Two infertile males with sex chromosomal abnormalities and mosaic karyotype, 45,X/46,X,dic(Yq) and 45,X/46,X,ring(Y), had considerably changed physical findings, including tooth sizes and craniofacial dimensions. Spermatogenesis was preserved with abnormal meiotic chromosomal behaviour. Mosaic karyotype and structurally changed Y chromosome in both cases had an influence on physical parameters. Tests were normally developed and spermatogenesis was preserved but depressed in later stages.

It has been conclusively shown that a majority of XX males has acquired Y-chromosome-specific DNA (Y-DNA) sequences from their father's Y chromosome. Testicular differentiation in these XX males is very likely induced by the testis-determining factor, TDF, normally located on Yp. The phenotypic expression of TDF in the presence of two X chromosomes is a male habitus, dysgenetic scrotal testes and no ambiguity of the genitalia. Among those XX males who do not have Y-DNA detectable by presently used methods, some, but not all, have ambiguity of the genitalia with or without hypospadias. XX true

A 37 year old second gravida received amniocentesis, because of advanced maternal age. Cytogenetic investigations of the fetal cells revealed a male karyotype with a pericentric inversion of the Y chromosome. Chromosome analysis of the father was also undertaken in order to decide whether this inversion was inherited or of de novo origin. The results of cytogenetic examinations of the mother as well as of two alleged fathers enabled us to detect an exclusion of paternity for one man and a positive hint of paternity for the other. The cytogenetic markers used were the inversion itself and a hig

Dicentric Y chromosomes are rare in man. Four new cases of dicentric Y chromosomes are described. The cases of the literature so far reported are reviewed. Among the cases, a wide range of variation in phenotype, external genitalia, histology, and chromosomal findings was observed. The relationship of the clinical picture and structural abnormalities of the Y chromosomes is discussed.

BACKGROUND: Although Y chromosomal genes are involved in male sex development, spermatogenesis, and height growth, these genes play no role in the survival or mitosis of somatic cells. Therefore, somatic cells lacking the Y chromosome can stay and proliferate in the body.
METHODS: Several molecular technologies, including next-generation sequencing and multiplex PCR-based assays, are used to detect mosaic loss of the Y chromosome (mLOY) in the blood of men.
MAIN FINDINGS: Accumulating evidence suggests that mLOY represents the most common acquired chromosomal alteration in humans, affecting

A case study involving an intentional homicide case in November 2018, in which the autosomal genotypes of the suspect were unavailable and only part of deletions of Y-STR loci were identified by Y-chromosomal typing. The suspect, male, was charged with beating the decedent, female, over the head with an iron water pipe to death. The use of standard autosomal DNA profiling to identify the suspect was unattainable due to the extensive volume blood of the decedent on the murder weapon which was inevitably cleaned by running water at the crime scene. As a result, autosomal genotypes of the suspect

OBJECTIVE: To determine the frequencies and the characteristics of Y chromosome microdeletions in infertile males.
MATERIAL AND METHODS: The records of 1616 infertile males were included in the study. The cases were divided into groups according to the infertility etiology and semen analysis. The frequencies and the characteristics of Y chromosome microdeletions were investigated in groups.
RESULTS: Y chromosome microdeletion was detected in 54 (3.3%) of 1616 cases. Microdeletions in the azoospermia factor (AZF) region were the most common (48.1%). When the cases were grouped according to ca

Turner syndrome (TS) is one of the most common types of aneuploidy among humans, and is present in 1:2000 newborns with female phenotype. Cytogenetically, the syndrome is characterized by sex chromosome monosomy (45,X), which is present in 50-60% of the cases. The other cases present mosaicism, with a 45,X cell line accompanied by one or more other cell lines with a complete or structurally abnormal X or Y chromosome. The presence of Y-chromosome material in patients with dysgenetic gonads increases the risk of gonadal tumors, especially gonadoblastoma. The greatest concern is the high risk of

A newborn male infant presented with midshaft hypospadias, chordee, and undescended left testis. Both gonads lacked the tunica albuginea and appeared to be adjacent to structures resembling fallopian tubes. On biopsy, there was marked dysgenesis of both gonads, with a paucity of testicular tubules and foci of ovarian-like stroma. Peripheral blood karyotype was 46,X,mar(Y) [39]/45,X [5]. Right gonadal biopsy material showed the same mosaicism but with a higher proportion of 45,X cells (46%). PCR and FISH analyses with primers/probes from different Yp, Yq, and Ycen loci defined the structure of

A dicentric Y chromosome was detected in a 30-year-old azoospermic male patient who was found to be mosaic for 45,X/46,X,dic Y(qter----p11::p11----qter). The dicentric iso (Y) chromosome was identified conclusively with C-banding, G-banding and Q-banding techniques. The relationship of structural abnormalities of the Y chromosome and azoospermia is discussed.

Pericentric inversion of the human Y chromosome has been estimated to occur with a frequency of 1-2 per thousand in various populations, and the results of this study, derived from over 12 000 prenatal diagnosis cases, is 1.15 per 1000. In these cases, it was concluded that there was no clinical significance because the fathers and male fetuses had the same pericentric inversion. Chromosome analysis of the father is advisable to determine whether or not the inversion is familial in order to be able to provide genetic counselling.

Twelve presumptive structurally altered Y chromosomes were studied with Q-, G-, G-11, C-, Cd, and lateral asymmetric banding techniques and were compared with normal X and Y chromosomes and with an abnormal [i(Yq)] Y chromosome that exhibited intact fluorescence. Significant to this work is the fact that the Y chromosome has a small block of Giemsa-11 heterochromatin adjacent to the centromere on the long arm, while the X chromosome does not, which allows a distinction between the X- and Y-derived chromosomes. Two of the twelve altered chromosomes of either X or Y origin are small nonfluoresce

The YFF sperm assay, which is a quantification of the incidence of sperm with two fluorescent bodies (YFF = two fluorescent bodies), was performed to measure Y chromosomal nondisjunction. Three categories of human subjects were analyzed: 1) nonexposed, 2) exposed to antineoplastic agents - ie, chemo- and radiation therapy, and 3) dibromochloropropane (DBCP)-exposed. The nonexposed individuals demonstrated a relatively constant mean YFF value of 1.3%, which was consistent with historical controls in this laboratory and with the results of other investigators. Further, a one-way analysis of vari

Using chromosome banding techniques, a phenotypically normal male was found to have an abnormal banding pattern of the Y chromosome. By the constitutive heterochromatin staining method, a darkly stained band was located on the short arm and the proximal region of the long arm. The quinacrine staining method also showed a similar abnormal banding pattern: a brightly fluorescing band was seen on the short arm and the proximal region of the long arm. By the conventional Giemsa staining method, however, no specific morphological abnormality was detected in the aberrant Y. On detailed karyotype ana

The Y chromosome in the XY sex-determination system is often shorter than its X counterpart, a condition attributed to degeneration after Y recombination ceases. Contrary to the traditional view of continuous, gradual degeneration, our study reveals stabilization within large mating populations. In these populations, we demonstrate that both mutant and active alleles on the Y chromosome can reach equilibrium through a mutation-selection balance. However, the emergence of a new species, particularly through the founder effect, can disrupt this equilibrium. Specifically, if the male founders of
