Effect of a novel xanthine derivative on experimental ulcers in rats.

The effects of the novel xanthine derivative 3-ethyl-1-(6-hydroxy-6-methylheptyl)-7-propylxanthine (A90 6119, CAS 134072-58-5) on various experimentally induced ulcers was investigated in rats. A90 6119 produced a dose-dependent inhibition of gastric ulcers induced by water immersion stress and absolute ethanol with ED50- values of 2.4 and 2.8 mg/kg, p.o., respectively. The erosion induced by oral administration of 1.5% NH4OH (3 ml/rat) was significantly reduced by A90 6119 at 10 and 30 mg/kg, p.o. Likewise, A90 6119 caused a dose-dependent inhibition of gastric erosions and intestinal hemorrh

Bronchodilatory activity and pharmacokinetics of new xanthines in guinea-pigs.

1. The in vitro biological activities and the effect of protein binding on the relaxant effects in vivo of N-3-alkylxanthine and N-3-alkyl-N-1-methylxanthine derivative were investigated in guinea-pigs. 2. A significantly positive correlation was observed among the in vitro muscle relaxant activity, the cyclic adenosine monophosphate (cAMP) phosphodiesterase (PDE) inhibitory activity and the protein-binding potency of xanthine derivatives. However, there was a weak relationship between these activities and affinity for adenosine receptors. 3. When theophylline, enprofylline and 1-methyl-3-prop

[An aged case of hereditary xanthinuria with xanthine urinary calculi].

A 74-year-old female with hereditary xanthinuria and xanthine stones is reported. She has a family history of consanguineous parents and a past history of right side nephrectomy due to a xanthine renal stone and vesicolithotomy of 3 bladder stones approximately 5 X 4 X 4 cm in size at the age of 58 and 71, respectively. Her young brother exhibited a slightly elevated urinary excretion of oxypurines. Laboratory examination showed a low serum level (0.3 mg/dl) and urinary excretion (1.56 mg/day) of uric acid, and high plasma and urine levels of oxypurines. No xanthine oxidase activity was detect

Metabolism of pyrazinamide and allopurinol in hereditary xanthine oxidase deficiency.

The metabolism of pyrazinamide and allopurinol was studied in three xanthinuric patients from two families with hereditary xanthinuria to determine whether both substrates were oxidized only by xanthine oxidase or by other oxidases as well. One xanthinuric patient could neither metabolize pyrazinamide into 5-hydroxypyrazinamide nor allopurinol into oxypurinol. Two xanthinuric patients could metabolize both pyrazinamide into 5-hydroxypyrazinamide and allopurinol into oxypurinol but could not oxidize pyrazinoic acid to 5-hydroxypyrazinoic acid. These findings suggest that xanthinuria comprises a

Efficacy of enprofylline, a new bronchodilating xanthine, in acute asthma.

Five different doses of enprofylline, a 3-propylxanthine, were compared with a standard dose of theophylline in 135 episodes of acute asthma in a double-blind study. The drugs were administered intravenously during ten minutes. Enprofylline induced dose-related bronchodilation and the 2.5 mg/kg dose produced significantly better effects than theophylline.

[Xanthines in apnea of premature infants. Influence on gastroesophageal reflux].

Theophylline and caffeine are two xanthine-derivated drugs frequently administered for their stimulating effects on the respiratory center in premature babies presenting with "idiopathic apnea". These drugs are known to increase the gastric acid secretion and to decrease the lower esophageal sphincter pressure, that in turn possibly increase gastroesophageal reflux. We studied 14 premature babies presenting with "idiopathic apnea", treated with caffeine at recommended dose (2.5 mg/kg/day). At 24 hour continuous esophageal pH monitoring was performed 3 to 5 days after starting the treatment. In

Relationships between adenosine, cyclic nucleotides, and xanthines in asthma.

Methylxanthines have been used for the treatment of asthma for more than 60 years, but their mechanism of action is poorly understood. Their ability to inhibit cyclic adenosine monophosphate phosphodiesterase has attracted much attention. However, this is clearly demonstrable only in high doses and is more likely to be related to toxicity. An alternative mechanism is antagonism of adenosine receptors in the lung. Adenosine has been shown to be released in asthma and cause bronchoconstriction in patients with asthma. Its effects are selectively inhibited by concentrations of theophylline that d

Copper-catalyzed xanthine-chalcogenide hybrids: synthesis, DNA binding and docking.

Copper-catalyzed C(sp2)-H chalcogenylation was employed for the efficient synthesis of a series of xanthine-chalcogenide hybrids using diaryl dichalcogenides as chalcogen sources. The reactions proceeded under mild conditions, providing the desired selenium- and sulfur-containing xanthine derivatives in moderate to good yields. The structures of the synthesized compounds were unambiguously confirmed by spectroscopic techniques, including 1H- and 13C-NMR spectroscopy. The DNA-binding behavior of the obtained hybrids was investigated by UV-visible absorption titration with calf thymus DNA, revea

Xanthine Oxidase Pathway and Muscle Damage. Insights from McArdle Disease.

