Two experiments were performed to examine the acute effects of thirst-inducing stimuli upon the intake of tap water and ethanol solutions by golden hamsters, a species which avidly consumes ethanol solutions. In Experiment 1, three groups of adult male hamsters (n = 6/group) were maintained on Purina chow and tap water; hamsters in two of the groups also had access to one of two ethanol solutions (15% or 30%, v/v). Animals were deprived at various times of either one or both fluids for 24 hr, and then either one or both fluids were presented during a 2-hr drinking test. Total water intake incr

This study examined the effect of subcutaneous administration of the neurohormone oxytocin on water intake of ad lib-fed (with or without sodium availability in the diet) and food-deprived animals. Results of the first experiment showed that oxytocin increased water intake and urine excretion in food-deprived but not in ad lib-fed animals. However, oxytocin treatment did not modify the reduced water "balance" (fluid intake minus urine volume) resulting from food deprivation or the daily food intake (Experiment 1). The dose-dependent polydipsic effect of oxytocin on food-deprived rats was alway

BACKGROUND: Humans and animals are frequently subjected to food deprivation or starvation. However, the adaptation of the kidney to this condition is not well understood. The purpose of these studies was to examine the effects of food deprivation on water handling by the kidney, the expression levels of collecting duct (CD) water channel aquaporin-2 (AQP2), and to determine the role of vasopressin in the adaptation of AQP2 to food deprivation.
METHODS: Sprague-Dawley (SD) and Brattleboro rats were placed in metabolic cages and deprived of food but had free access to water for 72 hours. Water

Groups of nine to ten male rats fed diets deficient in calcium, iron, magnesium, phosphorus, potassium, or all minerals (i.e. no mineral mix) were given 0.3 M NaCl and water to drink. Over the first 30 days of dietary treatment, 0.3 M NaCl intake was progressively increased by deprivation of calcium or iron, unaltered by deprivation of phosphorus, and increased and then decreased by deprivation of magnesium, potassium, or all minerals. The changes in NaCl intake of rats deprived of potassium or all minerals were matched by corresponding changes in water intake. Water intakes of the other group

BACKGROUND: The indirect water-deprivation test is the current reference standard for the diagnosis of diabetes insipidus. However, it is technically cumbersome to administer, and the results are often inaccurate. The current study compared the indirect water-deprivation test with direct detection of plasma copeptin, a precursor-derived surrogate of arginine vasopressin.
METHODS: From 2013 to 2017, we recruited 156 patients with hypotonic polyuria at 11 medical centers to undergo both water-deprivation and hypertonic saline infusion tests. In the latter test, plasma copeptin was measured when

The involvement of the L-arginine-nitric oxide (NO) pathway in brain, in the regulation of drinking behaviour, has been evaluated by injecting L-arginine and N omega-nitro-L-arginine methyl ester (L-NAME) into the lateral cerebral ventricle (i.c.v.). L-Arginine (5 and 10 micrograms/rat), but not D-arginine, was antidipsogenic when administered to 24 hr water-deprived rats but did not change the intake of water in normally hydrated rats. However, L-NAME (5 and 10 micrograms/rat) did antagonize the effect of L-arginine in water-deprived animals but, by itself, did not increase thirst. L-Arginine

The function of taurine in mammalian osmoregulation was studied in the Brattleboro rat with hereditary hypothalamic diabetes insipidus (DI). DI rats are chronically dehydrated because of their inability to synthesize vasopressin. One day of water deprivation did not affect the water balance in rats with normal vasopressin synthesis, whereas DI rats were markedly dehydrated and lost considerably body weight. Taurine content and 3H-taurine accumulation by platelets were significantly higher in DI rats, with a further increase after one day of water deprivation. In DI rats, water deprivation also

Since platelets accumulate taurine, they provide a model for studying the taurine transport in anisosmotic disorders. Thus, in this work we studied the taurine concentration and uptake in the platelets of Brattleboro rats, homozygous (DI) and heterozygous (HZ) for hereditary hypothalamic diabetes insipidus, and Long Evans (LE) normal rats after free water intake and after dehydration induced by water deprivation for 24 h. The decreased ability of the DI rats to concentrate urine led to plasma hypernatremia and hyperosmolality despite excessive drinking. Water deprivation in the DI rats induced

