, particularly noroviruses, are recognized as the leading cause. In The Netherlands, the surveillance of gastroenteritis outbreaks with suspected viral etiologies (as determined by Kaplan criteria) was established by the National Institute for Public Health and the Environment in 1994. This paper presents an overview of viral gastroenteritis outbreaks reported from 1994 through 2005. A minimum epidemiological data set consisting of the associated setting(s), the probable transmission mode, the da

Establishment of a nanoparticle-assisted RT-PCR assay to distinguish field strains and attenuated strains of porcine epidemic diarrhea virus.

Porcine epidemic diarrhea virus (PEDV) can cause serious disease and even death in neonatal piglets, resulting in serious damage to the swine industry worldwide. Open reading frame 3 (ORF3) is the only accessory gene in the PEDV genome. Previous studies have indicated that PEDV vaccine strains have a partial deletion in ORF3. In this study, a nanoparticle-assisted polymerase chain reaction (nanoparticle-assisted RT-PCR) assay targeting the ORF3 of PEDV was developed to distinguish PEDV field strains from attenuated strains by using a specific pair of primers. The PCR products of field strains

Isolation and Characterization of a Porcine Transmissible Gastroenteritis Coronavirus in Northeast China.

Transmissible gastroenteritis virus (TGEV) is a coronavirus (CoV) that is a major pathogenity of viral enteritis and diarrhea in suckling piglets, causing high morbidity and mortality. In this study, a TGEV strain HQ2016 was isolated from northeast China and characterized its genome sequence and pathogenicity. The phylogenetic analysis indicated that the TGEV HQ2016 strain was more similar to the TGEV Purdue cluster than to the Miller cluster. Both recombination and phylogenetic analysis based on each structural and non-structural gene revealed no recombination event in the HQ2016 strain. Expe

Plant-Produced Vaccines for Protection from Human and Veterinary Coronaviruses.

Coronaviruses are responsible for numerous diseases causing substantial morbidity and mortality to both humans and animals. In particular, they have caused three significant outbreaks in humans including severe acute respiratory syndrome 1 (SARS-CoV-1), Middle East respiratory syndrome (MERS-CoV), and, most recently, the severe acute respiratory syndrome 2 (SARS-CoV-2) pandemic. Two relevant veterinary diseases are porcine transmissible gastroenteritis virus (TGEV) and porcine epidemic diarrhea virus (PEDV), which cause severe losses in the pork industry. While efficacious vaccines were develo

Molecular epidemiology of human enteric caliciviruses in The Netherlands.

Caliciviruses are among the most common causes of gastroenteritis in people of all age groups. These antigenetically and genetically diverse viruses have been grouped into two genera within the family Caliciviridae, designated Norwalk-like viruses (NLV) and Sapporo-like viruses (SLV). To gain more insight in their epidemiology, we have developed a tentative genotyping scheme, which was used to differentiate the viruses detected in a set of epidemiological studies. NLVs and SLVs were detected by generic RT-PCR in stool specimens from 5.1% and 2.4% of cases with acute gastroenteritis for which a
