Detergent assisted lipid extraction from wet yeast biomass for biodiesel: A response surface methodology approach.

The lipid extraction from the microbial biomass is a tedious and high cost dependent process. In the present study, detergent assisted lipids extraction from the culture of the yeast Yarrowia lipolytica SKY-7 was carried out. Response surface methodology (RSM) was used to investigate the effect of three principle parameters (N-LS concentration, time and temperature) on microbial lipid extraction efficiency % (w/w). The results obtained by statistical analysis showed that the quadratic model fits in all cases. Maximum lipid recovery of 95.3±0.3% w/w was obtained at the optimum level of process

Biosynthesis of Sciadonic Acid Derived from Gymnosperms with Anti-Colitis Activity.

Sciadonic acid (SCA), Δ5-unsaturated fatty acid with anti-inflammatory and lipid-regulatory properties, is predominantly derived from seeds of gymnosperms. This study established a systematic engineered Yarrowia lipolytica platform for efficient SCA production and assessing the anti-colitis efficacy of SCA as a novel therapeutic lipid. First, production of eicosadienoic acid (EDA) was biosynthesized by iterative expression of endogenous polyunsaturated fatty acid desaturase and Δ9 elongase. Different Δ5 desaturases were screened to identify high-activity enzymes capable of catalyzing EDA-CoA s

Bioreduction of 4'-Hydroxychalcone in Deep Eutectic Solvents: Optimization and Efficacy with Various Yeast Strains.

4'-dihydrochalcones are secondary metabolites isolated from many medicinal plants and from the resin known as 'dragon's blood'. Due to their biological potential, our research objective was to determine the possibilities of using biocatalysis processes carried out in deep eutectic solvents (DESs) to obtain 4'-dihydrochalcones as a model compound. The processes were carried out in a culture of the yeast Yarrowia lipolytica KCh 71 and also in cultures of strains of the genera Rhodotorula and Debaryomyces. Based on the experiments carried out, an optimum process temperature of 35 °C was chosen, a

In situ engineering of synthetic yeast consortia for cross-species metabolic conversion of crude glycerol and byproducts into circular renewable bioenergy.

The rapid growth of biodiesel production generates large amounts of crude glycerol, a low-value byproduct with environmental and economic challenges. Here, we present an engineered yeast coculture system combining Saccharomyces cerevisiae and Yarrowia lipolytica to convert crude glycerol into isopropanol, a liquid organic hydrogen carrier. The system integrates metabolic engineering, cell surface display pairing, immobilization, and continuous cultivation in fibrous bed bioreactors. In S. cerevisiae, glycerol use was improved by transporter optimization, pathway redirection, and flux shift fro

Engineering nonconventional yeasts to produce carboxylic acids: Advances and Prospects.

Due to their versatile carboxy group, carboxylic acids can be used as bio-based monomers for high-value-added industrial products with great market demand. In recent years, metabolic engineering of nonconventional yeasts to produce carboxylic acids from renewable raw materials is showing increasing promise. In this review, we first summarize the characteristics of nonconventional yeasts (e.g., Yarrowia lipolytica, Candida tropicalis), and then focus on metabolic engineering strategies for the synthesis of mono-, di- and tricarboxylic acids. Finally, the development trends of low-cost, green an

Promoter engineering in oleaginous yeasts for value-added chemical production.

Oleaginous yeasts, particularly Yarrowia lipolytica, are increasingly used as microbial platforms for producing lipids and other value-added compounds from renewable feedstocks. Their biotechnological utility derives from active lipid metabolism, broad substrate flexibility, and expanding engineering tools, but efficient production requires transcriptional programs that match changing metabolic states during fermentation. Because carbon flux is redistributed across growth, nutrient limitation, lipid accumulation, and production phases, static constitutive expression is often insufficient for o

Synthesis and Biological Evaluation of Novel 2-(Piperidin-4-yl)-1,2,3,4-tetrahydroisoquinoline and 2-(Piperidin-4-yl)decahydroisoquinoline Antimycotics.

Piperidine antimycotics like fenpropidin are well established in agrochemistry. On the other hand, numerous isoquinoline derivatives show remarkable antimycotic effects. Here we present a series of 13 hybrid molecules of both lead structures, which were prepared using reductive amination as key reaction step. Pre-screening against Yarrowia lipolytica resulted in a batch of promising candidates whose antifungal efficacy was further evaluated against clinically relevant species. In these assays, complete growth inhibition was seen for five or six compounds against C. albicans or C. krusei, respe

Sec14 family of lipid transfer proteins in yeasts.

