The aryl hydrocarbon receptor (AhR) is a ligand-activated transcription factor with a basic region/helix-loop-helix (bHLH) motif. AhR has been sequenced and the functional domains defined and there is information on the formation of complexes with other peptides and interactions with DNA, although these areas continue to be investigated. AhR mediates many biological effects such as developmental toxicity, including induction of cleft palate and hydronephrosis. This regulatory protein is expressed in embryonic liver and has been immunohistochemically localized in cells of human and mouse second

The expression pattern of tissue nonspecific alkaline phosphatase (TNAP) in the developing neural tube of mouse is reported. Homogeneous AP activity in the neuroepithelium becomes prominent at E8.5. At E9.5, distinctly AP-positive cells appear in the brain and spinal cord area. At stages E10.5 to E12.5, AP positivity is observed between the mesencephalon and the rhombencephalon, along the entire spinal cord and cranial nerves emerging from the myelencephalon. At E13.5, strongly AP positive fibers become prominent in the pons. At E14.5, AP expression in brain tissue is considerably reduced and

Cartilage matrix protein (CMP) is a non-collagenous component of cartilage with a yet unknown function. In this study we used in situ hybridization to investigate the temporal and spatial distribution of CMP transcripts during human embryonic and early fetal development, and compared it to the pattern of expression observed for collagen types I, II, X, and decorin. The distribution of CMP and collagen type II transcripts followed a similar pattern in the embryonic bone anlage, the fetal growth plate, and the developing vertebral column. Expression was highest in the upper hypertrophic and lowe

Development of the node and formation of the notochordal plate in gestational day 7-9 mice (Theiler stages 10-14) has been documented principally with scanning electron microscopy (SEM) and cell fate analyses utilizing DiI and/or CFSE as a cell label. With SEM, cells composing these two populations are initially identifiable at stage 10 at the ventral midline of the rostral half of the embryo. They can be recognized by their relatively small ventral surface area, as compared to that of the peripherally adjacent prospective gut endodermal cells, and by the presence on the ventral side of each c

Fluorescent dyes were used in the present study to map three rostrocaudal levels of the primitive streak of mouse embryos at 7.5 days of gestation. Injections at the most rostral level, the 1/4 streak level, principally labeled cells of the paraxial mesoderm, including mesenchyme in the head, and somites and segmental plate mesoderm in the trunk. Injections at the intermediate level, the 1/2 streak level, principally labeled cells of the lateral plate mesoderm of the trunk. Injections at the most caudal level, the 3/4 streak level, principally labeled cells of the extraembryonic mesoderm at th

Mice which are homozygous for the limb deformity (ld) mutation also manifest an incompletely penetrant unilateral or bilateral renal agenesis phenotype. Intercross experiments suggest that the differences in penetrance of the renal agenesis phenotype between homozygous mice with different ld alleles are due to intrinsic differences in the strength of the mutant alleles or to one or more closely linked modifying loci, and not to generalized differences in genetic background. Analysis of ld/ld embryos between embryonic days 11-13 reveals delayed outgrowth or complete absence of the ureteric bud,

As part of an investigation into the role of FGF in lens development, we have studied the distribution of both aFGF and bFGF during eye morphogenesis from embryonic days 10 to 18 (E10-E18) in the rat. For aFGF, reactivity was found only in ectoderm at E10, prior to contact between the optic vesicle and presumptive lens ectoderm. During lens placode formation (E11) there was a transient, diffuse reactivity for aFGF in anterior optic vesicle cells directly apposed to the labelled ectoderm of the lens placode. At E12 the diffuse reactivity of the lens placode had changed to a discrete localisatio

Cultured stellate reticulum cells isolated from rat mandibular molars respond to incubation in EGF by increasing their level of expression of TGF-beta 1 mRNA. Northern blots showed that incubation in EGF for 6 hours stimulated over a two-fold increase in TGF-beta 1 mRNA in the cells. In contrast, incubating the cells in TGF-beta 1 did not enhance the expression of TGF-beta 1 mRNA in the cells, indicating that TGF-beta 1 does not have an autocrine effect on these cells. Immunocytochemistry showed that EGF receptor was present on the surface of many but not all of the cultured stellate reticulum

DiI, a fluorescent lipophilic dye, was micro-injected into the brachial somites of 10.5 day rat embryos to determine whether these somites can contribute cells to the development of the fore-limb bud. The injected embryos were cultured and harvested at the 20-25-somite stage. The dye did not interfere with somitogenesis because, at the injection site, the DiI-labelled somites were able to differentiate into dermomyotome and sclerotome. We have analyzed cryo-sections of 20-21-somite stage embryos and were unable detect the presence of DiI-labelled cells in the fore-limb buds. However, at the 22

Recently, we isolated a novel mouse gene, Etl-1 (Enhancer-trap-locus-1), whose deduced amino acid sequence shows in its C-terminal portion striking homology to the brahma protein (BRM), a transcriptional regulator of homeotic genes in Drosophila, and to SNF2/SWI2, a transcriptional regulator of various genes in Saccharomyces cerevisiae. Here we report the generation of antibodies against the Etl-1 gene product (ETL-1) and describe the subcellular localization as well as the expression and distribution of the ETL-1 protein during mouse pre- and early post-implantation development. ETL-1 is a nu

