Bovine tuberculosis source attribution using decision tree analysis: breakdown investigations in Italy (2022-2023).

In this study, we present an overview of 348 Bovine Tuberculosis (bTB) breakdowns reported in Italy between January 2022 and December 2023, and declared resolved between January 2022 and February 2025. The main objective of this study is to investigate the most probable sources of these bTB breakdowns using decision tree analysis, and to compare the findings with conclusions drawn by official veterinarians. Most of the studied breakdowns (332; 95.4%) involved cattle herds only, 11 (3.1%) involved water buffalo herds only, and five (1.4%) involved multiple species. bTB was primarily detected in

Cost-effectiveness assessment of alternative surveillance measures for bovine tuberculosis using stochastic modeling simulation.

To respond to the increasing bovine Tuberculosis (bTB) infection rates observed in France since 2010, national authorities are considering alternative surveillance measures for bTB in Reinforced Screening Areas, defined as municipalities within 5 or 10 km range of a bTB outbreak. The objective of this study was to determine the cost-effectiveness, from the farmer expenses point of view, of three alternatives to the current mandatory surveillance scenario applied on French dairy, beef and small (<10 heads) farms located in Reinforced Screening Areas (RSA), using stochastic economic and epidemio

Dairy farmers have minimal knowledge of bovine tuberculosis: A cross-sectional study in Bhutan.

Bovine tuberculosis (bTB) is a chronic disease of cattle and the leading cause of zoonotic tuberculosis. In Bhutan, there is no dedicated program for bTB despite the country being situated in the bTB enzootic region, where a large section of population relies on cattle and raw milk and, dairy products are widely consumed. To contribute to the design of future education, surveillance, mitigation, and control programs, we investigated the knowledge, attitudes, and practices relevant to bTB among dairy farmers. We surveyed 264 farmers in Thimphu, Paro, and Haa Dzongkhags. These farmers contribute

Inferring bovine tuberculosis transmission between cattle and badgers via the environment and risk mapping.

Bovine tuberculosis (bTB), caused by Mycobacterium bovis, is one of the most challenging and persistent health issues in many countries worldwide. In several countries, bTB control is complicated due to the presence of wildlife reservoirs of infection, i.e. European badger (Meles meles) in Ireland and the UK, which can transmit infection to cattle. However, a quantitative understanding of the role of cattle and badgers in bTB transmission is elusive, especially where there is spatial variation in relative density between badgers and cattle. Moreover, as these two species have infrequent direct

A genome wide association scan of bovine tuberculosis susceptibility in Holstein-Friesian dairy cattle.

BACKGROUND: Bovine tuberculosis is a significant veterinary and financial problem in many parts of the world. Although many factors influence infection and progression of the disease, there is a host genetic component and dissection of this may enlighten on the wider biology of host response to tuberculosis. However, a binary phenotype of presence/absence of infection presents a noisy signal for genomewide association study. METHODOLOGY/PRINCIPAL FINDINGS: We calculated a composite phenotype of genetic merit for TB susceptibility based on disease incidence in daughters of elite sires used for

Field evaluation of the protective efficacy of Mycobacterium bovis BCG vaccine against bovine tuberculosis.

The protective efficacy of Mycobacterium bovis BCG (1 x 10(6) single dose) was evaluated under field conditions. A total of 140 male Holstein Friesian calves, one to two week-old were selected. Two groups of 70 each were formed, one group was vaccinated and the other was injected with a placebo during their second week of age and followed until 12 months of age. The study considered a positive case of tuberculosis to be an animal that had a positive reaction to the three following tests in a row: tuberculin, IFNgamma PPD-B and IFNgamma ESAT6-CFP10 during the 12 months of exposure. The results

Experimental Fasciola hepatica infection alters responses to tests used for diagnosis of bovine tuberculosis.

Fasciola hepatica is a prevalent helminth parasite of livestock. Infection results in polarization of the host's immune response and generation of type 2 helper (Th2) immune responses, which are known to be inhibitory to Th1 responses. Bovine tuberculosis (BTB) is a bacterial disease of economic and zoonotic importance. Control polices for this disease rely on extensive annual testing and a test-and-slaughter policy. The correct diagnosis of BTB relies on cell-mediated immune responses. We established a model of coinfection of F. hepatica and Mycobacterium bovis BCG to examine the impact of he

Bovine tuberculosis history as a risk factor for recurrence in English cattle herds experiencing a breakdown.

