In a study of swine congenital anomalies, nine newborn piglets with varying degrees of optic hypotelorism including cyclopia were collected. Nasal and maxillary development were abnormal in all animals regardless of the degree of eye fusion. All animals except one had intact upper lips and hard palates that carried two or three small extopic teeth. The "snout" was only a medial wedge-shaped rudiment projecting from the upper lip. It was distally covered by a typical snout-like glabrous epithelium and carried small vibrissae. Six animals also had a tubular proboscis dorsal to the eye. The dista

Huntington's disease (HD) is a debilitating, genetically inherited neurodegenerative disorder that results in early loss of medium spiny neurons from the striatum and subsequent degeneration of cortical and other subcortical brain regions. Behavioral changes manifest as a range of motor, cognitive, and neuropsychiatric impairments. It has been established that replacement of the degenerated medium spiny neurons with rat-derived fetal whole ganglionic eminence (rWGE) tissue can alleviate motor and cognitive deficits in preclinical rodent models of HD. However, clinical application of this cell

The 27th annual Barrels meeting highlighted the latest advances in this rapidly growing field. The Barrels meeting annually focuses on the role of the posterior medial thalamus in somatosensation, dendritic processing, and the cortical dynamics involved during touch perception. Speakers utilized diverse molecular, physiological, computational techniques to understand the development, sensory processing, and motor commands that are involved with the rodent mystacial vibrissae. The meeting was held Thursday, 13 November through Friday, 14 November 2014 on the Homewood campus of Johns Hopkins Uni

Although the neurobiology of rodent facial whiskers has been studied intensively, little is known about sensing in other vibrissae. Here we describe the under-investigated submandibular "whisker trident" on the rat's chin. In this three-whisker array, a unique unpaired midline whisker is laterally flanked by two slightly shorter whiskers. All three whiskers point to the ground and are curved backwards. Unlike other whiskers, the trident is not located on an exposed body part. Trident vibrissae are not whisked and do not touch anything over long stretches of time. However, trident whiskers enga

PURPOSE: Supramaximal facial nerve stimulation is an applied current sufficient to evoke a maximal electromyographic response of facial musculature. It is used during cerebellopontine angle surgery for prognostication of postoperative nerve function. We utilized a rat model to examine safe parameters for intracranial electrical stimulation.
MATERIALS AND METHODS: Intracranial facial nerve stimulation with electromyographic monitoring of 14 rats was performed. Supramaximal current level was determined and 50 additional pulses of supramaximal (4 rats), 3 times supramaximal (4), 10 times suprama

BACKGROUND AND PURPOSE: The promotion of hair regeneration and growth heavily depends on the activation of Wnt/β-catenin signalling in the hair follicle, including dermal papilla (DP). KY19382, one of the newly synthesized analogues of indirubin-3'-monoxime (I3O), was identified as a Wnt/β-catenin signalling activator via inhibition of the interaction between CXXC-type zinc finger protein 5 (CXXC5) and dishevelled (Dvl). Given the close relationship between the Wnt/β-catenin signalling and hair regeneration, we investigated the effect of KY19382 on hair regrowth and hair follicle neogenesis.

Seals, sea lions, and other aquatic animals rely on their whiskers to identify and track underwater targets, offering valuable inspiration for the development of low-power, portable, and environmentally friendly sensors. Here, we design a single seal-whisker-like cylinder and conduct experiments to measure the forces acting on it with nine different upstream targets. Using sample sets constructed from these force signals, a convolutional neural network (CNN) is trained and tested. The results demonstrate that combining the seal-whisker-style sensor with a CNN enables the identification of obje

All small mammals have prominent facial whiskers that they employ as tactile sensors to guide navigation and foraging in complex habitats. Nocturnal, arboreal mammals tend to have the longest and most densely packed whiskers, and semi-aquatic mammals have the most sensitive. Here we present evidence to indicate that many small mammals use their whiskers to tactually guide safe foot positioning. Specifically, in 11, small, non-flying mammal species, we demonstrate that forepaw placement always falls within the ground contact zone of the whisker field and that forepaw width is always smaller tha

Motor and EMG responses were studied in thirty rats craniotomized under thiopental, in conditions of intracortical microstimulation and surface anode macrostimulation of the parietal and frontal neocortical areas. Strict contralaterality and distinct separation was found in motor representation (MRs) of the fore and hind limbs, of vibrissae fields and even of some joints and functional muscle groups. On the territory of the hind leg MR which was studied in greater details, centrally lies the compact MR zone of toes, mediocaudally from it - a vast MR of the foot, and further still more caudally

