Identification and molecular characterization of five putative toxins from the venom gland of the snake Philodryas chamissonis (Serpentes: Dipsadidae).

Philodryas chamissonis is a rear-fanged snake endemic to Chile. Its bite produces mild to moderate symptoms with proteolytic and anti-coagulant effects. Presently, the composition of the venom, as well as, the biochemical and structural characteristics of its toxins, remains unknown. In this study, we cloned and reported the first full-length sequences of five toxin-encoding genes from the venom gland of this species: Type III snake venom metalloprotease (SVMP), snake venom serine protease (SVSP), Cysteine-rich secretory protein (CRISP), α and β subunits of C-type lectin-like protein (CLP) and

Assembling an arsenal: origin and evolution of the snake venom proteome inferred from phylogenetic analysis of toxin sequences.

We analyzed the origin and evolution of snake venom toxin families represented in both viperid and elapid snakes by means of phylogenetic analysis of the amino acid sequences of the toxins and related nonvenom proteins. Out of eight toxin families analyzed, five provided clear evidence of recruitment into the snake venom proteome before the diversification of the advanced snakes (Kunitz-type protease inhibitors, CRISP toxins, galactose-binding lectins, M12B peptidases, nerve growth factor toxins), and one was equivocal (cystatin toxins). In two others (phospholipase A(2) and natriuretic toxins

A survey of gene expression and diversity in the venom glands of the pitviper snake Bothrops insularis through the generation of expressed sequence tags (ESTs).

In order to produce a global panorama of the transcriptional activity of snake venom glands and to correlate with its venom composition, we constructed a DNA complementary to RNA library from the venom glands of the Viperidae snake Bothrops insularis for the generation of expressed sequence tags (ESTs). Sequences from 610 independent clones were grouped in 297 clusters, revealing the putative identification of 210 distinct gene products. Toxin sequences correspond to 56% of all transcripts (85 clusters), being the metalloproteinases (23%) and the bradykinin-potentiating peptides (11%) the majo

Timeline of key events in snake venom metalloproteinase research.

It is reasonable to state that snake venom toxinology has been actively pursued for at least the past 400 to 500 years. Early on it was appreciated that the venoms of the Viperidae produced profound local effects, notably hemorrhage. For the past 100 years, with the advent of modern chemistry and biochemistry significant progress has been gained regarding the function, structure and role of the snake venom metalloproteinases (SVMPs) in viperid venom pathogenesis. In this review we provide a concise, chronological presentation of the key significant studies that have led to our current understa

Structure-function correlations of snake venom disintegrins.

Disintegrins represent a family of polypeptides released in the venoms of Viperidae and Crotalidae snakes (vipers and rattlesnakes) by the proteolytic processing of multidomain metalloproteinases, which selectively block the function of beta(1) and beta(3) integrin receptors. Research on disintegrins is relevant for understanding the biology of viper venom toxins, but also provides information of new structural determinants involved in integrin recognition that may be useful in both, basic and clinical research. The role of the composition, conformation and concerted dynamics of the integrin i

Distribution of low molecular weight platelet aggregation inhibitors from snake venoms.

An assay of platelet aggregation inhibitors measured by the turbidimeter using Aggregometer PAM 8C (Mebanix) was performed after each crude snake venom (57 species) was subjected to ultrafiltration using MILLIPORE UFP 1 LGC. The snake venoms of Viperidae (three species), Elapidae (11 species), and Hydrophiidae (two species) inhibited ADP-induced rabbit platelet aggregation. In particular, six venoms of Bitis gabonica, Pseudocerastes persicus, Dendroaspis angusticeps, D. polylepis, Ophiophagus hannah, and N. nigricollis crawshawii strongly inhibited platelet aggregation. Furthermore, adenosine

Identification and cloning of snake venom vascular endothelial growth factor (svVEGF) from Bothrops erythromelas pitviper.

