OBJECTIVE: To establish a rat model with paradoxical vocal fold movement to understand the detailed etiology and physiology of laryngeal synkinesis by evaluating vocal fold movement and by electromyography.
METHODS: Adult Wistar rats were used. The recurrent laryngeal nerve was transected, anastomosed, and the anastomotic portion was placed in a silicone tube. At 2, 4, and 10 weeks after the treatment (n = 30), we performed laryngoscopy and electromyography. The vocal fold movement was recorded, the hemiglottal area was measured, and the Δarea was calculated by subtracting the area during exp

Male and female rats emit ultrasonic vocalizations in reproductive encounters. While estrous bedding has been used to elicit vocalizations of males, the number of responses is variable. We report a reliable method to assess vocalizations using exposure to a stimulus animal. The stimulus rat is placed behind a wire barrier for 5 min, then removed. Vocalizations are then recorded for 5 min. Experiment 1 validated this method and it was used for subsequent experiments. In Experiment 2, male rats were castrated and tested for the restoration of vocalizations. In one group, males were allowed to co

OBJECTIVES: The comparative fine structures of the rabbit vocal fold (VF) were investigated using the technique combined with correlative light and electron microscopy (CLEM) and transmission electron microscopy (TEM) to elucidate the functional mechanisms.
METHODS: Seven normal New Zealand White Rabbit vocal folds were examined using CLEM technique and TEM to investigate their fine structures.
RESULTS: In contrast to the human VF, the rabbit VF lacked the macula flava and vocal ligament. However, the distribution of fibrous components of the lamina propria (LP) of the rabbit VF was similar

OBJECTIVES/HYPOTHESIS: Menopause adversely affecs power and endurance of the limb muscles. However, despite clinical observations that menopause corresponds to negative changes of the voice, the direct effects of estrogen deprivation on the thyroarytenoid muscles are unknown. The purpose of this study was to investigate the effects of estrogen deprivation via ovariectomy on three neuromuscular parameters of the thyroarytenoid muscles using a rat model.
STUDY DESIGN: Animal model.
METHODS: Cryosections of vocal folds of 20 (10 control and 10 ovariectomized) female rats were stained to label n

Inappropriate vocal expressions, e.g., vocal tics in Tourette syndrome, severely impact quality of life. Neural mechanisms underlying vocal tics remain unexplored because no established animal model representing the condition exists. We report that unilateral disinhibition of the nucleus accumbens (NAc) generates vocal tics in monkeys. Whole-brain PET imaging identified prominent, bilateral limbic cortico-subcortical activation. Local field potentials (LFPs) developed abnormal spikes in the NAc and the anterior cingulate cortex (ACC). Vocalization could occur without obvious LFP spikes, howeve

The purpose of this study is twofold. First, to introduce a new phenomenon--abductor vocal cord spasm, or abductor laryngospasm. Second, to reconcile a longstanding discrepancy involving the effect of lung inflation on vocal cord position. Abductor laryngospasm was induced in six anesthetized, tracheotomized dogs by the administration of continuous positive airway pressure (CPAP) after occlusion of the trachea for 60 seconds. Abductor laryngospasm was characterized by sustained vocal cord abduction with concomitant massive, continuous posterior cricoarytenoid muscle (PCA) electromyography (EMG

BACKGROUND: Vocal fold fibrosis (VFF), characterized by excessive extracellular matrix deposition, leads to permanent voice impairment, with current therapies often failing to restore vocal function. Current treatments are limited by suboptimal efficacy and high recurrence rates. Dysregulated energy metabolism is emerging as a critical driver of fibrosis. AMP-activated protein kinase (AMPK), a central cellular energy sensor, exerts broad antifibrotic effects in multiple organs, yet the therapeutic potential of novel AMPK activators, such as Aldometanib, in VFF remains to be fully characterized

It has been shown that stimulation-produced-analgesia (SPA) in the cat elicited from the periaqueductal gray matter (PAG) is obtained from sites located in the ventral part, particularly the dorsal raphé nucleus (DRN). These data contrast with the numerous studies performed in the rat in which efficient sites seem widely distributed throughout the PAG. These discrepancies led us to reinvestigate SPA from PAG and adjacent structures in the rat. Central stimulation was delivered through bipolar concentric electrodes (one for each animal). Analgesia was evaluated (before and during central stimul

Vocal fold tissues in animal and human species undergo deformation processes at several types of loading rates: a slow strain involved in vocal fold posturing (on the order of 1 Hz or so), cyclic and faster posturing often found in speech tasks or vocal embellishment (1-10 Hz), and shear strain associated with vocal fold vibration during phonation (100 Hz and higher). Relevant to these deformation patterns are the viscous properties of laryngeal tissues, which exhibit non-linear stress relaxation and recovery. In the current study, a large strain time-dependent constitutive model of human voca