The direct measurements of AVP during water deprivation and salt loading demonstrate the inability of the direct water deprivation test to distinguish accurately between several forms of polyuria. Polyuria and polydipsia are commonly formed disorders in the dog and can be caused by osmotic diuresis; deficient release of anti-diuretic hormone-arginine vasopressin (AVP); a decreased renal response to AVP; excessive water intake. The differentiation between these forms often requires a water deprivation test followed by administration of vasopressin. The latter test is an indirect one and relies

Clinical depression is often characterized by a loss of interest or pleasure in formerly enjoyable activities. Analogs of anhedonia are established in rats, but the generality of this phenomenon to other species is unknown. Maternally-deprived rhesus macaques show a wide range of behavioral abnormalities that are reversed by chronic antidepressant treatment. We tested consumption by maternally deprived versus control macaques of sweetened (seven sucrose concentrations) or bitter water (four quinine concentrations) versus plain water to evaluate a non-human primate model of depression for signs

Effects of the pyrazoloquinoline CGS 8216 (a partial benzodiazepine receptor inverse agonist) and the opiate antagonist, naltrexone, were compared in several tests of ingestion in non-deprived and deprived male rats. Both naltrexone (0.1-10.0 mg/kg, SC) and CGS 8216 (1.25-10.0 mg/kg, IP) significantly reduced the consumption of a highly palatable saccharin-glucose solution by non-deprived rats. Both compounds were also effective in reducing, dose-dependently, the intake of palatable sweet or oily mash by non-deprived animals. Hence, naltrexone and CGS 8216 attenuated palatability-induced inges

The use of fat to support the energy needs of reproduction (i.e. capital breeding) has been studied in a diversity of taxa. However, despite reproductive output (i.e. young or eggs) being approximately 70% water, little is known about the availability of internal resources to accommodate the hydric demands of reproduction. Recent research suggests that dehydration increases the catabolism of muscle as a means of maintaining water balance. Accordingly, we investigated the interactive effects of reproductive investment and water deprivation on catabolism and reproductive output in female Childre

This study analyzed the effects of systemic oxytocin (OT) administration and 48-h food deprivation on the polydipsia, hyperphagia, and polyuria produced by electrolytic lesions of the mediobasal hypothalamus (MBH). In a first experiment, food deprivation transiently decreased the polydipsic response, whereas food deprivation plus OT administration reduced the water intake and urine excretion of polydipsic animals but not their subsequent food intake. These results were replicated in a second experiment (20 days), which also showed that OT potentiates sodium excretion, reducing the estimated pl

Primary polydipsia is characterized by a marked increase in water intake and secondary polyuria, and in dogs often is described as a behavioral problem or a psychological disorder. We describe 4 dogs with primary polydipsia, diagnosed on the basis of a modified water deprivation test, in which further examination included serial measurements of urine osmolality (UOsm) and plasma vasopressin (VP) measurements during water deprivation and hypertonic saline infusion. The dogs, ranging in age from 4 months to 4 years, all were presented for evaluation of polyuria and polydipsia. Physical examinati

Colchicine was injected into the region of the organum vasculosum lamina terminalis/anterior region of the third cerebral ventricle (OVLT/AV3V) to produce a temporary disruption of the nervous connections of this area and the rest of the anterior hypothalamus, but to maintain the vascular connections intact. Rats were kept in metabolism cages throughout the experiment and food and water intake plus urine and electrolyte excretion and body weight were measured each day. Food intake, body weight gain and urine and sodium excretion were reduced for several days after the injection of colchicine a

The physiological stimulus for deprivation-enhanced ingestion was studied in developing rats. During an overnight deprivation period, continuous gastric infusions of isotonic saline or milk were made to 6- and 15-day-old rat pups in order to preferentially maintain hydrational or hydrational and nutritional status, respectively. Pups' ingestion was then studied in oral-infusion tests. In 6-day-old pups that received either milk or saline infusions, ingestion was depressed relative to intake in pups that were simply deprived. But in 15-day-old pups, only milk infusions reduced intake. These fin

This study investigated the effects of excess cortisol on physiological mechanisms that resist dehydration in Bos indicus steers (n = 31, 2 yr of age, 193 +/- 21.47 kg mean BW) during a 90-h period. Steers were assigned randomly to one of four groups: 1) no water/no cortisol (n = 8), 2) water/no cortisol (n = 8), 3) no water/cortisol (n = 8), and 4) water/cortisol (n = 7). Animals allocated to cortisol treatment groups were given 0.1 mg x kg BW(-1) x h(-1) of hydrocortisone suspended in isotonic saline for the duration of the study. Total body water, osmolality, hematocrit, urine output, feed