We know that, once rostral neurulation is completed in the neuroaxis of the chick embryo, the caudal neurocoel becomes occluded and the brain rapidly expands. However, very little is known about the mechanisms maintaining occlusion. Studies had shown that occluded neurocoels reopened in embryos treated with chelators of cations, but the reasons remained unclear and the cations unidentified. To begin defining the role of cations, this study explored the effect of Ca2+, calmodulin, and cAMP on maintaining the occluded neurocoel. Chick embryos during the natural phase of neurocoel occlusion (stag

Congenital megacolon develops in transgenic mice that overexpress the homeobox-containing gene, Hoxa-4. The current study was done to identify abnormalities of the terminal colon that might account for the phenotype. The terminal bowel of transgenic mice was compared with that of control and lethal spotted (ls/ls) mice, a strain in which megacolon also develops. The terminal colon of the transgenic mice contained fewer ganglia than that of controls, but was hypoganglionic, rather than aganglionic like that of ls/ls mice. The neurons present in the adult transgenic colon were significantly incr

Immunoperoxidase localization of epidermal growth factor receptors (EGFR) and epidermal growth factor (EGF) itself was examined in rat first and second mandibular molars postnatally from day 0 to 12. The results showed that the dental follicle stained heavily for EGFR from day 0 to 8, declined in staining at day 9, and was devoid of stain from day 10 onward. Preosteoblasts and osteoblasts of alveolar bone also stained and lesser staining of ameloblasts and odontoblasts was observed. Except for staining of occasional isolated cells, the stellate reticulum did not stain. Light staining of the de

Fibronectin mRNA and protein content were examined during embryonic implantation in the rat uterus. Content of total fibronectin mRNA at day 6 of pregnancy increased relative to the non-pregnant uterus. In contrast, fibronectin protein content of the subepithelial stroma was relatively decreased except in the region directly surrounding the lumen, and this fibronectin immunoreactivity was sensitive to hyaluronidase treatment. These changes are likely to reflect the degradation and subsequent remodeling of the previously stable uterine extracellular matrix in preparation for embryonic implantat

Although there is considerable interest in the possible role of olfactory glia in the pathfinding abilities of olfactory nerve axons, the complete development of these glia in vivo has not been described. Using a specific Schwann cell marker, the 1E8 antibody, we have found that olfactory nerve glia can be identified throughout development. These glia appear to originate in the olfactory placode and migrate initially into the periphery of the olfactory nerve, and later into the center of the nerve. Olfactory nerve glia enter the presumptive olfactory bulb with the olfactory receptor neuron axo

p35, a Ca(++)-phospholipid-binding protein that serves as a substrate for the EGF receptor tyrosine kinase, is expressed by primitive glial ependymal cells to define a raphe occupying the ventral midline in the spinal cord and hindbrain of rat embryos (McKanna and Cohen, Science 243:1477-1479, 1989). p35 appears transiently in the median one-third (80 microns) of the floor plate at precisely the time and place where axons cross to form the ventral commissure. We postulated that if p35 is involved with commissure development, homologous p35 raphes might be found at decussation sites rostral to

Renal diseases such as acute and chronic kidney disease cause loss of nephrons and kidney function, often leading to renal failure. Current treatments like dialysis and transplantation only manage symptoms and extend survival for a few years. Regenerative medicine offers promising approaches for repairing and regenerating renal tissue by targeting underlying disease mechanisms. For this narrative review, relevant original research articles and review articles were retrieved from online databases such as PubMed, Google Scholar, and ScienceDirect by using relevant keywords such as "renal regener

BACKGROUND: Pharyngeal pouch formation is fundamental to vertebrate craniofacial development, but molecular mechanisms underlying posterior pouch formation remain incompletely understood. Retinoic acid (RA) signaling is required for posterior pharyngeal development, and Hox genes are established RA targets. However, their genetic requirement in posterior pouch formation has not been established.
RESULTS: Zebrafish lacking both hoxba and hoxbb clusters-teleost-specific duplicates of the ancestral HoxB cluster-exhibited severe loss of posterior pharyngeal pouches and associated pharyngeal skele

BACKGROUND: Precise control of DYRK1A dosage is essential for embryonic development, including craniofacial morphogenesis. While LZTS2 is among the most consistently identified DYRK1A-interacting proteins, its roles in embryonic development remain incompletely understood, and its potential contribution to craniofacial development has not been examined. Xenopus laevis was used to test the role of LZTS2 in craniofacial development and its functional relationship with DYRK1A.
RESULTS: Lzts2 and Dyrk1a showed overlapping expression during craniofacial development, with both proteins present in de

BACKGROUND: Sclerostin domain-containing 1 (SOSTDC1) deficiency enhances BMP and Wnt signaling, causing supernumerary teeth formation. Competing hypotheses of this formation at different sites have been proposed, and the underlying molecular mechanisms and cell types remain poorly understood.
RESULTS: We examined Sostdc1 knockout effects on spatiotemporal expression of Wnt pathway genes in developing maxillary incisors. In wild type mice, we observed an inverse spatiotemporal expression of Sostdc1 and Wnt10a. The knockout caused ectopic expression of Wnt10a, Wnt6, Lrp4, Dkk1, and Sost and pro