OBJECTIVES/HYPOTHESIS: Vocal fold collagen composition is an important determinant of material properties and mucosal wave propagation. Collagen alignment and straightness are quantitatively characterized by second harmonic generation (SHG) imaging. We examined leporine, canined and porcine vocal folds showing collagen composition variation that is species, location, and strain specific.
STUDY DESIGN: Animal model.
METHODS: Leporine (n = 5), canine (n = 5), and porcine (n = 5) larynges were harvested and fixed in situ. Samples were transversely sectioned, and SHG images were collected for tw

Pig welfare constitutes a strategic pillar of sustainability within the pork industry. Consequently, there is a need to identify, develop and/or validate indicators for assessing pig wellbeing under commercial conditions. A systematic review following PRISMA guidelines identified 95 pig welfare indicators (PWIs) categorized into physiological, behavioral, health and post-mortem, and product quality. The review evaluated their validity and feasibility (V&F) for use in abattoirs to measure welfare during transport and slaughter. Thirty V&F indicators were found: one physiological (body temperatu

Morphine has been widely used for the treatment of acute, chronic, and cancer pain and is considered the strongest analgesic in clinical care. Conversely, morphine-induced analgesia may be accompanied by several side effects. Animal studies have demonstrated that low doses of morphine administered intrathecally can produce reliable analgesia for thermal, mechanical, and chemical nociceptive stimulation. On the other hand, high doses of morphine administered intrathecally may induce spontaneous nociceptive responses such as scratching, biting, and licking in mice as well as agitation and vocali

Encoding of emotional arousal in vocalisations is commonly observed in the animal kingdom, and provides a rapid means of information transfer about an individual's affective responses to internal and external stimuli. As a result, assessing affective arousal-related variation in the acoustic structure of vocalisations can provide insight into how animals perceive both internal and external stimuli, and how this is, in turn, communicated to con- or heterospecifics. However, the underlying physiological mechanisms driving arousal-related acoustic variation remains unclear. One potential driver o

Huntington's disease (HD) is an autosomal dominant, neurodegenerative disorder with a characteristic triad of cognitive, affective and motor symptoms. Transgenic HD mice show excellent construct and face validity for many of these symptoms, however the decline in some facets of every day activity in humans is difficult to model. One approach is the assessment of species-relevant behaviors. Here we described three ethologically appropriate tests in the mouse-olfactory sensitivity, nest-building and sexually-motivated vocalizations. In R6/1 HD mice, olfactory and nest-building tests were sensiti

Melanin concentrating hormone (MCH) is an orexigenic hypothalamic neuropeptide, which plays an important role in the complex regulation of energy balance and body weight. Here we show that SNAP-7941, a selective, high-affinity MCH1 receptor (MCH1-R) antagonist, inhibited food intake stimulated by central administration of MCH, reduced consumption of palatable food, and, after chronic administration to rats with diet-induced obesity, resulted in a marked, sustained decrease in body weight. In addition, after mapping the binding sites for [(3)H]SNAP-7941 in rat brain, we evaluated its effects in

Fifty-kHz ultrasonic vocalizations have been proposed to reflect a positive appetitive affective state in rats, being consistently linked to the positive appetitive behavior. In the first study, we examined the brain substrates of 50-kHz ultrasonic vocalizations (USVs) by using localized electrical stimulation of the brain (ESB) at various sites that are known to mediate reward. We found that the brain areas that produced ESB-induced 50-kHz calls are the areas that have previously been shown to support the most vigorous self-stimulation behavior (prefrontal cortex, nucleus accumbens, ventral p

Deterioration in the quality of a person's voice and speech is an early marker of Parkinson's disease (PD). In humans, the neural circuit that supports vocal motor control consists of a cortico-basal ganglia-thalamo-cortico loop. The basal ganglia regions, striatum and globus pallidus, in this loop play a role in modulating the acoustic features of vocal behavior such as loudness, pitch, and articulatory rate. In PD, this area is implicated in pathogenesis. In animal models of PD, the accumulation of toxic aggregates containing the neuronal protein alpha-synuclein (αsyn) in the midbrain and st

Human imaging studies implicate the insular cortex in processing complex sounds and vocal communication signals such as speech. In addition, lesions of the insula often manifest as deficits in sound or speech recognition (auditory agnosia) and speech production. While models of acoustic perception assign an important role to the insula, little is known about the underlying neuronal substrate. Studying a vocal primate, we identified a predominantly auditory region in the caudal insula and therein discovered a neural representation of conspecific communication sounds. When probed with natural so
