A subpopulation of nervus terminalis neurons projects to the olfactory mucosa in Xenopus laevis.

Biocytin application to the normal or zinc sulfate-treated nasal cavity of Xenopus laevis was used to trace retrogradely neurons associated with the terminal nerve (TN). Immunocytochemistry was conducted to identify the relationship of gonadotropin-releasing hormone-immunoreactive (GnRH-ir) TN neurons with biocytin-labeled neurons. Neurons that accumulated biocytin were located in olfactory nerve fascicles close to the olfactory mucosa lining the caudal, medial, and rostral walls of the principal cavity. GnRH-ir fibers were observed only in the olfactory nerve fascicle projecting to the rostra

Pharmacological characterization of the CGRP receptor in the lateral line organ of Xenopus laevis.

Calcitonin gene-related peptide (CGRP) is a neurotransmitter candidate colocalized with acetylcholine in efferent fibers innervating hair cell organs. We have used the Xenopus laevis lateral line organ to investigate the responses of a hair cell organ to the CGRP family of peptides. Two isoforms of CGRP, r alpha-CGRP and r beta-CGRP, and a human analog of alpha-CGRP, h(Tyro) alpha-CGRP, produced dose-dependent increases in afferent nerve fiber discharge rate with EC50 values of approximately 1 microM. Rate increases were 31.2, 18.9, and 10.3%, respectively. The peptide fragment rCGRP8-37 a sel

[Regeneration of the optic nerve of Xenopus laevis after argon laser injury].

Following mechanical lesion of the optic nerve (ON), lower vertebrates are capable of regeneration of the ON and regain visual function. We have studied, by light and electron microscopy, regeneration of the larval ON of Xenopus laevis after treatment with the argon laser. The laser energy is absorbed by the neurothelial pigment and provokes a burned crater and vesiculation in the interior of the ON. In contrast to the mechanically induced process, axons regenerate, after laser treatment, within the nerve and do not sprout into the neighboring tissue. The regenerating ON is not vascularized. I

Characterization of the fourth component of complement in the serum of the clawed frog Xenopus laevis.

The 202,000 MW protein XC, which is distinct from C3 but is essential for antibody-dependent haemolytic activity against SRBC, was obtained from Xenopus plasma following polyethylene glycol precipitation, ion-exchange chromatography and gel-filtration. The protein required other components of the Xenopus serum in order to lyse sensitized SRBC, but did not lyse unsensitized RRBC through the alternative pathway. The protein, contained at 0.17 mg/ml in the original plasma, comprised three distinct subunits of 96,000, 76,000 and 26,000, which were linked by disulphide bonds. Digestion by trypsin r

[Glomus cell in controlling vascular tone of the carotid labyrinth (Xenopus laevis)].

To clarify the physiological significance of g-s connection (intimate apposition of the glomus cell to the smooth muscle), vascular responses of the carotid labyrinth to both catecholamines and chemoreceptor stimulants were investigated using Xenopus laevis. The results obtained were as follows. Density of dense-cored vesicles in the glomus cell was significantly varied in three different parts of the cytoplasm (N, M, P). In the part of the cytoplasm containing the nucleus (N) the density was lowest, and in the processes (P) it was highest. This bias in distribution was intensified by efferent

Effects of light-deprivation on development of photopositive behavior in Xenopus laevis tadpoles.

The development of amphibian sensory systems and behavior is generally considered to proceed normally without reference to sensory experience during embryonic or larval stages. Most of the supporting research, however, has concentrated on the retinotectal (visual) systems of anurans and has ignored behaviors directed by other sensory systems. We demonstrate that early exposure to light is necessary for the development of photopositive behavior in Xenopus laevis tadpoles, a behavior probably directed by the pineal complex. Light-deprivation during the tadpoles' first 10 days of development resu

Ocular microvasculature in adult Xenopus laevis: Scanning electron microscopy of vascular casts.

The microvascular anatomy of choriocapillaris, iris, ciliary body, and superficial vascular hyaloid system of eyes was studied in the permanent aquatic Xenopus laevis by scanning electron microscopy of vascular casts and was compared with that published in two semiaquatic ranid species (Rana esculenta and Rana temporaria), and the urodelian species Triturus criststus carnifex. Results showed that the choriocapillaris in Xenopus consisted of a dense meshwork of wide capillaries displaying polygonal arrays at the scleral side with venules leaving the centers and arterioles supplied from the peri

Translational profiling of retinal ganglion cell optic nerve regeneration in Xenopus laevis.

Unlike adult mammals, adult frogs regrow their optic nerve following a crush injury, making Xenopus laevis a compelling model for studying the molecular mechanisms that underlie neuronal regeneration. Using Translational Ribosome Affinity Purification (TRAP), a method to isolate ribosome-associated mRNAs from a target cell population, we have generated a transcriptional profile by RNA-Seq for retinal ganglion cells (RGC) during the period of recovery following an optic nerve injury. Based on bioinformatic analysis using the Xenopus laevis 9.1 genome assembly, our results reveal a profound shif

Characterization of pax1, pax9, and uncx sclerotomal genes during Xenopus laevis embryogenesis.

BACKGROUND: The axial skeleton develops from the sclerotome, a mesenchymal cell population derived from somites. Sclerotomal cells migrate from somites to the perinotochordal and perineural space where they differentiate into chondrocytes to form cartilage and bone. In anurans, little is known about the way how the sclerotome changes as development proceeds and how these events are regulated at the molecular level. Pax1, Pax9, and Uncx4.1 genes play a central role in the morphogenesis of the axial skeleton in vertebrates, regulating cell proliferation and chondrogenic specification of the scle

Oocytes from Xenopus laevis contain an intrinsic sigma 2-like binding site.

