Cardiovascular adaptations, fluid shifts, and countermeasures related to space flight.

Significant progress has been made related to understanding cardiovascular adaptations to microgravity and development of countermeasures to improve crew re-adaptation to gravity. The primary ongoing issues are orthostatic intolerance after flight, reduced exercise capacity, the effect of vascular-smooth muscle loss on other physiologic systems, development of efficient and low-cost countermeasures to counteract these losses, and an understanding of fluid shift mechanisms. Previous animal studies of cardiovascular adaptations offer evidence that prolonged microgravity remodels walls of blood v

[Countermeasuring effect of lower body negative pressure against orthostatic intolerance induced by 21 d -6 degrees head down tilt in humans].

Objective. To investigate whether LBNP during 21 d -6 degrees head-down tilt (HDT) would modify orthostatic tolerance. Method. 12 healthy males, age 23.7 +/- 5.0, were exposed to -6 degrees HDT for 21 d. 6 of them received -4.0 kPa LBNP sessions for 1 h/d from 15th day to 21st day of HDT. The other 6 served as control. HUT + 75 degrees, 20 min orthostatic tolerance test were done before, on day 10 and on day 21 of HDT. Result. During HUT + 75 degrees, 20 min orthostatic tolerance test on day 10 of HDT, 5 subjects of the control group and 4 of the LBNP group presented presyncopal or syncopal si

Atractylodes macrocephala polysaccharide AMP1-1 ameliorates weightless-induced cognitive dysfunction via the microbiota-gut-brain axis.

Atractylodes macrocephala Koidz., a traditional Chinese medicine known for "strengthening the spleen and replenishing qi", has been recognized for its gastrointestinal protective and immunomodulatory properties, suggesting a potential role in gut-brain axis regulation. This study investigated the protective effect and underlying mechanism of a purified inulin-type polysaccharide from Atractylodes macrocephala, AMP1-1, against weightlessness-induced cognitive impairment via the microbiota-gut-brain axis. Using a tail-suspension rat model to simulate microgravity, we assessed: (i) cognitive func

LBNP-induced changes in plasma cGMP with and without head down tilt bed rest.

This investigation was conducted to test the following hypotheses: 1) If simulated orthostasis (LBNP) reduces plasma cGMP; 2) if simulated microgravity alters any such LBNP-induced effect; and 3) if simulated microgravity reduces resting plasma cGMP levels. In addition, we studied the time-course of thoracic impedance during, LBNP and asked if there is heart rate/blood pressure reduction after LBNP. During real and simulated spaceflight, blood is re-distributed throughout the vasculature along the body axis, vascular mechanoreceptor loads are altered, "excess" fluid is lost from the organism,

Influence of suspension on the expression of protective immunological memory to murine Listeria monocytogenes infection.

Female BDF1 mice were immunized with virulent Listeria monocytogenes, which resulted in the generation of a long-lived state of protective immunological memory for this facultative intracellular bacterial pathogen. The influence of antiorthostatic suspension, a ground-based model employed to simulate certain aspects of weightlessness that occur during spaceflight, on the capacity of these mice to express memory immunity was evaluated. Memory-immune mice were suspended by the tail in the orthostatic or antiorthostatic position and were reinfected with a lethal dose of virulent L. monocytogenes.

A direct approach to the study of the effect of gravity on axis formation in birds.

A system has been developed to enable the normal development of aborted very early uterine avian embryos, outside the female's uterus. The shell-less aborted egg was put into a foster shell of a sister egg, previously laid by the same female. The empty space between the shell and aborted egg was filled with artificial uterine fluid. The reconstructed eggs were incubated at 42 degrees C for 30 hours in a vertical position. The atmosphere contained a high concentration of CO2 (8-10%). At the termination of the 30 h the eggs were transferred to incubation at 37 degrees C in normal atmospheric con

Ultrasound evaluation of sinus fluid levels in swine during microgravity conditions.

BACKGROUND: Acute rhinosinusitis is a common problem that could occur in space secondary to absence of gravity-dependent drainage or odontogenic or external sources of infection. The purpose of this study was to determine the efficacy of ultrasound to determine sinus fluid distribution levels in swine and to assess the accuracy of ultrasound in the animal during normal and microgravity conditions. METHODS: Anesthetized swine had a catheter placed through a frontal bone window to allow aliquots of a viscous solution to be injected at 1 G (N = 4) or during brief microgravity parabolic flights (

From aging to space: A comparative biology of skeletal muscle degeneration.

Sarcopenia, the progressive loss of skeletal muscle mass and function with age, represents a major clinical concern, particularly in the context of disuse and unloading conditions such as simulated microgravity. This review explores the molecular, cellular, and physiological responses of skeletal muscle to simulated microgravity and compares them with those observed during aging-associated sarcopenia. A central focus is placed on impaired excitation-contraction coupling, altered calcium homeostasis, and dysregulation of signalling pathways critical for muscle maintenance. Simulated microgravit

Simulated microgravity predisposes kidney to injury through promoting intrarenal artery remodeling.

