The human polyomavirus JC virus (JCV) is responsible for the CNS demyelination observed in cases of progressive multifocal leukoencephalopathy. The JCV regulatory region (promoter) is a hypervariable, noncoding, nucleotide sequence positioned between the early and late protein-coding regions in the viral genome. Selective binding of cellular transcription factors to this promoter region participates in the control of viral tropism. Hence, further study of these proteins might provide new insights into JCV tropism and associated pathogenesis. This review gives an overview of viral proteomics -

Poliovirus replication is limited to a few organs, including the brain and spinal cord. This restricted tropism may be a consequence of organ-specific differences in translation initiation by the poliovirus internal ribosome entry site (IRES). A C-to-U mutation at base 472 in the IRES of the Sabin type 3 poliovirus vaccine strain, known to attenuate neurovirulence, may further restrict tropism by eliminating viral replication in the CNS. To determine the relationship between IRES-mediated translation and poliovirus tropism, recombinant human adenoviruses were used to express bicistronic mRNAs

This study compares the findings of plain film X-ray and computed CT examination in the diagnosis of facet orientation and the presence of tropism. Twenty consecutive patients having lumbar disc disease with sciatica were studied utilizing plain X-ray as well as CT scanning. A chiropractic radiologist read the films to determine if facet facings were sagittally, semi-sagittally or coronally oriented on both CT and plain X-ray study. CT was accepted as the most accurate method to determine the true facet orientation, and plain X-ray interpretation of facet orientation was compared to the CT rea

One mechanism for expanding the cellular tropism of a virus is through the formation of phenotypically mixed particles or pseudotypes, a process commonly occurring during viral assembly in cells infected with two or more viruses. We report here that dual infection of cells with human immunodeficiency virus (HIV) and a murine amphotropic retrovirus leads to the production of HIV pseudotypes that have acquired the host range of the amphotropic retrovirus and are capable of infecting not only CD4- human cells but also mouse cells. The replication of the HIV pseudotypes in the various CD4- cells w

In recent years, high pathogenicity avian influenza viruses (HPAIVs) have spread among wild, captive, and domestic birds, as well as mammals. Beyond the resulting economic and ecological losses, spillover into mammals has raised concerns about a potential pandemic. Viral tropism refers to the spectrum of host species, organs, and cells susceptible and permissive to viral infection. It is a potent driver of infection dynamics and shedding patterns, which presents important variations both between and within hosts: in poultry, HPAIV leads to systemic endothelial infection in domestic chickens, w

The SARS-CoV-2 virus causes severe acute respiratory syndrome (COVID-19) and has rapidly created a global pandemic. Patients that survive may face a slow recovery with long lasting side effects that can afflict different organs. SARS-CoV-2 primarily infects epithelial airway cells that express the host entry receptor Angiotensin Converting Enzyme 2 (ACE2) which binds to spike protein trimers on the surface of SARS-CoV-2 virions. However, SARS-CoV-2 can spread to other tissues even though they are negative for ACE2. To gain insight into the molecular constituents that might influence SARS-CoV-2

Oncolytic adenovirus has been applied in cancer therapy because of several advantages such as cost-effective production, high transduction efficiency and low toxicity. Recent efforts have been focused on the modification of oncolytic adenovirus by encoding transgenes within the viral genome to efficiently and selectively replicate within cancer cells, destroy cancerous cells, induce tumor cell apoptosis, and stimulate the recruitment of immune cells to the tumor site. Nevertheless, there are still big challenges for translational research of oncolytic virotherapy in clinical cancer management.

Avian influenza viruses from migratory birds have managed to cross host species barriers and infected various hosts like human and swine. Epidemics and pandemics might occur when influenza viruses are adapted to humans, causing deaths and enormous economic loss. Receptor-binding specificity of the virus is one of the key factors for the transmission of influenza viruses across species. The determination of host tropism and understanding of molecular properties would help identify the mechanism why zoonotic influenza viruses can cross species barrier and infect humans. In this study, we have co

The cellular and molecular mechanisms underpinning the unusually high virulence of highly pathogenic avian influenza H5N1 viruses in mammalian species remains unknown. Here, we investigated if the cell tropism of H5N1 virus is a determinant of enhanced virulence in mammalian species. We engineered H5N1 viruses with restricted cell tropism through the exploitation of cell type-specific microRNA expression by incorporating microRNA target sites into the viral genome. Restriction of H5N1 replication in endothelial cells via miR-126 ameliorated disease symptoms, prevented systemic viral spread and

Ebola viruses and Marburg viruses include some of the most virulent and fatal pathogens known to humans. These viruses cause severe haemorrhagic fevers, with case fatality rates in the range 25-90%. The diagnosis of filovirus using formalin-fixed tissues from fatal cases poses a significant challenge. The most characteristic histopathological findings are seen in the liver; however, the findings overlap with many other viral and non-viral haemorrhagic diseases. The need to distinguish filovirus infections from other haemorrhagic fevers, particularly in areas with multiple endemic viral haemorr