In preparation for expression studies for rat brain sigma-binding sites, Xenopus oocytes were tested for the presence of [3H]di-o-tolylguanidine (DTG)-binding sites. Native oocytes were found to contain two intrinsic [3H]DTG-binding sites, a high-affinity site (Kd = 32 +/- 6 nM, Bmax of 45.7 +/- 19 pmol/mg protein) and a low-affinity binding site (Kd = 1.3 +/- 0.7 microM, Bmax of 3.2 +/- 0.7 nmol/mg protein). In a series of radioligand-binding-displacement studies, the high-affinity binding sites were found to have a binding profile which has a similar Kd to that of the mammalian sigma 2-bindi

Embryonic and skeletal development of the albino African clawed frog (Xenopus laevis).

The normal stages of embryonic development for wild-type Xenopus laevis were established by Nieuwkoop and Faber in 1956, a milestone in the history of understanding embryonic development. However, this work lacked photographic images and staining for skeleton structures from the corresponding stages. Here, we provide high-quality images of embryonic morphology and skeleton development to facilitate studies on amphibian development. On the basis of the classical work, we selected the albino mutant of X. laevis as the observation material to restudy embryonic development in this species. The low

Influence of systemic copper toxicity on early development and metamorphosis in Xenopus laevis.

The primary objective of the present study was to examine the influence of early systemic toxicity resulting from copper (Cu) exposure on metamorphic processes in Xenopus laevis. A 28-day exposure study with copper, initiated at developmental stage 10, was performed using test concentrations of 3.0, 9.0, 27.2, 82.5, and 250 μg Cu/L. The primary endpoints included mortality, developmental stage, embryo-larval malformation, behavioral effects, hindlimb length (HLL), growth (snout-vent length [SVL] and wet body weight), and histopathology. The 28-day LC50 value with 95% confidence intervals was 6

The effect of food properties on grasping and manipulation in the aquatic frog Xenopus laevis.

The ability to grasp an object is fundamental from an evolutionary perspective. Involved in many daily activities, grasping has been extensively studied in primates and other mammals. Yet other groups of tetrapods, including anurans, have also evolved significant forelimb prehensile capacities that are often thought to have originated in an arboreal context. In addition, grasping is also observed in aquatic species. But how aquatic frogs use their forelimbs to capture and manipulate prey remains largely unknown. The aim of this study is to explore how the grasping and manipulation of food item

Neuronal degeneration and regeneration induced by axotomy in the olfactory epithelium of Xenopus laevis.

The olfactory epithelium (OE) has the remarkable capability to constantly replace olfactory receptor neurons (ORNs) due to the presence of neural stem cells (NSCs). For this reason, the OE provides an excellent model to study neurogenesis and neuronal differentiation. In the present work, we induced neuronal degeneration in the OE of Xenopus laevis larvae by bilateral axotomy of the olfactory nerves. We found that axotomy induces specific- neuronal death through apoptosis between 24 and 48h post-injury. In concordance, there was a progressive decrease of the mature-ORN marker OMP until it was

Comparative studies of apoptosis in Xenopus laevis and mouse thymoma cell lines.

With the use of in vitro methods and cell lines, functional aspects of apoptosis in the Xenopus laevis B3/B7 and mouse EL4 thymoma cell lines are revealed. Moreover, by using information gleaned from digital imaging and immunocytochemistry, changes in locations of key proteins implicated in apoptotic anti-cancer responses, e.g. p53 and Mdm2, are shown. Suggestions are offered as to what these results might mean with respect to the evolutionary conservation of the function and structure of these two molecules and to cancer resistance in amphibians. Finally, studies are described on resveratrol

The embryonic development of Xenopus laevis under a low frequency electric field.

The aim of this study was to determine the effects of a low frequency electric field on the early embryonic development of frogs. The embryos of African clawed toads, Xenopus laevis, were exposed to a 20-μA electric current during the cleavage stages. The developmental processes of embryos during and after electric field exposure were monitored for teratogenic effects. All the embryos continuously exposed to the electric field died without undergoing any developmental processes. However, when the embryos were exposed to the electric field for 20-min periods (four times/over 2 d), the embryos d

Developmental expression of the fermitin/kindlin gene family in Xenopus laevis embryos.

Fermitin genes are highly conserved and encode cytocortex proteins that mediate integrin signalling. Fermitin 1 (Kindlin1) is implicated in Kindler syndrome, a human skin blistering disorder. We report the isolation of the three Fermitin orthologs from Xenopus laevis embryos and describe their developmental expression patterns. Fermitin 1 is expressed in the skin, otic and olfactory placodes, pharyngeal arches, pronephric duct, and heart. Fermitin 2 is restricted to the somites and neural crest. Fermitin 3 is expressed in the notochord, central nervous system, cement gland, ventral blood islan

Glycolytic activation at the onset of contractions in isolated Xenopus laevis single myofibres.

Intracellular pH (pHi) was measured in isolated Xenopus laevis single myofibres at the onset of contractions, with and without glycolytic blockade, to investigate the time course of glycolytic activation. Single myofibres (n=8; CON) were incubated in 2',7'-bis(carboxyethyl)-5(6)-carboxyfluorescein acetoyxmethyl ester (10 microM; for fluorescence measurement of pHi) and stimulated for 15 s at 0.67 Hz in anoxia in the absence (control condition; CON) and presence of a glycolytic inhibitor (1 mM iodoacetic acid; IAA). Intracellular pHi and tension were continuously recorded, and the differences i