BACKGROUND: Bovine tuberculosis (TB) threatens cattle farming in England. Supplementing tuberculin skin testing with the interferon-gamma blood test during TB incident (breakdown) management can enhance diagnostic sensitivity. We aimed to identify herds at greater risk of recurrence depending on their recent TB history to inform targeted interferon-gamma test deployment. METHODS: Using observational data, we analysed TB breakdowns in herds with a lesion-positive or bacteriologically positive (officially tuberculosis free-withdrawn [OTF-W]) breakdown in the High-Risk Area or six-monthly survei

A restatement of the natural science evidence base relevant to the control of bovine tuberculosis in Great Britain.

Bovine tuberculosis (bTB) is a very important disease of cattle in Great Britain, where it has been increasing in incidence and geographical distribution. In addition to cattle, it infects other species of domestic and wild animals, in particular the European badger (Meles meles). Policy to control bTB is vigorously debated and contentious because of its implications for the livestock industry and because some policy options involve culling badgers, the most important wildlife reservoir. This paper describes a project to provide a succinct summary of the natural science evidence base relevant

Improved skin test for differential diagnosis of bovine tuberculosis by the addition of Rv3020c-derived peptides.

A peptide cocktail derived from the mycobacterial antigens ESAT-6, CFP-10, and Rv3615c allowed differentiation between Mycobacterium bovis-infected and M. bovis bacillus Calmette-Guérin (BCG)-vaccinated cattle when used as a skin test reagent for a "DIVA" test (i.e., a test capable of differentiating infected and uninfected vaccinated animals). Addition of the antigen Rv3020c improves the diagnostic sensitivity without compromising specificity in the face of BCG or Johne's disease vaccination.

Global prevalence and risk factors of bovine tuberculosis in cattle: a systematic review and meta-analysis.

INTRODUCTION: Bovine tuberculosis (bTB), caused by Mycobacterium bovis, is a chronic zoonotic disease of major global importance, particularly in low- and middle-income countries where surveillance and control remain inadequate. OBJECTIVES: This systematic review and meta-analysis aimed to estimate the global prevalence of bTB, identify key risk factors, and evaluate diagnostic approaches to support evidence-based control strategies within a One Health framework. METHODS: A systematic search following PRISMA 2020 guidelines was conducted across eight electronic databases (PubMed, Web of Scie

Risk Factors Influencing the Specificity of Skin Tests for Bovine Tuberculosis in a Low-Prevalence Region.

BACKGROUND: Skin tests are the main official diagnostic method for granting, maintaining, and regaining the officially tuberculosis free (OTF) herd status in the European Union. Although highly specific, repeated testing may lead to the culling of animals not infected with bovine tuberculosis (bTB) and the suspension of OTF status, a problem especially relevant in low-prevalence regions where most herds are OTF. Identifying risk factors that affect the specificity of the single (SIT) and comparative (CIT) intradermal tuberculin tests is therefore essential to optimize control measures. MATERI

Strategic use of enzyme-linked immunosorbent assay as a complementary tool for the diagnosis of bovine tuberculosis.

Bovine tuberculosis (bTB) is a global chronic disease. Argentina's bTB eradication program consists of the Caudal-Fold Tuberculin test (CFT) and the sacrifice of positive animals as the principal measure. However, despite the implementation of the control program, CFT-positive animals continue to appear in some herds, which generates skepticism about the potential to eradicate tuberculosis. The objective of this work was to evaluate the suitability of an ELISA test for the detection of M. bovis specific antibodies (bTB-ELISA) in CFT-negative animals as a complementary strategy to control bTB.

A systematic review and meta-analysis of bovine tuberculosis occurrence and burden in Bangladesh, 1970-2023.