Spaceflight-induced multi-organ dysfunction affects the health of astronauts and the safety of in-orbit flight. However, the effect of microgravity on the kidney and the underlying mechanisms are unknown. In the current study, we used a hindlimb unweighting (HU) animal model to simulate microgravity and employed histological analysis, ischemia-reperfusion experiments, renal ultrasonography, bioinformatics analysis, isometric force measurement, and other molecular experimental settings to evaluate the effects of microgravity on the kidneys and the underlying mechanisms involved in this transiti

Effect of simulated microgravity on testosterone and sperm motility in mice.

We examined changes in the serum testosterone level and in sperm in the testis and epididymis by using tail-suspended mice, which are a simulation model of the body fluid shift in space, to evaluate the possibility of spermatogenesis failure in space environment. We also studied pathological disorders of the testis in the tail-suspended mice. Tail suspension was imposed with a tail harness to a degree at which the hindlegs of mice did not touch the floor of the housing unit. In control mice, the tail was similarly fixed with a tail harness to impose the same stress, except that a hindleg remai

Clinorotation-induced weightlessness influences the cytoskeleton of glial cells in culture.

During and after spaceflight astronauts experience neurophysiological alterations. To investigate if the impairment observed might be traced back to cytomorphology, we undertook a ground based research using a random positioning machine (clinostat) as a simulation method for microgravity. The outcome of the study was represented by cytoskeletal changes occurring in cultured glial cells (C(6) line) after 15 min, 30 min, 1 h, 20 h and 32 h under simulated microgravity. Glia is fundamental for brain function and it is essential for the normal health of the entire nervous system. Our data showed t

Simulated microgravity triggers a membrane adaptation to stress in E. coli REL606.

Investigating the evolution of Escherichia coli in microgravity offers valuable insights into microbial adaptation to extreme environments. Here the effects of simulated microgravity (SµG) on gene expression and genome evolution of E. coli REL606, a strain evolved terrestrially for 35 years, is explored. The transcriptomic changes for glucose-limited and glucose-replete conditions over 24 h illustrate that SµG increased the expression of genes involved in stress response, biofilm, and metabolism. A greater number of differentially expressed genes related to the general stress response (GSR) an

Loss and recovery of myocardial mitochondria in mice under different tail suspension time: Apoptosis and mitochondrial fission, fusion and autophagy.

Long-term weightlessness in animals can cause changes in myocardial structure and function, in which mitochondria play an important role. Here, a tail suspension (TS) Kunming mouse (Mus musculus) model was used to simulate the effects of weightlessness on the heart. We investigated the effects of 2 and 4 weeks of TS (TS2 and TS4) on myocardial mitochondrial ultrastructure and oxidative respiratory function and on the molecular mechanisms of apoptosis and mitochondrial fission, autophagy and fusion-related signalling. Our study revealed significant changes in the ultrastructural features of car

Radix astragali and orthostatic response: a double-masked crossover study.

AIM: Recent results from animal experiments have shown that radix astragali (RA), a traditional Chinese herbal tonic, alleviates muscle atrophy under simulated weightlessness conditions, rendering RA a candidate for human use as a countermeasure against muscular atrophy. Possible cardiovascular side effects have not yet been investigated. We analyzed the effects of RA on the orthostatic stability of healthy men. METHODS: There were 10 test subjects who were assigned to a double-blinded, randomized crossover design using RA or placebo (PL) for 14 d each, respectively. Test runs were separated

Microgravity Modulates Effects of Chemotherapeutic Drugs on Cancer Cell Migration.

Microgravity or the condition of apparent weightlessness causes bone, muscular and immune system dysfunctions in astronauts following spaceflights. These organ and system-level dysfunctions correlate with changes induced at the single cell level both by simulated microgravity on earth as well as microgravity conditions in outer space (as in the international space station). Reported changes in single bone cells, muscle cells and white blood cells include structural/morphological abnormalities, changes in gene expression, protein expression, metabolic pathways and signaling pathways, suggesting

Long non-coding RNAs Kcnq1ot1 and Lncpint are involved in skeletal muscle atrophy induced by the space exposome.

Long non-coding RNAs (lncRNAs) play an important role in the regulation of skeletal muscle transcriptional processes, but their involvement in spaceflight- or inactivity-induced muscle atrophy remains poorly understood. To address this gap we simulated the space environment by combining microgravity, irradiation and stress in a mouse model. This simulation resulted in the differential expression (threshold set at P < 0.01) of 6191 protein-coding genes (3525 downregulated and 2666 upregulated compared to controls) and 465 lncRNAs, of which 27% were downregulated and 73% upregulated compared to

The impact of skeletal unloading on bone formation.

Skeletal unloading leads to decreased bone formation and decreased bone mass. Bone resorption is uncoupled from bone formation, contributing to the bone loss. During space flight bone is lost principally from the bones most loaded in the 1 g environment. Determining the mechanism(s) by which loading of bone is sensed and translated into a signal(s) controlling bone formation remains the holy grail in this field. It seems likely that matrix/cell interactions will underlie much of the mechanocoupling. Integrins are a prime mediator of such interactions. The role for systemic hormones such as PTH
