Effect of Dietary Fructooligosaccharide (FOS) Supplementation on Ileal Microbiota in Broiler Chickens.

The dietary effect of fructooligosaccharide (FOS) supplementation as an alternative to antibiotics on ileal mucosa and digesta microbiota was investigated in broiler chickens (n = 180). The study included three dietary treatments from d1 to 21: 1) positive control (PC), a wheat-corn-soybean meal based diet containing antibiotics (virginiamycin and monensin); 2) negative control (NC), as wheat-corn-soybean meal based diet without antibiotics; and 3) NC + FOS, as NC diet supplemented 0.5% of FOS. Ileal mucosa and digesta were collected and subjected to 16S rRNA-based next generation sequencing.

Dissemination of Quinupristin-Dalfopristin and Linezolid resistance genes among hospital environmental and healthy volunteer fecal isolates of Enterococcus faecalis and Enterococcus faecium.

BACKGROUND: Streptogramins and linezolid are important in the treatment of infections caused by vancomycin-resistant enterococci. PURPOSE: Then, we aimed to evaluate the resistance rates against these drugs and the prevalence of genes involved in hospital environmental and fecal normal-flora isolates of Enterococcus faecalis and Enterococcus faecium. METHODS AND RESULTS: The strains were isolated from the stool samples and hospital environments by culturing on M-Enterococcus (ME) agar, and identified by phenotypic and genotypic microbiological tests. The disk agar diffusion method was used t

In vitro post-antibiotic effect of fluoroquinolones, macrolides, beta-lactams, tetracyclines, vancomycin, clindamycin, linezolid, chloramphenicol, quinupristin/dalfopristin and rifampicin on Bacillus anthracis.

OBJECTIVES: The aim of this study was to investigate in vitro the post-antibiotic effect (PAE) of 19 antibacterial agents against two strains of Bacillus anthracis (ST-1 and Sterne strains). METHODS: PAE was determined by calculating the time required for the viable counts of antibiotic-exposed bacteria (at concentrations of 10x MIC and exposure for 2 h) at 37 degrees C to increase by 1 log10 above the counts observed immediately after antibiotic removal compared with the corresponding time for controls not exposed to antibiotics. RESULTS: The PAEs of the fluoroquinolones (ciprofloxacin, ofl

Implications of multiple antimicrobial-resistant enterococci associated with the poultry environment.

Poultry are increasingly being associated with carriage of multiresistant organisms that may cause disease in humans. Among these organisms are the enterococci, not regarded as a common cause of hospital-acquired infections. The use of antimicrobials for growth promotion in poultry production envronments may facilitate the dissemination of resistance to Enterococcus spp. that have the potential to be clinically significant. To assess descriptively the degree of multiresistant enterococci in the poultry environment of the Delmarva (Delaware-Maryland-Virginia) East Coast region of the USA, litte

Activity of quinupristin/dalfopristin against Streptococcus pneumoniae in vitro and in vivo in the rabbit model of experimental meningitis.

We tested the efficacy of quinupristin/dalfopristin, an antibiotic made up of dalfopristin (70%) and quinupristin (30%) against a large panel of Streptococcus pneumoniae strains. The pneumococcal isolates (217) included 200 penicillin-resistant and 17 penicillin-susceptible clinical isolates. Eighty-nine of the 200 resistant bacteria showed an intermediate level and 111/200 showed a high level of resistance to penicillin. Of the highly resistant strains, 56/111 belonged to the multidrug-resistant Spanish/USA epidemic clone of S. pneumoniae, as defined by appropriate genetic techniques. The res

Activity of quinupristin/dalfopristin against gram-positive bacteria: clinical applications and therapeutic potential.

In recent years there has been a dramatic worldwide increase in the prevalence of multiple drug-resistant strains of common Gram-positive bacteria. This highlights the need for a new class of antibiotic with activity against these organisms. Quinupristin/dalfopristin, the first injectable streptogramin antibiotic, has a unique spectrum of activity, encompassing most Gram-positive cocci (including multi-drug-resistant strains), respiratory pathogens and anaerobes, Gram-positive, and a prolonged post-antibiotic effect. Quinupristin/ dalfopristin is active in vitro against multi-drug-resistant is

[Contact eczema in patients with leg ulcers].

Patients with leg ulcers or varicose eczema suffer much more often from contact eczema due to the local application of pharmaceutical preparations than patients suffering from other dermatological problems (even those of eczematous origin). This contact allergy may concern not only the active ingredient but also the excipient, the preservative, or even the perfume. In all cases of leg ulcers, of varicose eczema, but also of badly healed ulcers, epicutaneous tests should be carried out with all the components of the pharmaceutical preparations concerned. Moreover, the pharmaceutical industry re

Effects of Medium-Chain Fatty Acid Glycerides on Nutrient Metabolism and Energy Utilization in Weaned Piglets.

