Serum xanthine oxidase: origin, regulation, and contribution to control of trypanosome parasitemia.

African trypanosomiasis is caused by Salivarian trypanosomes, tsetse fly-transmitted protozoa that inhabit the blood plasma, lymph and interstitial fluids, and, in the case of Trypanosoma brucei species, also the cerebrospinal fluid of mammal hosts. Trypanosomiasis in people and domestic animals manifests as recurring waves of parasites in the blood and is typically fatal. In contrast, trypanosomiasis in Cape buffaloes, which are naturally selected to resist the disease, is characterized by the development of only one or a few waves of parasitemia, after which the infection becomes cryptic, be

Xanthine oxidase inhibitors suppress testicular germ cell apoptosis induced by experimental cryptorchidism.

Apoptotic degeneration of germ cells in cryptorchid testis is associated with an increased level of reactive oxygen species, and may be prevented by antioxidant treatment. The objective of this study was to investigate whether xanthine oxidase inhibitors could confer such protection. Unilateral cryptorchidism was surgically induced in the immature rats, which were then left untreated or treated with xanthine oxidase inhibitors, and the testes were evaluated 7 days after the operation. In the control group, the weight of the cryptorchid testis was decreased to 47% of that of the contralateral s

Gut ischemia mediates lung injury by a xanthine oxidase-dependent neutrophil mechanism.

Neutrophils (PMNs) are believed to play a key role in the pathogenesis of postinjury adult respiratory distress syndrome. We have previously shown that gut ischemia/reperfusion (I/R) produces lung injury by a process that requires PMNs. More recently, we have shown that xanthine oxidase (XO) plays a role. The purpose of this study was to characterize the mechanistic sequencing of XO activity versus the PMN in this model of gut I/R-induced lung injury. Normal and XO-inactivated (tungsten enriched, molybdenum depleted diet) rats underwent 45 min of superior mesenteric artery occlusion. After 6 h

Effect of xanthine-related compounds on a theophylline assay using theophylline oxidase.

OBJECTIVE: This study investigated a new, nonimmunologic, enzymatic-based theophylline assay for the possible interference of a clinically achievable serum theophylline concentration by four xanthine-related compounds: pentoxifylline, 1-methylxanthine, allopurinol, and oxypurinol. RATIONALE: The enzyme theophylline oxidase is used in this assay kit to convert theophylline to 1,3 dimethyluric acid in the presence of cytochrome C. Cytochrome C is reduced to ferrocyte C, which is then measured by ultraviolet spectrophotometry at one minute and again at five minutes. The rate of appearance of fer

Targeted degradation of xanthine oxidase via PROTAC technology for the treatment of hyperuricaemia.

Hyperuricemia, characterized by an elevated serum uric acid level due to the abnormal xanthine oxidase (XOD)-mediated uric acid production, is generally associated with multiple systemic disorders. In this study, XOD-targeting proteolysis-targeting chimera (PROTAC) is developed to degrade the XOD and investigate the subsequent cellular behaviors. Febuxostat is a selective XOD inhibitor, designed to target XOD, and thalidomide functions as a ligand for the Cereblon (CRBN) E3 ubiquitin ligase. The two moieties are covalently linked via a polyethylene glycol (PEG) spacer. The synthesized chimera,

The activities of serum adenosine deaminase and xanthine oxidase enzymes in Behcet's disease.

BACKGROUND: Adenosine deaminase (AD) and xanthine oxidase (XO) are enzymes of purine catabolism that catalyze the conversion of adenosine to inosine, deoxyadenosine to deoxyinosine, hypoxanthine to xanthine and xanthine to uric acid, respectively. AD is known to be an important enzyme in the maturation and function of T lymphocytes. The aim of this prospective study was to evaluate whether there are changes in serum AD activity as an index of T lymphocyte function in Behcet's disease (BD) which is known as having T cell-mediated immune response. METHODS: A total of 32 patients and 26 sex- and

Cytotoxicity of, and DNA damage by, active oxygen species produced by xanthine oxidase.