The intent of this article is to summarize current body of knowledge on the potential implication of the xanthine oxidase pathway (XO) on skeletal muscle damage. The possible involvement of the XO pathway in muscle damage is exemplified by the role of XO inhibitors (e.g., allopurinol) in attenuating muscle damage. Reliance on this pathway (as well as on the purine nucleotide cycle) could be exacerbated in conditions of low muscle glycogen availability. Thus, we also summarize current hypotheses on the etiology of both baseline and exertional muscle damage in McArdle disease, a condition caused

Xanthine oxidase inhibitory and antioxidant potential of Indian Muscodor species.

Xanthine oxidase is a key enzyme responsible for hyperuricemia, a pre-disposing factor for Gout and oxidative stress-related diseases. Only two clinically approved xanthine oxidase inhibitors Allopurinol and Febuxostat are currently used for treatment of hyperuricemia. However, owing to their side effects there is a need for new non-purine-based selective inhibitors of xanthine oxidase. In the process of exploring novel xanthine oxidase inhibitors and anti-oxidants, we screened the culture filtrate of 07 novel species of Muscodor, a sterile endophytic fungi isolated from Cinnamomum and Aegle m

Xanthine oxidase inhibitory activity of extracts prepared from Polygonaceae species.

The xanthine oxidase (XO) inhibitory activity of aqueous and organic extracts of 27 selected species belonging in five genera (Fallopia, Oxyria, Persicaria, Polygonum and Rumex) of the family Polygonaceae occurring in the Carpathian Basin were tested in vitro. From different plant parts (aerial parts, leaves, flowers, fruits and roots), a total of 196 extracts were prepared by subsequent extraction with methanol and hot H2O and solvent-solvent partition of the MeOH extract yielding n-hexane, chloroform and 50% MeOH subextracts. It was found that the chloroform subextracts and/or the remaining

The xanthine oxidase inhibitor oxypurinol reduces cancer cachexia-induced cardiomyopathy.

BACKGROUND: Cachexia is a common complication of cancer and may be responsible for 22% of all cancer-related deaths. The exact cause of death in cancer cachexia patients is unknown. Recently, atrophy of the heart has been described in cancer cachexia animal models, which resulted in impaired cardiac function and is likely to contribute to mortality. In cancer patients hyperuricaemia independent of tumour lysis syndrome is often associated with a worse prognosis. Xanthine oxidase (XO) metabolizes purines to uric acid and its inhibition has been shown to improve clinical outcome in patients with

Identification of xanthine oxidase activity following reperfusion in human tissue.

In this series of experiments, we surveyed xanthine oxidase activity after microvascular transfer in the venous effluent after reperfusion of human rectus abdominis muscle (n = 8) and jejunum (n = 4). Enzyme activity was correlated with duration of ischemia and biochemical markers of cellular injury. Xanthine oxidase (XO) activity was measured spectrofluorometrically using a pterin assay, whereas cellular injury was measured with commercial creatinine phosphokinase activity assay and lipid peroxidation products using a spectrophotometer. The data demonstrated that XO activity was statistically

Pyrazinamide-induced hepatotoxicity mediated by aldehyde oxidase and xanthine oxidase.

Pyrazinamide (PZA) has been approved for the treatment of tuberculosis in clinical practice. However, its adverse effects, particularly hepatotoxicity, have raised concerns. The present study aimed at exploring the potential relationship between PZA-induced hepatotoxicity and its metabolites resulting from metabolic activation. Glutathione (GSH) conjugates with confirmed structures were detected in mouse cytosol incubations containing PZA or pyrazinoic acid (POA, a major metabolite of PZA) supplemented with glutathione (GSH). Such GSH metabolites were also observed in both liver homogenates fr

Xanthine oxidase inhibitors for gout: Applications and novel drug development.

Hyperuricemia is a well-established direct trigger of gouty arthritis. Xanthine oxidase (XO), the rate-limiting enzyme in uric acid production, represents a core therapeutic target for gout. Although XO inhibitors like allopurinol and febuxostat are widely-used clinically, their limitations, including severe hypersensitivity and cardiovascular risks, necessitate the continuous pursuit of safer and more efficient agents. This review provides a systematic and critical assessment of recent advances in XO inhibitor research, proposing for the first time a unified classification framework based on

Synthetic heterocyclic derivatives as promising xanthine oxidase inhibitors: An overview.

Inhibition of xanthine oxidase (XO) is an effective and most prominent therapeutic approach for the management of gout. Discovery of its association in the pathophysiology of diabetes, cardiovascular disorders, etc., widened its therapeutic horizons. Limited drug candidates in clinical practice along with side effects forced researchers to develop more efficacious and safer XO inhibitors for the management of gout and other disorders associated with XO hyperactivity. In this regard, this review focus on (a) various drug candidates in clinical practice and under clinical trials, (b) Development