We aimed to estimate the overall apparent prevalence, true prevalence, and the spatial, temporal, and test-specific burden of bovine tuberculosis in Bangladesh. PubMed, Web of Science, Scopus, Google Scholar, and BanglaJOL were searched for bovine tuberculosis publications in Bangladesh from 1 January 1970 to 23 June 2023. Of 142 articles screened, systematic review and meta-analysis were performed on 22 (15.5%) articles. The apparent estimated bovine tuberculosis prevalence was 7%. The apparent Bayesian pooled mean bovine tuberculosis prevalences based on caudal fold test and single intraderm

Efficient modelling of infectious diseases in wildlife: A case study of bovine tuberculosis in wild badgers.

Bovine tuberculosis (bTB) has significant socio-economic and welfare impacts on the cattle industry in parts of the world. In the United Kingdom and Ireland, disease control is complicated by the presence of infection in wildlife, principally the European badger. Control strategies tend to be applied to whole populations, but better identification of key sources of transmission, whether individuals or groups, could help inform more efficient approaches. Mechanistic transmission models can be used to better understand key epidemiological drivers of disease spread and identify high-risk individu

Bovine brucellosis and tuberculosis at a livestock-wildlife interface in Zimbabwe: A nexus for amplification of a zoonosis or a myth?

BACKGROUND: Bovine brucellosis and tuberculosis are important zoonoses affecting both livestock and wildlife. OBJECTIVES: The study aimed to investigate seroprevalence of brucellosis and to isolate Brucella spp. and Mycobacterium bovis in cattle from livestock-wildlife interface areas. METHODS: Three sites were selected from high, medium and low interface. The high interface was adjacent to the park and separated by a broken fence, while the medium and low interface were 15-20 and 50 km from the perimeter fence, respectively. Cattle aged ≥2 years were randomly selected and bled for serology.

Deletion in KARLN intron 5 and predictive relationship with bovine tuberculosis and brucellosis infection phenotype.

Kalirin (gene: KALRN) is a Rho-GEF kinase linked to neurodegenerative diseases in humans. Unexpectedly, various polymorphisms in KALRN gene were previously associated with resistance to bacterial infections in ruminants. In this study, we evaluated the effect of the rs384223075 (RS-075) deletion in KALRN intron 5 on the occurrence of Mycobacterium bovis and Brucella abortus infections in cattle. We performed two separate case-control association analyses: one for bovine tuberculosis (bTB) using 308 Holstein and Jersey cows from three herds with prevalence between 5 and 15% for this infection;

The immunoregulatory effects of co-infection with Fasciola hepatica: From bovine tuberculosis to Johne's disease.

Fasciola hepatica (liver fluke) is a parasite prevalent in much of the world that causes the economically-important disease of fasciolosis in livestock. The threat that this disease poses extends beyond its direct effects due to the parasite's immunomodulatory effects. Research at this laboratory is focusing on whether this immunoregulation can, in animals infected with liver fluke, exert a bystander effect on concurrent infections in the host. It has already been established that F. hepatica infection reduces cell mediated immune responses to Mycobacterium bovis in cattle, and that the intera

The Case for Live Attenuated Vaccines against the Neglected Zoonotic Diseases Brucellosis and Bovine Tuberculosis.

Vaccination of humans and animals with live attenuated organisms has proven to be an effective means of combatting some important infectious diseases. In fact, the 20th century witnessed tremendous improvements in human and animal health worldwide as a consequence of large-scale vaccination programs with live attenuated vaccines (LAVs). Here, we use the neglected zoonotic diseases brucellosis and bovine tuberculosis (BTb) caused by Brucella spp. and Mycobacterium bovis (M. bovis), respectively, as comparative models to outline the merits of LAV platforms with emphasis on molecular strategies t

Development and evaluation of an enzyme-linked immunosorbent assay for use in the detection of bovine tuberculosis in cattle.

As a consequence of continued spillover of Mycobacterium bovis into cattle from wildlife reservoirs and increased globalization of cattle trade with associated transmission risks, new approaches such as vaccination and novel testing algorithms are seriously being considered by regulatory agencies for the control of bovine tuberculosis. Serologic tests offer opportunities for identification of M. bovis-infected animals not afforded by current diagnostic techniques. The present study describes assay development and field assessment of a new commercial enzyme-linked immunosorbent assay (ELISA) th