The hydrophobicity of lipids prevents their free movement across the cytoplasm. To achieve highly heterogeneous and precisely regulated lipid distribution in different cellular membranes, lipids are transported by lipid transfer proteins (LTPs) in addition to their transport by vesicles. Sec14 family is one of the most extensively studied groups of LTPs. Here we provide an overview of Sec14 family of LTPs in the most studied yeast Saccharomyces cerevisiae as well as in other selected non-Saccharomyces yeasts-Schizosaccharomyces pombe, Kluyveromyces lactis, Candida albicans, Candida glabrata, C

Recombinant production of a diamine oxidase in Komagataella phaffii and in vitro degradation of histamine in simulated intestinal fluid.

Diamine oxidase (DAO-1) from Yarrowia lipolytica was recombinantly produced in Komagataella phaffii in fed-batch bioreactor cultivations using the constitutive GAP and methanol-inducible AOX1 promoters. Utilizing an exponential glucose feed, a maximum DAO-1 activity of 43 ± 7 µkat/Lculture was achieved after 37 h of cultivation using the GAP promoter. When the AOX1 promoter was used, a maximum DAO-1 activity of 230 ± 13 µkat/Lculture was achieved after 90 h of cultivation with a stepwise increase in the methanol feed. Thus, approximately 15- (GAP promoter) and 82-fold (AOX1 promoter) higher DA

Metabolic engineering of microbial competitive advantage for industrial fermentation processes.

Microbial contamination is an obstacle to widespread production of advanced biofuels and chemicals. Current practices such as process sterilization or antibiotic dosage carry excess costs or encourage the development of antibiotic resistance. We engineered Escherichia coli to assimilate melamine, a xenobiotic compound containing nitrogen. After adaptive laboratory evolution to improve pathway efficiency, the engineered strain rapidly outcompeted a control strain when melamine was supplied as the nitrogen source. We additionally engineered the yeasts Saccharomyces cerevisiae and Yarrowia lipoly

Recent advances in genetic engineering and chemical production in yeast species.

Yeasts have emerged as well-suited microbial cell factory for the sustainable production of biofuels, organic acids, terpenoids, and specialty chemicals. This ability is bolstered by advances in genetic engineering tools, including CRISPR-Cas systems and modular cloning in both conventional (Saccharomyces cerevisiae) and non-conventional (Yarrowia lipolytica, Rhodotorula toruloides, Candida krusei) yeasts. Additionally, genome-scale metabolic models and machine learning approaches have accelerated efforts to create a broad range of compounds that help reduce dependency on fossil fuels, mitigat

Binding properties of sterol carrier protein 2 (SCP2) characterized using Laurdan.

Sterol carrier protein 2 (SCP2) binds lipids with high affinity and broad specificity. The overall hydrophobicity, fluidity, and dipolar dynamics of the binding site of SCP2 from Yarrowia lipolytica were characterized using the environmentally-sensitive fluorescent probe Laurdan. The study revealed a binding site with an overall polarity similar to that of dichloromethane and an internal phase comparable to that of phospholipid membranes with coexisting solid-ordered and liquid-crystalline states. The fluorescence properties of bound Laurdan also revealed that the binding site of SCP2 can acco

Yeast vaccine production platform for human and animal infectious diseases.

Yeasts have contributed to human and animal health through functional antigen production for vaccine formulations. Some yeast-made vaccines have become a reality for humankind because they have reached commercialization (hepatitis B, HPV, and tick parasitosis). Many other vaccine prototypes are under preclinical and clinical evaluations, hoping for their usage soon. Currently, genomes, genetic modification techniques, and industrial vaccine manufacturing have been successfully developed for Saccharomyces cerevisiae, Komagataella phaffii (formerly Pichia pastoris), and Hansenula polymorpha. Advances in glycerol metabolism comprehension in yeasts: from regulation to metabolic engineering strategies.

Glycerol is a polyol that can be produced either chemically from oils or propylene, or biologically by yeasts, mainly under osmotic stress. Currently, glycerol is an abundant byproduct generated during biodiesel manufacturing that can be used as substrate in fermentative processes. Its efficient assimilation varies widely among yeast species; therefore, understanding the regulation of both transport and catabolism is pivotal for optimizing biotechnological processes based on this carbon source. This review addresses current knowledge on the regulatory networks controlling glycerol metabolism i