Toxicity to Raji cells of the xanthine oxidase/hypoxanthine system is related to the formation of single-strand DNA breaks. DNA damage was proportional to the concentration of xanthine oxidase and to the time of exposure. It was prevented by the absence of hypoxanthine, or by the presence of allopurinol, or both superoxide dismutase and catalase. The release of 51Cr from damaged cells was detectable 12 h after the inhibition of cloning efficiency and the production of DNA breakage. These data suggest that DNA damage induced by the oxygen products precedes the severe lesion to the cellular memb

Role of xanthine dehydrogenase and oxidase in focal cerebral ischemic injury to rat.

The role of xanthine dehydrogenase and oxidase as a source of free radicals contributing to focal cerebral ischemic injury was evaluated in Long-Evans rats after the middle cerebral artery was permanently occluded and both carotid arteries were clamped for 90 min. The fraction of xanthine dehydrogenase present as the free radical producing oxidase increased slightly from 22% in control cortex to 30% in the ischemic right cortex during the first 3 h of reperfusion and then remained relatively unchanged over the next 24 h. This increase may in part be due to entrapped plasma, which contained 4.5

NAD+ deficiency plays essential roles in the hyperuricemia of stroke-prone spontaneously hypertensive rat via xanthine dehydrogenase to xanthine oxidase conversion.

Inhibition of xanthine oxidoreductase (XOR) was shown to ameliorate the stroke susceptibility in the stroke-prone spontaneously hypertensive rat (SHRSP), suggesting hyperuricemia had a pathological role in this rat model. In this study, we thus aimed to explore mechanisms inducing hyperuricemia in SHRSP. XOR is known to have two forms, xanthine dehydrogenase (XDH) as the prototype and xanthine oxidase (XO) as the converted form through cleavage of a peptide bond or through formation of disulfide bonds in the enzyme. XO was shown to have a greater activity to produce UA and oxidative stress. We

Maternal Electronic Cigarette Inhalation Exposure During Gestation: Impacts on Prolactin and Xanthine Oxidase.

Pregnancy requires metabolic and endocrine changes that must occur to support fetal growth and development. Aberrations in these necessary modifications can impact maternal health and fetal growth. Exposure to toxicants during pregnancy can negatively affect fetal health and development, but studies on electronic cigarette (e-cig) exposure is limited. We hypothesized that maternal e-cig exposure during gestation leads to hormonal and redox imbalance and negatively impacts fetal development. Pregnant Sprague-Dawley rats were exposed to e-cig aerosols (1227 ± 131 mg/m3) or HEPA-filtered air for

Searching for potential xanthine oxidase inhibitors from Vietnamese herb plants via computational studies.

Gout, a prevalent form of inflammatory arthritis, affects individuals of all ages and is increasing worldwide. It is closely associated with hyperuricemia, a condition caused by excessive uric acid production. Xanthine oxidase (XO) inhibitors play a key role in managing hyperuricemia, making the discovery of new and effective inhibitors a priority in drug development. In this study, computational methods were employed to screen natural compounds for their potential to inhibit XO. A total of 4947 compounds derived from 71 Vietnamese medicinal herbs were subjected to molecular docking simulation

Uric acid and xanthine oxidase in heart failure - Emerging data and therapeutic implications.

The role of hyperuricaemia as cardiovascular risk factor has exhaustingly been debated for decades. While the association of elevated uric acid (UA) levels with increased mortality risk as convincingly been shown, the question whether UA is independently predictive of just a related effect within a more complex risk factor profile (including metabolic, inflammatory and haemodynamic risk factors) is still a matter of dispute. In heart failure the independent prognostic and functional impact of elevated UA has not only been shown but also the pathophysiologic mechanism(s) and the potential of ta

The xanthine oxidase-NFAT5 pathway regulates macrophage activation and TLR-induced inflammatory arthritis.