Weaning stress induces the depressed digestive and absorptive capacity and insufficient intestinal energy supply. Medium-chain fatty acid glycerides have shown to improve the growth performance and intestinal barrier function of weaned piglets in the previous study. This study was aimed to investigate the regulation of medium-chain fatty acid glyceride on the nutrient absorption and energy utilization of weaned piglets. Nighty healthy weaned piglets were randomly assigned into five treatments: NP (Normal protein, normal-protein diet no antibiotics included); NC (Negative control, low-protein d

Multiplex immunoassay based on biochip technology for the screening of antibiotic residues in milk: validation according to the European guideline.

The Infiniplex for milk® (IPM) kit is a quick method for the simultaneous and qualitative detection of more than 100 molecules including antibiotic residues, mycotoxins, anti-inflammatories and antiparasitic drugs into a single test that does not require milk treatment. The IPM® kit was validated according to the European decision EC/2002/657 and according to the European guideline for the validation of screening methods (2010). Our validation was focused only on antibiotic residues. The washing step was identified as the most critical step of the assay. Insufficient washes could cause a signi

Nationwide German multicenter study on prevalence of antibiotic resistance in staphylococcal bloodstream isolates and comparative in vitro activities of quinupristin-dalfopristin.

Antibiotic-resistant gram-positive bacteria have become an increasing problem in the last two decades. In order to evaluate the prevalence of antibiotic resistance in staphylococcal bloodstream isolates in Germany, 2,042 staphylococci collected in 21 tertiary-care hospitals were investigated during a 3-year period (March 1996 to March 1999). Altogether, 1,448 S. aureus isolates and 594 coagulase-negative staphylococci (CoNS) that comprised 13 different species were included. Furthermore, the antistaphylococcal activities of quinupristin-dalfopristin were compared with those of eight other comp

Epidemiologic trends in nosocomial and community-acquired infections due to antibiotic-resistant gram-positive bacteria: the role of streptogramins and other newer compounds.

The Gram-positive cocci have clearly re-emerged as important pathogens world-wide in the past two decades. Staphylococci, including the coagulase-negative staphylococci and Staphylococcus aureus, and the enterococci account for approximately one-third of all blood stream infections and as much as 50% of nosocomial blood stream infections. Although Streptococcus pneumoniae is often considered a community-acquired pathogen, it is also an important cause of nosocomial infection. The hallmark of these Gram-positive pathogens is increasing resistance to available antimicrobial agents. Of particular

Antimicrobials and competitive exclusion.

The compatibility of commonly used antimicrobial feed additives with CE treatment are examined. It appears that additives permitted in the U.K. are without effect on CE, with the possible exception of avoparcin. However, a sharp difference has been found with the effect of antimicrobials on CE treatment cultures in vitro, and the in vivo effect; and it is postulated that these differences may be accounted for by the fact that the substances may not be 'caecum available' in the bird. When effects are observed in the bird they appear to be largely due to dose variation. In the case of adult bird

Suppressive effects of peptide antibiotics against proliferation and cytokine production in mitogen-activated human peripheral-blood mononuclear cells.

Certain kinds of peptide antibiotics are suggested to have immunomodulatory effects; however, few studies have been carried out systemically to evaluate the antiproliferative effects of peptide antibiotics in human lymphoid cells. The suppressive efficacies of nine peptide antibiotics and seven non-antibiotic peptides against proliferation of human peripheral-blood mononuclear cells (PBMCs) stimulated with T cell mitogen were examined in vitro. Nigericin (CAS 28643-80-3), valinomycin (CAS 2001-95-8), gramicidin D (CAS 1405-97-6), and tyrothricin (CAS 1404-88-2) strongly inhibited the prolifera

Dietary fiber concentration modulates the efficacy of phytogenic and antimicrobial rumen modifiers on ruminal fermentation and methane emissions in cattle.

This study evaluated the effects of dietary fiber level and rumen modulators on feeding behavior, methane emissions, and ruminal fermentation parameters in cattle. A single 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments was used, involving four ruminally cannulated cattle evaluated over four experimental periods. The treatments combined two dietary neutral detergent fiber (NDF) concentrations (12% and 50% of dry matter) with two feed additive strategies: monensin plus virginiamycin (VM) or a phytogenic additive (LM+). Diets containing 12% NDF represented forage-free