BACKGROUND: Posttraumatic facial paralysis is a disabling condition. Current surgical management by faciofacial nerve suture provides limited recovery. To improve the outcome, the authors evaluated an add-on strategy based on a syngeneic transplantation of nasal olfactory stem cells in a rat model of facial nerve injury. The main readouts of the study were the recording of whisking function and buccal synkinesis.
METHODS: Sixty rats were allocated to three groups. Animals with a 2-mm facial nerve loss were repaired with a femoral vein, filled or not with olfactory stem cells. These two groups

Pseudoxanthoma elasticum (PXE) is an autosomal recessive disorder characterized by ectopic mineralization of connective tissues and shows considerable intra- and inter-familial phenotypic variability. PXE is caused by mutations in the ABCC6 gene, and targeted ablation of Abcc6 in mouse recapitulates PXE. In this study, we examined the hypothesis that the GGCX gene encoding gamma-glutamyl carboxylase may interfere with the mineralization process in Abcc6 ( -/- ) mice. Thus, Abcc6 ( -/- ) and Ggcx (+/-) mice were generated on 129S1;C57 and 129S1;129X1;C57 genetic backgrounds, respectively, and b

The selenoprotein cytosolic glutathione peroxidase (cGPx) is ubiquitously distributed in a variety of organs, and its primary function is to protect oxidative damage. To investigate the spatial and temporal expression pattern of cGPx mRNA in embryogenesis, as this has not been studied before, reverse transcription-polymerase chain reaction (RT-PCR) was carried out in a thermal cycler using mouse-specific cGPx primers, and in situ hybridization was performed in whole embryos or embryonic tissues using digoxigenin-labeled mouse cGPx riboprobes. Expression of cGPx mRNA was detected in all the emb

Treatment of pregnant Sprague-Dawley rats with etretinate or retinoic acid on pregnancy days 8.5-9.0 resulted in craniofacial defects in all embryos. A morphological investigation of the malformations occurring in the nasal region is described. The external nose was deformed with a depressed nasal bridge and pronounced hump. Fusion between nasal processes was defective, resulting in fistulas and clefts. The nasal septal cartilage was reduced in size, the paranasal cartilage was missing, the parietothecal cartilage was deformed and the paraseptal cartilage was dislocated. Aberrant vessels were

Trimming of vibrissae in dogs (Canis lupus familiaris) for cosmetic reasons remains a common practice, particularly among poodle breeds and show dogs. In discussions surrounding this practice, it has been claimed that domestic dogs would have fewer and structurally less developed sinus hairs than other mammals, and that, especially in poodles, vibrissae would lack functional relevance, although such narratives represent popularized assumptions rather than systematically supported peer-reviewed evidence. The follicle-sinus complexes (FSCs) of various species, including cats (Felis catus), rats

Many people suffer from hair loss and abnormal skin pigmentation, highlighting the need for simple assays to support drug discovery research. Current assays have various limitations, such as being in vitro only, not sensitive enough, or unquantifiable. We took advantage of the bilateral symmetry and large size of mouse whisker follicles to develop a novel in vivo assay called "whisker follicle microinjection assay". In this assay, we plucked mouse whiskers and then injected molecules directly into one side of the whisker follicles using microneedles that were a similar size to the whiskers, an

Psychological stress affects a wide spectrum of brain functions and poses risks for many mental disorders. However, effective therapeutics to alleviate or revert its deleterious effects are lacking. A recently synthesized psychedelic analog tabernanthalog (TBG) has demonstrated anti-addictive and antidepressant potential. Whether TBG can rescue stress-induced affective, sensory, and cognitive deficits, and how it may achieve such effects by modulating neural circuits, remain unknown. Here we show that in mice exposed to unpredictable mild stress (UMS), administration of a single dose of TBG de

Stroke is a worldwide leading cause of death and long-term disability, with ischemic strokes making up approximately 85% of all cases. There is a significant need for an ideal animal model that accurately replicates the disease's pathology to study the molecular mechanisms of brain injury. Various experimental models have been created to induce middle cerebral artery occlusion (MCAO), including intraluminal MCAO, photothrombotic models, endothelin-1 injections, and electrocoagulation. However, these often result in large infarct or lesion volumes accompanied by considerable variability. In thi

Spontaneous neuronal activity strongly impacts stimulus encoding and behavioral responses. We sought to determine the effects of neocortical prestimulus activity on stimulus detection. We trained mice in a selective whisker detection task, in which they learned to respond (lick) to target stimuli in one whisker field and ignore distractor stimuli in the contralateral whisker field. During expert task performance, we used widefield Ca2+ imaging to assess prestimulus and post-stimulus neuronal activity broadly across frontal and parietal cortices. We found that lower prestimulus activity correla