NFAT5 (nuclear factor of activated T cells), a well-known osmoprotective factor, can be activated by isotonic stimuli such as Toll-like receptor (TLR) triggering. However, it is unclear how NFAT5 discriminates between isotonic and hypertonic stimuli to produce different functional and molecular outcomes. Here, we identified a novel XO-ROS-p38 MAPK-NFAT5 pathway (XO is xanthine oxidase, ROS is reactive oxygen species) that is activated in RAW 264.7 macrophages upon isotonic TLR stimulation. Unlike what is seen under hypertonic conditions, XO-derived ROS were selectively required for the TLR-ind

Xanthine oxidase and lens oxidative stress markers in diabetic and senile cataract patients.

Xanthine oxidase (XOD) is a prooxidant enzyme possibly implicated in diabetic lens injury and genesis of senile cataract (SC). We evaluated the impact of diabetes on XOD activity and its relationships with lens oxidative stress markers in patients operated on for SC. Serum and lens XOD activities, lens malondialdehyde (MDA), conjugated dienes, superoxide dismutase (SOD), glutathione peroxidase (GPx) and reduced glutathione (GSH) levels were measured in 62 non-diabetic and 29 diabetic patients operated on for SC. Lens XOD, SOD, GPx and GSH levels were gradually declining, while MDA and serum XO

Inhibition of xanthine oxidase by some Chinese medicinal plants used to treat gout.

The enzyme xanthine oxidase catalyses the oxidation of hypoxanthine to xanthine and then to uric acid, which plays a crucial role in gout. A total of 122 traditional Chinese medicinal plants, selected according to the clinical efficacy and prescription frequency for the treatment of gout and other hyperuricemia-related disorders, have been evaluated for the enzyme inhibitory activity. Among the 122 methanol extracts derived from these species, 69 were shown to be inhibitory at 100 microg/ml, with 29 having greater than 50% inhibition. As to the equal amount of water extracts, 40 were disclosed

Xanthine oxidase inhibitory activity of northeastern North American plant remedies used for gout.

Xanthine oxidase (xanthine: oxygen oxidoreductase EC 1.2.3.2) inhibitory activity was assayed from 26 species belonging to 18 families traditionally used for the treatment of gout and related symptoms by Indigenous people of northeastern North America. The degree of inhibition was determined by measuring the increase in absorbance at 295 nm associated with uric acid formation. Eighty-eight percent of the plants were found to have inhibitory activity at 100 microg/ml, with 20% having greater than 50% inhibition. Larix laricina exhibited the highest activity with an inhibition of 86.33%. Of the

Subcellular distribution of xanthine oxidase during cardiac ischemia and reperfusion: an immunocytochemical study.

Oxygen-derived free radicals are known to take part in cardiac injury during post-ischemic reperfusion (I/R). Xanthine oxidase (XO) is closely associated with the generation of superoxide radicals. We have determined the distribution of XO in rat myocardium after ischemia (I) and I/R by immunocytochemical method using murine monoclonal antibody against XO (bovine milk) and by enzyme histochemistry (EHC) in situ. Frozen sections of periodate-lysine-paraformaldehyde (PLP) fixed myocardium after 15, 60 and 90 min ischemia and 15 min ischemia and 30 min reperfusion were processed for immunocytoche

Decreased xanthine oxidase activities and increased urinary oxypurines in heterozygotes for hereditary xanthinuria.

Two brothers with hereditary xanthinuria (xanthine oxidase deficiency) and several members of their family were studied. In both subjects, plasma and urinary concentrations of uric acid were low whereas xanthine and hypoxanthine concentrations were markedly elevated. Xanthine oxidase activity was virtually absent in the patients' duodenal mucosa, a finding that established the diagnosis of hereditary xanthinuria. In their parents (obligate heterozygotes), the duodenal xanthine oxidase activity was about 50% of that in control subjects (father 9.3 and mother 12.8 mU/g tissue compared with 21.3
