Organometal-induced antinociception: a time- and dose-response comparison of triethyl and trimethyl lead and tin.

Recent reports have demonstrated that organolead and -tin compounds can alter behavioral reactivity to noxious stimuli. To further define the dose response and temporal characteristics of these neurobehavioral effects, male Fischer 344 rats were injected sc with either one-fourth, one-half, or three-fourths the acute LD50 of triethyl lead (TEL), triethyl tin (TET), trimethyl lead (TML), trimethyl tin (TMT), or distilled water and tested on a 57.5 degrees C hot plate 1, 7, 14, 21, and 28 days after dosing. All four organometals altered hot plate latencies, but the magnitude and time course of t

Adsorption and degradation processes of tributyltin and trimethyltin in landfill leachates treated with iron nanoparticles.

Biotic and abiotic degradation of toxic organotin compounds (OTCs) in landfill leachates is usually not complete. In this work adsorption and degradation processes of tributyltin (TBT) and trimethyltin (TMeT) in leachate sample treated with different iron nanoparticles (FeNPs): Fe(0) (nZVI), FeO and Fe3O4 were investigated to find conditions for their efficient removal. One sample aliquot was kept untreated (pH 8), while to the others (pH 8) FeNPs dispersed with tetramethyl ammonium hydroxide (TMAH) or by mixing were added and samples shaken under aerated conditions for 7 days. The same experi

Coordination polymers containing ferrocene backbone. Synthesis, structure and electrochemistry.

The reaction of 1,1'-ferrocenedicarboxylic acid (LH(2)) with bis(triphenyltin) oxide afforded a molecular heterobimetallic compound [(Ph(3)Sn)(2)L]. In the latter the two carboxylate units of [L](2-) are involved in an anisobidentate chelating coordination mode to two triphenyl tin units. The reaction of LH(2) with trimethyltin hydroxide or bis(tri-n-butyltin) oxide afforded 2D-coordination polymers [(Me(3)Sn)(2)L](n) and [(n-Bu(3)Sn)(2)L](n) which are formed as a result of anisobidentate bridging coordination action of the two carboxylate units of [L](2-). Attenuation of trimethyltin-evoked glutamate (GLU) efflux from rat cortical and hippocampal slices.

Trimethyltin (TMT) is a toxic alkyltin that produces neuronal necrosis in the CNS. TMT stimulates the efflux of the excitatory amino acid glutamate (GLU) from rat cortical slices. This release is concentration dependent, partially calcium dependent, but not inhibited by calcium channel blockers or a NMDA antagonist. In the present study the compounds furosemide, bumetanide, 4,4'-diisothiocy-anatostilbene-2,2'-disulfonic acid (DIDS), and DL-threo-beta-hydroxyaspartic acid (HAA) were tested for their ability to attenuate TMT-stimulated GLU efflux from rat cortical and hippocampal slices. Trimethyltin-induced apoptosis is associated with upregulation of inducible nitric oxide synthase and Bax in a hippocampal cell line.

Trimethyltin (TMT) produces selective neuronal degeneration in the central nervous system (CNS), in which the hippocampus is the most sensitive area. Since previous studies have been conducted in either non-neural cells or mixed primary cultures, an immortalized hippocampal neuronal cell line (HT-22 cell) was used to assess the mechanism and mode of death produced by TMT. The compound produced a time- and concentration-dependent apoptotic death that was caspase-mediated. Excessive generation of reactive oxygen species (ROS) and subsequent reduction of mitochondrial membrane potential (DeltaPsi

A system for assessing toxicity of chemicals by continuous monitoring of homecage behaviors.

A noninvasive system is described for continuous recording of behaviors in the home cages of rats. Commercially available mesh cages were used so as to conform with housing conditions in most toxicological studies. A minicomputer controlled environmental lighting and recorded eating, drinking, rearing, and horizontal activity. The system's sensitivity was comparable to more complex systems. Validity was demonstrated through manipulation of environmental lighting, food deprivation, and the effects of amphetamine, scopolamine, ethanol, methylscopolamine, triethyltin, and trimethyltin. Electron spin resonance measurements of the effect of ionophores on the transmembrane pH gradient of an acidophilic bacterium.

The delta pH in ionophore-treated cells of an acidophile has been determined by electron spin resonance spectroscopy. The values obtained were comparable to those obtained using the more conventional techniques involving radiolabeled probes. No binding of the spin-labeled probe was observed as determined by two independent control experiments and by the characteristics of the probe signal. These results led us to conclude that the delta pH measured in protonophore/ionophore-treated cells is a result of a Donnan potential, which may be a physical property of all intact bacterial cells at low pH

Pioglitazone protects against trimethyltin hippocampal injury by reducing pyroptosis, mitochondrial dysregulation and ER stress.

Hippocampus-specific neurotoxic trimethyltin (TMT) is routinely used to mimic a reliable murine phenotype of neurodegeneration as well as cognitive loss and is accordingly appropriate to analyze pathogenesis of the prevalent neurodegenerative disorders, i.e. Alzheimer's disease (AD), and to examine the effectiveness of novel therapeutics. Antidiabetic medication pioglitazone has exhibited neuroprotective effects with promising clinical indications for neurodegeneration-based illnesses. This study was accomplished for studying the neuroprotective effect of pioglitazone against TMT-initiated cog

ARMC10 (S43) phosphorylation mediates trimethyltin chloride-induced cognitive impairment through mitochondrial dynamics disruption.

