Protoporphyrin IX and verteporfin potently inhibit SARS-CoV-2 infection in vitro and in a mouse model expressing human ACE2.

The SARS-CoV-2 infection is spreading rapidly worldwide. Efficacious antiviral therapeutics against SARS-CoV-2 is urgently needed. Here, we discovered that protoporphyrin IX (PpIX) and verteporfin, two Food and Drug Administration (FDA)-approved drugs, completely inhibited the cytopathic effect produced by SARS-CoV-2 infection at 1.25 μmol/L and 0.31 μmol/L, respectively, and their EC50 values of reduction of viral RNA were at nanomolar concentrations. The selectivity indices of PpIX and verteporfin were 952.74 and 368.93, respectively, suggesting a broad margin of safety. Importantly, PpIX an

The clinical ionizable lipid SM-102 enhances photochemical immunogenic cell death by verteporfin-conjugated liposomes and improves survival in vivo.

Light activation of photosensitizers, including verteporfin-conjugates entrapped in liposomes, can induce photochemical immunogenic cell death (ICD) in cancer cells by exposing damage-associated molecular patterns (DAMPs) under specific conditions. In this study, we investigated how incorporation of the clinical ionizable lipid SM-102 into light-activated liposomes containing the verteporfin-conjugate 20:0 BPD-PC effects key photochemical and photobiological attributes we previously identified to be strongly associated with photochemical ICD. We show that SM-102 incorporation increases the rat

Oxygen-Generating Transdermal Nanoplatform Codelivering BRD4 Proteolysis-Targeting Chimera/Verteporfin/CaO2 Synergistically Remodels Immunosuppressive Melanoma Microenvironment to Potentiate Combination Immunotherapy.

Melanoma relapse and metastasis remain formidable clinical challenges, with the inadequate immunogenicity and highly immunosuppressive tumor microenvironment (ITME) presenting serious obstacles to current postsurgical immunotherapies. Herein, an oxygen self-supplying core-shell microneedle patch (AV@LDL&CaO2 MNs) featuring multiple innovative anticancer therapies and multimodal immunomodulatory properties is developed to facilitate melanoma postoperative management. Specifically, recombinant low-density lipoprotein nanoparticles (AV@LDL NPs) embedding both the bromodomain containing protein 4

Mesenchymal stromal cells modulate YAP by verteporfin to mimic cartilage development and construct cartilage organoids based on decellularized matrix scaffolds.

The mechanical elasticity or stiffness of the ECM modulates YAP activity to regulate the differentiation of stem cells during the development and defect regeneration of cartilage tissue. However, the understanding of the scaffold-associated mechanobiology during the initiation of chondrogenesis and hyaline cartilaginous phenotype maintenance remains unclear. In order to elucidate such mechanisms to promote articular cartilage repair by producing more hyaline cartilage, we identify the relationship between YAP subcellular localization and variation of the cartilage structure and organization du

The photosensitizer verteporfin has light-independent anti-leukemic activity for Ph-positive acute lymphoblastic leukemia and synergistically works with dasatinib.

Cell lines have been used for drug discovery as useful models of cancers; however, they do not recapitulate cancers faithfully, particularly from the viewpoints of microenvironmental independence. Patient-derived xenografts (PDX) are established by the transfer of primary tumor cells directly from patients into immunodeficient mice and can provide primary-like tumor cells of the amount needed at the desired time. We developed a high-throughput drug screening system using PDX cells and performed drug screening using the PDX cells of Philadelphia chromosome-positive acute lymphoblastic leukemia

Photodynamic therapy with verteporfin for choroidal neovascularization due to age-related macular degeneration and other causes: a New Zealand outcomes study.

PURPOSE: To assess the efficacy and safety of photodynamic therapy (PDT) for choroidal neovascularization (CNV) secondary to age-related macular degeneration (AMD) and other conditions by analysing visual acuity changes. METHODS: A retrospective review of patients treated with PDT was conducted. CNV was confirmed on fluorescein angiography. Visual acuity outcomes were recorded at 3-monthly intervals to a maximum of 48 months. The primary outcome measure was the proportion of patients avoiding moderate visual loss (losing less than three lines of visual acuity relative to baseline) at 12 month

Choroidal neovascularization reduced by targeted drug delivery with cationic liposome-encapsulated paclitaxel or targeted photodynamic therapy with verteporfin encapsulated in cationic liposomes.

PURPOSE: Intravitreal antivascular endothelial growth factor (anti-VEGF) application has revolutionized the treatment of choroidal neovascularization (CNV), a hallmark of wet age-related macular degeneration. However, additional treatment options are desirable as not all CNV lesions respond to anti-VEGF injections. Here, we assessed the feasibility of targeted delivery of cationic liposome-encapsulated paclitaxel (EndoTAG-1) in treating CNV. Furthermore, we investigated whether a new formulation of verteporfin encapsulated in cationic liposomes (CL-VTP) enhances the effect of photodynamic ther

A carrier-within-a-carrier system with calcium nanoparticles encapsulated in β-cyclodextrin-polysaccharide hydrogel for verteporfin delivery and radiosensitization in orthotopic osteosarcoma.

Osteosarcoma remains a clinical challenge due to its high invasiveness, early metastasis, and intrinsic radioresistance, which collectively limit the efficacy of conventional treatments and immunotherapy. Here, we developed a pH-responsive hydrogel reservoir, referred to as VP/CaNPs@Gel, by integrating verteporfin (VP)-loaded CaCO3 nanoparticles (CaNPs) into a β-cyclodextrin-crosslinked polysaccharide hydrogel to achieve precise radiosensitization and immunomodulation. The hydrogel undergoes rapid in-situ crosslinking to form a robust scaffold, ensuring the sustained and pH-triggered release o

Effect of photodynamic therapy with verteporfin on tumor thickness and best corrected visual acuity in patients with circumscribed choroidal hemangioma: a single-center experience.

The management of circumscribed choroidal hemangioma (CCH) with photodynamic therapy (PDT) using verteporfin has resulted in significant functional and clinical improvement compared with the pre-PDT era. Literature data on factors influencing clinical outcomes and predictors of response to PDT in symptomatic CCH are inconsistent. The aim of this study was to investigate the efficacy of PDT with verteporfin in patients with CCH depending on symptom duration and tumor thickness at baseline. We analyzed the medical records of 37 patients with symptomatic CCH divided into 3 groups according to sym

Cost-effectiveness of pegaptanib compared to photodynamic therapy with verteporfin and to standard care in the treatment of subfoveal wet age-related macular degeneration in Canada.

BACKGROUND: Age-related macular degeneration (AMD) is characterized by loss of central vision and is the leading cause of blindness among persons over the age of 50 years in Canada. The wet form of AMD has 3 subtypes-occult, minimally classic, and predominantly classic. Photodynamic therapy (PDT) with verteporfin is indicated only for the category of predominantly classic wet AMD. Currently, there are no treatments available for the other AMD subtypes. Pegaptanib sodium was the first pharmacologic therapy approved in Canada for the treatment of subfoveal wet AMD regardless of subtype. OBJECTI