The phylum Apicomplexa is home to a variety of parasites of significant medical and economic relevance, including the coccidian species Toxoplasma gondii, Neospora caninum and Eimeria tenella. In spite of their shared ancestry, the aforementioned coccidian species exhibit highly variable host and tissue tropisms; whilst T. gondii invades a broad spectrum of cell types and host organisms, E. tenella infection is restricted to the caecum in chicken. apicomplexans are obligatory intracellular parasites, and are uniquely adapted for host cell invasion via several conserved features. The process of

The accurate diagnosis of HIV infection demands that to consider a positive result, at least three assays with different antigenic base should be used, one of them, Western-Blot being mandatory for confirmation. Fourth generation ELISAs shorten the window phase to 13-15 days, as they now include p24 antigen detection. Proviral DNA or Viral RNA detection by molecular methods have proved useful for addressing complex situations in which serology was inconclusive. Viral load (HIV-RNA) is routinely used to follow-up HIV infected patients and is used for treatment initiation decisions. It is also u

The clinical picture of viral encephalitis is determined by the affinity and persistence of the virus to different brain regions. Therefore, the present study was aimed to investigate the neuropathological changes following Japanese encephalitis virus (JEV) infection in rat at different time points. Twelve days old Wistar rats were infected by intracerebral inoculation of JEV. Presence of JEV antigen was detected in thalamus, striatum, cortex and mid brain on 3, 6, 10 and 20 days post inoculation (d.p.i.). Histopathological changes were also studied in different brain regions at different time

Microglia, alveolar macrophages, and Langerhans cells are representatives of cells of macrophage lineage that are susceptible to infection with HIV-1 and they play important roles in the pathogenesis of AIDS dementia, lymphoid interstitial pneumonia, and systemic viral invasion from mucosal surfaces, respectively. In contrast, elimination of CD4+ T cells with resultant development of immunosuppression and AIDS is thought to be reflective of the exclusive tropism of the virus for CD4+ T cells. Examination of these concepts in macaques infected with molecularly cloned strains of SIVmac suggested

Human polyomavirus JC virus (JCV) is the causative agent of the demyelinating disorder progressive multifocal leukoencephalopathy (PML). In vivo the cellular tropism of JCV has been shown to be very narrow, and replication appears to be essentially restricted to oligodendrocytes. To investigate the detail cellular tropism of JCV, we employed transfection, microinjection and CAT assays using JCV permissive cells, and several non-permissive cell lines. IMR-32 (human neuroblastoma cells) was permissive for IMR-32 adapted JC virus. A431 (human epidermoid carcinoma) and COS-7 (a SV40 transformed Af

Hepatitis E virus (HEV) is a leading cause of acute viral hepatitis, yet the virus has been associated with extrahepatic complications, including neurological and renal disorders. These extrahepatic manifestations pose challenges for the diagnosis and treatment of HEV and are considered a reservoir for disease relapse. In addition to recent in vivo patient data, several animal and in vitro models have been established in the last few years, confirming the broad HEV tissue tropism. However, it remains unclear whether extrahepatic complications arise from viral replication within the tissue or f

The points of Chongmai (thoroughfare vessel) recorded in Huangdi Neijing (The Yellow Emperor 's Inner Classic) were shared with those of foot-shaoyin meridian in Zhenjiu Jiayi Jing (The Systematic Classic of Acupuncture and Moxibustion). Since the Song Dynasty, all of these points were gradually reassigned to the foot-shaoyin meridian, in the way of "the points of foot-shaoyin meridian, and the confluence points of Chongmai ". The paper presents the evolution of the meridian tropism of these points and analyzes the underlying causative factors. It is proposed that Chongmai was developed in the

Avian reoviruses (ARVs) within genetic cluster (GC) 2 represent a large, heterogenous group of currently circulating ARVs that have been isolated from clinical cases of tenosynovitis and malabsorption. In this study, the pathogenesis of an ARV GC 2 isolate was investigated via quantitative reverse transcriptase PCR (RT-qPCR), in situ hybridization (ISH), and histopathology, following oral or footpad inoculation. RT-qPCR detected ARV within the digital flexor tendon, heart, lung, liver, spleen, kidney, duodenum, cecum, cloacal bursa, and thymus. The highest viral RNA load was observed within th

Lumbar spine pathologies have been linked independently to both neutral zone (NZ) properties and facet joint anatomical characteristics; however, the effect of facet joint orientation (FO) and tropism (FT) on NZ properties remains unclear. The aim of the present study was to investigate how axial plane FO and FT relate to NZ range and stiffness in the human lumbar spine and porcine cervical spine. Seven human lumbar functional spine units (FSUs) and 94 porcine cervical FSUs were examined. FO and FT were measured, and in vitro mechanical testing was used to determine anterior-posterior (AP) and