Vascular endothelial growth factors (VEGFs) are among the most important angiogenic proteins found on vertebrates. In the last years, some reports of the occurrence of such proteins in snake venoms are rising the importance of this family of proteins as toxins, since they appear to be involved in many features of Viperidae envenoming, such as hypotension and venom spread through increase in vascular permeability. Here we describe the occurrence of snake venom VEGF in Bothrops erythromelas, a clinical important snake from Northeast of Brazil, through immunodetection and cloning of its cDNA and

Biochemical characterization and molecular cloning of a plasminogen activator proteinase (LV-PA) from bushmaster snake venom.

The protein (LV-PA) from bushmaster (Lachesis muta muta) venom is a serine proteinase which specifically activates the inactive proenzyme plasminogen. LV-PA is a single chain glycoprotein with an apparent molecular mass of 33 kDa that fell to 28 kDa after treatment with N-Glycosidase F (PNGase F). Approximately 93% of its protein sequence was determined by automated Edman degradation of various fragments derived from a digestion with trypsin. A cDNA library of L. m. muta was constructed to generate expressed sequence tags (ESTs) and the plasminogen activator precursor cDNA was sequenced. The c

Inhibition of hemorragic snake venom components: old and new approaches.

Snake venoms are complex toxin mixtures. Viperidae and Crotalidae venoms, which are hemotoxic, are responsible for most of the envenomations around the world. Administration of antivenins aimed at the neutralization of toxins in humans is prone to potential risks. Neutralization of snake venom toxins has been achieved through different approaches: plant extracts have been utilized in etnomedicine. Direct electric current from low voltage showed neutralizing properties against venom phospholipase A2 and metalloproteases. This mini-review summarizes new achievements in venom key component inhibi

[Venomous and poisonous animals. III. Elapidae snake envenomation].

Envenomation by Elapidae snakes is less frequent than by Viperidae snakes but represents a true medical emergency due to rapid progression of cobra syndrome. Elapidae venom contains neurotoxins that paralyze striated muscles especially in the thoracic cavity. Respiratory paralysis can occur within a few hours and is preceded by neurological symptoms (local paresthesia and paresis progressing to the cranial nerves). When cobra envenomation is suspected, antivenom administration by the direct venous route must be undertaken as quickly as possible to stop the envenomation process. Artificial vent

Novel in vitro assays for assessing the haemorrhagic activity of snake venoms and for demonstration of venom metalloproteinase inhibitors.

Standard methods used for assessing the haemorrhagic toxicity of snake venoms and the effectiveness of antivenoms are laborious, expensive and involve the use of large numbers of laboratory animals. This paper examined the feasibility of using a gelatin degradation ELISA for preliminary screening of snake venom metalloproteinases (MPs). Potent gelatinolytic activity was observed in venoms from snakes of the family Viperidae and, as expected, little or no activity was evident in the venoms of snakes that induce neurotoxic pathology (most elapids). A reverse gelatin zymography assay was used on

Bites by exotic snakes reported to the UK National Poisons Information Service 2009-2020.

Introduction: Snakebite is recognised as a neglected tropical disease and a cause of substantial morbidity and mortality. Whilst the most medically important snakes are typically native of Asia, Africa, Latin America and Oceania, the possibility of encountering these snakes is no longer limited by geography due to an increasing number of exotic (non-native) snakes being held in captivity. Methods: A retrospective review of snakebite enquiries to the UK National Poisons Information Service (NPIS) between 2009 and 2020. Mechanical properties, microstructure, and strike biomechanics of tubular fangs in neotropical vipers.

Viper fangs represent one of the most mechanically sophisticated biological penetration devices found in nature. Despite extensive interest in their venom-delivery function, a comprehensive characterization of the structure, composition, and biomechanics of neotropical viper fangs remains lacking. This study presents the first integrated analysis of the tubular fangs of three Latin American viper species - Lachesis acrochorda, Crotalus durissus cumanensis, and Bothrops asper - combining optical microscopy, scanning electron microscopy, high-resolution X-ray computed tomography, attenuated tota

Clinical aspects of bites by viper in Spain.