Trimethyltin chloride (TMT) is a well-established environmental neurotoxin known to induce hippocampal damage and cognitive impairment. Abnormal mitochondrial dynamics are involved in TMT-induced neurotoxicity, but the underlying molecular mechanisms remain unclear. Here, our study revealed that TMT-induced mitochondrial fragmentation led to mitochondrial dysfunction, nerve cell death and cognitive deficit. We assessed the expression of mitochondrial dynamics‑related proteins (OPA1, MFN1/2, DRP1, and FIS1). Using phosphoproteomics, we further identified Ser43 (S43) as a phosphorylation site in

Early and late stage of neurodegeneration induced by trimethyltin in hippocampus and cortex of male Wistar rats.

BACKGROUND: Trimethyltin (TMT), a potent neurotoxicant, elicits neuronal death in the limbic system and causes damage particularly in the hippocampus. Current interest relates to the opportunity to use TMT as an experimental model of neurodegeneration in the study of Alzheimer-like diseases. OBJECTIVE: In light of recently found species-specific and strain-specific differences in TMT intoxication, the aim of this study was to characterise the model of TMT-induced neurodegeneration in the brain of Wistar rats during early (days 1-3) and late (days 22-24) stage of neuronal damage. RESULTS: Red

Differential activation of microglia and astrocytes following trimethyl tin-induced neurodegeneration.

We have investigated the response of astrocytes and microglia to trimethyl tin intoxication in the septum, hippocampus, olfactory bulb, and pyriform cortex of the rat. Microglia were studied qualitatively using lectin histochemistry, and astrocytes were examined both qualitatively with immunohistochemistry, and quantitatively using an immunoassay for glial fibrillary acidic protein. Our results show that activated microglia first appeared 2 days after trimethyl tin intoxication in the lateral septum and hippocampus. Four days after trimethyl tin intoxication, the same regions revealed a most i

Atomic modelling and systematic mutagenesis identify residues in multiple drug binding sites that are essential for drug resistance in the major Candida transporter Cdr1.

The ABC (ATP-Binding Cassette) transporter Cdr1 (Candida drug resistance 1) protein (Cdr1p) of Candida albicans, shows promiscuity towards the substrate it exports and plays a major role in antifungal resistance. It has two transmembrane domains (TMDs) comprising of six transmembrane helices (TMH) that envisage and confer the substrate specificity and two nucleotide binding domains (NBDs), interconnected by extracellular loops (ECLs) and intracellular loops (ICLs) Cdr1p. This study explores the diverse substrate specificity spectrum to get a deeper insight into the structural and functional fe

Alteration in glutathione homeostasis and oxidative stress during the sequelae of trimethyltin syndrome in rat brain.

Trimethyltin (TMT), a by-product of tin, is used in a wide variety of industrial and agricultural purposes which serves as a model neurotoxicant in hippocampal neurodegeneration, and this could, in turn, be exploited for various therapeutic compounds essential for hippocampal neurodegeneration. Therefore, the present investigation explores the sequential changes in behavior, oxidative burden, and apoptosis following TMT administration in rat hippocampus. Male SD rats weighing 250 g were given single dose of 8.5 mg/kg TMT (i.p.) that resulted in "TMT syndrome" which begins at the third post-TMT

Integrated network toxicology and multiomics analyses reveal the mechanisms underlying trimethyltin chloride-induced epilepsy and the ameliorative effects of astaxanthin via NFKB1 and IL6.

Trimethyltin chloride (TMT) is a ubiquitous neurotoxic organotin pollutant that induces epilepsy, while the natural marine product astaxanthin (AST) exerts potent antioxidant, anti-inflammatory and neuroprotective effects. However, the causal molecular mechanisms of TMT-induced epilepsy and the targeted intervention mechanism of AST remain unclear. In this work, we adopted a multidimensional analytical framework encompassing an National Health and Nutrition Examination Survey (NHANES) population cohort analysis, network toxicology, machine learning, single-cell RNA sequencing, Mendelian random

Interactions of trimethyl tin (TMT) with rat primary astrocyte cultures: altered uptake and efflux of rubidium, L-glutamate and D-aspartate.

Studies were undertaken to assess the effects of trimethyl tin (TMT) on metabolic functions in primary neonatal rat cultured astrocytes. Concentrations as low as 10(-5) M TMT significantly inhibited the initial rate (1 min) of uptake of 86RbCl, used as a tracer for K+. TMT also markedly inhibited the initial rate (1 min) of Na(+)-dependent uptake of L-[3H]glutamate and D-[3H]aspartate, and stimulated the release of intracellular 86Rb+, -[3H]glutamate and D-[3H]aspartate in a dose-dependent fashion. These observations support the hypothesis that the astrocyte plasma membrane is potentially an i

Neuropeptide Y and somatostatin participate differently in the seizure-generating mechanisms following trimethyltin-induced hippocampal damage.

Trimethyltin (TMT) is an organic metal known to induce neuronal degeneration in the hippocampus, and abnormal behavior characterized by seizures, increased aggression and memory deficits. We administered TMT to rats and studied the changes of neuropeptide Y (NPY) and somatostatin (SOM) in the hippocampus. Phenobarbital (PB) was administered as an anticonvulsant to assess the effect of seizures on neuropeptide expressions in both dorsal and ventral hippocampus. Histochemically, NPY-immunoreactivity increased 4 days after TMT treatment in the hilus of the hippocampus, then progressively decrease

Effects of in vivo treatment of rats with trimethyltin chloride on respiratory properties of rat liver mitochondria.

Liver mitochondria isolated from rats treated in vivo with trimethyltin chloride show stimulation of respiration using glutamate/malate as substrate, and a transient inhibition on rates of respiration using palmitoyl-L-carnitine as substrate. This phenomenon was observed with both ADP- and FCCP-stimulated respiration. In contrast, rates of respiration by liver mitochondria isolated from rats treated in vivo with trimethyltin chloride, following prior treatment with clofibrate, were inhibited when glutamate/malate was respiratory substrates. With palmitoyl-L-carnitine no effect of trimethyltin
