Obesity and employment: Causal evidence from Genetic Instruments in the UK Biobank.

This study explores the causal impact of obesity on employment using data from the UK Biobank, a comprehensive dataset combining genetic, health, and socioeconomic information. We apply a Mendelian Randomisation (MR) approach, using a genetic risk score for a higher body mass index (BMI) as an instrumental variable. This method leverages genetic predispositions determined at conception, enabling us to isolate the exogenous variation in obesity while addressing key endogeneity concerns, such as omitted variable bias and reverse causation. To account for the heterogeneity in obesity's impact acr

Shared Neurocardiac Pathways Linking Atrial Fibrillation and Depression: A UK Biobank Analysis.

BACKGROUND: Atrial fibrillation (AF) and major depressive disorder (MDD) frequently co-occur and are each associated with adverse cardiovascular outcomes, yet the biological pathways linking these conditions remain poorly defined. Using the UK Biobank, we evaluated shared neurocardiac, inflammatory, and cardiovascular correlates underlying the AF-MDD association. OBJECTIVES: To assess bidirectional associations between AF and MDD and determine whether shared inflammatory, cardiovascular, autonomic, and neuroimaging correlates characterize their comorbidity. METHODS: We analyzed individuals w

Insomnia, proteomic characteristics, and mortality risk in depression: a UK Biobank study.

BACKGROUND: Although insomnia is associated with increased mortality in patients with depression, the underlying biological pathways remain unclear. As functional gene products, proteins are well suited to bridge behavioural risk factors and clinical outcomes. OBJECTIVE: To explore the associations between insomnia, proteomic characteristics, and mortality risk in depressed individuals. METHODS: We included 4,498 patients with depression from the UK Biobank. Insomnia was self-reported; all-cause mortality was the primary outcome. Cox models, LASSO regression, single- and multi-mediator joint

Blood Pressure Phenotypes and Cardio-Kidney Outcomes: Findings From UK Biobank Data.

BACKGROUND: Hypertension is a leading cause of cardio-kidney outcomes (CKO). However, distinct blood pressure (BP) phenotypes and their effect on CKO remain underexplored. This study aimed to evaluate the independent contributions of BP phenotypes to CKO. METHODS: This prospective, population-based study included UK Biobank participants enrolled between 2006 and 2010 with baseline systolic BP ≥90 mm Hg and estimated glomerular filtration rate ≥60 mL/min per 1.73 m2, excluding those with prior chronic kidney disease and cardiovascular disease. Five BP phenotypes were constructed based on the 2

Factors associated with drug-drug interactions involving citalopram in the UK Biobank.

BACKGROUND: Adults with mood and/or anxiety disorders have increased risks of comorbidities, chronic treatments and polypharmacy, increasing the risk of drug-drug interactions (DDIs) with antidepressants. AIMS: To use primary care records from the UK Biobank to assess DDIs with citalopram, the most widely prescribed antidepressant in UK primary care. METHOD: We classified drugs with pharmacokinetic or pharmacodynamic DDIs with citalopram, then identified prescription windows for these drugs that overlapped with citalopram prescriptions in UK Biobank participants with primary care records. We

Resting-state network analysis of suicide attempt history in the UK Biobank.

BACKGROUND: Prior research has identified altered brain structure and function in individuals at risk for self-directed violence thoughts and behaviors. However, these studies have largely utilized healthy controls and findings have been inconsistent. Thus, this study examined differences in resting-state functional network connectivity among individuals with lifetime suicide attempt(s) v. lifetime self-directed violence thoughts alone. METHODS: Using data from the UK Biobank, this study utilized a series of linear regressions to compare individuals with lifetime suicide attempt(s) (n = 566)

Genetically Predicted Blood Pressure and Cognition in Midlife: A UK Biobank Study.

BACKGROUND: This UK Biobank study uses a mendelian randomization approach to mitigate the variability and confounding that has affected previous analyses of the relationship between measured blood pressure (BP) and cognition and thus delineate the true association between the two. METHODS: Systolic BP (SBP) and diastolic BP polygenic risk scores (PRSs) were calculated using summary statistics from the International Consortium of Blood Pressure-Genome Wide Association Study (n=299 024). Adjusted nonlinear mixed-effects regression models were used, including a natural splines term for BP-PRS wi

Cumulative life course adversity, mental health, and cognition in the UK biobank.

The association between adversity and cognition varies according to the specific adversity, when the adversity was experienced, and the cognitive domains investigated. Disentangling the effect of adversity and the underlying mechanistic pathway is therefore difficult. The association between adversity (i.e., maltreatment) accumulated over the life course and cognitive flexibility, as well as two potential mediators (i.e., intra-individual variability in reaction time and depression) of this association, were investigated. Data stem from the baseline population of the UK Biobank study (N = 73,4

Multi-omics integration predicts the incidence of 17 diseases in the UK Biobank.

Multi-omics technologies, such as metabolomics and proteomics, offer deep molecular perspectives that could enhance risk prediction, but large-scale studies integrating both are scarce. Here we show the predictive values of these two omics across 17 incident diseases in 23,776 UK Biobank participants with complete baseline for 159 NMR-based metabolites and 2,923 Olink affinity-based proteins. We found that adding omics data significantly improved risk prediction for all 17 diseases compared to clinical predictors alone. Proteomics-only models generally outperformed metabolomics-only models for