Poisonings due to Viperidae bites are quite common in our country. There is a mortality of 3 to 7 cases out of 35 million inhabitants and a variable morbidity which increases in some years. From 1965 to 1980, about 125 cases of bites by Vipers latasti, 80 by V. aspis and 23 by V. seoanei received treatment. Fifty per cent of the cases received small doses of venom and presented proteolytic and coagulant reactions. With stronger doses the reactions were anticoagulant and hemolytic. Two cases affected by bites of V. aspis developed a neurotoxic syndrome with ptosis, dysphagia, dysphasia and para

Snake Venom PLA2, a Promising Target for Broad-Spectrum Antivenom Drug Development.

Snakebite envenomation is a neglected global health problem, causing substantial mortality, disability, and psychological morbidity, especially in rural tropical and subtropical zones. Antivenin is currently the only specific medicine for envenomation. However, it is restricted by cold storage, snakebite diagnosis, and high price. Snake venom phospholipase A2s (svPLA2s) are found in all kinds of venomous snake families (e.g., Viperidae, Elapidae, and Colubridae). Along with their catalytic activity, svPLA2s elicit a wide variety of pharmacological effects that play a pivotal role in envenomati

Snake venom proteins acting on hemostasis.

The venoms of Viperidae and Crotalidae snakes are a rich source of proteins with activity against various factors involved in coagulation and fibrinolysis. These proteins are very specific for their molecular targets, resistant to physiological inhibitors and stable in vitro and in vivo. They have therefore proved to be useful for diagnostic tests. Based on sequence similarities, these snake venom proteins have been classified into various families, such as serine proteinases, metalloproteinases, C-type lectins, disintegrins and phospholipases A(2). The various members of a given family, altho

Venomous snakes of Costa Rica: biological and medical implications of their venom proteomic profiles analyzed through the strategy of snake venomics.

UNLABELLED: In spite of its small territory of ~50,000km(2), Costa Rica harbors a remarkably rich biodiversity. Its herpetofauna includes 138 species of snakes, of which sixteen pit vipers (family Viperidae, subfamily Crotalinae), five coral snakes (family Elapidae, subfamily Elapinae), and one sea snake (Family Elapidae, subfamily Hydrophiinae) pose potential hazards to human and animal health. In recent years, knowledge on the composition of snake venoms has expanded dramatically thanks to the development of increasingly fast and sensitive analytical techniques in mass spectrometry and separ

Replacement of fangs in a free-ranging desert viperid, Cerastes vipera.

Venomous viperid snakes possess relatively large and fragile hollow fangs that are an integral part of the envenomation apparatus for predation. We hypothesized that fangs serve like disposable needles and predicted a high loss rate and, hence, high replacement rate in free-ranging snakes. Snakes also possess smaller rear teeth that aid in gripping and swallowing the prey. We reasoned that these teeth are less delicate than fangs and predicted that their loss would be at a slower rate than fangs. To test our predictions, we analyzed fecal samples of free-ranging Saharan sand vipers, Cerastes v

Lethal herpesvirosis in 16 captive horned vipers (Vipera ammodytes ammodytes): pathological and ultrastructural findings.

Sixteen captive adult horned vipers (Vipera ammodytes ammodytes) were submitted for necropsy examination following a 2-week history of lethargy, anorexia and dyspnoea. Gross lesions included widespread haemorrhage, serosanguineous effusions in the body cavities and multiple pinpoint white to yellow foci in the liver. Microscopically, there was multifocal hepatic coagulative necrosis associated with intranuclear acidophilic inclusion bodies in sinusoidal endothelial cells. Similar endothelial lesions were observed in the myocardium, fat bodies, kidneys and spleen. Transmission electron microsco
